Photodynamic therapy (PDT) is a minimally invasive treatment that uses light-activated photosensitizers to selectively destroy diseased tissue through the generation of reactive oxygen species. However, conventional in vitro models often fail to accurately reproduce the pharmacokinetics of photosensitizer accumulation and clearance observed in vivo. This study investigated protoporphyrin IX (PpIX) accumulation following 5-aminolevulinic acid (ALA) exposure under physiologically relevant conditions using two-dimensional (2D) monolayers and three-dimensional (3D) spheroids. By comparing continuous ALA exposure with transient exposure followed by ALA washout, this work provides a more biologically relevant framework for studying PpIX kinetics and optimizing photodynamic therapy.

This thesis investigates the relationship between macroscopic rheological signals and underlying microscopic dynamics in complex fluids, specifically focusing on ”living polymers” within the cetyltrimethylammonium bromide (CTAB) and sodium salicylate (NaSal) surfactant system. The research addresses the inverse parameterization problem, demonstrating how bulk measurements like zero-shear viscosity can mask fundamentally different physical topologies, such as purely cylindrical, reptating micelles versus highly branched networks. Through the successful synthesis of viscoelastically ”degenerate” samples, the study utilizes an array of characterization techniques including frequency sweeps, Large Amplitude Oscillatory Shear (LAOS) to resolve these unique underlying states. Distance-selective interactions in Ising systems give rise to a range of rich and unexpected phenomena. A notable example is the so-called Grasshopper Ising model, a conserved spin Ising model with interactions only between spins at a fixed distance. This model is the discrete version of the following mathematical problem: A grasshopper lands at a random point on a planar lawn of area one. This thesis develops a documented, reproducible procedure for building diffraction-limited microscope objectives entirely from catalog singlets, for imaging ultracold 6Li atoms in a ring-trap experiment. The objectives must resolve micron-scale features through a 5 mm fused-silica vacuum window, operate at multiple wavelengths, fit inside a 48 mm magnet bore, and contain no conductive or magnetic material, because the surrounding coils switch 0.1 T fields on microsecond timescales. Commercial long-working-distance objectives are universally housed in metal, which the eddy-current constraint rules out, and custom fabrication of a suitable non-conductive matched pair is estimated at nearly $200k.

Numerically solving the electronic structure problem is a fundamentally difficult problem due to the exponential growth in the dimension of the Hilbert space as the system size increases. In order to solve problems at a chemically relevant accuracy, both the choice of basis set and numerical method are important factors that are intrinsically connected.

In this thesis, we study the discretization and resulting compression of electronic Hamiltonians using diagonal basis sets. A diagonal basis set approximately diagonalizes the matrix and tensor representations of the one- and two-body potentials. The muon is a high-energy particle that can be produced by cosmic rays and trigger upper-atmospheric particle cascades and energetic processes. It has been hypothesized that such cascades are at the onset of lightning. As such, muons serve as a valuable probe for investigating the underlying mechanisms of its initiation, whose full governing dynamics remain vastly unknown despite extensive research. Quantum sensors achieve exceptional measurement sensitivity through coherent, well-isolated quantum systems, but practical deployment is hindered by destructive readouts that require repeated state preparation and create long system dead times. Optical cavities enable continuous, nondestructive measurements with minimal back-action, yet integrating high-finesse cavities into scalable quantum devices remains limited by existing fabrication methods. To overcome these constraints, an adaptive CO₂ laser-milling platform was developed. Guided by glass thermodynamics and melt dynamics, this closed-loop system uses in-situ phase-shifting interferometry to sculpt complex fused-silica surfaces with sub-nanometer root-mean-square roughness. We present the emergence of coherent two-photon backscattering, a manifestation of weak localization, in multiple scattering of maximally entangled pure and fully mixed two-photon states and examine the effect of entanglement and classical correlations. Quantum correlations in backscattering are investigated for finite three-dimensional disordered structures in the weak localization regime, as well as systems of a small number of scatterers with specified spatial arrangements. No assumptions are made about the statistical behavior of the scattering matrix elements. Humidity sensors are pivotal in numerous sectors, such as environmental monitoring, industrial automation, and health-care. This study presents the fabrication and detailed evaluation of an advanced humidity sensor using ethyl cellulose (EC)-coated LaFe0.925Ti0.075O3 (LFTO) perovskite nanoparticles synthesized via the sol-gel method. The LFTO nanopar-ticles exhibited a mesoporous structure with an enhanced surface area, which significantly improved their moisture adsorption capabilities. The innovative application of EC coating addresses critical substrate adhesion issues, enhancing mechanical stability without compromising sensor sensitivity. We present a study of the inclusive charmless semileptonic b decay, bXuℓv at next-to-next-to-leading order (NNLO) in perturbative QCD, with the primary aim of extracting the hadronic structure functions Wi at NNLO. The analysis is based on a numerical calculation of the relevant kinematic distributions using a phase-space slicing method to handle infrared-sensitive contributions from real gluon emissions. We use known results from Heavy Quark Effective Theory and Soft-Collinear Effective Theory to extract the singular terms and construct a model for the regular contributions to the structure functions at NNLO, then perform a fit to the …

