Background: The COVID-19 pandemic has shed new light on inequities in healthcare access faced by immigrant and refugee communities. To address ongoing disparities, there is an urgent need for ecological approaches to better understand the barriers that hinder and resources that facilitate access to healthcare. This study investigates barriers to healthcare system access faced by Yazidi refugees in the Midwestern United States.

Barriers To Summer Food Service Program (Sfsp) Implementation Before And After Covid-19: A Qualitative, Collective Case Study, Masoomeh Hajizadeh Oghaz, Falah N. Rashoka, Megan S. Kelley

Objectives: SFSP, as a federal food assistance program, could ensure that children have sufficient nutritious food during summer. According to a 2016 USDA report, only 0.4–0.6% of eligible children participated in this program in Nebraska, which makes for one of the lowest SFSP participation rates in the U.S. This study aims to better understand how the COVID-19 pandemic has impacted SFSP implementation in urban and rural settings across the state.
Conclusions: Insights from this study may inform the development of policies that expand food access to vulnerable families. On the organizational level, this includes providing participating kids and their parents …

Sustained inflammatory responses have been implicated in various neurodegenerative disorders. Cleistocalyx nervosum var. paniala (CN), an edible berry, has been reported to exhibit several beneficial properties. However, only limited research is available on this fruit seed, which is classified as agricultural food waste. We therefore focused on the anti-neuroinflammatory effects and underlying mechanism of C. nervosum var. paniala seed extract (CNSE) on BV-2 mouse microglial cells induced by lipopolysaccharide (LPS) or tumor necrosis factor-alpha (TNF-α). ในปัจจุบันโรคมะเร็งเต้านมเป็นมะเร็งที่มีอุบัติการณ์และอัตราการตายสูงที่สุดในเพศหญิงทั่วโลกรวมถึงในประเทศไทยด้วย โดยพบว่า LINE-1 ถือว่าเป็น Retrotransposon element ที่พบมากที่สุดถึง 17 % ของจีโนมและมีความสามารถในการคัดลอกแล้วแทรกตัวเองในบริเวณอื่นของจีโนมซึ่งเป็นสาเหตุของทำให้เกิดความไม่สเถียรของจีโนมอันเป็นสาเหตุของการเกิดโรคมะเร็ง และยังพบว่าการแสดงออกของLINE-1 มีผลต่อการลุกลาม, ขนาด, และระยะของโรคมะเร็งเต้านมด้วย นอกจากนี้ยังพบว่าผู้หญิงที่การได้รับฮอร์โมนเอสโตรเจนต่อเนื่องเป็นระยะเวลานานทำให้ความเสี่ยงของโรคมะเร็งเต้านมเพิ่มมากขึ้น ถึงกระนั้นยังคงไม่ทราบถึงกลไกการทำงานของเอสโตรเจน ดังนั้นผู้วิจัยจึงตั้งสมมุติฐานว่า เอสโตรเจนสามารถเหนี่ยวนำให้เกิดการแทรกของ LINE-1 ดีเอ็นเอเข้าไปในจีโนมของเซลล์มะเร็งเต้านม โดยทำการศึกษาบผลของเอสโตรเจนที่ได้รับต่อการแสดงออกของ LINE-1 mRNA และ LINE-1 โปรตีน รวมถึงผลของเอสโตรเจนที่ได้รับต่อการแทรกของ LINE-1 ดีเอ็นเอโดยวิเคราะห์จากปริมาณดีเอ็นเอในเซลล์มะเร็งเต้านม T47D ผลการทดลองพบว่าที่เอสโตรเจนมีผลทำให้การแสดงออกของ LINE-1 mRNA เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ความเข้มข้นของเอสโตรเจน 5 nM ด้วยวิธี Realtime RT-PCR ในเซลล์มะเร็งเต้านม T47D และผลทำให้การแสดงออกของ LINE-1 mRNA เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ 12 ชม. ด้วยวิธี Realtime RT-PCR ในเซลล์มะเร็งเต้านม T47D และเอสโตรเจนมีผลทำให้การแสดงออกของโปรตีน LINE-1 ORF1p เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ 48 และ 72 ชม. ในเซลล์มะเร็งเต้านม T47D ด้วยวิธี Western blot โดยพบว่าการแสดงออกของโปรตีน LINE-1 ORF1p เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญ 72 ชม. นอกจากนี้เอสโตรเจนมีผลทำให้ปริมาณของ LINE-1 ดีเอ็นเอบริเวณ ORF2 เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ 72 ชม.ในเซลล์มะเร็งเต้านม T47D มากที่สุดอย่างมีนัยสำคัญด้วยวิธี Real-time PCR แสดงให้เห็นว่าเอสโตรเจนมีผลทำให้เกิดการแทรกของ LINE-1 ดีเอ็นเอเข้าไปในจีโนมได้ จากผลการทดลองทั้งหมดจึงสรุปได้ว่า เอสโตรเจนมีผลต่อการเพิ่มแสดงออกของ LINE-1 mRNA และ LINE-1 โปรตีน รวมถึงเพิ่มปริมาณ LINE-1 ดีเอ็นเอ อย่างมีนัยสำคัญในเซลล์มะเร็งเต้านม T47D แสดงว่าเอสโตรเจนสามารถเหนี่ยวนำให้เกิดการแทรกของ LINE-1 ดีเอ็นเอเข้าไปในจีโนมนำไปสู่การกลายพันธุ์ของยีนและความไม่เสถียรของจีโนม

