The Role Of Sn-1,2-Diacylglycerol In Insulin-Stimulated Glucose Transport In Rat Skeletal Muscle, Kevin Paul Boggs

Insulin stimulates glucose transport into skeletal muscle upon binding to its cell surface receptor. The signals which stimulate transport are unknown. Studies of insulin's effects on muscle cell lines and adipocytes suggest that increases in sn-l,2-diacyglycerol (DAG) content may mediate insulin-stimulated glucose transport. To test this, the effect of insulin on muscle DAG content was measured. DAG increased in diaphragm and soleus muscles exposed to insulin in vitro and in diaphragm in vivo. However, the supraphysiologic insulin dose (60 nM) and incubation time required (10 min) are not consistent with a role for DAG as a mediator of insulin stimulated …

The power of molecular biology is unleashed with the ability to clone and sequence genes, and then express these genes in heterologous systems. This sets the stage for the full analysis of proteins that are otherwise difficult to isolate and/or purify, especially when present at very low copy number per cell or when isolated from relatively precious materials. Overexpression of protein is now possible in a number of systems including prokaryotes (e.g., E. coli) and various eukaryotes (yeast, insects, and plants). Pain is a subjective phenomenon and is, in varying degrees, an inherent part of the childbirth experience. However, most Western societies view suffering as unacceptable and as such, regard analgesia as a necessity. Many women do not anticipate the intensity of pain experienced in childbirth and are, therefore, not ready to manage this amount of pain when it occurs. This prospective study compared the expected and experienced labour pain of 99 primiparous women, aged 17-40 years. The purpose of this qualitative study is to discover and examine the coping strategies concerning death and dying from the perspectives of hospice patients and hospice volunteers. A review of applicable literature, informal but structured interviews and participant observation are procedures for the study. The focus of the study lies in three areas: what are the internal attributes used to cope?; what are the external support systems used to cope? and, what are the adjustments in personal goals? The effect of ethyl alcohol (10%) on the postnatal development of cerebellar cortex was studied using histological and morphometric methods. The external granular layer was much increased in thickness in alcohol fed rat than those of the control while the molecular and granular layer were reduced. The cells of themolecular layer showed neuronal degeneration and many spaces were present in this layer esp < /strong>ecially late postnatal. The Purkinje cells failed to reach their final position up the tenth day and still arranged in more than one row. The degenerative changes appeared at five days and increased with age. The extent …

Provides clinical details in the form of 11 case vignettes from a prospective epidemiological study that found the combination of childhood 'soft' signs and anxious behavior to be a strong risk factor for adolescent emotional disorders. The original study conducted a neuropsychiatric assessment of 180 African-American adolescents (aged 16–18 yrs) who had been followed through childhood. The at-risk Ss exhibited a persistent and specific pattern of both motor abnormalities and anxiety, obsessional compulsive, or depressive symptoms over time. The purpose of this study was to determine the relationship between locus of control, altruism, occupational risk, and personal factors to the frequency of universal precautions practices of prehospital emergency medical care providers. The effects of OSHA's Occupational Exposure to Bloodborne Pathogens; Final Rule on reducing prehospital emergency medical care providers occupational exposure to bloodborne pathogens were also examined. Prehospital emergency medical care providers from municipal and private EMS agencies in the Tidewater Emergency Medical Services region participated in this study. Patients with undiagnosed haemostatic defects seen at The Aga Khan Hospital and Fatimid Blood Transfusion Centre during the period of 7 years (1 985-1 992) were screened with routine tests including bleeding time (BT), whole blood clotting time (CT), platelet count, activated partial thromboplastin time (APTT), prothrombin time (PT) and 5 molar urea test. Nine patients had a positive 5 molar urea test indicating factor XIII deficiency. Rest of the screening tests were normal in these patients. Intake and coefficient of absorption of nutrients wefe measured in 72 children dufing acute diarrhoea and 2 weeks after fecovery. No diarrhooeal pathogens could be identified in 18 (25%) childfen (group I). Aetiology of diafrhoea was identified in fest of the 54 children (group II). Absofption of oalofie, fat and cafbohydrate during the recovery stage were similaf in all 72 children, In group I, absorption of nutrients improved from acute stage to recovery stage as follows: fat from 62 to 85%, calofies from 68 to 86% and carbohydrate from 81 to 91%. A cDNA has been isolated from human hippocampus that appears to encode a novel Na(+)-dependent, Cl(-)-independent, neutral amino acid transporter. The putative protein, designated SATT, is 529 amino acids long and exhibits significant amino acid sequence identity (39-44%) with mammalian L-glutamate transporters. Expression of SATT cDNA in HeLa cells induced stereospecific uptake of L-serine, L-alanine, and L-threonine that was not inhibited by excess (3 mM) 2-(methylamino)-isobutyric acid, a specific substrate for the System A amino acid transporter. SATT expression in HeLa cells did not induce the transport of radiolabeled L-cysteine, L-glutamate, or related dicarboxylates. Northern blot hybridization revealed high levels …

To test the hypothesis that epidermal rather than dermal mosaicism determines Blaschko's lines in hypomelanosis of Ito (HI), we studied the distribution of chromosomal mosaicism in four patients. In two, mosaicism had not been detected in lymphocytes or dermal fibroblasts, but was clearly shown in epidermal keratinocytes; furthermore, the abnormal cell line was confirmed to the hypopigmented epidermis and the normal epidermis contained only normal cells. Negative findings in the other two patients might be because of mosaicism which was undetected either because it was submicroscopic or because it was present in melanocytes, which have not yet been studied. The effects of ice and compression wrap applications on intramuscular temperature at the skin surface, 1 cm below the fat layer, and 2 cm below the fat layer were recorded in 11 subjects. Each subject was tested under all treatment conditions including control, compression only, ice only, and ice+ compression. Subjects underwent a 5-minute pre-application, a 3 ominute application, and a 20-minute post-application. Using thermocouples, surface and intramuscular temperatures were recorded at 30-second intervals for the entire study. A repeated measures ANOVA and Duncan Post Hoc tests were used to evaluate temperature differences between the treatment conditions and the depths …

Trimethyltin (TMT) is a neurotoxin occurring in the environment. Exposure to (TMT) is known to destroy specific neuronal components of the hippocampus in the rat and to cause clinical symptoms in exposed humans, including mnemonic deficits, that indicate hippocampal involvement. In addition to hippocampal cell loss TMT causes significant increases in cholinergic markers such as acetylcholinesterase (AChE) stain density and choline acetyltransferase (ChAT) activity in the hippocampus of rats. However, despite these observations the effect of TMT on hippocampal cholinergic system has not been investigated in detail.
