Deep brain photoreceptors (DBPs) are non-visual photoreceptors found in the brains of non-mammalian vertebrates like zebrafish. While using optogenetic techniques in our lab to photo-activate the ear (by expressing the light-gated channel: channelrhodopsin) in efforts of identifying optic tectum (OT) neurons that respond to and integrate auditory and visual stimuli, we found that violet light was able to elicit OT neuronal activity. OT activity was also detected in cases when visual and auditory stimulation was not possible due to removal of the eyes and absence of the exogenously expressed channelrhodopsin, respectively. This thesis discusses the history of catharanthine and related compounds, and their potential anti-addictive properties. Current research is exploring possible mechanisms of these properties. Past studies have found catharanthine has effects on neurons that project to the mesocorticolimbic system, an area implicated in addiction. We have seen that catharanthine decreases evoked dopamine (DA) release but increases basal DA release. This is the first study to investigate catharanthine’s effect on DA transmission in vivo. Early exposure to hypoxia is related to a variety of physiological and metabolic changes that have lasting effects on organisms’ physiology and life history. We measured the effects of maternal and embryonic mild, intermittent hypoxia on the life history of four clones of microcrustacean Daphnia magna, an emerging model organism for the studies of senescence and longevity. Daphnia individuals were produced parthenogenically, maintained in individual vials, and fed standard algal concentration daily. The cohort consisted of 189 individuals. We measured body size at first reproduction, fecundity (including late-life fecundity peak), offspring sex ratio, and longevity. We found no effect …

This thesis gives an overview of the relationship between diabetes, sleep apnea, obesity, and heart disease. It then addresses evidence that the traditional understanding of this relationship is incomplete or misleading. In the process, there is a brief discussion of the evolutionary rationale for the development and retention of sleep apnea in light of blood sugar dysregulation, as an adaptive mechanism in response to environmental stressors, followed by a brief overview of the general concepts of epigenetics. Women with Autism Spectrum Conditions (ASCs) may be more likely to experience symptoms of trauma exposure due to greater likelihood of facing adverse childhood experiences (ACEs). In order to gain a better understanding of the neural mechanisms involved with ASC and PTSD in women who experience difficulty with social interactions, I examined the ability of two vision tasks-- Binocular Rivalry Perceptual Alternation (BR) and Visual Approach/Avoidance by the Self Task (VAAST)-- to predict symptoms of both conditions as well as differentiate between them. Intrinsic foot muscles maintain foot structural integrity and contribute to functional movement, posture and balance. Thus, assessing intrinsic foot muscle size and strength are important. Magnetic resonance imaging (MRI) has been shown to accurately image the individual muscles but is costly and time consuming. Kinetoplastid parasites all require glycolysis for survival, with host glucose key for ATP production. One such parasite, Trypanosoma brucei, exclusively metabolizes glucose in its bloodstream form. Heart failure (HF) is a common cause of morbidity and mortality in the United States that may disproportionately affect certain racial/ethnic groups. Compared with White individuals, HF may affect Black individuals at a younger age with less favorable prognosis, and this excess risk may be partially explained by differences in HF risk factor burden. This thesis explores the synthesis of fluorinated α,β-dehydroamino acids, specifically a fluorinated dehydrovaline derivative. Previous work has been done on the equivalent dehydrovaline derivative without fluorine present and this work builds toward the fluorinated version with the goal of comparing the two structurally. The synthesis presented here pulls from previous synthetic strategies employed for dehydrovaline while also exploring the synthetic impact of the electronegative fluorine atoms.

Hypofunctionality at the N-Methyl-D-aspartic acid receptor (NMDAR) is a commonly used model of the neurodevelopmental disorder schizophrenia due to the complex circuitry changes that follow NMDAR blockade. While these animal models are very popular for modeling the cognitive deficits seen in schizophrenia, actual treatments for this disorder remain sparse. In recent decades, Danaus plexippus (the monarch butterfly) has experienced a drastic decrease in population size due to urbanization, climate change, and habitat loss. The primary habitat and food source of D. plexippus is plant species that are within the genus Asclepias (milkweed). The preservation and propagation of Asclepias spp. is necessary to combat D. plexippus’s population declines. The pedunculopontine nucleus (PPN) comprises the mesencephalic locomotor region (MLR) of the midbrain. It contains glutamatergic, cholinergic, and GABAergic interneurons, but only glutamatergic neurons influence locomotion, which is our focus here. The PPN receives upstream and downstream input from the basal ganglia. It projects to the reticular formation of the brainstem, which connects to the spinal cord. The PPN is involved in a wide range of physiological and behavioral processes, but our focus is locomotion. 8-hydroxyguanine (8-oxoG) is one of the most important products resulting from the oxidation of guanine in DNA. 8-oxoG is known as a biomarker of oxidative stress such as lung cancer. 2,5-diamino-4H-imidazol-4-one (Iz) and its hydrolysis product 2,2-diamino-4-[(2-deoxy-β-D-erythro-pentofuranosyl)-amino]-2,5-dihydrooxazol-5-one (Oz) are also important products of oxidation of guanine in DNA. Iron is extremely important for many organisms. Despite its abundancy, it exists in insoluble forms that makes its usability difficult. Some organisms secrete siderophores, low molecular weight compounds, that can chelate iron and convert it into usable forms for cells. One such organism, Rhizobium leguminosarum, is a nitrogen fixing symbiont proteobacteria that infects leguminous plants. The genome of Rhizobium leguminosarum ATCC 14479, which infects the red clover, Trifoli pratense, has previously been completely sequenced in our lab. X-ray diffraction is a robust method for determining the detailed 3D structures of specific proteins. However, this requires the formation of well-ordered protein crystals, a process that is time-consuming, expensive, and only has about a 10-30% success rate. New methods are needed to enable the efficient crystallization of challenging proteins. One such technique is explored here, which utilizes a protein polymer (the sterile alpha motif domain of the human protein translocation Ets leukemia, or TELSAM) as a crystallization chaperone to form a more ordered crystal lattice of target proteins and drive crystallization. Several bulk proteomics studies characterize differences between tumor cells and healthy B cells, yet the transition to symptomatic disease is not well understood. In recent studies small nucleolar RNAs (snoRNA) have been shown to be processed into smaller microRNA-like fragments known as sno-derived RNA (sdRNA). We recently identified 9 snoRNAs that contribute to prostate malignancy using The Cancer Genome Atlas (TCGA) patient prostate tumor next-generation sequencing datasets. These snoRNA were found to be processed into sdRNAs, heavily misexpressed in prostate cancer cell types 8140 and 8550, and shown to function by binding Ago proteins in order to impact mRNA translation. The purpose of the project presented in this thesis is to assess the contribution of inflammasome activation in mediating vascular wall remodeling that occurs following carotid artery injury due to decreased blood flow. To examine this, we used a mouse model of carotid artery ligation in mice lacking the expression of a key mediator of the inflammasome signaling pathway. Following ligation, we completed a histological assessment to compare the degree of the neointima formation and expression of inflammasome related proteins in both injured and the non-injured contralateral carotid artery in both wild-type (WT) and Caspase 1 knockout mice (Cas1 -/-).

This was assessed using students enrolled in introductory psychology courses during either Fall 2019 or Fall 2020 semesters who completed the Narcissistic Personality Inventory during a subject pool registration process. Predictions were made that the NPI scores would be lower for the Fall 2020 sample compared to the Fall 2019 sample.
