Chronic opioid therapy is widely used for managing persistent pain, yet its long-term efficacy is questionable and often associated with adverse outcomes such as opioid misuse and dependence. This study investigated whether central sensitization (CS), a condition of heightened neural response to pain stimuli, better predicts opioid-related outcomes compared to self-reported pain severity. Among adults with chronic pain currently using long-term opioid therapy, participants completed measures assessing CS, pain severity, and opioid-related outcomes including readiness and confidence to change opioid use, misuse, and dependence. BACKGROUND: Both bleeding and adverse ischemic events increase with age, compounding the benefit-risk balance of anticoagulants in older patients. We present analyses using benefit-risk methods to better understand the age-dependence of the benefit-risk profile of rivaroxaban in patients with nonvalvular atrial fibrillation (NVAF) or venous thromboembolism (VTE).
METHODS: Randomized controlled trial data from the ROCKET-AF (NVAF) and EINSTEIN DVT, EINSTEIN PE, EINSTEIN-Extension, and EINSTEIN CHOICE in (VTE) were used. Schistosomiasis, a parasitic disease caused by flatworms of genus Schistosoma, is a neglected tropical disease that causes significant morbidity in the developing world. Despite numerous efforts to eradicate the disease, the parasite remains a significant global burden, particularly within Sub-Saharan Africa. Schistosoma species possess an arsenal of potent mechanisms to defend against direct attack from host immune cells and a remarkable ability to modulate the host immune system through proximal and systemic modes that facilitate its stage-specific development, survival, and reproduction. As artificial intelligence (AI) transforms drug development, regulatory frameworks are evolving to oversee its implementation, particularly at the US Food and Drug Administration (FDA) and the European Medicines Agency (EMA). This paper makes three contributions to understanding emerging regulatory approaches. First, we offer a comparative analysis of how these agencies have responded to AI-driven advances, incorporating new US executive orders and the European Union (EU)’s AI Act. Second, we propose a novel analytical framework to understand regulatory divergence: the FDA’s flexible, dialog-driven model contrasts with the EMA’s structured, risk-tiered approach, reflecting broader institutional and political-economic differences. Background Artificial intelligence (AI) is transforming healthcare, including pharmacy practice, by enhancing access to information, reducing errors, and supporting decision-making. However, its integration into pharmacy education remains underexplored. This study investigated pharmacy students' perceptions of AI's impact on their profession and the need for curriculum reform. Materials and Methods A cross-sectional survey was conducted among 245 Bachelor of Pharmacy (BPharm) students from Jamia Hamdard, India. Data were collected using a structured questionnaire covering knowledge, perceptions, and opinions on AI in pharmacy. Heart failure (HF) affects 6 million people in the US, and type 2 diabetes (T2D) is a significant risk factor for HF. Since 2008, regulatory agencies have required cardiovascular safety trials for new T2D therapies, and these have consistently demonstrated the cardiovascular benefits of glucose-lowering drugs. Sodium-glucose cotransporter inhibitors (SGLTi) and glucagon-like peptide-1 (GLP-1) receptor agonists (RAs)/glucose-dependent insulinotropic polypeptide (GIP) reduce major adverse cardiovascular events and deaths, regardless of diabetes status, and improve cardiorespiratory fitness. SGLTis, such as dapagliflozin and empagliflozin, benefit patients with reduced and preserved ejection fraction, significantly reducing HF hospitalizations and cardiovascular deaths. TU100 (14-methyl-5H-5,12-epiminobenzo[4,5]cyclohepta[1,2-b]naphthalene-6,11,13(12H)-trione) exhibits exceptional chemotherapeutic properties. It operates via a distinct mechanism that triggers cell death to create cytotoxic effects while concurrently inhibiting topoisomerase I and II. Its efficacy rivals that of well-known chemotherapeutic drugs like Daunorubicin. The distinctive characteristics of TU100 have led to initiatives aimed at creating structurally similar compounds to discover even more effective bioactive alternatives.

Understanding plasma-liquid interactions and the impact of the electrical conductivity (σ) of liquid on plasma is important for plasma applications in water treatment and medicine. Ultrapure water, phosphate-buffered saline (PBS) solution, biological media, and sodium chloride (NaCl) solutions at various concentrations were used in this study to provide a broad range of conductivity values ranging from 0.055 μS cm−1 to 222 mS cm−1. Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has caused a global health crisis, marked by high transmissibility and virulence. Despite widespread vaccination efforts, effective treatments for COVID-19, particularly for severe cases leading to Acute Respiratory Distress Syndrome (ARDS), remain limited. This study investigates the efficacy of KVX-053, a protein tyrosine phosphatase type IVA (PTP4A3) small molecule inhibitor, in modulating SARS-CoV-2-induced inflammation and lung injury using in vitro cell models and in vivo K18-hACE2 transgenic mice. KVX-053 reduced in vitro pro-inflammatory cytokine expression, including TNFα, CXCL10, and CXCL11, without impacting viral replication or cell viability. Nearly 67% of pediatric patients treated for bronchiolitis received steroids with no underlying indication, which deviates from clinical guidelines. Patients who received steroids without an indication had an apparent trend toward more oxygen support and had an increase in maximum flow rate compared to patients with an indication for steroid use.
