The boletes are conspicuous stipito-pileate macrofungi that are generally defined by their tubulose hymenophores. Molecular phylogenetic analyses have informed extensive taxonomic revisions across the boletes. Despite this, the boletes in the southeastern USA have been largely absent from the literature. This dissertation seeks to understand the microbiome inhabiting the tuberculate ectomycorrhizae of the bolete Hemileccinum rubropunctum, and addresses taxonomic revisions of boletes found in the Boletaceae and Suillaceae. The introduction provides a brief background on boletes, as well as our understanding of tuberculate ectomycorrhizae. The major forces that govern social groups, namely fission-fusion dynamics, cohesion and maintenance, are nearly ubiquitous across animal groups. The field of animal collective behaviour has recently been married with automated radiotracking producing a ‘re-wilding’ of field research into sociality. The combination of this with Social Network Analysis has led to discoveries such as population wide information transfer and the flexibility of animal groups to change social connectivity based on environmental context. Galectins are a family of -galactoside-binding proteins involved in cell stress responses and differentiation. Galectins are multifunctional proteins widely studied in many cell models including acute myeloid leukemia HL-60 cells where they mediate numerous intra- and extracellular functions in response to many stress-inducing stimuli. O-GlcNAcylation is a dynamic post-translational modification implicated in the regulation of many cellular diseases including cancers. The O-GlcNAc mediated expression and secretion of galectins during neutrophilic differentiation was examined in HL-60 cells. Galectin gene (LGALS), galectin protein expression, and galectin secretion were determined by RT-qPCR, immunoblotting, and ELISA, respectively. Abscisic acid (ABA) is a plant hormone that triggers a signaling transduction in response to drought, salinity, and cold. The core components in the ABA signaling pathway that lead to downstream ABA responses have been identified.

Phosphatidic acid (PA) is a membrane phospholipid that is involved in the signaling pathway for ABA-induced stomatal closure. We tested whether PA is involved in the signaling pathway for ABA-induced gene expression as well, using model plant Arabidopsis thaliana. Under the framework of the speciation cycle, the process begins by population subdivision creating structured populations that then diverge until two new species form. Cancer is well-recognized as an evolutionary system, as first proposed by Cairns and Nowell more than 60 years ago. In an evolutionary context, cancers growing in vivo typically consist of heterogeneous subpopulations of cells that interact with each other and with host cells through selection forces operating at many temporal and spatial scales. Polygyny, or the formation of colonies with multiple cooperating queens, has been observed in a variety of social Hymenoptera and likely exists as a convergent evolutionary strategy. Polygyne cooperation has been observed in several Vespula sp. and is correlated with a perennial social strategy. This perennial-polygyne behavior has been observed most commonly within the tropical and subtropical regions of the invasive Vespula pensylvanica and V. germanica, and rarely within their native temperate ranges. Prefrontal cortical regions play an essential role in generating appropriate cardiovascular adjustments, particularly in cardio-vagally mediated heart rate (HR) responses to active tasks. Functional imaging studies provide correlational evidence that this region coordinates HR responses to exercise, however, direct experimental evidence of prefrontal cortical HR regulation in humans is not available. Seven persons with epilepsy implanted with intracranial electroencephalography (iEEG) completed 2-second isometric handgrip (IHG) contractions at no-stimulation (NO-STIM) or sham-stimulation (SHAM) conditions, and during direct electrical stimulation (STIM) of the orbitofrontal and medial prefrontal cortex. The invasive red lionfish, Pterois volitans, has been shown to considerably reduce reef biomass via predation and competition in non-native ranges. Primary maintenance of lionfish is through human removal (e.g., culling), which does not sufficiently control the population due to deep-water spawning, recolonization of recently culled reefs, and ontogenetic migrations to mesophotic waters. Here, I investigated the diets and primary energy pathways during ontogeny in lionfish from the Florida Keys using stomach contents and stable isotope (δ13C and δ15N) analysis to better understand their role in the trophic food web. Within the last century anthropogenic effects have shifted fire resilient habitats that are adapted to thousands of years of frequent low intensity fires to landscapes that are susceptible to extreme fire behavior which threaten human assets and sensitive natural resources. The purpose of this study was to determine recovery from Tabata bodyweight high intensity interval training (HIIT) exercise using different recovery assessment methodologies across 24- and 48-hour time intervals. Participants (23.2 ± 3.1 years old, 163.1 ± 19.9 lbs., and 22.8 ± 9.6 % body fat) consisted of 3 females and 7 males (n=10) Individuals who were recreationally trained (4+ days per week, 30+ minutes per day at moderate to vigorous intensity) and conducted both Trial A (24-hours between HIIT sessions) and Trial B (48-hours between HIIT sessions). Microbial Dark Matter: Culturing The Uncultured In Search Of Novel Chemotaxonomy, Sarah J. Kennedy

Although <1% of environmental isolates can be cultured, insights into the culturable fraction refine future techniques to unlock the cryptic biodiversity. This cryptic biodiversity is omnipresent within culture-independent surveys of environmental bacterial populations. Subsequently, these elusive organisms are termed “microbial dark matter” due to the intrigue surrounding their potential taxonomic and biosynthetic impacts. To this end of investigating bacterial-specific dark matter, we employed high-throughput culturing techniques to examine Gulf of Mexico sediments and sponges for chemotaxonomically important bacteria. Through stepwise implementation of permissive and selective recovery techniques, we succeeded in recovering over 150 unique bacterial isolates. We observed several isolates that pose as chemotaxonomically interesting, including several strains that belong to critical bioremediation taxa. In addition to these biotechnological-amenable isolates, we have also succeeded in culturing taxa historically attributed with antibacterial discoveries. One such isolate was a previously sponge-associated organism belonging to the rarely cultured Verrucosispora genus. Through extensive comparative genomics, we identified it as a novel species and subsequently named it Verrucosispora sioxanthis sp. nov. with the published genome serving as a resource for continued microbial dark matter investigations. Following continued passaging of the organism, we observed improved growth, biomass, and liquid growth dispersal within each increasingly sub-passaged generation of the isolate. As such, the laboratory attenuation, or domestication, of the organism warranted in-depth whole genomic analyses to generate insight into the increased laboratory sustainability. Through phenotypically-guided bioinformatics, we determined that the domestication was mediated through genome-wide selective pseudo- or non-pseudogenization of genes. As the previously sponge-associated organism evolved with the selection pressures of the static laboratory, we demonstrate that isolate effectively underwent genomic atrophy to resolve into a largely stable, sustainable laboratory-domesticated strain. Collectively, our chemotaxonomic discovery pipeline expands on the culturable bacterial biodiversity and ultimately provides a previously uncultured model organism for initial forays into resolving the microbial dark matter knowledge gap.

Carnosol, a naturally occurring Phyto polyphenol found in sage, oregano, and rosemary, has been extensively studied by our laboratory for its anticancer effects in various types of cancer. In human Triple-Negative Breast Cancer (TNBC), carnosol was shown to inhibit cellular viability, colony growth, induced cell cycle arrest, autophagy, and apoptosis. Nonetheless, very little is known about the molecular mechanism of action. In the current study, the ability of carnosol to inhibit metastasis and tumour growth was examined.
