The Role Of D-Gluconic Acid In The Regulation Of The Synthesis Of The Enzymes Of The Entner-Doudoroff Pathway In Pseudomonas Fluorescens, Steven C. Quay

Induction of Entner-Duodoroff pathway enzymes in Pseudomonas fluorescens was investigated to study the role of gluconate as a possible inducer. Glucose oxidase-deficient mutants were isolated and characterized. One of these mutants, gox-7, was deficient in particulate glucose oxidase; another mutant, gox-17, was deficient in particulate glucose and gluconate oxidase activities. Gluconate, but not glucose, induced synthesis of gluconokinase and 6-phosphogluconate dehydratase in both mutants. High constitute levels of 2-keto-3-deoxy-6-phosphogluconate aldolase were found when both mutants were grown on glucose. Growth of parent and both mutant strains on glycerol also resulted in high levels of Entner-Doudoroff pathway enzymes. It was concluded …

The nature of the action of penicillin on the cell walls of bacteria is reviewed. The composition of bacterial cell walls is compared to cell walls of blue-green algae.
The test organisms used were thermophilic: Anacystis nidulans grew optimally at 35⁰C, whereas Synechococcus lividus grew best at 45⁰ C. Growth was recorded by reading optical densities.
Cells of these two thermophilic blue-green algae were treated with varying dosages of penicillin and streptomycin. Penicillin inhibited growth of Anacystis nidulans and Synechococcus lividus in concentrations of 3.0 µg/ml and 0.03 µg/ml respectively. However, when 0.3 µg/ml of penicillin was added to Anacystis …

The problem with which this study was concerned was the development of criteria that could be used to determine when urinary catheter changes were necessary. By determining when encrustation occurred, it was felt that some complications could be eliminated by changing catheters before obstruction occurred due to encrustation.
It was the purpose of this study to: 1) explore some indications for catheter change, 2) determine if encrustation of urinary catheters necessitates catheter change, and 3) estimate potential criteria for a routine time for catheter changes.

Infiltration to an injury site or area of bacterial contamination by polymorphonuclear leucocytes (or neutrophils) is an early and essential phase of the early inflammatory reaction. Their action on foreign matter is the most critical step taken by body defense mechanisms in the first 24 hours. Anything which interferes with that action, phagocytosis, greatly hampers early inflammation. This study was designed to see the effect of elevated glucose levels on neutrophil phagocytosis in a unique strain of genetically diabetic mice.

Five new bacteriophages were isolated from lysogenic S. potsdam in this laboratory. Group B phages (P3 and P9a) were found to have unique properties, including their capability to infect strains of E. coli K-12 and E. coli C+/L. Passage of the phages through these hosts modified them (host-controlled modification) so that their ability to infect their original host, S. potsdam, was reduced and only 1-4 per million were able to form plaques.

A series of preliminary investigations was carried out to determine the factors promoting the dense, late summer waterblooms of Aphanizomenon flos-aguae in Hyrum Reservoir in northern Utah. Attempts were made to culture the Aphanizomenon in the ASM-8a medium of O'Flaherty and Phinney (J. Phycol. 6:95-97. 1970), but no growth was obtained and the algae soon lysed. Cultures were maintained, without growth, for more than four months in a lake water--lake sediment medium at 17 C under 1500 lux flourescent light.

The Salmonella potsdam group B bacteriophage P3, which grows in the rough variant strain of S. potsdam (SP2-R) and E. coli KCS11, KW117, and C+/L with similar efficiencies, was restricted by SP2-R after a passage though E. coli. The entrance of restricted E. coli-grown phage DNA into nonpermissive SP2-R cells was confirmed by the finding that after adsorption of restricted 32P-labeled P3-C1·C+/L to SP2-R and removal of the adsorbed phage heads from cells, 80.3% of the labeled DNA activity sedimented with the cells. The following members of the Rhizobiaceae were tested for their lytic response to various treatments: Agrobacterium tumefaciens, Chromobacterium violaceum, Rhizobium phaseoli, Rhizobium trifolii, Rhizobium lupini, and Rhizobium japonicum. Lysis was observed in spent growth media and as a function of saline concentration, pH, buffer composition, disodium (ethylenedinitrilo) tetraacetate (EDTA) concentration, and lysozyme.
Cells were harvested by centrifugation, washed, and resuspended in various treatment solutions. The lytic response was followed spectrophotometrically.
The different responses of the six cultures was applied to a summation of reclassification proposals evolved through modern taxonomic techniques such as DNA homology …