Bisphenol A (BPA) is widely used in the production of polycarbonate plastics, it has been reported that BPA can activate neuroinflammation. Neuroinflammation is a brain pathology that involves the high levels of pro-inflammatory mediators, including tumor necrosis factor-alpha (TNF-α). An excessive TNF-α expression could result in neuronal cell death and subsequently lead to neurodegeneration. Auricularia polytricha (AP) is an edible mushroom with several medicinal properties. Herein, the anti-neuroinflammatory effects of AP extracts against BPA-induced BV2 microglial inflammation were investigated. Objectives: The human gastrointestinal tract (GI)may play a critical role in the prevention of obesity and metabolic diseases. Considering the importance of a healthy diet for optimal health, this study examined associations of diet quality assessed by three a priori-defined dietary patterns (Healthy Eating Index 2010 [HEI-2010],Mediterranean Eating Pattern for Americans [MEPA], and Dietary Approaches to Stop Hypertension [DASH] with gut microbiota, short-chain fatty acids (SCFA) and intestinal permeabilitymarkers in adult obese participants.

Milk exosomes (MEs) and their microRNA cargos constitute novel bioactive food compounds, and bovine MEs (BMEs) are being considered for use in drug delivery. The internalization of MEs by macrophages and degradation in lysosomes (in all cells) limit biological activities. Here, we determined whether BMEs internalized by murine bone marrow-derived macrophages (BMDMs) retrofuse with the intralumenal membrane in the multivesicular body (MVB) for subsequent release into the extracellular space or whether BMEs are destined for degradation in lysosomes.
Conclusions: The retrofusion of BME-derived intralumenal MVBs and secretion of BME into the extracellular space is quantitatively minor in BMDMs. The majority …

Objectives: To develop BMEs that evade elimination by BMDMs.
Conclusions: The elimination of BMEs UNL1 and UNL2 is significantly reduced compared unmodified BMEs in BMDM cultures. This is of great importance when using BMEs for delivering therapeutics.

Neurodegenerative diseases (NDs) are the age-related diseases which represent a major cause of mortality in aging population nowadays. Previous studies have shown that glutamate, the most abundant excitatory neurotransmitter, significantly increases in extracellular fluid of the patient’s brain and is the cause of neurodegeneration in NDs. The increase of extracellular glutamate elevates reactive oxygen species (ROS) accumulation and calcium (Ca2+) which exacerbate mitochondrial function, followed by inducing MAPK pathway and endoplasmic reticulum (ER) stress, respectively. The importance of vitamin D toxicity (VDT) has been underestimated and under recognized. This study was done to determine the frequency toxicity (>150 ng/ml) in subjects for 25-hydroxyvitamin D (25OHD) and evaluate the vitamin D (VD) supplements used by these subjects.

Objectives: Dietary γ -glutamyl peptides (γ -GPs) found in dry edible beans exhibit biological activities (antioxidant, anti-inflammatory) with potential benefits against chronic metabolic disorders. γ -GPs are responsible for the desirable Kokumi flavor through allosteric activation of the Calcium Sensing Receptor (CaSR) present in multiple mammalian tissues. In this study, we investigated the metabolic effects of γ -glutamyl valine (γ -EV) in diabetic obese mice.
Conclusions: γ -EV improved the diabetic condition of db/db mice via modulation of glucose and lipid metabolism.

