Drug metabolism is the biochemical process of modifying drugs to detoxify and remove them through enzymatic transformations. These biotransformation’s occur primarily in the liver and are critical to understanding how pharmaceutical compounds are chemically altered inside the human body. Human carboxylesterases (CESs) catalyze the hydrolysis of esters, amides, thioesters, and carbamates. CES-mediated hydrolysis plays an important role in the metabolism of many drugs including the first FDA approved antiviral treatment for COVID-19, remdesivir (Veklury), the seizure control medication rufinamide (Banzel), and the flu antiviral drug oseltamivir (Tamiflu). Trichomonas vaginalis is responsible for 156 million new cases per year worldwide. When present asymptomatically, the parasite can lead to serious complications, such as development of cervical and prostate cancer. As infection increases the acquisition and transmission of HIV, the control of trichomoniasis represents an important niche for the discovery and development of new antiparasitic molecules. This urogenital parasite synthesizes several molecules that allow the establishment and pathogenesis of infection. Intranasal vaccines hold enormous potential for improving vaccine efficacy against respiratory pathogens by activating the mucosal immune system. The urgency for novel approaches in vaccine development is amplified by the recent COVID-19 pandemic. Most intranasal vaccines that were developed are either live-attenuated or inactivated vaccine candidates that demonstrated poor safety profiles. Protein-based subunit vaccines represent a safe and cost-effective alternative, but they are often weak immunogenically. TLR7/8 adjuvants such as INI-4001 have shown promise for enhancing the humoral and cellular immunogenicity of protein-based subunit vaccines. Epithelial-mesenchymal transition is a process by which cancer cells increase their capability for metastasis. EMT involves conversion of epithelial cells into a mesenchymal phenotype which makes cancer cells more invasive and motile. To investigate whether higher order packing interactions confer protein-DNA specificity, a modified Knob-Socket (KS) model was used to analyze the interface of bZIP-DNA crystal structures. The KS analysis identified a nine-residue quadripartite recognition core consisting of four contiguous KS pockets P1, P2, N3, and N4 that each pack one of the four DNA half-site bases in the target sequence. Human stem cells have revolutionized the study of early neural development, offering a powerful tool to investigate the complex processes involved in the formation of the nervous system. In this study, the human pluripotent stem cell line, TERA2.cl.SP12, was utilized to derive neuroglial cells and to determine their differentiation and functional maturation up to one year in culture; this model was then investigated as a new tool for the study of neurodevelopmental toxicity, for seizure modeling and as a screening method for the investigation of new anticonvulsants. Pharmacogenomics (PGx) is considered an emergent field in developing countries. Research on PGx in the Latin American and the Caribbean (LAC) region remains scarce, with limited information in some populations. Thus, extrapolations are complicated, especially in mixed populations. In this paper, we reviewed and analyzed pharmacogenomic knowledge among the LAC scientific and clinical community and examined barriers to clinical application. We performed a search for publications and clinical trials in the field worldwide and evaluated the contribution of LAC. Curcumin is a natural active ingredient from traditional Chinese medicine (TCM) that has multi-target characteristics to exert extensive pharmacological activities and thus has been applied in the treatment of various diseases such as cancer, cardiovascular diseases, nervous system, and autoimmune disorders. As an important class of membranous organelles in the intracellular membrane system, lysosomes are involved in biological processes such as programmed cell death, cell metabolism, and immune regulation, thus affecting tumor initiation and progression. ที่มาและความสำคัญ: การตรวจยีนแพ้ยา HLA-B*58:01 ก่อนเริ่มใช้ยา allopurinol สามารถช่วยป้องกันการแพ้ยาทางผิวหนังจากการใช้ยา allopurinol ได้ วัตถุประสงค์: เพื่อเปรียบเทียบอุบัติการณ์การแพ้ยาทางผิวหนังจากยา allopurinol โดยรวมทั้งชนิดที่ไม่รุนแรงและรุนแรง และศึกษาความแตกต่างของค่าใช้จ่ายทางตรงที่เกี่ยวข้องกับการรักษาเมื่อพบการแพ้ยาในกลุ่มผู้ป่วยที่ได้รับการตรวจยีนแพ้ยา (กลุ่มศึกษา) และผู้ป่วยที่ไม่ได้รับการตรวจยีนแพ้ยา (กลุ่มเปรียบเทียบ) วิธีการศึกษา: การวิจัยจากเหตุไปหาผลแบบย้อนหลัง ผู้ป่วยที่เข้ารับการรักษาในโรงพยาบาลเจริญกรุงประชารักษ์ กรุงเทพมหานคร ระหว่างวันที่ 1 มกราคม พ.ศ. 2562 ถึงวันที่ 31 มีนาคม พ.ศ. 2567 ผู้ป่วยที่ผ่านคุณสมบัติตามที่ผู้วิจัยกำหนดทั้งหมด 1,026 ราย แบ่งเป็นกลุ่มศึกษา 281 รายและกลุ่มเปรียบเทียบ 745 ราย ผลการศึกษา: ผู้ป่วยกลุ่มศึกษา 281 ราย ตรวจพบยีนแพ้ยาเป็นบวก 53 ราย (ร้อยละ 18.8) ได้รับการรักษาด้วยยาลดกรดยูริกในเลือดอื่น ๆ 32 ราย ตรวจพบยีนแพ้ยาเป็นลบ 228 ราย (ร้อยละ 81.14) ได้รับการรักษาด้วยยา allopurinol 163 ราย และกลุ่มเปรียบเทียบ 745 ราย ผู้ป่วยทุกรายได้รับการรักษาด้วยยา allopurinol ที่ระยะเวลาการติดตามการแพ้ยา 90 วัน ในกลุ่มศึกษาพบอุบัติการณ์การแพ้ยาทางผิวหนังโดยรวม และชนิดที่ไม่รุนแรงเท่ากันร้อยละ 1.4 และกลุ่มเปรียบเทียบร้อยละ 5.9 และ 5.0 ตามลำดับ อย่างมีนัยสำคัญทางสถิติ (p = 0.007, 0.02 ตามลำดับ) และในกลุ่มศึกษาไม่พบอุบัติการณ์การแพ้ยาทางผิวหนังชนิดที่รุนแรง แต่กลุ่มเปรียบเทียบพบร้อยละ 0.9 (p = 0.216) ความแตกต่างของค่าใช้จ่ายทางตรงที่เกี่ยวข้องกับการรักษาเมื่อพบการแพ้ยาระหว่างกลุ่มศึกษาน้อยกว่ากลุ่มเปรียบเทียบ 78,198.52 บาทต่อราย สรุปผลการศึกษา: การตรวจคัดกรองยีน HLA-B*58:01 ก่อนเริ่มใช้ยา allopurinol มีประสิทธิผลในการลดอุบัติการณ์แพ้ยาทางผิวหนัง และลดค่าใช้จ่ายทางตรงที่เกี่ยวข้องกับการแพ้ยา

Lung, breast, and colorectal cancers significantly contribute to the high incidence and mortality rates of cancer worldwide. Naphthoquinone is a well-known anticancer agent, including lawsone and napabucasin. The series of lawsone (bicyclic system) and napabucasin (tricyclic system) were designed to find the suitable target proteins though computational modelling, predicting the interactions between 172 naphthoquinone analogues and 12 potential cancer-related target proteins. The virtual study proposed the naphthoquinone analogues based on their superior docking scores, interactions, and absorption, distribution, metabolism, and elimination (ADME) predictions compared to lawsone and napabucasin. Interestingly, the PI3K/AKT/mTOR proteins emerged as the promising target for the naphthoquinones. …

An increase in lysosomal membrane permeabilization (LMP) impaired lysosome integrity causing release of cathepsin B, and subsequently may contribute to cell death. Indomethacin, which is a non-steroidal anti-inflammatory drugs (NSAIDs), caused apoptosis through generation of cell stress, and disruption of mitochondria and lysosome functions. Recently, it was reported that naringenin, a natural bioactive flavonoid, elicited cytoprotection against indomethacin toxicity in Caco-2 cells. A simple, accurate, and low-cost effective method has been introduced in this study for separation of valsartan enantiomers using ligand-exchange chromatography as chiral mobile phase additives. The optimized method was found using a mobile phase system of 57%(v/v) methanol, adding 1.5 mM of L-isoleucine as the chiral selector along with 0.5 mM of cupric sulfate and 10 mM of sodium acetate, adjusted to pH 6.1.
