Radiofrequency ablation (RFA) is a minimally invasive ablative modality for the treatment of thyroid nodules. Reports of RFA use have demonstrated an impressive safety profile and excellent volume reduction rates between 60-90%. Given its increased popularity in the United States as well as globally, numerous recent works have been published and a discussant of relevant complications incorporating recent insight may assist practitioners in minimizing complications and optimizing patient outcomes. Herein, we provide a comprehensive and updated review of the reported complications and side effects following RFA, summarizing their frequency and clinical presentation. The circadian system coordinates 24-hour cycles of internal biological processes with the environmental light-dark cycle. Abrupt shifts in the timing of the light-dark cycle misalign internal circadian clocks with the environment and cause jet lag until resynchronization occurs. The objective of this study was to investigate the mechanisms underlying sex differences in jet lag in mice. Female mice resynchronized faster than male mice to 6-hour advances and delays of the light-dark cycle, which mimicked eastward and westward travel, respectively. Circulating estradiol was necessary for rapid resynchronization in female mice since ovariectomized females resynchronized slower than mice treated with estradiol. We …

Horses are regularly transported across the United States; however, the reasons for travel, trip lengths, and management practices are not well characterized. While long distance transportation has been associated with increased incidences of health-related problems, the impact of short distance transportation on horse health is less understood. Notably, aged horses ( > 15 years) have an altered immune response following short-distance transportation, but this has yet to be compared to young horses. It is also unknown whether transportation stress would alter endocrine responses of horses with metabolic disorders, such as insulin dysregulation (ID) or pituitary pars intermedia dysfunction (PPID).

Titin, the third most abundant filament in muscle, serves many roles. It underlies muscle stiffness and functions as a signaling hub particularly during muscular growth. The PEVK region of titin contributes to muscle stiffness and is suspected to play a role in hypertrophic signaling. Regulation of small GTPases within the pancreatic beta cell is a highly complex and coordinated sequence of protein interactions and post translational modifications. Various families participate in the physiological secretion of insulin in roles ranging from cytoskeletal reorganization to secretory vesicle priming and docking. However the regulatory partners of GTPase function, while studied broadly, lack vital information relating to smgGDS, a protein chaperone of small GTPases to and from the prenylation machinery, and RhoGDI function. The neural mechanisms that control the onset of puberty are not completely understood. In livestock, the onset of puberty relies on an increase in gonadotropin releasing hormone (GnRH) and luteinizing hormone (LH). Kisspeptin, neurokinin B, and dynorphin neurons (KNDy) and their receptors, Kiss1r, NK3R, and KOR, respectively, are important regulators of puberty. Previous data from our laboratory has shown that KNDy neurons may be responding to other inputs, including the melanocortin system, that will affect the timing of puberty. The energetic state of an animal has vital consequences for its fitness, but it can be difficult to detect through behavioral observations. The development of techniques to determine an animals’ energetic state non-invasively would allow conservation planners to rapidly assess the energy balance and conservation potential of individuals, groups, and populations. Thyroid hormones like triiodothyronine (T3) regulate and reflect metabolic state in mammals. T3 is a non-invasive biomarker that can be detected in feces and could potentially be used for this purpose. In female sheep, the release of gonadotropin-releasing hormone (GnRH) from the hypothalamus triggers luteinizing hormone (LH) to be released from the pituitary. Luteinizing hormone, in turn, acts on the ovaries to elicit estradiol (E2) release. E2 acts in a negative feedback manner to control the amount of LH released. The increase in LH release that drives puberty onset results from a decrease in sensitivity to E2 as the animal ages. Biomanufactured 3D-organoids show promise for modeling and treating various conditions, particularly insulin-dependent diabetes mellitus (type-1 diabetes). Pancreatic islet organoids serve as sustainable sources of insulin-producing β-cells, potentially restoring insulin regulation in patients and providing a long-term solution for diabetes. Amidst the amphibian extinction crisis, in situ and ex situ amphibian species conservation initiatives utilize assisted reproductive technologies for optimal genetic management of captive and wild populations. Development of effective, simple, low-cost methods for obtaining gametes for artificial fertilization, sperm biobanking, or natural breeding alleviates obstacles for institutions or programs to apply reproductive technologies. Objectives herein include investigating the efficacy of alternative hormones for inducing spermiation, an alternative hormone administration route for inducing ovulation, exploring the physiological effects of hormone therapy, and utilizing sperm from deceased animals for biobanking.

Abstract Obesity-associated diabetes is linked to the accumulation of ceramide in various organs, including the liver. The exact mechanisms by which ceramide contributes to diabetic pathology are unclear, but one proposed scenario is that ceramide accumulation may inhibit insulin signaling pathways. It is unknown however whether the excess ceramide is generated proximal to the insulin receptor, that is, at the plasma membrane (PM), where it could affect the insulin signaling pathway directly, or the onset of insulin resistance is due to ceramide- induced mitochondrial dysfunction and/or lipotoxicity. Dogs were chosen as the model organism because testes are readily available. Canine testes were obtained from a local veterinary clinic. Thin sections were generated using a commercial electric slicer. Sleep is one of the body’s greatest and most basic needs. Chronic sleep loss (<6 hours a day) is linked to many cardiovascular issues, diabetes, and inflammatory conditions and has been shown to lower life expectancy. Obstructive sleep apnea (OSA) is a complication that causes sleep fragmentation in >8% of adults from developed countries, and many with OSA may be unaware of their condition. Having long-term OSA is linked to neuroinflammation caused by activated microglia in the brain, but the mechanics of this inflammation are not well understood. Non-alcoholic fatty liver disease (NAFLD) is a liver manifestation of metabolic syndrome, and is estimated to affect one billion individuals worldwide. An increased intake of a high-fat diet (HFD) and sugar-sweetened beverages are risk-factors for NAFLD development, but how their combined intake promotes progression to a more severe form of liver injury is unknown. Here we show that fructose metabolism via ketohexokinase (KHK) C isoform leads to unresolved endoplasmic reticulum (ER) stress when coupled with a HFD intake. PURPOSE: This study investigated changes in salivary α-amylase (sAA), cortisol, and interleukin-1β (IL-1β) concentrations following completion of an ultra-marathon trail run (UMT) to better understand the physiological stressors imposed by this extreme type of race. METHODS: Eight subjects participated in this study. Each subject completed a 50 km UMT. Two-minute oral salivary swabs were taken 10 minutes prior to race start and again within 1 minute of race finish. Samples were analyzed for sAA, cortisol and IL-1β using ELISA kits. RESULTS: Our results demonstrated a significant increase in both sAA (p = 0.002) and cortisol (p < 0.001) from baseline. No significant differences were observed for IL-1β. There was no significant relationship between the observed sAA increase and race speed. The observed increase in cortisol was significantly correlated with race speed (R2 = 0.582, p …

Background: Radiofrequency ablation (RFA) utilizes minimally invasive high-energy current to precisely ablate tumor cells. It has been utilized in many cancer types including thyroid, lung, and liver cancer. It has been shown to provide adequate ablative margins with minimal complications; however, incomplete RFA may lead to recurrence of tumor. The underlying cellular mechanism and behavior of ablated cancer tissue is poorly understood.
