Use Of A Pelleted Corn Residue Complete Feed In Receiving Diets, Sarah J. Peterson, Brandon L. Nuttelman, Dirk B. Burken Burken, James C. Macdonald, Matt K. Luebbe, Galen E. Erickson
Nebraska Beef Cattle Reports
The effects of feeding a complete pelleted feed to newly received steer calves (585 ± 4 lb; n = 1318 ) was compared to a control ration consisting of 32% (DM basis) wet or modified distillers grains, 32% alfalfa hay, 32% dry-rolled corn, and 4% supplement. The pelleted complete feed consisted of 35% corn residue and a blend of grain byproducts and minerals. Feeding the complete pelleted feed increased DMI but decreased ADG, thereby reducing feed efficiency. The pelleted feed numerically reduced morbidity. Feeding a complete pellet consisting of corn residue appears to be a viable option for receiving calves …

Comparison Of Long-Term Progestin-Based Synchronization Protocols On Fixed-Time Ai Pregnancy Rate In Beef Heifers, Rebecca A. Vraspir, Adam F. Summers, Doug O'Hare Hare, Larry D. Rowden, Rick N. Funston
Nebraska Beef Cattle Reports
Yearling Angus heifers at a commercial ranch in the Nebraska Sandhills were randomly assigned to one of two progestin-based fixed-time AI protocols (MGA or 14-day CIDR) to compare pregnancy rates. Heifers had similar fixed-time AI pregnancy rates between MGA and 14-day CIDR. A similar proportion of MGA and 14-day CIDR heifers displayed a second estrus; however, heifers previously synchronized with MGA tended to have a greater second AI pregnancy rate. Overall pregnancy rate was similar between MGA and 14-day CIDR treatments. The MGA system was the more cost effective synchronization protocol in this study.

Nebraska Beef Cattle Reports
Cattle fed a de-oiled wet distiller’s grains plus solubles (WDGS) diet were compared to cattle fed corn or traditional (full-fat) WDGS diets to determine effects on discoloration, oxidation, and tenderness of beef aged for seven and 21 days. At seven days of aging, dietary treatment had no effect on discoloration. At 21 days of aging, beef from cattle fed de-oiled WDGS had less oxidation than the corn control and several of the full-fat WDGS treatments. Although tenderness increased with aging and retail display, dietary treatment had no effect on tenderness. These findings suggest that these dietary treatments, followed by a …

Shelf Life Of Cooked Ground Beef Patties From Cattle Fed Wet Distillers Grains With Solubles, Nathan T. Dierks, Tommi F. Jones, Kimberly A. Varnold Varnold, Derek J. Schroeder, Amy L. Redfield, Gary A. Sullivan
Nebraska Beef Cattle Reports
Cattle were grazed without or with energy supplementation of wet distillers grains with solubles (WDGS) during backgrounding, and were finished on a corn-based diet with or without 35% WDGS. Ground beef patties were made from shoulder clods, cooked, and stored in a refrigerated or frozen state. Cattle supplemented with WDGS had greater lipid oxidation in cooked ground beef patties regardless of finishing diet or storage type.

Differences In Fecal Bacterial Community Composition Between Beef Steers Which Are High-Shedders And Low-Shedders Of Shiga Toxin-Producing Escherichia Coli (Stec), Nirosh D. Aluthge, Yoshitha A. Wanniarachchi, Brandon L. Nuttelman Nuttelman, Cody J. Schneider, Terry J. Klopfenstein, Galen E. Erickson, Samodha C. Fernando
Nebraska Beef Cattle Reports
The community composition of the fecal microbiota was compared between beef steers which were high-shedders and low-shedders of Shiga toxin-producing Escherichia coli. Based on Shannon and Chao 1 diversity indices, the high-shedders had a more diverse fecal bacterial community than the low-shedding steers. Members of the genus Prevotella were observed as being more abundant in the low-shedders compared to the high-shedders, while Succinivibrio were more abundant in the high-shedders. Isolation of specific bacteria which are significantly more abundant in low-shedders may pave the way to developing direct-fed microbials which are effective in reducing STEC shedding among high-shedding beef steers.

Optimal Marketing Date Of Steers Depends On Marketing Strategy, James C. Macdonald, Cody J. Schneider, Kelsey M. Rolfe Rolfe, Stephen D. Kachman, Terry J. Klopfenstein, Galen E. Erickson
Nebraska Beef Cattle Reports
Seven research trials conducted over five years at the University of Nebraska–Lincoln were summarized to determine how animal performance changes through the finishing period on a live and carcass weight basis. Live weight, carcass weight, carcass ADG, and carcass feed efficiency all changed quadratically throughout the feeding period; live ADG and live feed efficiency declined linearly. During times of negative profit margins, optimal profitability for steers marketed on a live-basis occurred by selling early, whereas optimal profitability was achieved by feeding steers marketed on a carcass-basis longer.

