Human Leukocyte Antigen (HLA) is a gene that codes for cell-surface proteins that are the basis of our bodies’ immune response, and are crucial for the combatting of pathogens or infections. For our experiment we observed two cell lines, a human Leukemia cell line (HL-60), and an epidermoid carcinoma cell line A-431. Our purpose for this experiment was to observe the effects of two drugs as well as the combination of them on the expression of HLA antigens in these cancerous cell lines. The drugs being used in this experiment are Gamma interferon, which is known to increase antigen presentation …

Abstract
Pyrrole-imidazole polyamides (PAs) are small molecules that typically develop H-bonds to bind to the minor groove of DNA. PAs are of interest because they can be designed to recognize DNA sequences. PAs have numerous biomedical applications in areas like regulation of gene expression and antimicrobial activity. Specifically, polyamide UMSL1011 (a polyamide synthesized in Dr. Bashkin's lab) inhibits replicating vesicular stomatitis virus (VSV) by binding the viral RNA inside the nucleocapsid, as indicated by research at Professor Ming Luo's lab at Georgia State University. However, some reports have revealed that polyamides have a low affinity for “simple” double-stranded RNA. Nevertheless, …

Tpt1, an essential component of the fungal and plant tRNA splicing machinery, catalyzes transfer of an internal RNA 2′-PO4 to NAD+ yielding RNA 2′-OH and ADP-ribose-1′,2′-cyclic phosphate products. Here, we report NMR structures of the Tpt1 ortholog from the bacterium Runella slithyformis (RslTpt1), as apoenzyme and bound to NAD+. RslTpt1 consists of N- and C-terminal lobes with substantial inter-lobe dynamics in the free and NAD+-bound states. ITC measurements of RslTpt1 binding to NAD+ (KD ∼31 μM), ADP-ribose (∼96 μM) and ADP (∼123 μM) indicate that substrate affinity is determined primarily by …

Alzheimer disease (AD) is recognized as the six leading cause of the death in the United States. As of now, there is no cure for this fatal disease. The current treatment methods can only temporarily slow the worsening of symptoms. Research data suggested that an excess generation of hydroxyl radical in the brain causing the aggregation of Amyloid-β (Aβ) peptide which is considered to be responsible for Alzheimer's disease. Thus, there is a pressing need to find a suitable drug which can quench hydroxyl radicals effectively and stop or slow down the formation of aggregation of Aβ peptide. The primary …

Since the discovery of G-quartet (G4)by M. Gellertin 1962, much attention has been given on G4and C4(also called i-motif) as important drug design targets for the treatment of various human disorders. G4 forming sequences are prevalent in human genome, which includes many important regions of the eukaryotic genome, such as telomere ends, regulatory regions of many oncogenes c-kit, proto-oncogene c-myc, Kirsten rat sarcoma viral oncogene homolog (KRas). Curcumin(diferuloylmethane), an antiinflammatory and antioxidant compound, is found in the rhizomes of the plant Curcuma longa. The phytopolyphenolic chemical curcumin has been in the prominence due to its diverse pharmacological activities. Here, we …

Undergraduate Research Conference
The enzyme human prolyl peptidase, or hPEP, has been linked to many processes within the brain, including breaking down neural peptides. A bovine milk protein, a-s1-casein (see above), has been found to inhibit hPEP in colon cancer cells. A 68 amino acid long section (bolded) of a-s1- casein containing the inhibitor region was cloned. The casein fragment expression was optimized for expression of the peptide. The purpose for optimizing expression is to have much of it on hand to utilize in further studies on its inhibitory properties on hPEP. By using this fragment of casein, one can study the structure …

As the technology for science develops, the research strategy in medicines and therapeutics also improves. In this paper, I will cover the process of Sonogashira cross-coupling and Amide Coupling reaction for an anticancer agent in both batch and flow chemistry. Continuous Flow Chemistry has advantages such as being more efficient, safer, and faster. This paper studies the synthesis of HSNO608, an anticancer lead compound for Acute Myeloid Leukemia (AML), which has a specific potent activity to FTL3 Kinase. Inhibition of FLT3 Kinase leads to inhibition of downstream pathways such as MPK and P13K pathways. In this two-step experiment, the Sonogashira …

Development Of A Rapid Drug Detection Method For Insects Using Paper Spray Ionization Mass Spectrometry (Psi-Ms), Alexandria Plyler
Undergraduate Research and Scholarship Symposium
Insects have the ability to reflect the chemical environment that they have developed in. Thus, necrogenous insects can reflect the chemical environment of cadavers they have fed on. Forensic scientists can potentially utilize insect larvae as an alternate biological matrix to detect drugs in cadavers that are skeletonized, burned, or otherwise badly damaged. Analysis of insects using paper spray ionization mass spectrometry (PSI-MS), an analytical method requiring little to no sample preparation, could prove a rapid, cost-effective, and non-destructive alternative form of toxicological analysis. Traditional tissue analysis via GC and LC-MS often require lengthy sample preparation, use expensive reagents, and …

Corruption In Capsules: How It Is Legal For Companies To Put Harmful Ingredients In Vitamins And Dietary Supplements, Emily Leggiero
English Department: Research for Change - Wicked Problems in Our World
The vitamin and supplement industry has increased exponentially in profits as well as potential products on the market since the turn of the century. However, these products are not regulated, nor do they undergo any premarket clinical research or testing. Public health is compromised by vitamins and supplements that are available for American consumption that is disproportionately unregulated to their chemically similar counterparts. This wicked problem is facilitated through the combination of historical legislative definitions that has since been distorted for corrupt administrative gain through the allotment of corporate expenditures. Company disbursements are made to the same policymakers that create …

Drugs have a serious effect on our mental health, and mental health has a major effect on drug abuse and addiction. There is a reason so many drug users usually need to keep going back to rehab. It is because drugs affect mental health to a point where you aren’t the same person during and even after drug addiction. Poor mental health also can be a major cause influencing people to start doing drugs. People with depression, anxiety disorders, mood disorders are more prone to drug use. That is why we need to make it a public policy to screen …

The problem of medication pricing and lobbying in the healthcare industry has caused millions of people within the United States to not be able to afford medications essential to their lives. Pharmaceutical companies are taking advantage of the need people have for certain drugs by setting the sale price far higher than it costs to produce. My suggested solution is to make lobbying illegal in the United States so that pharmaceutical companies would not have control over government officials who pass laws to help or hurt the issue of medication pricing. Illegalizing government lobbying will take away the incentive politicians …

We are investigating the effect of Crotalus adamanteus acidic phospholipase A2 on MCF-7 cells using the MTS assay. Understanding these interactions and isolated effects is critical to developing new ways to treat envenomation. By understanding the effects of individual toxins within a whole venom, we are set to better understand the effects of the whole venom and investigate synergistic actions between venom toxins. In this paper, we are quantifying the amount of MCF-7 cell death caused by Crotalus adamanteus phospholipase A2 on MCF-7 using the MTS assay. Analysis of the amount of cells death caused by the phospholipase …
