Salicaceae are an economically and ecologically important family of flowering plants. The family includes willows and cottonwoods and was recently enlarged to include a large number of tropical species formerly placed in the family Flacourtiaceae. Relationships of these tropical relatives to willows and cottonwoods have been explored at a basic level using morphology and plastid DNA data, but to date no molecular phylogenies have been constructed with significant sampling of nuclear DNA, which sometimes results in a different picture of relationships because of its biparental inheritance. Modelling The Role Of Chance In Mating Dynamics, Libby K. Boykin

This undergraduate honors thesis examines the role of chance in mating dynamics, focusing on the popular measures of Is and OSR and whether or not they are accurate measures of sexual selection. The first chapter is an overview of sexual selection and how biologists typically measure it, and the second chapter describes the modelling scenarios we created to assess the interaction of Is and OSR with both chance and parental investment. Our results show that Is and OSR have indeed been used incorrectly, and that we should shift our focus to trait-based methods when measuring sexual selection.

Four hundred and twenty-seven amphibian species are critically endangered. The fungal disease Batrachochytrium dendrobatidis (Bd) has been correlated with amphibian decline and extinction. Numerous studies concerning the nature of Bd have been conducted; however, additional studies are needed to adequately evaluate Bd in southeastern Tennessee. As a result of previous studies, the following hypothesizes were proposed: watercourses in southeastern Tennessee will test positive for Bd, non-impacted streams will have a greater presence of Bd than impacted streams, and increased canopy coverage has a positive correlation with the presence of Bd. Fragmentation divides continuous habitat into smaller patches. Fragmentation can also produce smaller populations of species, because fragmentation can split a population into smaller groups. Both terrestrial and aquatic ecosystems suffer from fragmentation. There are a variety of fishes that live along marsh, each depending upon the marsh for protection, food, and sometimes even competition. Species richness of fishes can be altered due to marsh fragmentation. I predict islands with a larger patch index should have a higher species richness of fishes. Regulated secretion of pro-inflammatory molecules (e.g, histamines, proteases) from mast cells plays critical roles in immunity, allergic reactions, cardiovascular disease and cancer. These molecules are stored in secretory granules inside the cell and are rapidly released into the extracellular environment when mast cells are activated. It is known that mast cell degranulation depends upon membrane anchored SNAREs (soluble N-ethylmaleidimide-sensitive factor attachment protein receptors) and accessory proteins that form the trans-SNARE complex, a 4 helical bundle central to exocytic fusion. In this project, a DNA construct was designed and developed to remove the first fifty amino acids of the CsoSCA protein in the chemolithotrophic bacterium Halothiobacillus neapolitanus. The csoS3 gene codes for a carbonic anhydrase enzyme (CsoSCA) that is unique to a structure called a carboxysome. Carboxysomes are polyhedral microcompartments where carbon fixation is housed. The carbonic anhydrase is a shell-associated protein that improves the catalytic efficiency of ribulose-bisphosphate carboxylase/oxygenase (RuBisCO), the enzyme that catalyzes the fixation of carbon. Laterality is defined as the occurrence of each cerebral hemisphere having asymmetric control over the different sides of the body, leading to preferences to use one side of the body over the other for various behaviors. Many types of laterality exist, but handedness is the form that is most predominantly assessed. The purpose of this study was to examine the relationship between physical activity levels in college students and health related quality of life scores. Additional analysis was used to determine whether more of a relationship exists between female or male students’ in terms of activity versus quality of life.

The development of therapeutic immune responses from the manipulation of embryonic stem cells (ESCs) via induction of synthetic RNA with 5’ capped and 3’ poly(A)tailed ends would lead to development of stem cell therapy. A necessary step in attaining such a goal is to first produce an mRNA transcript from a plasmid containing the open reading frame (ORF) for a transcription factor for cellular activation.

