Type 2 diabetes mellitus (T2D) is one of the leading causes of morbidity and mortality in the United States, with approximately 25% of healthcare expenditures directed toward its management and associated metabolic complications. In response to the urgent need for novel therapeutic strategies, recent research has focused on bioactive compounds derived from dietary sources. In light of that, we identified the plant lignan matairesinol (MA) and its gut-derived metabolite, enterolactone (ENL), as agents capable of enhancing glucose uptake in 3T3-L1 differentiated adipocytes. Maternal insulin resistance and hyperglycemia lead to gestational diabetes mellitus (GDM) which can cause fetal hyperinsulinemia, ultimately leading to excessive fetal growth associated with macrosomia and increased adiposity at birth. Increased placental macronutrient transport and epigenetic changes in growth and metabolic genes affected by GDM contribute to fetal overgrowth and can result in complications such as overweight/obesity, insulin resistance/type 2 diabetes, cardiovascular disease risk factors, as well as cognitive impairment later in life. Many HIV-positive (HIV+) males on Highly Active Anti-Retroviral Therapy (HAART) experience metabolic abnormalities, including Non-Alcoholic Fatty Liver Disease (NAFLD) and lipodystrophy. The intestinal microbiota and short chain fatty acids (SCFA), participate in bidirectional communication with their host. Dysbiosis in HIV+ males on HAART demonstrate a Prevotella-rich enterotype shaped by multiple factors including, medications, adiposity, diet, intestinal permeability, and lifestyle; our objective was to investigate these factors. 19 HIV+ and 21 HIV- males were enrolled. กลุ่มอาการออทิซึมสเปกตรัม (Autism spectrum disorder; ASD) เป็นปัญหาสุขภาพระดับโลกที่สำคัญ โดยพบว่ามีความชุกของโรคที่เพิ่มสูงขึ้น อย่างไรก็ตามสาเหตุและกลไกในการเกิดภาวะออทิซึมสเปกตรัมยังไม่ทราบแน่ชัดอาจเกี่ยวข้องกับปัจจัยด้านพันธุกรรมและสิ่งแวดล้อม โดยปัจจุบันมีรายงานถึงบทบาทของวิตามินกับภาวะออทิซึมสเปกตรัม วัตถุประสงค์ของการศึกษานี้คือศึกษาการเกิดภาวะพหุสัณฐานทางพันธุกรรมของยีนในวิถีเมทาบอลิกของวิตามินดีและความเสี่ยงต่อการเกิดภาวะออทิซึมสเปกตรัมและระดับความรุนแรงของโรคในเด็กไทย การศึกษานี้มีจำนวนอาสาสมัคร 276 ราย โดยเป็นกลุ่มเด็กที่มีภาวะออทิซึมสเปกตรัม 169 ราย และกลุ่มควบคุมเป็นเด็กที่ไม่มีโรคทางระบบประสาท 107 ราย ซึ่งได้ทำการตรวจวิเคราะห์ภาวะพหุสัณฐานทางพันธุกรรมของยีน CYP2R1, CYP27B1, GC และ VDR โดยเทคนิค TaqMan-based real-time Polymerase Chain Reaction (PCR) และตรวจวิเคราะห์ระดับวิตามินดีในกระแสเลือดโดยใช้เทคนิค Chemiluminescence Immunoassay (CLIA) โดยเด็กที่พบภาวะออทิซึมสเปกตรัมจะได้รับการตรวจระดับความรุนแรงของภาวะออทิซึมสเปกตรัมด้วย Childhood Autism Rating Scale, 2nd Edition (CARS2) โดยผลที่ได้จากการศึกษาพบว่า การเกิดภาวะพหุสัณฐานทางพันธุกรรมของยีน CYP2R1, CYP27B1, VDR และ GC (rs4588) ไม่มีความสัมพันธ์กับการเกิดภาวะออทิซึมสเปกตรัมและความรุนแรงของอาการออทิซึมสเปกตรัม อย่างไรก็ตามการศึกษาชี้ให้เห็นว่า การเกิดภาวะพหุสัณฐานทางพันธุกรรมของยีน GC (rs7041 (A>C)) มีความสัมพันธ์อย่างมีนัยสำคัญกับความเสี่ยงที่เพิ่มขึ้นต่อการเกิดภาวะออทิซึมสเปกตรัมและความรุนแรงของอาการออทิซึมสเปกตรัมด้วย โดยเฉพาะอย่างยิ่งบุคคลที่มีการขาดระดับวิตามินดีร่วมด้วย นอกจากนี้ผลศึกษาพบว่าไอโซฟอร์มของ GC1s และ GC1s-GC1s ฟีโนไทป์มีความสัมพันธ์อย่างมีนัยสำคัญกับความเสี่ยงต่อการเกิดภาวะออทิซึมสเปกตรัมตลอดจนความรุนแรงของอาการออทิซึมสเปกตรัม โดยสรุปการศึกษาวิจัยในครั้งนี้ชี้ให้เห็นว่า การเกิดภาวะพหุสัณฐานทางพันธุกรรมของยีน GC (rs7041 (A>C)) และไอโซฟอร์ม GC1s อาจเป็นตัวบ่งชี้ทางพันธุกรรมที่สามารถช่วยในการประเมินความเสี่ยงต่อการเกิดภาวะออทิซึมสเปกตรัมและความรุนแรงของอาการออทิซึมสเปกตรัมในเด็กไทยได้ในอนาคต อย่างไรก็ตามการศึกษาในอนาคตควรศึกษาในกลุ่มประชากรที่ใหญ่ขึ้นเพื่อยืนยันผลการศึกษาวิจัยครั้งนี้

Progesterone receptor (PR) extranuclear signaling mediates mainly through PR polyproline domain (PPD) interaction with SH3 domain of cytoplasmic signaling molecules. PR-PPD interfered with Grb2-SOS binding through SH3 interaction resulting in decreased cell proliferation and correlates with a good prognosis in PR positive breast cancer. Grb2 is a key adaptor molecule and expressed in both nucleus and cytoplasm. We hypothesized that change in Grb2 localization alters PR transcription resulting in better clinical outcomes in PR positive breast cancer. Maternal obesity has led to an increase in adverse offspring developmental outcomes and a greater risk for long-term metabolic diseases. Choline, a semi-essential nutrient, can be incorporated into phosphatidylcholine (PC) as well as sphingomyelin (SM) and donate its labile methyl group for the remethylation of homocysteine after choline is oxidized to betaine. Prenatal choline insufficiency has been related to maternal obesity and metabolic diseases, such as metabolic associated fatty liver disease (MAFLD). Cleistocalyx nervosum var. paniala (CN), an edible berry, has been reported to exhibit several beneficial properties. However, only limited research is available on this fruit seed, which is classified as agricultural food waste. We therefore focused on the anti-neuroinflammatory effects and underlying mechanism of C. nervosum var. paniala seed extract (CNSE) on