Nebraska’s Eastern Saline Wetlands are unique ecosystems endemic to the Salt and Rock Creek waters in Lancaster and Saunders County.

They provide an ecosystem services as well as habitat for endangered species such as the state endangered saltwort (Salicornia rubra) and federally endangered Salt Creek tiger beetle (Cicindela nevadica lincolniana).

Over 80 % of the saline wetlands are highly degraded and in recent years, the Saline Wetland Conservation Partnership has formed to conserve and restore the remaining saline wetland fragments, but there is limited information about inland saline wetland restoration. Information Thermodynamics Of Cytosine Dna Methylation, Robersy Sanchez Rodriguez, Sally Ann Mackenzie

Cytosine DNA methylation (CDM) is a stable epigenetic modification to the genome and a widespread regulatory process in living organisms that involves multicomponent molecular machines. Genome-wide cytosine methylation patterning participates in the epigenetic reprogramming of a cell, suggesting that the biological information contained within methylation positions may be amenable to decoding. Adaptation to a new cellular or organismal environment also implies the potential for genome-wide redistribution of CDM changes that will ensure the stability of DNA molecules. This raises the question of whether or not we would be able to sort out the regulatory methylation signals from the CDM background …

Arabidopsis thaliana is an important model organism with a robust network of resources that has been of enormous value to the plant science research community. The use of isogenic material as a reference point or control is critical for many types of experiments in plant molecular biology and genetics. Recently, we noticed that some seed from a common source of the widely used Columbia-0 (Col-0) strain gave rise to plants showing features atypical for this strain. Whole-genome DNA-sequencing and allelespecific PCR assays confirmed that the abnormal individuals contain multiple introgressions from the ecotype Wassilewskija-2 (Ws-2), as described below. This …

High school agricultural education teachers have expressed concern about the lack of easily accessible educational materials dealing with contemporary topics in sustainable agriculture. There are numerous textbooks and monographs available for farmers and students at the college level, including the highly practical resources available from the Sustainable Agriculture Research and Education (SARE) book series on soil fertility (Magdoff and van Es, 2010), cover crops (Bowman et al., 2007) and building a farm business (DiGiacomo et al., 2003), among others. Poor stand establishment, failure to recover after grazing, and premature plant death have reduced the utilization of arrowleaf clover (Trifolium vesiculosum Savi) as a forage crop in the southeastern United States in recent years. Clover plants collected from poor stands in east Texas pastures during the 1995 to 1996 and 1996 to 1997 seasons first exhibited root disease symptoms as young seedlings in the fall. Symptoms consisted of one or more of the following: tan discoloration of lateral roots and taproot; root pruning; and small, tan, sunken lesions on the taproot and crown. Fresh strawberries are highly perishable and have a short shelf-life especially when the cold chain is not maintained. Strawberries exported to Asia are currently fumigated with methyl bromide for phytosanitary purposes, which exposes strawberries to warm temperatures for several hours and air freight without temperature control, resulting in a shelf life of just a few days in the destination country. Irradiation offers an efficacious alternative to fumigation and can be performed on cold fruit. The interaction between lipid oxidation products and bovine sarcoplasmic (SP) and myofibrillar protein (MP) homogenates in the presence of green tea was investigated. To monitor the effect of green tea on lipid oxidation, aldehydes were measured while effect on protein was monitored via changes in myoglobin, thiols, and tryptophan fluorescence over nine days of refrigerated storage. The presence of SP and MP decreased free aldehydes in the buffers. The SP bound more aldehydes than MP. The tea compounds exhibited more favorable binding energies than aldehydes near histidine 64 close to the heme moiety of myoglobin. Addition of tea lowered tryptophan …

