Autism Spectrum Disorder (ASD) is a neurodevelopmental disorder typically diagnosedwhen a child displays social and communication impairments, repetitive behaviors, and rhythmic movements like hand flapping. This disorder is unique because of the high variability of symptoms displayed between each individual who is diagnosed. Because of this, no neurological or physiological test exists to diagnose a child with ASD. Bacteria respond to environmental cues through a variety of mechanisms. Two-component systems (TCSs) are a conserved mechanism used by bacteria to accurately respond and sense environmental changes. The monitoring of envelope perturbance is linked to TCS CpxA/CpxR in animal-infecting pathogens. The study of the TCS response regulator (RR) CpxR is largely unexplored in phytopathogens. This work focuses on the genetic linkage between cpxR and T3SS expression regulation. We identified the multiple roles of CpxR on several T3SS regulators and its participation in the bacterial second messenger (c-di-GMP) signaling cascade. All-atom molecular dynamics (MD) simulations combine the high temporal resolution of experimental methods like smFRET and spatial resolution of methods like x-ray crystallography, to provide a detailed dynamic picture of biomolecular processes. Here, microsecond-level atomistic MD simulations have been used to characterize chemomechanical couplings in human fibroblast growth factor 1 (hFGF1) and the spike proteins of SARS CoV-1 and SARS-CoV-2. hFGF1 is a globular signaling protein that is involved in several physiological processes ranging from cell proliferation to wound healing. Epigeal predatory arthropods were collected with pitfall-style traps in nine sandstone glades of varying ecological stages, and management histories, in the Ozarks of Northern Arkansas over three years. These sites were categorized into three Site Types; Intact, high quality glades; Degraded, forest-encroached glades, and Restored glades which had received woody vegetation removal and burning. Collections of ground spiders (Gnaphosidae), predatory ground beetles (Carabidae), and the sole species of scorpion present in the Ozarks, Centruroides vittatus, were reported on as well as the applicability of arthropod collection methods for glade habitat assessment. Immune checkpoint drugs have completely changed the way people treat metastatic melanoma and non-small-cell lung cancer. While the impacts of these immunological checkpoints and their suppression on T cell function are well characterized, their consequences on the tumor microenvironment are not. In a CT26 mouse colorectal cancer model, we employed diffuse reflectance spectroscopy to track in vivo tumor microenvironmental alterations in response to immune checkpoint inhibitors. On three separate days, animals bearing CT26 tumor xenografts were given anti-PD-L1, anti-CTLA-4, a combination of both inhibitors, and isotype control. Formaldehyde fumigation in poultry hatch cabinets has been utilized for sanitation of hatching eggs for over a century. Formaldehyde is utilized to minimize pathogenic microbes on the surface of the egg as well as the microbial bloom during the hatching process. While formaldehyde is effective, its use is regulated in the United States and Europe due to its carcinogenic nature. Formaldehyde has been shown to damage the cuticle of the egg, cause embryonic death, and damage the epithelial lining of the respiratory tract of freshly hatched chicks, predisposing them for respiratory infection. Tumor hypoxia plays an active role in mediating treatment resistance and has been linked to metastases and metastatic potential in cancer. Spectral platforms are increasingly being used to assess biodiversity due to the fact that light reflected from an organism’s surface carries much of information about that organism. This research compared the visual symptomology of four auxin herbicides applied at three rates to simulate tank contamination on non-auxin tolerant soybeans. The herbicides examined were aminocyclopyrachlor (ACP), 2,4-D, dicamba, and aminopyralid. The standard labeled rates (SLR) used to base the application rates were 0.12 kg ae ha-1 for ACP and aminopyralid while dicamba and 2,4-D used 1.12 kg ae ha-1. These four herbicides exhibit similar symptoms on soybeans which is problematic when diagnosing soybean response. The purpose of this study was to investigate the physiological and mechanical comparisons between clipless and flat pedals. Participants (n=4) completed two at-home 20-minute FTP tests: using clipless and flat pedals using Zwift. The order of conditions were randomized for each participant. Participants used their personal Smart Trainers, clipless pedals, and cycling shoes while flat pedals were provided (Syun-LP, Road Bike Platform Pedals). Power, heart rate and cadence were recorded and used for analysis. All dependent variables were compared using paired t-tests. This thesis reports the design and outcomes of several circuits intended for transcranial magnetic stimulation (TMS) and repetitive transcranial