BV-2 mouse microglial cells induced by lipopolysaccharide (LPS) or tumor necrosis factor-alpha (TNF-α). ในปัจจุบันโรคมะเร็งเต้านมเป็นมะเร็งที่มีอุบัติการณ์และอัตราการตายสูงที่สุดในเพศหญิงทั่วโลกรวมถึงในประเทศไทยด้วย โดยพบว่า LINE-1 ถือว่าเป็น Retrotransposon element ที่พบมากที่สุดถึง 17 % ของจีโนมและมีความสามารถในการคัดลอกแล้วแทรกตัวเองในบริเวณอื่นของจีโนมซึ่งเป็นสาเหตุของทำให้เกิดความไม่สเถียรของจีโนมอันเป็นสาเหตุของการเกิดโรคมะเร็ง และยังพบว่าการแสดงออกของLINE-1 มีผลต่อการลุกลาม, ขนาด, และระยะของโรคมะเร็งเต้านมด้วย นอกจากนี้ยังพบว่าผู้หญิงที่การได้รับฮอร์โมนเอสโตรเจนต่อเนื่องเป็นระยะเวลานานทำให้ความเสี่ยงของโรคมะเร็งเต้านมเพิ่มมากขึ้น ถึงกระนั้นยังคงไม่ทราบถึงกลไกการทำงานของเอสโตรเจน ดังนั้นผู้วิจัยจึงตั้งสมมุติฐานว่า เอสโตรเจนสามารถเหนี่ยวนำให้เกิดการแทรกของ LINE-1 ดีเอ็นเอเข้าไปในจีโนมของเซลล์มะเร็งเต้านม โดยทำการศึกษาบผลของเอสโตรเจนที่ได้รับต่อการแสดงออกของ LINE-1 mRNA และ LINE-1 โปรตีน รวมถึงผลของเอสโตรเจนที่ได้รับต่อการแทรกของ LINE-1 ดีเอ็นเอโดยวิเคราะห์จากปริมาณดีเอ็นเอในเซลล์มะเร็งเต้านม T47D ผลการทดลองพบว่าที่เอสโตรเจนมีผลทำให้การแสดงออกของ LINE-1 mRNA เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ความเข้มข้นของเอสโตรเจน 5 nM ด้วยวิธี Realtime RT-PCR ในเซลล์มะเร็งเต้านม T47D และผลทำให้การแสดงออกของ LINE-1 mRNA เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ 12 ชม. ด้วยวิธี Realtime RT-PCR ในเซลล์มะเร็งเต้านม T47D และเอสโตรเจนมีผลทำให้การแสดงออกของโปรตีน LINE-1 ORF1p เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ 48 และ 72 ชม. ในเซลล์มะเร็งเต้านม T47D ด้วยวิธี Western blot โดยพบว่าการแสดงออกของโปรตีน LINE-1 ORF1p เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญ 72 ชม. นอกจากนี้เอสโตรเจนมีผลทำให้ปริมาณของ LINE-1 ดีเอ็นเอบริเวณ ORF2 เพิ่มขึ้นสูงที่สุดอย่างมีนัยสำคัญที่ 72 ชม.ในเซลล์มะเร็งเต้านม T47D มากที่สุดอย่างมีนัยสำคัญด้วยวิธี Real-time PCR แสดงให้เห็นว่าเอสโตรเจนมีผลทำให้เกิดการแทรกของ LINE-1 ดีเอ็นเอเข้าไปในจีโนมได้ จากผลการทดลองทั้งหมดจึงสรุปได้ว่า เอสโตรเจนมีผลต่อการเพิ่มแสดงออกของ LINE-1 mRNA และ LINE-1 โปรตีน รวมถึงเพิ่มปริมาณ LINE-1 ดีเอ็นเอ อย่างมีนัยสำคัญในเซลล์มะเร็งเต้านม T47D แสดงว่าเอสโตรเจนสามารถเหนี่ยวนำให้เกิดการแทรกของ LINE-1 ดีเอ็นเอเข้าไปในจีโนมนำไปสู่การกลายพันธุ์ของยีนและความไม่เสถียรของจีโนม