Nebraska Beef Cattle Reports
Crossbred steers (n = 64) were grazed on warm- or cool-season grass-dominated pastures, without or with energy supplementation of wet distillers grains with solubles (WDGS), and were finished on a corn-based diet with or without 35% WDGS. Finishing on corn increased desirable flavor notes and decreasedundesirable flavor notes in both L. dorsi and B. femoris steaks. In addition, grazing on warm-season grasses increased the prevalence of undesirable flavors but was often dissipated by the addition WDGS supplementation. Longer aging periods tended to increase the prevalence of undesirable flavors, especially in B. femoris steaks. It is recommended producers provide WDGS supplementation, …

The Role Of Monocyte Chemoattractant Protein-1 In Innate Immunity Against Methicillin Resistant Staphylococcus Aureus Pneumonia, Mary Katherine Leissinger
LSU Master's Theses
Monocyte chemoattractant protein-1 is critical for monocyte recruitment to the lungs in response to bacterial infection. MCP-1 is also essential for protective neutrophil recruitment to the lungs during Escherichia coli and Klebsiella pneumoniae infection. Staphylococcus aureus pneumonia, specifically strain USA300, carries a high morbidity and mortality rate and is an important pathogen in hospital/ventilator and community acquired pneumonia. In the current study, we investigated the role of MCP-1 in pulmonary innate immunity to S. aureus in C57Bl/6, MCP-1-/- and MCP-1 AB blocked mice. As compared to C57Bl/6, MCP-1-/- mice showed increased concentrations of neutrophils in the airways and lung parenchyma …

Autologous Adipose Derived Adult Multipotent Stromal Cells Alter The Porcine Systemic Immune And Bone Biomarkers Response To Cancellous Bone Xenografts, Jonathan Francis Bova
LSU Master's Theses
The use of a porcine model in assessing bone grafts in vivo is common when a large animal model is necessary. In this thesis we aimed to improve the porcine model of facial reconstruction through the use of a local anesthesia and novel methods of assessing the immune response to and bone forming ability of adult adipose derived mesenchymal stromal cells (ASC). The goals of the research were: 1) evaluate the effect of a bupivacaine mandibular nerve block on blood pressure (BP) and heart rate (HR) in response to surgical stimulation and the need for systemic analgesics postoperatively, 2) quantify …
Influence Of Aedes Aegypti Saliva On The Vertebrate Host Response To Dengue Virus, Michael Kevin Mccracken
Influence Of Aedes Aegypti Saliva On The Vertebrate Host Response To Dengue Virus, Michael Kevin Mccracken
LSU Doctoral Dissertations
Dengue virus (DENV) is maintained in a primarily anthroponotic cycle between humans and the mosquito, Aedes aegypti. Investigations into DENV infection of the vertebrate host generally do not account for the contribution of vector saliva, an inherent part of the mosquito-borne viral inoculum. Feeding by mosquitoes on vertebrate hosts is initiated by probing, which results in physical damage to the skin and vasculature, and the simultaneous introduction of DENV and saliva into the skin. Saliva contains many individual proteins with the potential to modulate host hemostasis and immune responses, thereby facilitating blood feeding and virus transmission. As exogenous antigens, both …
Outer Surface Lipoprotein Layer Homeostasis And Gene Regulation In Borrelia Burgdorferi, Poonam Dadhwal
Outer Surface Lipoprotein Layer Homeostasis And Gene Regulation In Borrelia Burgdorferi, Poonam Dadhwal
LSU Doctoral Dissertations
The outer surface lipoprotein (osp) layer forms an interface between the internal and the external environment of the Lyme disease spirochete, Borrelia burgdorferi. The homeostatic maintenance of the osp layer effectuates adaptation of B. burgdorferi as it gets transmitted from the tick vector to a mammalian host and vice-versa. However, the regulation of the outer surface lipoproteins (osps) is still a conundrum for borrelia scientists. Part of this dissertation inquires about the homeostatic maintenance of the osp layer. We found that the deletion of the dominantly expressed tick phase osp, OspA, induces expression of two other osps. OspD, and BBJ41. …
Mechanisms Of Innate Immunity In Polymicrobial Sepsis, Liliang Jin
Mechanisms Of Innate Immunity In Polymicrobial Sepsis, Liliang Jin
LSU Doctoral Dissertations
Severe bacterial sepsis leads to a pro-inflammatory condition that can manifest as septic shock, multiple organ failure, and death. Neutrophils are critical for the rapid elimination of bacteria, however, the role of neutrophil chemoattractant CXCL1, pattern recognition receptors (PRRs)- NLR protein 3 (NLRP3) and alcohol in bacterial clearance during sepsis remains elusive. We demonstrate that CXCL1 plays a pivotal role in mediating host defense to polymicrobial sepsis following cecal ligation and puncture (CLP) in gene-deficient mice. CXCL1 appears to be essential for restricting bacterial outgrowth and preventing multiple organ failure and death in mice. Moreover, CXCL1 is essential for neutrophil …

