In induction experiments with naturally occurring and artificially lysogenic strains of S. potsdam, cells were irradiated in buffer, transferred to complete medium, and then assayed for free phage. Irradiation was followed by half hourly samples for free phage from the growing populations of irradiated cells. For an estimation of burst sizes, the irradiated cells were diluted out to a final concentration of 1 cell per ml and distributed to tubes in 0.5 ml quantities. All assays were done by the top-layer method with streptomycin incorporated in the top-layer and SP2-strr as the bacterial indicator. The non-lysogenic strain S. …

Biochemical Properties Of Salmonella Potsdam Bacteriophage P4/2, Clyde L. Roggenkamp

Over 90% recovery of phage P4/2 was realized with differential centrifugation concentration and purification, whereas only about 16% was recovered from the aqueous two-phase polymer - Genetron - ECTEOLA-cellulose column system. Purity of preparations was greatest with the density gradient centrifugation (A260 = 2.66 and A280 = 1.80 per 1012 plaque-forming units). With the chemical system of concentration and purification followed by an ECTEOLA anion exchange run, the A260 was 6.6 and the A280 was 4.3.

A series of experiments involving Listeria monocytogenes was undertaken to study first the fecal shedding of Listeria by conventional mice challenged intraperitoneally with Listeria and second the fecal shedding and ability to survive of germ free mice with similar challenge. In addition, mites infesting Listeria infected mice were cultured for their possible contamination by Listeria.
Conventional mice were challenged intraperitoneally with a single near LD50 dose of Listeria. Fecal Listeria were detected by collecting specimens from each mouse daily for the first 4 days and every second day thereafter. The feces was held at 7°C for 14 …

LUPINS AND SERRADELLA are not as extensively grown in Western Australia as some other legumes, but they are useful pasture species, particularly on poorer sandy soils.
Virgin soils may contain rhizobial bacteria capable of nodulating some members of the lupin group, but none of these bacteria have been found to nodulate serradella (Lange, 1961).
The use of inoculated seed is imperative except where lupins have been grown before.

By infection of the non-lysogenic strain of S. potsdam, SP2, with the phages P9a/2 and P9/c three artificially lysogenized strains were isolated: SP2-9a, SP2-9c, and SP2-9ca. SP2 lysogenized with P9a could not further be lysogenized with P9c.
There was evidence from the induction data that the presence of P9c influenced the induction of phage P9a, although P9a did not appear to influence the induction of P9c. Much greater levels of free phage P9a were achieved in induced cultures of SP2-9ca than in induced cultures of SP2-9a.
There were marked differences in induction of both phages P9a and P9c between …

Three lysogenic strains of Salmonella potsdam were studied to determine whether or not they were inducible by ultraviolet light under certain conditions. The strains examined were SP3, SP4, and SP9. SP3 and SP4 contained a single prophage each and SP9 contained two prophages P9a and P9c. The ultraviolet light sensitivities of each of these lysogenic strains and the indicator strain SP2 were also examined.
All three lysogenic strains were found to be inducible. SP3 was induced an average of 3.59% and SP4 an average of 2.21%. The difference between the induction percentages of SP3 and SP4 was not statistically significant. …

The fruiting myxobacteria and the cellular slime molds (Acrasiales) have many superficial resemblances with respect to fruiting behavior. Cells in the swarm are triggered by chemical substances to aggregate and form fruiting bodies (Bonner, 1947; Fluegel, 1963a). A chief difference between the two with respect to fruiting behavior, is that the myxoamoeba are free cells whereas the myxobacters are enmeshed in slime threads (Fluegel, 1963b). However, it is tacitly assumed that if parallel studies be undertaken in myxobacteria as has been done with the slime molds, the cells must be grown dispersed. ANGULAR leaf spot caused by the bactterium Pseudomonas lachrymans (E.F.Sm. & Bryan) Carsner, is common on cucumbers and gherkins grown late in the season.
The same disease has been recorded on rockmelons, watermelons and pumpkins but rarely occurs on these hosts in Western Australia.