The effect of surface or lattice modification of nanoparticles (NPs) on terrestrial plants is poorly understood. We investigated the impact of different zinc oxide (ZnO) NPs on green pea (Pisum sativum L.), one of the highest consumed legumes globally. Pea plants were grown for 65 d in soil amended with commercially available bare ZnO NPs (10 nm), 2 wt% alumina doped (Al2O3@ZnO NPs, 15 nm), or 1 wt% aminopropyltriethoxysilane coated NPs (KH550@ZnO NP, 20 nm) at 250 and 1000 mg NP/kg soil inside a greenhouse. Corn (Zea mays L.) N use is of continued interest due to agronomic performance and environmental issues. This 2-yr study evaluated era hybrid response to fertilizer nitrogen (FN) rate in a factorial arrangement of one popular hybrid per five decades (1960–2000 eras) and five N rates (0–224 kg N ha–1). An additional hybrid per era was grown at 168 kg N ha–1. Hybrid productivity and nitrogen use efficiency (NUE) increased across the eras, but not between the 1980 and 1990 eras. The objective of the trial was to compare foliar fungicides applied to three growth stages of corn for grey leaf spot (GLS) and southern rust (SR) efficacy. Irrigated corn was grown based on Nebraska Extension irrigation recommendations at the South Central Ag Lab near Clay Center, NE. Soils were a silt loam with 6.7 pH and 1.8% OM and the previous crop was soybean. Reduced tillage was performed to the field prior to planting. Some plant-parasitic nematodes occur in every [sunflower] production field. Nematode populations likely include multiple species, although not all species cause notable disease. Symptoms depend on the species, the population density, and the crop's interaction with environmental factors. Several nematode species are known to cause disease in sunflower, although little research has focused on their impact or management in the crop. The highest population densities of plant-parasitic nematodes often occur in isolated areas that are randomly distributed in fields, as do the most severe plant symptoms. Symptoms are not necessarily diagnostic for nematodes, and the problem may be misdiagnosed. The most …

Sweet sorghum [Sorghum bicolor (L.) Moench] has potential for bioenergy. Itis adapted to a variety of U.S. locations and the extracted juice can be directly fermented into ethanol. However, little research on fungal stalk rots, diseases that pose serious constraints for yield and quality of juice and biomass, has been reported. A greenhouse bioassay was designed to assess charcoal rot (Macrophomina phaseolina) and Fusarium stalk rot (Fusarium thapsinum) in plants at maturity, the developmental stage at which these diseases are manifested. Pathogen exposure to sublethal doses of fungicides may result in mutations that may represent an important and largely overlooked mechanism of introducing new genetic variation into strictly clonal populations, including acquisition of fungicide resistance. We tested this hypothesis using the clonal plant pathogen, Sclerotinia sclerotiorum. Nine susceptible isolates were exposed independently to five commercial fungicides with different modes of action: boscalid (respiration inhibitor), iprodione (unclear mode of action), thiophanate methyl (inhibition of microtubulin synthesis) and azoxystrobin and pyraclostrobin (quinone outside inhibitors). Ecological theory predicts that diversity decreases variability in ecosystem function. We predict that, at the landscape scale, spatial variability created by a mosaic of contrasting patches that differ in time since disturbance will decrease temporal variability in aboveground plant biomass. Using data from a multi-year study of seven grazed tallgrass prairie landscapes, each experimentally managed for one to eight patches, we show that increased spatial variability driven by spatially patchy fire and herbivory reduces temporal variability in aboveground plant biomass. This pattern is associated with statistical evidence for the portfolio effect and a positive relationship between temporal variability and functional …

Efforts to mitigate shoreline erosion through living shoreline methods along the USA Atlantic seaboard have often incorporated the cultivation and transplantation of smooth cordgrass, Spartina alterniflora. Assessments of these transplants at several sites in the Indian River Lagoon have shown that survival is variable after a year (survival: 10-93%). Lower survival has been attributed to environmental variables such as dislodgement by wave energy, and transplant shock due to salinity changes from cultivation to estuarine conditions. To improve living shoreline projects, we examined the effects of cultivation salinity (0 ppt, 15ppt) on transplantation success, and the success of anchoring plants …
