Although host cells are a rather rich source for co-opted host factors, lipids and metabolites, positive stranded RNA viruses vastly rewire cellular pathways and remodel cellular membranes to support viral replication. To accomplish such major changes, these viruses depend on the availability of different host factors and the ability to readily assemble viral replication organelles (VROs). Genome-wide screens and proteomics approaches with Tomato Bushy Stunt Virus (TBSV) in a yeast model host indicated that tombusviruses rely on the cellular cytoskeleton to reorganize the cellular environment of their hosts. During mycoparasitism, a fungus—the host—is parasitized by another fungus—the mycoparasite. The genetic underpinnings of these relationships have been best characterized in ascomycete fungi. However, within basidiomycete fungi, there are rare instances of mushroom-forming species parasitizing the reproductive structures, or sporocarps, of other mushroom-forming species, which have been rarely investigated on a genetic level. One of the most enigmatic of these occurs between Entoloma abortivum and species of Armillaria, where hyphae of E. abortivum are hypothesized to disrupt the development of Armillaria sporocarps, resulting in the formation of carpophoroids. Obtaining good accuracy and reliability of estimated breeding values is essential to increase the efficiency of a plant breeding program. Genetic variation was assessed for categorical (Virc) and binary (Virb) mosaic (caused by Panicum mosaic virus), dry matter (DMY) and predicted ethanol (Etoh) yields, and lignin content (Klason or KL, and acid-detergent or ADL) in a Summer–Kanlow switchgrass (Panicum virgatum L.) population. Breeding values were predicted with the restricted maximum likelihood–best linear unbiased prediction (REML-BLUP) approaches using a multivariate phenotypic (PBLUP) and animal (ABLUP) models, integrating a three-generation pedigree (1,622 half-sibs) in ABLUP and not in PBLUP. Promoters play critical roles in controlling the transcription of genes and are important as tools to drive heterologous expression for biotechnological applications. In addition to core transcription factor-binding motifs that assist in the binding of RNA polymerases, there are specific nucleotide sequences in a promoter region to allow regulation of gene expression. The allene oxide synthase (AOS) gene family are cytochrome P450s that are responsive to a variety of environmental stress, making them good candidates for the discovery of inducible promoters. Pratylenchus smoliki is a new species of root-lesion nematode described from corn-soybean production fields in the Central Great Plains of North America. It is characterized by populations with relatively abundant males, two lip annuli, females with a round functional spermatheca and a conoid to subcylindrical tail with a non-crenate, smooth terminus. In host preference tests, corn and wheat produce the largest nematode populations, whereas sorghum and soybeans produce less than 20% the numbers observed on corn. Several Bacillus amyloliquefaciens isolates obtained from wheat residue have been studied as biological agents to control wheat diseases. Notable traits of isolates 1BA and 1D3 include growth with high salt (10% NaCl), at temperatures up to 50°C and over a wide pH range. BIOLOG Gen III plates were used to study and further characterize Bacillus amyloliquefaciens 1BA and 1D3. Both isolates showed tolerance to high salt concentration supporting previous studies. They also grew in the presence of lithium chloride, potassium tellurite, and sodium bromate. Both isolates grew at pH 5, with almost identical carbon source utilization fingerprints. Brief history: In 1993, severe mosaic and necrosis symptoms were observed on corn (maize) and wheat from several Great Plains states of the USA. Based on the geographical location of infections, the disease was named High Plains disease and the causal agent was tentatively named High Plains virus. Subsequently, researchers renamed this virus as maize red stripe virus and wheat mosaic virus to represent the host and symptom phenotype of the virus. Positive-stranded (+) RNA viruses are the largest family of viruses that infect plants, causing important economic losses in different crops. Tomato bushy stunt virus (TBSV), a small positive-stranded RNA virus, has emerged as a model virus to study virus-host interactions. TBSV encodes for only five proteins, therefore, to infect the host cell TBSV co-opts selected host components and subverts specific molecular pathways.
Firstly, I performed a proteomic screening using Arabidopsis proteins. I found that TBSV viral replication proteins interact with 88 host proteins, including the ubiquitin-conjugating enzyme 2 (Ubc2), fructose 1,6 biphosphate aldolase (Fba1), and several members of the Hps70 …

Bipolaris leaf spot (BLS) disease emerged with the reintroduction of hemp as a crop in the United States following more than 60 years of prohibition. The causal agent was identified as Bipolaris gigantea (=Drechslera gigantea), a known minor pathogen of monocots which causes devastating disease on hemp. BLS has been confirmed throughout Kentucky and reported in 15 states. Morphology and growth characteristics of isolates from eight counties across Kentucky were similar with the exception of some isolates producing protoperithecial-like structures. Burley and dark tobacco production are important to agriculture in Kentucky, worth $145.1 and $94.4 million in 2020, respectively. The price received for a tobacco crop is influenced by leaf quality, determined by multiple characteristics including leaf damage. Frogeye leaf spot (FLS), caused by Cercospora nicotianae, has historically been a minor disease of tobacco. However, when FLS infections reach the upper canopy of tobacco, lesions are seen as damage that negatively influence leaf quality. Fungicides are regularly used to manage foliar diseases of tobacco, and the only systemic fungicide active ingredient labeled for tobacco is azoxystrobin. Azoxystrobin belongs to the …

White pine blister rust (WPBR) has been affecting New Hampshire’s white pines for more than a century, yet no data exist on the long-term effects of the non-native disease on the state’s forests, particularly with respect to the regeneration and sustainability of white pine, and forest succession. Alternatives to protect crops against diseases are desperately needed to secure world food production and make agriculture more sustainable. Genetic resistance to pathogens utilized so far is mostly based on single dominant resistance genes that mediate specific recognition of invaders and that is often rapidly broken by pathogen variants. Perturbation of plant susceptibility (S) genes offers an alternative providing plants with recessive resistance that is proposed to be more durable. S genes enable the establishment of plant disease, and their inactivation provides opportunities for resistance breeding of crops. However, loss of S gene function can have pleiotropic effects. Dry edible beans are a vital food source in Mozambique, East Africa—one that alleviates hunger and malnutrition and adds value to the economy. In recent years, root/crown rot (RCR) pathogens have emerged as limiting constraints in dry bean production. Not much has been characterized concerning the causal agents of RCR in Mozambique. The purpose of this study was to identify the primary pathogen(s) associated with RCR dry bean samples collected at breeder nursery sites and farmer fields in Mozambique using molecular sequencing and culture-based methods. Lichens are traditionally defined as a symbiotic relationship between fungi and algae and/or cyanobacteria. This union forms a unique structure called the thallus, which attaches to surfaces such as rocks and tree bark. Recent reports challenge the view that lichens are comprised of one fungus and one photobiont, and instead suggest that they are a consortium of microbes. Much of lichen biology remains unknown as most of our knowledge of lichens is limited to morphological characteristics with little to no functional analysis of lichen genes. Fall implementation of a rye (Secale cereale L.) cover crop (RCC) prior to spring corn (Zea mays L.) planting is a management practice used to improve soil conservation, water quality, and limit herbicide dependence. However, corn growth and yield following a RCC is often reduced due to early-season nitrogen (N) stress and decreased plant emergence, which can limit RCC adoption.