Bisphenol A (BPA) is widely used in the production of polycarbonate plastics, it has been reported that BPA can activate neuroinflammation. Neuroinflammation is a brain pathology that involves the high levels of pro-inflammatory mediators, including tumor necrosis factor-alpha (TNF-α). An excessive TNF-α expression could result in neuronal cell death and subsequently lead to neurodegeneration. Auricularia polytricha (AP) is an edible mushroom with several medicinal properties. Neurodegenerative diseases (NDs) are the age-related diseases which represent a major cause of mortality in aging population nowadays. Previous studies have shown that glutamate, the most abundant excitatory neurotransmitter, significantly increases in extracellular fluid of the patient’s brain and is the cause of neurodegeneration in NDs. The increase of extracellular glutamate elevates reactive oxygen species (ROS) accumulation and calcium (Ca2+) which exacerbate mitochondrial function, followed by inducing MAPK pathway and endoplasmic reticulum (ER) stress, respectively. มะเร็งท่อน้ำดีเป็นมะเร็งที่เกิดขึ้นบริเวณท่อทางเดินน้ำดี โดยมีความหลากหลายของเซลล์มะเร็งบริเวณเนื้อเยื่อมะเร็งและมีความรุนแรง มะเร็งท่อน้ำดีจัดเป็นสาเหตุหลักในการเสียชีวิตของผู้ป่วยในประเทศไทย เนื่องจากผู้ป่วยได้รับการวินิจฉัยเมื่อมะเร็งเจริญพัฒนาไปยังระยะท้ายและยังขาดวิธีการรักษาที่มีประสิทธิภาพ การรักษามะเร็งท่อน้ำดีด้วยวิธีมาตรฐานในปัจจุบันในผู้ป่วยที่ไม่สามารถผ่าตัดได้ หรือในผู้ป่วยหลังการผ่าตัด คือ การรักษาด้วยยาเคมีบำบัด ชนิดเจมไซทาบีนหรือยาเจมไซทาบีนร่วมกับยาซิสพลาติน แต่อย่างไรก็ตามผู้ป่วยส่วนใหญ่พบปัญหาการดื้อต่อยาเคมีบำบัดนำไปสู่การพยากรณ์โรคที่ไม่ดี ดังนั้นการพัฒนาการรักษาที่มีประสิทธิภาพมากยิ่งขึ้นถือว่ามีความจำเป็นเร่งด่วน โดยได้มีการศึกษาการตายของเซลล์มะเร็งแบบใหม่ที่ทำให้เซลล์มะเร็งตาย และกระตุ้นการทำงานของระบบภูมิคุ้มกันแบบจำเพาะ เรียกว่า การตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการตายของเซลล์แบบเนคโครปโทซิส จัดว่าเป็นการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยได้มีการศึกษาอย่างแพร่หลายในมะเร็งหลายชนิด โดยพบว่าการกระตุ้นให้เซลล์มะเร็งตายแบบเนคโครปโทซิสสามารถกระตุ้นระบบภูมิคุ้มแบบจำเพาะและยับยั้งการเจริญพัฒนาของก้อนมะเร็งในหนูทดลอง การศึกษาภายในห้องปฏิบัติการก่อนหน้าของคณะผู้วิจัยพบว่าการเหนี่ยวนำร่วมกันระหว่างยาเคมีบำบัดเจมไซทาบีนและสารสะแมคไมเมติกสามารถเหนี่ยวนำให้เซลล์มะเร็งท่อน้ำดีตายแบบเนคโครปโทซิสในสภาวะที่มีการยับยั้งการทำงานของเอนไซม์แคสเปส ทั้งนี้จากข้อจำกัดที่ยาเคมีบำบัดมีความเป็นพิษที่สูง การศึกษานี้จึงมีจุดมุ่งหมายเพื่อค้นหาสารที่สามารถนำมาทำการเหนี่ยวนำร่วมกับสารสะแมคไมเมติก โดยมีสมมติฐานของงานวิจัย คือ สารพฤกษเคมีที่มีฤทธิ์ทำให้เกิดการทำลายของดีเอ็นเอและหยุดวัฏจักรของเซลล์จะสามารถเสริมฤทธิ์กับสารสะแมคไมเมติก โดยกระตุ้นการตายของเซลล์แบบในคโครปโทซิสในเซลล์มะเร็งท่อน้ำดี โดยงานวิจัยนี้ได้ทำการศึกษาฤทธิ์ยับยั้งการแบ่งตัวและการตายเซลล์มะเร็งท่อน้ำดีของสารพฤกษเคมี ได้แก่ ชิโคนิน พลัมบาจิน เควอซิติน โดยพบว่าสารเควอซิตินมีค่าความเข้มข้นของสารที่ยับยั้งการรอดชีวิตของเซลล์ได้ 50% (IC50) ที่ต่ำที่สุดเมื่อเปรียบเทียบกับสารพฤกษเคมีชนิดอื่นๆ ในการวิเคราะห์ด้วยวิธีการทางชีวสารสนเทศน์พบว่าสารเควอซิตินมีเป้าหมายไปยังยีนที่มีบทบาทในการควบคุมวัฏจักรเซลล์ระยะ G2/M และยีนเป้าหมายของสารเควอซิตินมีการแสดงออกที่เพิ่มมากขึ้นอย่างมีนัยสำคัญในเนื้อเยื่อมะเร็งท่อน้ำดีเมื่อเปรียบเทียบกับเซลล์ปกติ การทดลองในเซลล์มะเร็งท่อน้ำดี (in vitro) พบว่าสารเควอซิตินสามารถหยุดวัฏจักรเซลล์ที่ระยะ G2/M นอกจากนี้สารเควอซิตินช่วงความเข้มข้นค่าต่ำที่มีความเป็นพิษน้อยในรูปแบบสารเดี่ยว เมื่อนำไปเหนี่ยวนำร่วมกับสารสะแมคไมเมติกชนิด LCL-161 แล้วพบว่าสามารถเสริมฤทธิ์กันกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดี โดยเมื่อทำการวิเคราะห์ชนิดการตายของเซลล์เพิ่มเติมพบว่ามีการแสดงออกที่เพิ่มขึ้นของโปรตีนเอ็มแอลเคแอลที่ถูกเติมหมู่ฟอสเฟต (phosphorylated MLKL; pMLKL) ซึ่งเป็นตัวบ่งชี้ที่จำเพาะต่อการตายแบบเนคโครปโทซิส นอกจากนั้นยังพบว่าการตายของเซลล์ถูกยับยั้งเมื่อมีการเติมตัวยับยั้งที่จำเพาะกับโปรตีนที่สำคัญต่อการเกิดเนคโครปโทซิส แสดงให้เห็นว่าการเหนี่ยวนำเซลล์ด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นให้เซลล์มะเร็งท่อน้ำดีตายผ่านเนคโครปโทซิส การศึกษาเพิ่มเติมเพื่อศึกษาการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน โดยการวิเคราะห์การเคลื่อนย้ายของโปรตีนเอชเอ็มจีบีวัน (HMGB-1) จากภายในนิวเคลียสมายังบริเวณไซโตพลาสซึมด้วยวิธีการย้อมแบบอิมมูโนฟลูออเรสเซนส์ พบว่าการเหนี่ยวนำเซลล์มะเร็งท่อน้ำดีด้วยสารเควอซิตินร่วมกับสารสะแมคไมเมติกสามารถกระตุ้นการตายของเซลล์ที่กระตุ้นภูมิคุ้ม กล่าวโดยสรุป การเหนี่ยวนำร่วมกันระหว่างสารเควอซิตินและสารสะแมคไมเมติกในสภาวะที่มีการยับยั้งเอนไซม์แคสเปสสามารถเสริมฤทธิ์กันโดยกระตุ้นการตายของเซลล์มะเร็งท่อน้ำดีแบบเนคโครปโทซิสและการตายของเซลล์ที่กระตุ้นภูมิคุ้มกัน ดังนั้นงานวิจัยนี้อาจนำไปสู่การพัฒนากลยุทธ์การรักษามะเร็งท่อน้ำดีแบบใหม่ที่เพิ่มประสิทธิภาพในการรรักษาในอนาคต อย่างไรก็ตาม งานวิจัยนี้ยังต้องมีการศึกษาเพิ่มเติม เช่น การศึกษากลไกการเสริมฤทธิ์และการกระตุ้นการตายของเซลล์ การศึกษาฤทธิ์ในเซลล์มะเร็งท่อน้ำดีชนิดอื่นๆ และการเมินความเป็นพิษในเซลล์ปกติ เป็นต้น

Two Pore Channels (TPCs) are endolysosomal ion channels that are permeable to sodium and calcium. Defects in TPCs have been implicated to impair vesicle trafficking, autophagy and cell metabolism control; however, the detailed mechanism remains largely unknown. In this study, I show that TPCs are critical for appropriate cargo delivery to the lysosomes and deletion of either TPC1 or TPC2 leads to delayed clearance of autophagosomes, resulting in enlarged lysosomes and accumulated contents inside the lysosomes. Cells with both TPC deleted also exhibit 50% reduction in lysosomal amino acids under normal culture conditions, leading to reduced homeostatic mTORC1 activation.

