Oxytocin is a neurohypophyseal hormone that plays a crucial role in regulating mother-infant bonding and maternal behavior. Oxytocin modulates sex-specific parental behaviors by binding to the oxytocin receptor (OXTR)-expressing neurons in the brain. This study investigates the sexually dimorphic OXTR neurons in the anteroventral periventricular nucleus (AVPV) to understand how oxytocin modulates behavior in females. Specifically, we explored the behavioral roles, neural projections, and intrinsic electrophysiological characteristics of these neurons in the female mice. Using immunohistochemistry revealed that the number of OXTR-expressing neurons was enhanced in postpartum females compared with virgin females, suggesting that these neurons are involved in female-specific …

Background: The average menstrual cycle of a healthy adult woman is a duration of approximately 28 days, during which in the follicular phase (first 14 days) the progesterone hormone is at a stable level while estrogen increases to its peak level on day 14. Day 14 is known as ovulation, during which progesterone increases during its Luteinizing Hormone (LH) surge while estrogen decreases. Hormonal contraceptives contain a synthetic estrogen (e.g., ethinyl estradiol/EE) and/or a synthetic progestin (e.g., levonorgestrel/LEVO) and can exacerbate nicotine intake. However, whether altered intake reflects changes in nicotine reinforcement or enhancement by nicotine of co-occurring reinforcers is unknown. This reinforcer-enhancement effect is evidenced in rats when nicotine self-administration is increased by the presence of a visual stimulus (VS) reinforcer. This dissertation examined the effects of EE and LEVO on nicotine reinforcement and reinforcer-enhancement in ovary-intact female Sprague-Dawley rats. The neural mechanisms that control the onset of puberty are not completely understood. In livestock, the onset of puberty relies on an increase in gonadotropin releasing hormone (GnRH) and luteinizing hormone (LH). Kisspeptin, neurokinin B, and dynorphin neurons (KNDy) and their receptors, Kiss1r, NK3R, and KOR, respectively, are important regulators of puberty. Previous data from our laboratory has shown that KNDy neurons may be responding to other inputs, including the melanocortin system, that will affect the timing of puberty. Amidst the amphibian extinction crisis, in situ and ex situ amphibian species conservation initiatives utilize assisted reproductive technologies for optimal genetic management of captive and wild populations. Development of effective, simple, low-cost methods for obtaining gametes for artificial fertilization, sperm biobanking, or natural breeding alleviates obstacles for institutions or programs to apply reproductive technologies. Objectives herein include investigating the efficacy of alternative hormones for inducing spermiation, an alternative hormone administration route for inducing ovulation, exploring the physiological effects of hormone therapy, and utilizing sperm from deceased animals for biobanking.

Herein we review our work involving dispersed adrenocortical cells from several lizard species: the Eastern Fence Lizard (Sceloporus undulatus), Yarrow's Spiny Lizard (Sceloporus jarrovii), Striped Plateau Lizard (Sceloporus virgatus) and the Yucatán Banded Gecko (Coleonyx elegans). Early work demonstrated changes in steroidogenic function of adrenocortical cells derived from adult S. undulatus associated with seasonal interactions with sex. However, new information suggests that both sexes operate within the same steroidogenic budget over season. The observed sex effect was further explored in orchiectomized and ovariectomized lizards, some supported with exogenous testosterone. Overall, a suppressive effect of testosterone was evident, especially in cells …

