Canine testes thin section culture

Testes tissue culture systems would provide a tool to elucidate spermatogenesis mechanisms, with the aim of genetic preservation of mammals, especially endangered species. Our experiment aims to develop a culture system capable of producing viable mammalian sperm cells in vitro.

Dogs were chosen as the model organism because testes are readily available. Canine testes were obtained from a local veterinary clinic. Thin sections were generated using a commercial electric slicer. They then were cleaned using Dulbecco’s Phosphate-Buffered Saline (DPBS) supplemented with antibiotics then cultured in a modified Tissue Culture Medium 199 (TCM-199). Sections were …

Metabolic disease and obesity affect millions of individuals worldwide and the prevalence of obesity is expected to continue to rise. Mechanisms underlying diet-induced metabolic disease and obesity are complex and are largely unknown. In this dissertation research, we aimed to make advances toward understanding how body temperature and high-fat diet consumption contribute to the development of obesity. Herein, we describe the progress that was made toward three research projects. Morphological And Functional Evidence Of Sexual Dimorphism In The Retrochiasmatic Area Population Of Nk3r-Containing Neurons In Sheep, Justin A. Lopez

Neurokinin B (NKB) and kisspeptin are critical for fertility and potent stimulators of gonadotropin-releasing hormone (GnRH) and luteinizing hormone (LH) secretion in sheep. We previously demonstrated that administration of NKB receptor (NK3R) agonist, senktide, within the retrochiasmatic area (RCh) resulted in surge-like release of LH and increased expression of kisspeptin neurons in the arcuate nucleus (ARC) of female lambs. We concluded that NKB-responsive neurons in the RCh stimulate GnRH secretion by inducing kisspeptin release from neurons in the ARC.

Hormone Therapy (HT) is used to increase the 17β-estradiol (E2) levels in women who are going through the menopausal transition to alleviate the negative symptoms associated with menopause and decrease the risk of chronic diseases. Clinical and basic science research had previously shown that HT provided protection from cognitive decline and reduced the risk of dementia. However the Women's Health Initiative (WHI) clinical trial demonstrated that HT had no beneficial effects for cognitive decline and, in some cases increased the risk of dementia in post-menopausal women.