Background: The COVID-19 pandemic brought new challenges affecting the wellbeing of individuals in communities with low income. Understanding where people live and how those environments can facilitate or hinder living a healthy lifestyle is essential for developing interventions that target behavior change and health promotion. Objective: This study compares Extension Nutrition Educators’ (NEs) perceptions of the barriers and facilitators impacting the healthfulness of the environment of communities with low income in eleven states before and during the COVID-19 pandemic.

มะเร็งท่อน้ำดีเป็นมะเร็งที่เกิดขึ้นบริเวณท่อทางเดินน้ำดี โดยมีความหลากหลายของเซลล์มะเร็งบริเวณเนื้อเยื่อมะเร็งและมีความรุนแรง มะเร็งท่อน้ำดีจัดเป็นสาเหตุหลักในการเสียชีวิตของผู้ป่วยในประเทศไทย เนื่องจากผู้ป่วยได้รับการวินิจฉัยเมื่อมะเร็งเจริญพัฒนาไปยังระยะท้ายและยังขาดวิธีการรักษาที่มีประสิทธิภาพ การรักษามะเร็งท่อน้ำดีด้วยวิธีมาตรฐานในปัจจุบันในผู้ป่วยที่ไม่สามารถผ่าตัดได้ หรือในผู้ป่วยหลังการผ่าตัด คือ การรักษาด้วยยาเคมีบำบัด ชนิดเจมไซทาบีนหรือยาเจมไซทาบีนร่วมกับยาซิสพลาติน แต่อย่างไรก็ตามผู้ป่วยส่วนใหญ่พบปัญหาการดื้อต่อยาเคมีบำบัดนำไปสู่การพยากรณ์โรคที่ไม่ดี ดังนั้นการพัฒนาการรักษาที่มีประสิทธิภาพมากยิ่งขึ้นถือว่ามีความจำเป็นเร่งด่วน โดยได้มีการศึกษาการตายของเซลล์มะเร็งแบบใหม่ที่ทำให้เซลล์มะเร็งตาย และกระตุ้นการทำงานของระบบภูมิคุ้มกันแบบจำเพาะ เรียกว่า การตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการตายของเซลล์แบบเนคโครปโทซิส จัดว่าเป็นการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยได้มีการศึกษาอย่างแพร่หลายในมะเร็งหลายชนิด โดยพบว่าการกระตุ้นให้เซลล์มะเร็งตายแบบเนคโครปโทซิสสามารถกระตุ้นระบบภูมิคุ้มแบบจำเพาะและยับยั้งการเจริญพัฒนาของก้อนมะเร็งในหนูทดลอง การศึกษาภายในห้องปฏิบัติการก่อนหน้าของคณะผู้วิจัยพบว่าการเหนี่ยวนำร่วมกันระหว่างยาเคมีบำบัดเจมไซทาบีนและสารสะแมคไมเมติกสามารถเหนี่ยวนำให้เซลล์มะเร็งท่อน้ำดีตายแบบเนคโครปโทซิสในสภาวะที่มีการยับยั้งการทำงานของเอนไซม์แคสเปส ทั้งนี้จากข้อจำกัดที่ยาเคมีบำบัดมีความเป็นพิษที่สูง การศึกษานี้จึงมีจุดมุ่งหมายเพื่อค้นหาสารที่สามารถนำมาทำการเหนี่ยวนำร่วมกับสารสะแมคไมเมติก โดยมีสมมติฐานของงานวิจัย คือ สารพฤกษเคมีที่มีฤทธิ์ทำให้เกิดการทำลายของดีเอ็นเอและหยุดวัฏจักรของเซลล์จะสามารถเสริมฤทธิ์กับสารสะแมคไมเมติก โดยกระตุ้นการตายของเซลล์แบบในคโครปโทซิสในเซลล์มะเร็งท่อน้ำดี โดยงานวิจัยนี้ได้ทำการศึกษาฤทธิ์ยับยั้งการแบ่งตัวและการตายเซลล์มะเร็งท่อน้ำดีของสารพฤกษเคมี ได้แก่ ชิโคนิน พลัมบาจิน เควอซิติน โดยพบว่าสารเควอซิตินมีค่าความเข้มข้นของสารที่ยับยั้งการรอดชีวิตของเซลล์ได้ 50% (IC50) ที่ต่ำที่สุดเมื่อเปรียบเทียบกับสารพฤกษเคมีชนิดอื่นๆ ในการวิเคราะห์ด้วยวิธีการทางชีวสารสนเทศน์พบว่าสารเควอซิตินมีเป้าหมายไปยังยีนที่มีบทบาทในการควบคุมวัฏจักรเซลล์ระยะ G2/M และยีนเป้าหมายของสารเควอซิตินมีการแสดงออกที่เพิ่มมากขึ้นอย่างมีนัยสำคัญในเนื้อเยื่อมะเร็งท่อน้ำดีเมื่อเปรียบเทียบกับเซลล์ปกติ การทดลองในเซลล์มะเร็งท่อน้ำดี (in vitro) พบว่าสารเควอซิตินสามารถหยุดวัฏจักรเซลล์ที่ระยะ G2/M นอกจากนี้สารเควอซิตินช่วงความเข้มข้นค่าต่ำที่มีความเป็นพิษน้อยในรูปแบบสารเดี่ยว เมื่อนำไปเหนี่ยวนำร่วมกับสารสะแมคไมเมติกชนิด LCL-161 แล้วพบว่าสามารถเสริมฤทธิ์กันกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดี โดยเมื่อทำการวิเคราะห์ชนิดการตายของเซลล์เพิ่มเติมพบว่ามีการแสดงออกที่เพิ่มขึ้นของโปรตีนเอ็มแอลเคแอลที่ถูกเติมหมู่ฟอสเฟต (phosphorylated MLKL; pMLKL) ซึ่งเป็นตัวบ่งชี้ที่จำเพาะต่อการตายแบบเนคโครปโทซิส นอกจากนั้นยังพบว่าการตายของเซลล์ถูกยับยั้งเมื่อมีการเติมตัวยับยั้งที่จำเพาะกับโปรตีนที่สำคัญต่อการเกิดเนคโครปโทซิส แสดงให้เห็นว่าการเหนี่ยวนำเซลล์ด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นให้เซลล์มะเร็งท่อน้ำดีตายผ่านเนคโครปโทซิส การศึกษาเพิ่มเติมเพื่อศึกษาการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการวิเคราะห์การเคลื่อนย้ายของโปรตีนเอชเอ็มจีบีวัน (HMGB-1) จากภายในนิวเคลียสมายังบริเวณไซโตพลาสซึมด้วยวิธีการย้อมแบบอิมมูโนฟลูออเรสเซนส์ พบว่าการเหนี่ยวนำเซลล์มะเร็งท่อน้ำดีด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นการตายของเซลล์ที่กระตุ้นภูมิคุ้ม กล่าวโดยสรุป การเหนี่ยวนำร่วมกันระหว่างสารเควอซิตินและสารสะแมคไมเมติกในสภาวะที่มีการยับยั้งเอนไซม์แคสเปสสามารถเสริมฤทธิ์กันโดยกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดีแบบเนคโครปโทซิสและการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน ดังนั้นงานวิจัยนี้อาจนำไปสู่การพัฒนากลยุทธ์การรักษามะเร็งท่อน้ำดีแบบใหม่ที่เพิ่มประสิทธิภาพในการรรักษาในอนาคต อย่างไรก็ตาม งานวิจัยนี้ยังต้องมีการศึกษาเพิ่มเติม เช่น การศึกษากลไกการเสริมฤทธิ์และการกระตุ้นการตายของเซลล์ การศึกษาฤทธิ์ในเซลล์มะเร็งท่อน้ำดีชนิดอื่นๆ และการเมินความเป็นพิษในเซลล์ปกติ เป็นต้น

