Hypothalamic proopiomelanocortin (POMC) neurons are critical for controlling homeostatic functions in the mammal. We used a transgenic mouse model in which the POMC neurons were labeled with enhanced green fluorescent protein to perform visualized, whole-cell patch recordings from prepubertal female hypothalamic slices. The mouse POMC-enhanced green fluorescent protein neurons expressed the same endogenous conductances (a transient outward K current and a hyperpolarization-activated, cation current) that have been described for guinea pig POMC neurons. In addition, the selective -opioid receptor agonist DAMGO induced an outward current (maximum of 12.8 1.2 pA), which reversed at K equilibrium potential (EK), in the majority …

The Anorexigenic Fatty Acid Synthase Inhibitor, C75, Is A Nonspecific Neuronal Activator, Kanji A. Takahashi, James L. Smart, Hongyan Liu, Roger D. Cone

C75, a recently derived compound that potently suppresses feeding and induces weight loss, has been proposed to act mainly by inhibiting fatty acid synthase (FAS) in central neurons that control feeding. For example, normal, fasting- associated, hypothalamic increases in neuropeptide Y (NPY)/Agouti-related protein (AGRP) expression and decreases in proopiomelanocortin (POMC)/cocaine and amphetamine regulated transcript (CART) expression were reported to be blocked by C75. Using loose-patch extracellular recording in acute slices, we tested the effect of C75 on anorexigenic POMC neurons and orexigenic NPY neurons of the hypothalamic arcuate nucleus, which were identified by promoter-driven GFP expression, as well as on …

Two receptors (CRH receptor type 1 and CRH receptor type 2) have been identified for the stress-induced neuropeptide, CRH and related peptides, urocortin, and urocortin II. We previously found marked down-regulation of cardiac CRH receptor type 2 expression following administration of bacterial endotoxin, lipopolysaccharide, a model of systemic immune activation, and inflammation. We postulated that inflammatory cytokines may regulate CRH receptor type 2. We show that systemic IL-1α administration significantly down-regulates CRH receptor type 2 mRNA in mouse heart. In addition, TNFα treatment also reduces CRH receptor type 2 mRNA expression, although the effect was not as marked as with …

Several classes of environmental contaminants have been demonstrated to disrupt reproductive function in wild populations and laboratory animals through interactions with endocrine pathways. Primary targets for these endocrine active contaminants are steroid hormone receptors and steroidogenic enzymes. The first study tests the hypothesis that aromatase represents a major target for environmental contaminants which disrupt reproductive function. The results indicate that the biosynthesis of estrogens can be inhibited by certain pesticides and that aromatase represents a target for contaminants disrupting reproductive function. The second study develops an estrogen sensitive and specific reporter gene assay responsive to steroid hormones and several estrogenic …

This study examines the present state of aging and longevity research as it applies to longevous elderly, those who reach their 10th and 11th decade of life. Their numbers, if not percentages, have risen considerably in the growing elderly population, The difference between the longevous and other elderly persons is not just one of degree, but of kind. Each of them is a living biological prototype of the human potential to live an extended life span without significant loss of vitality.

The existence of extrathyroidal monoiodotyrosine (MIT) has been debated because of uncertain results from measuring serum MIT. The reason is that the isotopic and chromatographic assays used have not been sensitive enough. This investigation undertook to establish the existence of MIT in serum utilizing a radioimmunoassay (RIA), to study the physiology of serum MIT, and to discover the pharmacologic effects of exogenous MIT.

A total of 26 pineal glands from the white-tailed deer were histologically examined. Five different stains were utilized to determine various cell types present in pineal tissue. Connective tissue septa divide the pineal gland into incompletely separated lobules which are supplied by a very extensive vascular system. The pinealocyte is the most numerous cell in the gland and is most easily recognized by a prominent nucleus approximately 6-8 microns in diameter. The pinealocyte cytoplasm contains many minute granules. A distinct difference was noted in the histology of the oldest age deer pineal (age 7 years) when globular granules were observed …

The adrenal physiology of the albino rat was studied through the measurement of adrenal ascorbic acid levels. A cyclic phenomenon was observed which follows the diel periodicity of the animal. Ascorbic acid values were found to be characteristically higher in males than in females. A similar difference was also seen in populations divided into young animals (body weight less than 175 grams) and old animals (body weight greater than 175 grams). The diurnal rhythm of adrenal ascorbic acid (DAR) displays an inverse relationship to the diurnal rhythm of circulating levels of plasma corticosteroids. Alloxan monohydrate, 175 mg/kg body weight was given subcutaneously to albino rats to determine the effect of the diabetic state on the DAR. A total of 200 female albino rats were divided into two study groups. Study Group I was composed of 150 animals divided into populations of 15, each including 3 controls. Western Australia has a million acres of medics including various cultivars of barrel medic, Medicago truncatula, and strand medic, M. littoralis. Both species contain coumestrol, a chemical thought to have caused delayed conception and reduced twinning rates among grazing ewes in New Zealand.
No infertility has been reported among ewes grazing medic pastures in Western Australia, but coumestrol levels in both species have often been high enough to suggest the possibility of oestrogenic responses among sheep grazing them.

A recent field experiment on established pastures at Esperance Downs Research Station has demonstrated a relationship between superphosphate application, pasture oestrogens and oestrogenic response in sheep.
Results of the experiment indicate that on phosphate deficient soils carrying oestrogenic clovers, the levels of oestrogenic materials in the pasture are increased if superphosphate applications are below those recommended for healthy pasture growth.

DURING the past five years there has been a marked increase in the incidence of breeding abnormalities of sheep associated with the grazing of subterranean clover pastures.
This complex of diseases first became a major problem in Western Australia in the years following 1940 and eventually became known as "clover disease".
