Caesar Creek Lake (CCL) in Warren County, OH has recently been experiencing harmful algal blooms (HABs) which are most likely attributed to an excess of phosphorus (P) from fertilizers and manures applied to surrounding fields. Sediments act as a sink for P later supplying a source of P in lakes for HABs when waters become thermally stratified and anoxic. This study seeks to determine the relationship between HABs in CCL and riparian cover at the main tributaries, Anderson Fork and Caesar Creek. This project focuses on the optimization of benzothiazole-based chromophores to utilize them as fluorescent tags functionalized onto stimuli-responsive, or “smart,” polymers as non-viral gene delivery vectors for gene therapy applications. Blue-fluorescent emissive chromophores will allow tracking capabilities for the transfection pathway of the vector to be monitored as it delivers the DNA payload within intercellular space. The most appropriate chromophore must be covalently bound to a free amine within the vector, which is accomplished through NAS chemistry from fluorophenyl-benzothiazoles (F-BTZ-CBz) derivatives to amidated hyperbranched poly(ethyleneimine)(HPEI-IBAm0.61). In this study, novel silver nanoparticles (AgNPs) were successfully synthesized and functionalized with an antibacterial agent, namely chlorothymol (denoted C@AgNPs). The resulting colloid (C@AgNPs) was purified by two comparative methods: ultrafiltration and ultracentrifugation. Ultrafiltration proved to be more efficient in purifying and size selecting (10 kD filter) and concentrating the C@AgNPs than ultracentrifugation. The physicochemical properties of the filtered C@AgNPs were then characterized by UV-Vis absorption spectroscopy, inductively coupled plasma optical emission spectroscopy (ICP-OES), Raman spectroscopy, Cytoviva hyperspectral imaging, and Scanning electron microscopy. The covalent attachment of a blue emitting, benzothiazole based TADF chromophore into poly(arylene ether) backbone polymers was explored. A benzothiazole derivative, 2-(2,4-difluorophenyl)-benzothiazole (2,4-diF-BTZ), was synthesized to form the electrophilic component of the host polymer. Two polymers were synthesized to have chromophore incorporated, using 2,4-diFBTZ and 4,4-dihydroxydiphenyl ether (DPE) or 4,4’-biphenol (BP). The chromophore was covalently incorporated at a 10% molar ratio into the polymer backbone by copolymerization with 2,4-diF-BTZ and BP, utilizing nucleophilic aromatic substitution (NAS). At local, state, and federal levels, crime laboratories analyze samples in a variety of mixtures and concentrations. Benzotriazoles are a class of contaminant of emerging concern which are commonly used as anticorrosive agents in aircraft deicer and anti-icing fluids (ADAFs). The analogues 1H-benzotriazole (BTZ), 4-methyl-1H-benzotrizole (4m-BTZ), and 5-methyl-1H-benzotriazole (5m-BTZ) are commonly found in environmental occurrence together. The two methylated isomers, collectively known as tolytriazole (TTZ), have different toxicity and stability. These contaminants are highly water soluble and resistant to biodegradation, making them persistent through water treatment. There have been limited studies on the analysis of the nucleation and precipitation behind calcium fluoride. Earlier studies support a nucleation mechanism in agreement to classical nucleation theory (CNT) in which a surface nucleation mechanism is required for calcium fluoride. This experiment devised using the ISE method to calcium fluoride to find evidence of a nucleation mechanism like that of calcium carbonate (prenucleation cluster pathway). The potential and pH were recorded versus time and the potential data was converted to nCa2+ data of free calcium ions. Amongst all known nanomaterials in use, silver is the leading metal, present in 531 different types of products, owing to their unique optical, electrical, antimicrobial, and thermal properties. Previous investigations into the presence of benzotriazole (BTZ) and corresponding analogs done in early 2019 found elevated levels near the Wilmington Air Park in Wilmington, Ohio. The analogs detected were 4-methyl-1H-benzotriazole and 5-methyl-1H-benzotriazole: known together as tolytriazole (TTZ). BTZ and TTZ are emerging environmental contaminants of concern that are often found in aircraft de-icing solutions, anti-icing solutions and detergents. The Wilmington Air Park has two facilities used to pre-treat runoff water before its subsequent release into surrounding streams. Three sites were chosen: Lytle Creek, Indian Run, and Cowan Creek. Oncogenic K-Ras is a driver protein found in specific cancers and was once thought to be “undruggable.” However, recent advances have suggested that mislocalizing K-Ras from the plasma membrane could be a viable strategy to target these cancers. The objective of this project was to identify a lead compound and determine a structure-activity relationship from chalcones bearing the 3,4,5-trimethoxy motif towards oncogenic K-Ras and MAO-B. This was accomplished by keeping the trimethoxy group consistent on one ring while varying the electron-donating or -withdrawing groups on the opposite ring. N-heterocyclic carbenes exhibit desirable metal bonding characteristics which allow for modulation of the metal center via back-bonding interactions. Additionally, this creates a system of increased bonding strength creating stable complexes. Tetrathiafulvalene exhibits strong redox-active and optical characteristics. The synthesis of poly(arylene ether)s, with a pendent aryl iodide, was achieved via nucleophilic aromatic substitution (NAS) polycondensation of DI-DFDPS monomer. The platform diiodo-difluorodiphenyl sulfone monomer, DFDPS-DI (3), was prepared in one-step, via EAS chemistry. Structural characterization was provided by