โรคออทิซึมสเปกตรัมเป็นความผิดปกติเกี่ยวกับพัฒนาการในช่วงเริ่มแรกของระบบประสาทและสมองซึ่งมีอาการทางคลินิกที่หลากหลายมาก ปัจจุบันยังไม่มีสารบ่งชี้เพื่อช่วยในการวินิจฉัยและแบ่งผู้ป่วยเป็นกลุ่มย่อย แต่การวินิจฉัยที่รวดเร็วเป็นสิ่งสำคัญที่จะช่วยให้ผู้ป่วยออทิซึมสเปกตรัมสามารถได้รับการเรียนรู้และพัฒนาพฤติกรรมที่จำเป็นในการดำรงชีวิตได้ การวินิจฉัยโรคออทิซึมสเปกตรัมตามมาตรฐานสากลใช้แบบสอบถามที่มีความซับซ้อนและใช้เวลามาก ซึ่งในปัจจุบันมีการนำเอาเทคโนโลยีปัญญาประดิษฐ์มาประยุกต์ใช้ในการวิเคราะห์ข้อมูลขนาดใหญ่ที่มีความซับซ้อน อย่างไรก็ตามการนำเอาปัญญาประดิษฐ์มาใช้วิเคราะห์อาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมยังไม่แพร่หลายนัก ในการศึกษานี้ผู้วิจัยต้องการนำเอาปัญญาประดิษฐ์มาใช้แบ่งผู้ป่วยออทิซึมสเปกตรัมจำนวน 85 ราย ออกเป็นกลุ่มย่อยตามอาการทางคลินิกจากข้อมูลแบบทดสอบในการวินิจฉัยผู้ป่วยออทิซึมสเปกตรัม ADI-R จำนวน 123 ข้อ จากนั้นได้ทำการสร้างโมเดลในการจัดกลุ่มและเปรียบเทียบประสิทธิภาพรวมถึงจำนวนข้อคำถามที่ใช้ในแต่ละโมเดล นอกจากนี้ผู้วิจัยได้ศึกษาหาสารบ่งชี้ทางชีวภาพที่เป็นตัวแทนของโรคออทิซึมสเปกตรัมกลุ่มย่อยจากลักษณะอาการทางคลินิกโดยอาศัยการบูรณาการโอมิกส์หลายระดับ จึงได้ดำเนินการวิเคราะห์รูปแบบทรานสคริปโตม และวิเคราะห์รูปแบบโปรตีโอมของผู้ป่วยออทิซึมสเปกตรัมโดยใช้เซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์เป็นต้นแบบ โดยใช้เทคนิคการแยกโปรตีนด้วยกระแสไฟฟ้า 2 มิติ (2D-GE) จากนั้นจะทำการระบุโปรตีนด้วยเทคนิค LC-MS/MS เพื่อหาโปรตีนที่ถูกรบกวนการทำงานอย่างมีนัยสำคัญ และบูรณาการข้อมูลเพื่อศึกษาความสอดคล้องและสัมพันธ์กันระหว่างการแสดงออกของโปรตีนและยีน และทำการยืนยันระดับการแสดงออกที่ผิดปกติไปของยีนด้วยเทคนิค Real-time qPCR และโปรตีนด้วยเทคนิค Western blot ในการศึกษาครั้งนี้จากผลการวิเคราะห์ข้อมูลลักษณะอาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมจากแบบทดสอบ ADI-R 123 ข้อ พบว่าวิธี K-means clustering ที่แบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อยนั้นให้ประสิทธิภาพในการแบ่งดีที่สุด และเมื่อทำการสร้างโมเดลในการจัดกลุ่มผู้ป่วยตามผลการแบ่งกลุ่มดังกล่าว พบว่าอัลกอริทึม Random forest ที่มีจำนวนต้นไม้ 10 ต้น โดยกระบวนการคัดเลือกข้อมูลแบบ Forward selection ให้ผลการทำนายถูกต้องมากที่สุด (100%) โดยใช้ข้อคำถาม ADI-R เพียง 5 ข้อ ในการแบ่งผู้ป่วยออกเป็นกลุ่มย่อย และจากการวิเคราะห์รูปแบบโปรตีโอมจากเซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์ของผู้ป่วยออทิซึมสเปกตรัมและออทิซึมสเปกตรัมกลุ่มย่อยด้วยเทคนิค 2D-GE ผู้วิจัยได้ทำการคัดเลือกจุดโปรตีนที่มีการแสดงออกผิดปกติอย่างมีนัยสำคัญและมีค่าสัดส่วนความเข้มแตกต่างกันอย่างน้อยสองเท่ามา 19 จุด เพื่อทำการระบุชนิดของโปรตีนด้วยเทคนิค LC-MS/MS โดยพบว่าจากโปรตีนทั้งหมด HNRNPA2B1 มีระดับการแสดงออกเพิ่มขึ้นในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่ไม่มีความผิดปกติด้านภาษารุนแรง โดยที่โปรตีน HNRNPA2B1 MDH2 DLD และ MSN มีระดับการแสดงออกลดลงในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่มีความผิดปกติด้านภาษารุนแรง ดังนั้นจากการศึกษาวิจัยในครั้งนี้ได้แสดงให้เห็นว่าการใช้เทคโนโลยีปัญญาประดิษฐ์ในการแบ่งผู้ป่วยออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามลักษณะอาการทางคลินิกนั้น สามารถลดความหลากหลายตามลักษณะอาการที่แสดงออกของโรคออทิซึมสเปกตรัมในการวิเคราะห์รูปแบบทรานสคริปโตมและโปรตีโอม รวมถึงการค้นหาโปรตีนที่จำเพาะต่อผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อย ได้อย่างมีประสิทธิภาพมากขึ้น ซึ่งโปรตีนเหล่านี้อาจใช้เป็นโปรตีนบ่งชี้ในการศึกษาเชิงลึกในประชากรผู้ป่วย เพื่อพัฒนาเป็นสารบ่งชีทางชีวภาพ (Biomarker) หรือศึกษากลไกพยาธิสภาพในระดับชีวโมเลกุลของโรคออทิซึมสเปกตรัมได้ต่อไปในอนาคต

Growth factors and hormones often mediate their biological effects through series of protein-protein interaction between cytoplasmic signaling molecules or transcription factors. Interfering of these protein-protein interactions has been shown to reduce or inhibit biological and growth responses to various growth factors and hormones. We previously demonstrated that progesterone receptor (PR) contains a polyproline domain (PPD) which directly interact to Src homology 3 (SH3) domain-containing molecules and expression of PR-PPD inhibits EGFR-mediated NSCLC cell proliferation. หนึ่งในสาเหตุสำคัญของการเกิดโรคความเสื่อมของระบบประสาทนั้นเกิดได้จากภาวะเครียดออกซิเดชันภายในเซลล์ประสาท โดยพบว่าสารสื่อประสาทชนิดกลูตาเมตเมื่อมีระดับสูงผิดปกติสามารถเหนี่ยวนำให้เกิดอนุมูลอิสระและภาวะเครียดออกซิเดชันนำไปสู่การสูญเสียและการตายของเซลล์ประสาทได้ โดยเฉพาะการตายของเซลล์จากการเกิดกระบวนการออโตฟาจีมากเกินไป ซึ่งเป็นกระบวนการที่มีบทบาทสำคัญในการควบคุมการตายของเซลล์ผ่านวิถีอื่นๆ รางจืด (Thunbergia laurifolia) เป็นพืชสมุนไพรไทยมีสรรพคุณในการถอนพิษ ต้านการอักเสบ และ ต้านอนุมูลอิสระได้ดี จุดประสงค์ในการวิจัยครั้งนี้ คือ ศึกษาคุณสมบัติการต้านอนุมูลอิสระของสารสกัดหยาบจากใบรางจืด และการปกป้องการตายของเซลล์ประสาทผ่านวิถีออโตฟาจี จากการเหนี่ยวนำภาวะเครียดออกซิเดชันด้วยกลูตาเมตโดยทดสอบในเซลล์ประสาทส่วนฮิปโปแคมปัสของหนูชนิด HT-22 ใบรางจืดถูกนำมาสกัดด้วยเอทานอล จากผลการทดสอบพบว่าสารสกัดหยาบจากใบรางจืดมีความสามารถในการต้านอนุมูลอิสระ และมีปริมาณสารประกอบ ฟลาโวนอยด์และฟีนอลิกสูง โดยจากการวิเคราะห์ด้วยเครื่อง LC-MS และเปรียบเทียบผลที่ได้กับฐานข้อมูล METLIN-(CA, USA) พบสารออกฤทธิ์สำคัญที่ยังไม่มีรายงานในสารสกัดเอทานอลของใบรางจืด ได้แก่ Apigenin 7-O-glucoside และ 7-Hydroxycoumarin ยิ่งไปกว่านั้นสารสกัดหยาบจากใบรางจืดยังสามารถลดการตายของเซลล์ประสาทจากการเหนี่ยวนำความเป็นพิษด้วยกลูตาเมต โดยสามารถยับยั้งการเกิดอนุมูลอิสระภายในเซลล์ และเพิ่มการแสดงออกของยีนต้านอนุมูลอิสระที่ทำหน้าที่สำคัญในกระบวนการต้านอนุมูลอิสระในร่างกาย ได้แก่ superoxide dismutase 1 และ 2 (SOD1 and SOD2) catalase (CAT) และ glutathione peroxidase (GPx) ซึ่งสัมพันธ์กับการลดการเกิดกระบวนการออโตฟาจีภายในเซลล์และเพิ่มปริมาณ ไมโตคอนเดรียโปรตีน โดยพบว่าสารสกัดหยาบจากใบรางจืดสามารถลดอัตราส่วนการแสดงออกของโปรตีน LC3B-II/LC3B-I และเพิ่มการแสดงออกของไมโตคอนเดรียโปรตีน TOM20 เมื่อทำการตรวจหาตำแหน่งที่จำเพาะต่อการแสดงออกของโปรตีนในกระบวนการออโตฟาจี (LC3B) และไมโตคอนเดรียด้วยกล้องจุลทรรศน์ชนิดคอนโฟคอล พบว่า สารสกัดหยาบจากใบรางจืดสามารถลดการเกิด LC3-puncta ซึ่งเป็นลักษณะเด่นของการเกิดกระบวนการออโตฟาจี นอกจากนี้สารสกัดหยาบจากใบรางจืดยังลดการ colocalization ของ LC3B และไมโตคอนเดรีย จากผลการศึกษานี้เราพบสารออกฤทธิ์สำคัญใหม่ที่ยังไม่มีรายงานมาก่อนของใบรางจืด โดยสารออกฤทธิ์นี้มีความสามารถในการปกป้องเซลล์ประสาทชนิด HT-22 จากภาวะเครียดออกซิเดชันและเป็นปัจจัยสำคัญทำให้สารสกัดจากใบรางจืดมีความสามารถลดการตายของเซลล์ประสาทผ่านวิถีออโตฟาจีได้ ซึ่งอาจสามารถนำไปพัฒนาเป็นสมุนไพรทางเลือกหรือยาเพื่อใช้ในการป้องกันหรือรักษาการเกิดโรคความเสื่อมของระบบประสาทได้ในอนาคต