Conventional therapy approaches often fail to elicit significant responses in patients, resulting in low survival rates compared to other cancer types. Low-hierarchy selenoproteins are sensitive to selenium (Se) deficiency and are proposed to confer the protection of body Se against age- and development-related diseases. Selenoprotein H (SELENOH), a low-hierarchy selenoprotein, is greatly downregulated by both dietary Se deficiency and age. To explore physiological roles of SELENOH, Selenoh knockout mice were employed.
Segregation analyses demonstrated reduced frequencies of homozygotes and heterozygotes among the neonates of the breeding combinations of Selenoh+/- males vs. Selenoh+/- or Selenoh-/- females, demonstrating essential roles of SELENOH in embryogenesis. How regular physical activity is able to improve health remains poorly understood. The release of factors from skeletal muscle following exercise has been proposed as a possible mechanism mediating such systemic benefits. We describe a mechanism wherein skeletal muscle, in response to a hypertrophic stimulus induced by mechanical overload (MOV), released extracellular vesicles (EVs) containing muscle-specific miR-1 that were preferentially taken up by epidydimal white adipose tissue (eWAT). In eWAT, miR-1 promoted adrenergic signaling and lipolysis by targeting Tfap2α, a known repressor of Adrβ3 expression. Many of the molecular and cellular mechanisms discovered to regulate skeletal muscle hypertrophy were first identified using the rodent synergist ablation model. This model reveals the intrinsic capability and necessary pathways of skeletal muscle growth in response to mechanical overload (MOV). Reminiscent of the rapid cellular growth observed with cancer, we hypothesized that in response to MOV, skeletal muscle would undergo metabolic programming to sustain increased demands to support hypertrophy. Aberrant activation of the nucleotide-binding domain and leucine-rich repeat related (NLR) family, pyrin domain containing 3 (NLRP3) inflammasome drives the development of many complex inflammatory diseases, such as obesity, Alzheimer’s disease, and atherosclerosis. However, no medications specifically targeting the NLRP3 inflammasome have become clinically available. Therefore, we aim to identify new inhibitors of the NLRP3 inflammasome in this study. Lipid-soluble micronutrients may be beneficial to non-alcoholic fatty liver disease due to their important roles in metabolism and maintaining tissue functions. Utilizing 2017–2018 National Health and Nutrition Examination Survey, this study examined the potential overall and race/ethnicity-specific (black, Hispanic and white) associations of dietary lipid-soluble micronutrients (α-tocopherol, retinol, vitamin D, β-carotene and total carotenoids) with hepatic steatosis. The analysis included 4376 adults (1037 blacks, 981 Hispanics, 1549 whites) aged ≥20 years who completed the transient elastography examination with dietary data available. We sought to characterize the lipid profile of skeletal muscle cell-derived Extracellular Vesicles (EVs) to determine if a hypertrophic stimulus would affect the lipid composition of C2C12 myotube-derived EVs. Analyses included C2C12 murine myoblasts differentiated into myotubes and treated with Insulin-Like Growth Factor 1 (IGF-1) for 24 h to induce hypertrophic growth. EVs were isolated from cell culture media, quantified using Nanoparticle Tracking Analysis (NTA) and analyzed using Transmission Electron Microscopy (TEM). Background: A significant portion of lactating women in less developed countries have vitamin A (VA) deficiency. Lactation has substantial effects on a mother’s metabolism and VA is known to be needed in multiple biological processes, including growth, vision, immunity, and reproduction.

