Glutaredoxin 2 (Grx2) Knockout Increases Sensitivity To Oxidative Stress In Mouse Lens Epithelial Cells, Hongli Wu, Liren Lin, Frank Giblin, Ye-Sheh Ho, Marjorie F. Lou

School of Veterinary and Biomedical Sciences: Faculty Publications

Glutaredoxin belongs to the oxidoreductase family with cytosolic glutaredoxin 1 (Grx1) and mitochondrial gluraredoxin 2 (Grx2) isoforms. Of the two isozymes, the function of Grx2 is not well understood. This paper studied the effect of Grx2 deletion on cellular function using primary lens epithelial cell cultures isolated from Grx2 gene knockout (KO) and wild type (WT) mice. We found that both cell types showed similar growth patterns and morphology, and comparable mitochondrial glutathione pool and complex I activity. Cells with deleted Grx2 did not show affected Grx1 or thioredoxin (Trx) expression but exhibited high sensitivity to oxidative stress. Under treatment …

Controlling Viral Immuno-Inflammatory Lesions By Modulating Aryl Hydrocarbon Receptor Signaling, Tamara Veiga-Parga, A Suryawanshi, B T. Rouse Dec 2012

Controlling Viral Immuno-Inflammatory Lesions By Modulating Aryl Hydrocarbon Receptor Signaling, Tamara Veiga-Parga, A Suryawanshi, B T. Rouse

Tamara Veiga-Parga

Ocular herpes simplex virus infection can cause a blinding CD4+ T cell orchestrated immuno-inflammatory lesion in the cornea called Stromal Keratitis (SK). A key to controlling the severity of SK lesions is to suppress the activity of T cells that orchestrate lesions and enhance the representation of regulatory cells that inhibit effector cell function. In this report we show that a single administration of TCDD (2, 3, 7, 8- Tetrachlorodibenzo-p-dioxin), a non-physiological ligand for the AhR receptor, was an effective means of reducing the severity of SK lesions. It acted by causing apoptosis of Foxp3- CD4+ T cells but had …

The Thai Journal of Veterinary Medicine

Two semen samples of Siamese Eld,s deer (Cervus eldii siamensis: SED) and seven semen samples of Thamin Eld,s deer (Cervus eldii thamin: TED) were collected by using electroejaculation. Each ejaculate was diluted and cryopreserved with 2 extenders; 1) TRIS; and 2) modified BF5F supplemented with 5% final glycerol concentration. The movement patterns of frozen-thawed spermatozoa were assessed using computer-assisted sperm analysis (CASA). In TED, the results revealed a significantly higher (p< 0.05) percentage of total motility (%TM), percentage of progressive motility (%PM), track speed (VCL) and lateral amplitude (ALH) in TRIS extender. The results also revealed a significantly low (p< 0.05) progressive velocity (VSL), straightness (STR) and linearity (LIN) in TRIS extender. However, for path velocity (VAP) and beat frequency (BCF), there were no differences between both extenders. In SED, sperm motility and sperm movement parameters did not show differences between TRIS and modified BF5F extenders. Findings from this study will aid in Eld’s deer semen banking and artificial insemination success in the future.

However, in many species, oxidative damage due to generation of reactive oxygen species (ROS) and long term storage which specifically cause DNA damage, and lipid peroxidation during preservation techniques can cause a decline of sperm motility and fertility capacity. In this study, the effect of antioxidants catalase (CAT) and superoxide dismutase (SOD) on motility, viability and acrosomal integrity of chilled canine semen were investigated. Semen was collected by digital manipulation from 4 dogs (two ejaculates/dog). After removal of seminal plasma, spermatozoa were diluted in egg yolk …

Structure-Function Analysis Of A Protein Encoded By The Bhv-1 Latency Related Gene, Devis Sinani Dec 2012

Structure-Function Analysis Of A Protein Encoded By The Bhv-1 Latency Related Gene, Devis Sinani

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Bovine herpes virus 1 (BHV-1) is a significant viral pathogen in cattle that induces a myriad of clinical symptoms. These symptoms include: conjunctivitis, upper respiratory tract infections, genital disorders, and abortions. BHV-1 infection can also lead to transient immune-suppression, which predisposes cattle to secondary bacterial infection leading to life-threatening pneumonia referred to as bovine respiratory disease (BRD). Following acute infection, BHV-1 establishes latency in sensory neurons within trigeminal ganglia. Reactivation of the virus can occur periodically, resulting in virus transmission. The latency-related (LR) RNA is the only abundantly expressed transcript in latently infected sensory neurons and it encodes several proteins, …

Socioeconomic Determinants Of Geographic Disparities In Campylobacteriosis Risk: A Comparison Of Global And Local Modeling Approaches, Jennifer Weisent, Barton Rohrbach, John Dunn, Agricola Odoi Oct 2012

Socioeconomic Determinants Of Geographic Disparities In Campylobacteriosis Risk: A Comparison Of Global And Local Modeling Approaches, Jennifer Weisent, Barton Rohrbach, John Dunn, Agricola Odoi

Agricola Odoi

Background: Socioeconomic factors play a complex role in determining the risk of campylobacteriosis. Understanding the spatial interplay between these factors and disease risk can guide disease control programs. Historically, Poisson and negative binomial models have been used to investigate determinants of geographic disparities in risk. Spatial regression models, which allow modeling of spatial effects, have been used to improve these modeling efforts. Geographically weighted regression (GWR) takes this a step further by estimating local regression coefficients, thereby allowing estimations of associations that vary in space. These recent approaches increase our understanding of how geography influences the associations between determinants and …

Background

We previously established a congenic mouse strain with TALLYHO/Jng (TH) donor segment on chromosome 6 in a C57BL/6 (B6) background that harbors an obesity quantitative trait locus, tabw2. The B6.TH-tabw2 congenic mice developed increased adiposity that became exacerbated upon feeding a high fat-high sucrose (HFS) diet. To fine map the tabw2, in this study we generated and characterized subcongenic lines with smaller TH donor segments.

Results

We fixed four subcongenic lines, with maximum size of donor segment retained in the lines ranging from 10.8 – 92.5 Mb. For mapping, all the subcongenic mice, along with B6.TH- …