Social context alters cognitive performance, but the mechanisms for this remain unclear. One possibility is that stressors associated with social contexts mediate changes in cognitive performance by influencing attention. We conducted two experiments to investigate how social context affects performance on an attention task in capuchin monkeys (Cebus [Sapajus] apella). We additionally validated and measured physiological stress through salivary cortisol. In experiment one, capuchins were exposed to three conditions where they were either isolated or paired with an affiliative or agonistic social partner separated by a barrier. The southern Appalachian Mountains have among the highest salamander diversity in the world, largely due to local speciation in the family Plethodontidae. Plethodontid salamanders (i.e., lungless salamanders) are particularly sensitive to habitat climate conditions due to their reliance on cutaneous respiration, and their immediate environmental conditions (microhabitat) likely influence their dispersion and activity more than the large-scale atmospheric conditions. Recent studies suggest that the use of thermoregulatory treatments that affect brown fat may help curb obesity. However it is unknown how these treatments may affect bone homeostasis. Our work has focused on the transient receptor potential melastatin (TRPM8) protein, which is responsible for detecting colder temperatures in sensory neurons. Previous work within the Motyl laboratory has found that Trpm8 plays a role in bone acquisition. Mice with a global deletion of the Trpm8 gene have reduced trabecular bone volume fraction due to reduced bone formation by osteoblasts. Female mimicry in the red-sided garter snake, Thamnophis sirtalis parietalis, is described in several well-studied evolutionary advantages, but an aspect that remains elusive is how reproductive traits are linked to the 3-fold higher circulating androgens in males with this phenotype. In this study, we implanted male garter snakes (n=15 per group) with either a blank implant (SHAM; control) or a T implant, the latter simulating the female mimic phenotype. In a cellular model, we demonstrate that the non-receptor protein tyrosine kinase 2 beta (PTK2B) plays a critical role in mouse cultured beige adipocyte differentiation. CRISPR/Cas9-mediated knock-out of Ptk2b results in non-differentiating white adipocytes and differentiated beige adipocytes with significantly reduced thermogenic gene and protein expression, enlarged lipid droplet size, and altered mitochondrial respiration. Together, these data in a cell culture system provide evidence for a role of PTK2B in the differentiation of mouse beige adipocytes.

I tested the hypothesis that recombinant leptin protein can be introduced to zebrafish in vivo through non-invasive soaking in a solution containing the protein. One way to study various molecules’ effects in vivo is through intraperitoneal or intracerebroventricular injections during the embryonic or larval stage, which is invasive, difficult to administer, and can have a high mortality rate. 48 hours post fertilization (hpf) zebrafish were soaked in a His-tagged recombinant leptin protein solution at 10 nM and 100 nM concentrations (produced by Genscript). After soaking, zebrafish larvae were washed extensively to remove all recombinant protein on their exterior before homogenization. …

Habitat creation is an important tool for conservation to counteract habitat loss and degradation. Vernal pools are susceptible to destruction due to limited detection, protection, and regulation. These wetlands provide fishless breeding habitat for many amphibian species including spotted salamanders (Ambystoma maculatum) in eastern North America. Determining whether created vernal pool habitat is successful is often determined by demographic data of colonizing populations. Non-invasive drug therapies are not available for bone fractures which represents a problem for this population. Vasoactive intestinal peptide (VIP) and Pituitary Adenylate Cyclase-Activating Polypeptide (PACAP), two neuromodulator peptides in the glucagon superfamily, have demonstrated positive regulation of osteoblast proliferation and activity. Using acidic oligopeptides, we have developed ligands that target to and accumulate at fracture sites. Molting in crustaceans is a highly complex physiological process involving negative regulation by two paired endocrine glands, the X-organ/sinus gland complex (XO/SG) and the Y-organ (YO). The XO/SG complex is responsible for making molt-inhibiting hormone (MIH) which negatively regulates synthesis of the molting hormones, ecdysteroids, by the YO. Analysis of gene expression in the XOs and YOs has led to the development of a proposed molecular signaling pathway which regulates ecdysteroidogenesis and subsequent molting in crustaceans. The adrenal physiology of the albino rat was studied through the measurement of adrenal ascorbic acid levels. A cyclic phenomenon was observed which follows the diel periodicity of the animal. Ascorbic acid values were found to be characteristically higher in males than in females. A similar difference was also seen in populations divided into young animals (body weight less than 175 grams) and old animals (body weight greater than 175 grams). The diurnal rhythm of adrenal ascorbic acid (DAR) displays an inverse relationship to the diurnal rhythm of circulating levels of plasma corticosteroids. Alloxan monohydrate, 175 mg/kg body weight was given subcutaneously to albino rats to determine the effect of the diabetic state on the DAR.