magnetic stimulation (rTMS) research. In simple terms, TMS circuits are composed of four main blocks: high voltage power source, energy storage bank, control switch, and coil. Each one of these blocks has characteristics that influence how well the circuit will perform for TMS procedures. Background: Within a large subset of heart failure, cardiac ubiquitin-proteasome system (UPS) inadequacy is causative. A vital UPS regulator is the COP9 signalosome (CSN). The CSN holocomplex is formed by 8 unique protein subunits (COPS1~COPS8) and regulates Cullin-RING ligases via Cullin deneddylation. Cardiomyocyte-restricted knockout (cko) of Cops8 causes massive cardiomyocyte necroptosis via the RIPK1-RIPK3-MLKL pathway, resulting in dilated cardiomyopathy (DCM) and shortened lifespan in mice. CSN’s deneddylase resides in COPS5 and Cops5-cko has not been explored. Climate change and anthropogenic stressors have contributed to rapid declines among various taxonomic groups; however, amphibian declines have been particularly intense and primarily stemmed from warming temperatures, habitat loss, exposure to contaminants, disease, and their subsequent interactions. For plant species, such anthropogenic changes have resulted in increasingly fragmented populations, desynchronized interactions with mutualists, and shifted geographic ranges, among other effects. However, despite numerous examples of human impacts on plant populations, the consequences of these changes on plant reproduction remain poorly understood. The purpose of this project was to determine if different oak/pine arrangements elicited potential beneficial interactions that affected seedling growth, mortality and overall protection from deer browsing. Northern red oak (Quercus rubra L.) and shortleaf pine (Pinus echinata Mill.) were planted together and alone in six different planting patterns and spacings, replicated over three blocks in recent clearcuts in east Tennessee, USA. In vitro produced (IVP) bovine embryos provide enhanced genetic value and a means for studying early embryonic development. However, IVP embryos are developmentally compromised and are lower quality compared to in vivo derived (IVD) embryos. Consequently, after transfer pregnancy rates from IVP embryos are consistently lower, as they exhibit the highest amounts of pregnancy failure during the early embryonic phase (prior to day 28). Production of industrial hemp, Cannabis sativa L., has increased tremendously in the U.S. after its legalization through the Agricultural Improvement Act of 2018, known as the 2018 Farm Bill, which allowed commercial hemp production. By grower number, in 2020, Tennessee was the nation’s largest hemp program with over 1,800 producers licensed to grow up to 6,500 hectares (16,000 acres) according to the Tennessee Department of Agriculture. The native range of Viburnum species spans much of the Northern Hemisphere and extends into the mountains of South America and Southeastern Asia. Their wide geographic range has made Viburnum particularly interesting to phylogeographers and phylogeneticists. Uranium processing and waste storage in unlined waste ponds leached contaminants into the groundwater at Y-12, Oak Ridge, Tennessee, from the 1950s to 1980s. Groundwater wells near the S-3 ponds have had the highest nitrate concentrations of groundwater anywhere in the world (>10,000 mg/L). For reference, the maximum contaminant level for nitrate in drinking water set by the U.S. Environmental Protection Agency is 10 mg/L. With the increase of antibiotic resistant bacteria, current established therapeutic treatment methods are rapidly demonstrating decreasing efficacy. For this project, we are studying a novel alternate mechanism of treating antibiotic resistant bacteria by utilizing magnetic iron oxide nanoparticles (IONPs) coated with a heterobifunctional polyethylene glycol polymer (PEG) and functionalized with glycan structures that can interact with Neisseria gonorrhoeae FA1090. The overall objective of this research was to determine the effects of cashew nut shell extract (CNSE; 59% anacardic acid and 18% cardol) on nutrient digestibility and rumen fermentation profile under in vitro batch culture (Chapter 2) and continuous culture conditions (Chapter 3). Presented research was conducted at Clemson University’s Edisto Research and Education Center to quantify harvest related losses associated with the effects of peanut digger blade geometry, the effects of the peanut digger inversion assembly, and the effects of vine load on digging and strategies to address vine load. There were an estimated 20 million new cancer cases worldwide in 2020, resulting in nearly 1000 deaths per hour [1]. Oral cancer exemplifies the difficulties of treating cancer patients. The first line for oral cancer treatment is surgery and radiation that can lead to patient disfigurement and decreased quality of life in cancer survivors [2-4]. Though there have been many developments in chemotherapy in the last 30 years, the 50% mortality rate associated with oral cancer has not changed [4, 5].