NMR spectroscopy, GC/MS, and elemental analysis. As “pre” approach didn’t work here, so “post” approach was used to synthesize N-heterocycle functionalized copolymers. Acid dissociation constants, often expressed as pKa values, afford vital information with regards to molecular behavior in various environments and are of significance in fields of organic, inorganic, and medicinal chemistry. Several quantitative structure-activity relationships (QSARs) were developed that correlate experimental pKas for a given class of compounds with a descriptor(s) calculated using density functional theory at the B3LYP/6-31+G** level utilizing the CPCM solvent model. A novel organic TADF chromophore was synthesized via a 3-step process to emit a blue fluorescent light upon excitation. The chromophore was covalently attached to a 61% isobutyryl amidated poly(ethyleneimine) (PEI-IBAm0.61) non-viral gene vector utilized in gene transfection experiments. The gene vectors are loaded with a green fluorescent protein (GFP) and are monitored via fluorescence microscopy. The addition of the blue chromophore therefore allows for the monitoring of the movements of the PEI-IBAm0.61 through the full delivery mechanism of the GFP inside the cells. In order to develop a better understanding heterogeneous nucleation of barite, barite precipitation was studied under varying experimental parameters. Hydrothermal atomic force microscopy (HAFM) and optical microscopy were used to investigate the effect of change in temperature, supersaturation and varying ratios of ions on heterogeneous nucleation of barite. In the experiments conducted at higher temperatures, the particles thus nucleated were found to display characteristic hexagonal and rhomboidal shapes. In comparing results of particle densities among different ion ratios, there is evidence suggesting that barium to sulfate ratio plays a role of promoter. Illicitly manufactured fentanyls (IMFs) have become a global and local epidemic and have recently caused an increase in the number of accidental exposures and overdoses in first responders and the community. The aim of the research is to explore a method to discriminate between IMFs and cutting agents in mock street samples using Raman Spectroscopy. Predominant Raman bands corresponding to the structural moieties of each individual illicit, licit drugs, and cutting agents were tracked, then used to discriminate between varying components in a mock street sample. Agricultural runoff poses a threat to the Wilmington, Ohio municipal water supply and its source waters. A harmful algal bloom (HAB) was documented in June 2019 in Caesar Creek Lake, Waynesville, Ohio. This study seeks to establish baseline nutrient concentrations in Caesar Creek Lake (Warren County, Ohio) and its tributaries, and to identify potential non-point sources of excess nitrogen and phosphorous that contributed to the HAB event. The number of consumer products containing nanomaterials over the last eight years has increased over two-fold. Silver is one of the most commonly used materials in consumer goods nanoparticle fabrication.1 Thus, the uptake and biodistribution of silver nanoparticles (AgNPs) within mammalian cells can provide insight into their possible toxicological effects. An important research element in the Turnbull laboratory has been the one-pot synthesis of 5-methyl-3-phenyl-1,3,4-oxadiazol-2(3H)-one from sydnone. The present study reexamined the reaction of 5-methyl-3-phenyl-1,3,4-oxadiazol-2(3H)-one with electrophiles and optimized methods for synthesis of oxadiazolinone derivatives. Further the oxadiazolinone derivatives were reacted with electrophiles to better comprehend the donating ability of the oxadiazolinone ring system in general with the expectation that the oxadiazolinone ring system is a slightly donating system. The synthesis of N-heterocycle functionalized triphenylphosphine oxide based PAEs, utilizing 3,5 difluorinated diphenyl sulfone systems was achieved. 3’-Iodo 3,5-difluorodiphenylsulfone was subjected to copper and palladium catalyzed coupling reactions with carbazole, phenothiazine, and indole. The syntheses of 3’-(9-carbazole) 3,5-difluorodiphenylsulfone, 3’-(9-phenothiazine) 3,5-difluorodiphenylsulfone, and 3’-(9-indole) 3,5-difluorodiphenylsulfone were successfully carried out. Copolymers with 4,4’ difluorotriphenylphosphine oxide, as well as 4,4’ dihydroxydiphenyl ether (DPE), as the nucleophilic partner, were prepared via standard nucleophilic aromatic substitution (NAS) polycondensataion reactions. One goal of the Turnbull research group has been the synthesis of dihalogenated arylsydnones; i.e. sydnones with halogen substitution on the sydnone ring itself, as well as at the position on the aryl ring ortho to the sydnone. Further, it has been of interest to explore the reactivity of these compounds, once obtained. The present study examined the synthesis of 4-iodo-3-(2-iodoaryl)-, or “diiodo”, and 4-bromo-3-(2-bromoaryl)-, or “dibromo”, sydnones and the reactions of these compounds with Na2SO3 (sodium sulfite) and NaBH4 (sodium borohydride) to create a better understanding of the reactivity of diiodo and dibromo arylsydnones in general. Poly(aryl ether ketone)s (PAEK) are high performance thermoplastics, which are chemically robust, semi-crystalline, and stable at high temperatures. Of the family of PAEKs, poly(ether ether ketone) (PEEK) is a well-known semi-crystalline thermoplastic widely used for electronics, energy, industrial, and medical applications due to its resistance to solvents, radiation, heat, and other environmental factors. The traditional PEEK is prepared from 4,4’-difluorobenzophenone (2) and disodium hydroquinone. However, a challenge is processability, since due to its highly crystalline nature, PEEK possesses very limited solubility.
