G protein-coupled receptors (GPCRs) play a vital role in cell signaling, and imaging the internalization of these proteins will allow for better understand of protein trafficking within the cell, as well as identification of potential drug targets. However, there is a need for further development of fluorescent protein labels that are able to effectively monitor GPCR internalization in the near-infrared (NIR) range. Acid dissociation constants, often expressed as pKa values, afford vital information with regards to molecular behavior in various environments and are of significance in fields of organic, inorganic, and medicinal chemistry. Several quantitative structure-activity relationships (QSARs) were developed that correlate experimental pKas for a given class of compounds with a descriptor(s) calculated using density functional theory at the B3LYP/6-31+G** level utilizing the CPCM solvent model. A set of carbon acids provided a good final QSAR model of experimental aqueous pKas versus ΔEH2O (R2 = 0.9647) upon removal of three aldehydes as outliers. The project covers the synthesis of two bio-based polymers. One aspect of the research investigates the synthesis of materials for use in vegetable oil-based UV-curable coating, and the subsequent polymerization of the materials into a coating film. The second aspect of the research follows the synthesis of a functionalized cellulose material for use as a reinforcer in a vegetable oil-based composite. Both products are subsequently analyzed by various methods.

The synthesis of poly(arylene ether)s, with a pendent aryl iodide, was achieved via nucleophilic aromatic substitution (NAS) polycondensation of DI-DFDPS monomer. The platform diiodo-difluorodiphenyl sulfone monomer, DFDPS-DI (3), was prepared in one-step, via EAS chemistry. Structural characterization was provided by NMR spectroscopy, GC/MS, and elemental analysis. As “pre” approach didn’t work here, so “post” approach was used to synthesize N-heterocycle functionalized copolymers. The DI-copolymers were subjected to copper-catalyzed C-N coupling reaction with carbazole, phenoxazine, and indole and the syntheses of chromophore based copolymer were successfully carried out. Oncogenic K-Ras is a driver protein found in specific cancers and was once thought to be “undruggable.” However, recent advances have suggested that mislocalizing K-Ras from the plasma membrane could be a viable strategy to target these cancers. The objective of this project was to identify a lead compound and determine a structure-activity relationship from chalcones bearing the 3,4,5-trimethoxy motif towards oncogenic K-Ras and MAO-B. This was accomplished by keeping the trimethoxy group consistent on one ring while varying the electron-donating or -withdrawing groups on the opposite ring. Separately, N-Heterocyclic carbene and Tetrathiafulvalene compounds show great promise for a variety of applications. N-heterocyclic carbenes exhibit desirable metal bonding characteristics which allow for modulation of the metal center via back-bonding interactions. Additionally, this creates a system of increased bonding strength creating stable complexes. Tetrathiafulvalene exhibits strong redox-active and optical characteristics. These are owed to an increased aromaticity as oxidation of the compound occurs and thus, is stable through a multitude of oxidative states. A New England community college reported that 60% of General Chemistry college students, who were science, technology, engineering, and mathematics (STEM) majors, did not advance in the STEM curriculum. To potentially increase student persistence in STEM curriculum, this qualitative case study project explored the self-efficacy perceptions of General Chemistry students after participating in project-based service-learning with elementary-school students. Bandura's social cognitive theory provided the conceptual framework for the study, supporting an understanding of learner self-efficacy. Research questions focused on chemistry students perceived self-efficacy after interactions with elementary-school students, teamwork, and the development of a project. The dissociative photoionization reactions of two small, oxygenated organics, namely 1,3-dioxolane and methyl vinyl ketone, were studied by photoelectron photoion coincidence (PEPICO) spectroscopy. Experiments involving 1,3-dioxolane were carried out in the photon energy range of 9.5‒13.5 eV. The statistical thermodynamics model shows that a total of six dissociation channels are involved in the formation of three fragment ions, namely C3H5O2+ (m/z 73), C2H5O+ (m/z 45) and C2H4O+ (m/z 44), with two channels contributing to the formation of each. For the past three decades, the coordination-driven self-assembly of three-dimensional structures has undergone rapid progress; however, parallel efforts to create large discrete two-dimensional architectures—as opposed to polymers—have met with limited success. The synthesis of metallo-supramolecular systems with well-defined shapes and sizes in the range of 10–100 nm remains challenging. Here we report the construction of a series of giant supramolecular hexagonal grids, with diameters on the order of 20 nm and molecular weights greater than 65 kDa, through a combination of intra- and intermolecular metal-mediated self-assembly steps. In modern organic chemistry, hypervalent (HV) iodine(III) compounds are frequently used as oxidizing agents but application of λ3-iodanes in polymer and material chemistry is still underexplored. This dissertation describes the preparation of dynamic and self-healing materials by employing ligand exchange reactions involving HV iodine(III) compounds of the type ArIL2 (Ar = Aryl, L = ligand, e.g., carboxylate or (pseudo)halide). Hydrogen sulfide (H2S) is an important biological signaling molecule that has been recognized alongside nitric oxide and carbon monoxide as being a small gasotransmitter that is enzymatically produced and impacts multiple physiological functions. To detect hydrogen sulfide, there has been a focus on developing fluorescent probes to target particular analytes; however, fluorescent probes lack sensitivity and depth penetration due to background autofluorescence and light scattering. Chemiluminescence does not require light excitation, which greatly reduces the amount of autofluorescence and photoactivation.