Weak value amplification (WVA) has emerged as a powerful technique that enhances measurement precision. However, traditional table-top WVA setups and on-chip demonstrations have not achieved an amplification over 20 dB. The practical limit of the amplification is the fidelity of photon post-selection. To address this limit, we design a weak value device with an over 30 dB interferometer extinction ratio, where the stray light in the dark port is minimized with thermally tunable phase shifters. Background: Breast cancer progression and metastasis are closely linked to alterations in the mechanical properties of tumor cells and their microenvironment. Softer, more deformable cells are often associated with higher metastatic potential. While atomic force microscopy (AFM) is the current gold standard for mechanical characterization, it is limited to surface measurements and can damage 3D cultures. This dissertation presents two experimental investigations at the intersection of quantum sensing and precision optical instrumentation. The primary project demonstrates optically detected nuclear magnetic resonance (NMR) of 13C nuclear spins in diamond, using state-selective Landau-Zener transitions under microwave frequency sweeping to bidirectionally transfer spin polarization between nitrogen-vacancy (NV) electron spins and remote 13C nuclear spins. This enables optical polarization and readout of large ensembles of polarized nuclear spins at low magnetic fields and room temperature, with spin dephasing times limited by longitudinal relaxation of nearby NV electron spins. Frequency metrology and quantum control, which uses light as a tool for measurement and manipulation, relies on spectrally narrow, stable lasers. Often thought of as being perfectly monochromatic and coherent, in reality, the free-running instantaneous linewidth of lasers can be on the order of hundreds of kHz in the millisecond time-frame and drift on the order of tens of MHz over hours. To correct for these instabilities in real-time, a variety of laser stabilization methods have been implemented, including the Pound-Drever-Hall (PDH) method, saturation absorption spectroscopy (SAS), and dichroic atomic vapor laser locking (DAVLL). The growing demand for energy-efficient optical information processing motivates compact nonlinear photonic devices that can operate at low power. Silicon photonics is a mature platform for linear optical functions, but nonlinear operation remains challenging because of its weak Kerr response, two-photon absorption at telecommunication wavelengths, and limited compatibility with deeply subwavelength plasmonic confinement. This thesis computationally investigates epsilon-near-zero thin films integrated into plasmonic waveguide architectures as a route toward stronger light–matter interaction in compact nonlinear devices.

Event-based vision sensors (EVSs) provide unique frequency analysis opportunities due to their event data output and high temporal resolution. Anomaly detection methods used in hyperspectral analysis can be used on the event frequency spectra to detect targets. However, the introduction of a strong, flickering interfering source can reduce the EVS sensitivity and obscure targets of interest. Previous work presented a method showing that targets could still be detected through an overwhelming source using frequency analysis, background suppression, and statistical filtering. Southern Adventist University’s Beamline for Ionic and Neutral Collisions (BLINC) is investigating the interactions between hydrogen molecules and fusion particles. To perform in-house theoretical calculations, the BLINC Theory and Analysis Group is testing Psi4 as a viable computational method for BLINC fusion research. Successful calculations of the potential energy curves and dissociation energies of H2 and H2+ have been completed and compared to known values. Preliminary results align with known values, demonstrating Psi4’s potential for BLINC research. Future work will focus on the vibrational energies, wavefunctions, and transition probabilities of H2 and H2+.

This work presents the desig and development of an alkali metal oven used for charge-transfer collision experiments within the Beamline for Ionic and Neutral Collisions (BLINC) at Southern Adventist University. This oven is engineered to produce a stable and controllable alkali vapor source (K/Cs), enabling interactions between the H2+ ions and neutral alkali atoms for the study of rovibrational energy transfer.

Deep Virtual Compton Scattering (DVCS), the exclusive electron-proton scattering process ep →e′p′γ, provides access to generalized parton distributions (GPDs), which correlate informa tion about the longitudinal momentum and transverse spatial structure of quarks inside the nucleon. Experiment E12-13-010 in Hall C at Jefferson Lab was designed to take high-precision measure ments of the DVCS cross section over an extended kinematic range using the newly commis sioned Neutral Particle Spectrometer (NPS).
