Background: The average menstrual cycle of a healthy adult woman is a duration of approximately 28 days, during which in the follicular phase (first 14 days) the progesterone hormone is at a stable level while estrogen increases to its peak level on day 14. Day 14 is known as ovulation, during which progesterone increases during its Luteinizing Hormone (LH) surge while estrogen decreases. Menses, known colloquially as a woman’s “period,” begins approximately at day 14 after ovulation and lasts an average of 7 days. Canine testes thin section culture

Testes tissue culture systems would provide a tool to elucidate spermatogenesis mechanisms, with the aim of genetic preservation of mammals, especially endangered species. Our experiment aims to develop a culture system capable of producing viable mammalian sperm cells in vitro.

Dogs were chosen as the model organism because testes are readily available. Canine testes were obtained from a local veterinary clinic. Thin sections were generated using a commercial electric slicer. They then were cleaned using Dulbecco’s Phosphate-Buffered Saline (DPBS) supplemented with antibiotics then cultured in a modified Tissue Culture Medium 199 (TCM-199). Sections were …

Amphibian biodiversity has greatly diminished in recent years due to disease. Panzootic pathogenic fungi Batrachochytrium dendrobatidis (Bd) causes the deadly disease chytridiomycosis (chytrid), a primary driver of catastrophic global amphibian declines and extinctions. The pathogenesis of chytrid is still unclear, as certain species and individuals within a species are differentially affected. Susceptibility and mortality of Bd are influenced by prolonged chronic stress. Prolonged glucocorticoid activity deleteriously affects many of the same physiological processes as Bd infections, and corticosterone is the primary glucocorticoid released by amphibians in response to stress. Recent studies suggest that the use of thermoregulatory treatments that affect brown fat may help curb obesity. However it is unknown how these treatments may affect bone homeostasis. Our work has focused on the transient receptor potential melastatin (TRPM8) protein, which is responsible for detecting colder temperatures in sensory neurons. Previous work within the Motyl laboratory has found that Trpm8 plays a role in bone acquisition. Mice with a global deletion of the Trpm8 gene have reduced trabecular bone volume fraction due to reduced bone formation by osteoblasts. Life history morph, sex, and body condition are traits that may influence stress within salamander populations because of differences in physiology and environmental conditions. Given widespread declines and the effects chronic stress can have on amphibian health, it is important to understand within-population drivers of stress and how population level variation may influence population viability. In patients over the age of 65 especially, bone fractures represent a significant disease burden. Non-invasive drug therapies are not available for bone fractures which represents a problem for this population. Vasoactive intestinal peptide (VIP) and Pituitary Adenylate Cyclase-Activating Polypeptide (PACAP), two neuromodulator peptides in the glucagon superfamily, have demonstrated positive regulation of osteoblast proliferation and activity. Using acidic oligopeptides, we have developed ligands that target to and accumulate at fracture sites. These targeting ligands can be synthesized in sequence with bone anabolic peptides to minimize off target effects and increase potency at the fracture site to create safer and …

This project will explore the impact of environmental stressors on the cognitive abilities of birds. Predator perception has been demonstrated to elicit a stress response by elevating stress hormones which can alter the behaviour of birds. The aim of this study is to observe if chronic stress from differently perceived threats of predation in an individual’s environment will lead to differences in the spatial memory abilities in two species of birds.