Alzheimer’s Disease (AD) is a progressive neurodegenerative disorder characterized by extensive memory loss and cognitive deficits, which occur due to severe neuronal loss. Two hallmark lesions, intracellular neurofibrillary tangles composed of hyperphosphorylated tau and extracellular neuritic plaques formed by the aggregation of amyloid-β (Aβ), are responsible for the progressive neuronal loss seen in AD brains. Neurotoxic Aβ aggregates are also known to cause inflammation within the brain. There are many transcriptional regulators found in pathogenic Staphylococcus aureus. Many of these regulators are essential for the organism’s ability to switch from commensal form to the virulent pathogenic form. One of these main regulators is CodY. This regulator has been shown to be responsive to nutrient availability during phases of growth. Additionally, we have recently found that MsaB, the only protein coding ORF of the msaABCR operon, is a putative co-regulator of capsule along with CodY. Dengue Virus (DENV) is a flavivirus spread by mosquitoes that are endemic in tropic and subtropic climates. It is the causative pathogen of Dengue Fever, Dengue Hemorrhagic Fever, and Dengue Shock Syndrome. Besides mosquito transmission, DENV can also be transmitted through blood transfusion. Research regarding the effectiveness of these garments has shown an increase in variables such as increased blood flow, decreased post-exercise soreness, and increased joint position awareness. However, little research is available concerning their possible use to increase joint stability. I examined the presence and absence of the amphibian pathogen Batrachochytrium dendrobatidis in three survey areas in Eastern Tennessee. Amphibians were swabbed to assess for Bd and other ecological qualities like canopy cover, and air and water temperature are a few of the measurements evaluated. DNA was extracted from the swabs using a Qiagen DNeasy Blood and Tissue Kit and samples were PCRed in triplicate to confirm presence or absence. Enzymes can have many different functions within an organism. One such common function is methylation. The SABATH family of enzymes perform methylation of various organic acids. Many of the members of this family of enzymes have become specialized in the acid they methylate. For example: salicylic acid methyl transferase (SAMT) will primarily methylate salicylic acid, and benzoic acid methyl transferase (BAMT) will primarily methylate benzoic acid. Investigation of cancer-inducing molecules in cooked foods has led to the discovery of mutagenic heterocyclic amines (HCAs) in meat. The amino acids creatin(in)e and L-phenylalanine form the precursors for these HCAs. Recent research here at Andrews University have found similar mutagenic HCAs produced from plant-based proteins when L-arginine, rather than creatin(in)e, is substituted in high temperature reactions (simulated cooking) with L-phenylalanine. Our research focused on developing HPLC methods to isolate individual candidates that are then screened for mutagenicity via the Ames test. Mass spectroscopy methods were also developed to identfy the molecular structure of these lead mutagenic L-arginine-based HCAs.

Despite their lack of conjugation, polyamidoamine (PAMAM) dendrimers are intrinsically fluorescent, and this fluorescent is somewhat manipulable through altering the dendrimer's solvent. This level of fluorescence is dependent on a number of the dendrimer's qualities: the pH of the solvent, the mass percent concentration of the dendrimer in solvent, and the generation size of the dendrimer. In past studies, fluorescent intensity increased as generation size increased and pH decreased. Thus, this research project will attempt to replicate those results as well as further analyze the efficiency of fluorescence of PAMAM dendrimer in comparison to fluorescein.

West Indian manatees (Trichechus manatus latirostris) are tropical and subtropical aquatic mamals that can develop cold stress syndrome (CSS) when subjected to water temperatures below 20'C for prolonge dperiods. This study investigtes the potential impact that cold winter water temperatures in Florida may have on manatee body shape as a selective force to reduce surface-area-to-volume ratio (SA:Vol; i.e. Bergmann's and Allen's Rule). Morphometric measurements collected from state-mandated necropsies (1974-2014) were used to calculate surface area, volume, and mass. We found that manatees that died of CSS on average had significantly greater SA:Vol and less mass compared to those that died …

Mustard seed meal (MSM) has been shown to control weeds and pests in crop fields. Prior work showed that a sandy loam soil was superior to a silt loam soil in supporting velvetleaf growth suppression by MSM. To explore this difference, myrosinase enzyme activity was measured in both soils. Results showed no significant difference between the soils. Sand content in Michigan silt loam soil was manipulated in order to test the role of sand content on MSM efficacy in suppressing short-term velvetleaf growth. Results showed that sand enhanced the effect of MSM in suppressing seedling growth and germination.

Female crickets respond phonotactically to the calls of conspecific males. Females' phonotaxis has been reported to be variable, ranging from unselective to selectve in response to calls with varying syllable periods (30-90 ms). Octopamine, an intertebrate neurotransmitter, has been reported to increase aggressive behavior in crickets, (Stevanson et al. 2005) but the effects of octopamine on behaviors such as phonotaxis have not been investigated. The goal of this study is to determine the effects of octopamine on the syllable-period selective phonotactic response of females. Results suggest a decrease in phonotactic responsiveness shown by 5-10 day-okds after prothroacic nanoinjection of octopamine.

Past research has shown that alyl isothiocyanate (AICT) and its sister ITC compounds are effective alternatives to herbicides in reducing growth in weed seedlings. In this project the effects of mustard seed meal (MSM), a source of AITC, on plant-associated soil bacteria populations was assessed along with the effect of AITC on velvetleaf seed in vitro germination. Results show that MSM reduces bacterial levels in the soil. Velvetleaf seed germination was inbibited by micromolar levels of AITC. These results suggest that MSM has the potential to impact velvetleaf growth in the field as well as becoming a weed management tool.

Selective phonotaxis by female crickets has been shown to be variable. Mechanisms which underlie such behavioral variability are being studied. Juvenile Hormone III is a neuromodulator that has been shown to increase selectivity in phonotactic behavior of female crickets. In an attempt to further explore its effects on the behavioral responses of femaile crickets, experiments have been performed with chelerythrine cloride that is known to have an opposite effect of Juvenile Hormone III. Post-traumatic stress disorder (PTSD) is a psychological disorder with limited treatment options. Molecular mechanisms that cause PTSD remain unclear due to a lack of viable animal models. Considering the behavioral similarities between zebrafish (Danio rerio) and humans, their social nature, and functional parallels between select brain regions, we hypothesized that zebrafish can demonstrate PTSD and that antioxidant treatment will prevent PTSD induction. To test this hypothesis, adult zebrafish were exposed to a traumatic event then were tested immediately and three weeks after the event.