Genetics And Functions Of Herpes Simplex Virus Type-1 Membrane Proteins Ul20/Gk In Virion Envelopment And Entry, Anu Susan Charles
LSU Doctoral Dissertations
The envelopment and egress of the Herpes Simplex Virus is an important event in the life cycle of the virus. The important membrane proteins required for the envelopment and egress of the virus are glycoproteins gM, gK, gE and non-glycosylated membrane proteins UL20p and UL11. Among them one of the most critical protein is the UL20 protein which has four transmembrane domains with amino and carboxyl termini are predicted to lie within the cytoplasmic side of cellular membranes. Studies done in our laboratory have shown that deletion of UL20 or other lethal mutations have an adverse effect on envelopment and …

North American Lyme Borreliae: New Distributions And An Insight Into Borrelia Bissettii Infection, Immune Response And Transmission In A Murine Model, Brian Francis Leydet Jr
LSU Doctoral Dissertations
Lyme borreliosis is an emergent threat to human health. It is estimated that 300,000 cases of Lyme disease are diagnosed annually in the United States. In recent years, non-traditional areas are starting to report more and more cases. While much research has gone into the tick and bacteria responsible for the disease, frequently new Borrelia species are being described and implicated in human illness. These species often go unstudied, and their threat to human health is not known. In the hopes of closing knowledge gaps on Lyme borreliosis in the US, we surveyed a non-endemic state, Louisiana, for Lyme Borrelia …

The Role Of Viral Glycoproteins And Tegument Proteins In Herpes Simplex Virus Type 1 Cytoplasmic Virion Envelopment, Dmitry Vladimirovich Chouljenko
LSU Doctoral Dissertations
Herpes simplex virus type 1 (HSV-1) is a ubiquitous neurotropic alphaherpesvirus transmitted by contact with mucocutaneous surfaces of infected individuals. HSV-1 enters the host by fusion of the viral envelope with the host cell plasma membrane, followed by translocation of the viral capsids to the nucleus where viral DNA is injected into the host cell nucleus to initiate viral replication. To generate infectious virions, newly assembled capsids travel to the cytoplasm and undergo a process called secondary envelopment by budding into cytoplasmic vesicles derived from the trans-Golgi network. Cytoplasmic envelopment is a complex process involving interactions between a multitude of …

Turkish Journal of Veterinary & Animal Sciences
To determine sequence characteristics and phylogenetic relationships of the nonstructural (NS) gene, the NS genes of 9 avian influenza (AI) viruses were sequenced and compared with other Iranian AI H9N2 viruses available in GenBank. The similarity among the earliest Iranian H9N2 isolate (A/chicken/Iran/ZMT-101/1998) and recent (2007–2011) studied isolates was low (94%). Some of the studied viruses revealed substitutions in RNA-binding and effector domains as well as antigenic regions of the NS protein. Recently studied isolates have a high similarity (97.4%-98.7%) with some Pakistani isolates (like A/chicken/Sawabi/NARC-2434/2006) in the NS gene. In the phylogenetic analysis, recently studied isolates composed a cluster …

Chitosan (CH) has been shown to be a promising natural antimicrobial agent. This study examined the effects on rumen metabolism when providing diets containing high forage (HFD), low forage (LFD), and maize silage (MSD) to a 24-h batch culture in which the diets were not supplemented (control) or were supplemented with CH (100 mg/L), sunflower oil (SF, 35.0 g/kg dry matter), rapeseed oil (RP, 35.0 g/kg dry matter), or SF and RF with chitosan combinations (SFCH and RPCH). Dry matter digestibility in the batch culture was influenced by CH and was decreased in HFD compared with the control (P < 0.01). Decreasing total number of protozoa and the number of Entodinium spp. was evident, especially in LFD and MSD, with all additives. RPCH had a predominantly adverse effect on Dasytricha ruminantium in MSD. Interactions of the diets × additives in the majority of rumen ciliate populations were detected. The concentrations of trans-vaccenic acid and conjugated linoleic acid (c9, t11-CLA) were unchanged by chitosan. However, the combination of SF and RF with chitosan (SFCH and RPCH) positively affected fatty acid profiles, which might be related to differences in diets, SF or RP composition, and different microbial activity in the batch cultures.

One hundred and two samples of mussels (Mytilus galloprovincialis), harvested from approved shellfish coastal water in northern Greece, were screened for the presence and antimicrobial resistance of Listeria monocytogenes. Listeria spp. were isolated according to International Organization for Standardization method 11290-1: 1996/FDAM 1: 2004(E) and identified using a multiplex polymerase chain reaction (PCR) system. The serovar identity of L. monocytogenes isolates was also determined with a multiplex PCR assay. The antimicrobial profile of the isolates was determined by the disk diffusion method. Listeria spp. were present in 8 of 102 samples tested (8%) and only 1 (1%) yielded L. monocytogenes. …

Ractopamine (RAC) causes fat deposition and/or fatty acid synthesis reduction and increases the rate of protein synthesis and muscle growth. However, there are few scientific studies detailing the mechanism of action of RAC and its possible metabolic pathways in swine. The objective of this study was to evaluate the effect of RAC on lipid metabolism of finishing pigs. Subcutaneous and retroperitoneal fat, muscle, and blood samples were collected at slaughter. Forty pigs were fed different RAC levels (ppm): 0, 5, 10, 15, and 20. RAC did not affect lipoprotein lipase activity in any of the tissues. There were no changes …