Bovine milk exosomes (BMEs) have attracted attention as vehicles for delivering RNA therapeutics. BMEs originate in mammary alveolar cells. Here, we determined whether bovine mammary alveolar MAC-T cells are a promising tool to assess RNA delivery by BMEs. MAC-T cells exosomes (MAC-T BMEs) and BMEs were harvested by differential ultracentrifugation. Exosome size, morphology, microRNA content and marker proteins were assessed using nanoparticle tracking analysis, transmission electron microscopy, real-time PCR and immunoblot analysis, respectively. Background: Healthy dietary patterns are related to better cognitive health in aging populations. While levels of individual nutrients in neural tissues are individually associated with cognitive function, the investigation of nutrient patterns in human brain tissue has not been conducted.
Methods: Brain tissues were acquired from frontal and temporal cortices of 47 centenarians from the Georgia Centenarian Study. Background
Previous work in HEK-293 cells demonstrated the importance of amino acid-induced mTORC1 translocation to the lysosomal surface for stimulating mTORC1 kinase activity and protein synthesis. This study tested the conservation of this amino acid sensing mechanism in human skeletal muscle by treating subjects with chloroquine—a lysosomotropic agent that induces in vitro and in vivo lysosome dysfunction.

Zika virus (ZIKV) infection in pregnancy is associated with the development of microcephaly, intrauterine growth restriction, and ocular damage in the fetus. ZIKV infection of the placenta plays a crucial role in the vertical transmission from the maternal circulation to the fetus. Our previous study suggested that ZIKV induces endoplasmic reticulum (ER) stress and apoptosis of placental trophoblasts. Here, we showed that palmitoleate, an omega-7 monounsaturated fatty acid, prevents ZIKV-induced ER stress and apoptosis in placental trophoblasts.