A series of semi-fluorinated and non-fluorinated Diels−Alder step-growth polyarylene polymers and co-polymers were synthesized via typical oil bath heating (days/weeks) and more rapid microwave-assisted polymerization (hours). The polymers were characterized by multi-nuclear (1H, 13C, and 19F) NMR and ATR−FTIR spectroscopy, thermal analysis (TGA, DSC, and DMA), GPC, XRD, water contact analysis (WCA), and refractive index (RI) measurements. The NMR spectra indicated a mixture of para and meta conformations through the polymer backbone increasing to more para with greater fluorine content. Thermal gravimetric analysis revealed the fluorine-containing polyarylenes possessed the highest char yields at almost 80% at …

Identity has been theorized to aid in student persistence toward science, engineering, technology, and math (STEM) degrees. However, before we can explore the impacts of learning environments on identity, and subsequently persistence, robust measures that can aid in understanding how identity is fostered need to be available. This research study aims to create measures of identity by expanding an existing physics identity framework and contextualizing it to science and chemistry identities. The atmosphere is a key component of the biogeochemical cycle of mercury, acting as a reservoir, transport mechanism, and facilitator of chemical reactions. The chemical and physical behavior of atmospheric mercury determines how, when, and where emitted mercury pollution impacts ecosystems. In this review, we provide current information about what is known and what remains uncertain regarding mercury in the atmosphere. 3D-printing is a relatively inexpensive additive manufacturing method used to create tangible objects. As a relatively new technology, the true capability of 3D-printing has not yet been fully exploited, especially in academic and research/teaching settings. Through this work, 3D-printing in a classroom and research environment is employed as a tool for creation of fully customizable, three-dimensional molecular models and for the visualization of theoretical abstract concepts.

Heterotrophic protists play pivotal roles in aquatic ecosystems by transferring matter and energy, including lipids, from primary producers to higher trophic predators. Using Oxyrrhis marina as a model organism, changes to the non-saponifiable protist lipids were investigated under satiation and starvation conditions. During active feeding on the alga Cryptomonas sp., the O. marina hexane soluble non-saponifiable fraction lipid profile reflected its food source with the observed presence of long chain mono-unsaturated fatty alcohols up to C25:1. Evidence of trophic upgrading in O. marina was observed with long chain mono-unsaturated fatty alcohol accumulation of up to C35:1. To the best of …

Eventual depletion and environmental toxicity of natural non-renewable energy resources are two issues pushing research toward alternative energy resources. Nature’s process of renewable, unlimited, clean energy production is photosynthesis and porphyrins are a critical component due to their ability to undergo photoinduced electron transfer. This property also makes porphyrins excellent candidates for applications such as solar cells, water splitting, and optical sensing. Researchers have previously tethered porphyrins to fullerene (C60), an excellent electron acceptor, to mimic the reaction centers in photosynthesis. This research encompasses many aspects of chemical education research including curriculum and pedagogical changes to the freshman and sophomore courses. Curriculum changes included the addition of recitations to the general chemistry and organic chemistry lectures and the creation of four new classes, CHEM 1001, 1002, 3091, and 3092. The addition of recitations was not limited to but was focused on improving DFW rates for these courses.

Toll like receptors (TLRs) are found on B cells, macrophages, monocytes, and dendritic cells, and these cells belong to the innate immune system that recognizes antigens and induces multiple cell responses through the release of cytokines. TLR1, TLR2 and TLR6 function as heterodimers, either as TLR1/TLR2 or TLR2/TLR6 to recognize lipopeptides. TLR1/2 dimer activation releases inflammatory cytokines, while TLR2/TLR6 dimer activation releases immunomodulatory cytokines. Based on the size of the binding pocket between TLR2 and TLR6, it was hypothesized that lipopeptides, such as FSL1, could be simplified while keeping overall activity. Rare earth elements, or REE, are used in modern society in televisions, computers, cellphones, military equipment, and smart weapons systems. These metals are also used by the medical industry in magnetic resonance imaging and in medical products.

The igneous rocks in the Western Kentucky Fluorspar District of the New Madrid Rift System are considered alkaline ultramafic rocks that are slightly enriched in REE. These rocks are rare and only occur in several hundred locations in the world. They have a complex history of emplacement, fractionation, metasomatism, and alteration, and are overprinted with Mississippi Valley-type mineralization. They are classified as lamprophyres …