There is concern in the cheese industry that residues from new chemical sterilisers used by dairy farmers could affect cheese making processes.
The sterilisers could also affect tests for antibiotic residues in milk. The responsibility of seeing that chemical sterilisers do not enter the milk belongs to the dairy farmers.
They should follow the manufacturers' recommendations and use the chemicals with care.

The development and description of a quantitative method of evaluating streptococcal antibodies by inhibition of fluorescent staining is reported. Suspensions of Streptococcus pyogenes group A, treated with methyl alcohol to reduce auto-fluorescence, were used as antigen. The degree of inhibition of uptake of fluorescent labeled antibody produced by the presence of antibodies in the immune test sera was measured after elution of the labeled antibody. Comparisons were made with controls using normal sera containing no inhibiting antibodies. A method of reporting the results in terms of titer is described.

Because pathogens such as streptococcus and staphylococcus have been transferred in many hospitals, the source of the infections is of concern to doctors, nurses, and other medical personnel. As shown by the review of literature, good techniques are necessary to stop the spread of disease.

Over three decades ago Rogers and Greenbank (1) published observations on intermittent growth in a bacterial culture. These interesting data are analyzed further in this report by computational procedures ( 2-8) designed for detecting and evaluating the significance of nearly periodic phenomena. By such methods, circadian (about 24-hour) periodicity in a culture of E. coli is unmasked as a significant component of the recorded changes. These results extend the already broad scope of circadian temporal organization in microbiology.

During the fall of 1959, monolayer behavior studies were made on two lakes in the southwest; Boulder Basin of Lake Mead, Nevada, and Lake Sahuaro near Phoenix, Arizona. These studies were made to evaluate the effect of geographical and climatological conditions on movement and behavior of monolayers. The Lake Sahuaro tests also were used to provide data for design and development of improved methods of application and maintenance of a film on this lake in anticipation of full scale, evaporation-reduction tests to be performed there during the summer of I960. EACH season, Bacterial Wilt causes heavy losses to individual growers in several of the potato districts throughout the South-West of the State.
The disease is incited by the bacterium Pseudomonas solanacearum E.F.Sm. and is most prevalent in the districts of Donnvbrook. Pemberton and Manjimup as well as in the area between Waroona and Dardanup.
It is a tuber-borne disease which is extremely difficult to eradicate, once it has been introduced on to a property.

Whether restaurant eating utensils serve as vectors in the transmission of respiratory and related diseases is still a matter for question, in spite of the overwhelming evidence in support of this contention.This is due to largely to the lack of epidemiological evidence, the tracing of an outbreak of a disease directly and unquestionably to contaminated eating utensils.The very nature of the vectors concerned obviates any possibility of obtaining this epidemiological evidence, since the contaminating agent exists as such only momentarily at the best. An attempt was made to determine the minimum concentration of the different peptones which are necessary to support the growth of a group of aerobic microorganisms. The medium for this experiment was semi-solid glucose peptone agar, with the peptone being varied in amount and kind.
- The growth supporting properties of the peptones under investigation varied a great deal.
- The minimum concentration of peptone necessary for the growth of Shigella dysenteriae, Proteus vulgaris, Streptococcus lactis, and Salmonella pullorum was determined.

Early studies on the bacterial content of milk were made mainly to satisfy the interest of people who wished to determine the various materials that contained bacteris. Soon the value of bacterial counts, as an indication of the general conditions of production, of handling, and of the keeping qualities of milk, became evident and bacterial counts were used to obtain information concerning these problems. Numbers of bacteria in milk have been used also in the studies of the desirable and undesirable changes in milk.

Variation in quality of plants of the same species has been known to exist for a long time. The influences bringing about such variation have been studied only comparatively recently. Quantity production has been given the greatest amount of attention as a rule, the beneficial results from the use of manures being judged by the quantity of produce. On August 29, 1912, I was requested by the Board of Health of the city of Lincoln to make an investigation to determine if possible the origin and cause of an outbreak of a disease reported as typhoid fever which was then prevailing. This outbreak was said to be most prevalent in one particular portion of the city and consequently attention was turned to that portion.
