CXCR4 is a chemokine receptor that is overexpressed in multiple disease states, including cancer. Understanding the mechanisms by which cells regulate CXCR4 expression is of high importance, as they can reveal downstream effectors that can potentially be targeted pharmacologically to more effectively treat diseases with fewer side effects.
CXCR4 is internalized in response to stimulation by its ligand CXCL12, and localizes to early endosomes as part of a homologous desensitization mechanism. Heparin, a highly sulfated glycosaminoglycan (GAG), is used extensively as an anticoagulant. It consists of repeating disaccharide units, containing iduronic acid (or glucuronic acid) and glucosamine, exhibiting variable degrees of sulfation. Heparin, and its analogues, are used during surgery and dialysis, and are often used to coat indwelling catheters and other devices where the vascular system is exposed. Administered parenterally, often continuously due to its short half-life, over 0.5 billion doses are required per year. Isoquinoline alkaloids, a plant secondary metabolites present in many plant species including lotus (Nelumbo nucifera Gaertn.). Lotus leaf contains high amount of bioactive nuciferine and N-nornuciferine. A set of enzyme including NCS, 6OMT and CNMT and WRKY transcription factors (WRKY TFs) has been reported to have corresponding function in the biosynthesis isoquinoline alkaloids. Thus, we performed quantitative analysis on expression level of these corresponing genes and on nuciferine and N-nornuciferine content in lotus leaf using mechanical wounding method. The objectives of this observational descriptive cross-sectional study were to determine the prevalences of depression and metabolic syndrome and to examine the relationship between depression and metabolic syndrome and dietary pattern in working age people in Bangkok. The samples were 446 participants (123 males and 323 females) aged 20 and over who received an annual health examination 2017. The metabolic syndrome was defined by NCEP ATP III criteria. Autoxidation of polyethylene glycol (PEG) 600, which is a hydrophilic liquid commonly filled in soft gelatin capsules, results in formaldehyde which is an oxidizing agent. The objectives of the present study were to study the effect of formaldehyde on fading of brilliant blue dye and crosslinking in the soft gelatin capsules; also to investigate possibility of inhibition of PEG 600 autoxidation by a selected antioxidant. The main objective of this study was to focus on the preparation of X-ray amorphous salbutamol sulfate (SS) with mesoporous silica (S244 silica) carrier by using incipient wetness impregnation approach and further investigated on the utilization of poly (vinylpyrrolidone) (PVP) K12 for the stabilization of amorphous produced. Initially, SS was prepared in solution state and later loaded on S244 silica. Two hundred and thirty-eight strains of lactic acid bacteria (LAB) isolated from several plant-materials and fermented food products collected in Thailand were screening for antibacterial activity. Fifty-three strains which exhibited antimicrobial activity against Helicobacter pylori clinical strains, were selected and identified based on the phenotypic, chemotaxonomic characteristics and 16S rRNA gene sequence analysis. A biocellulose wound dressing composed of silk sericin as an accelerative wound healing component and polyhexamethylene biguanide (PHMB) as an antimicrobial agent which showed effective and safe wound treatment. A prospective, single-blinded, randomized controlled matched-pair study was designed to evaluate the safety and the clinical efficacy of the biocellulose wound dressing containing silk sericin and PHMB compared with Bactigras® in healthy volunteers (phase I clinical study) and in split-thickness skin graft (STSG) donor site wound treatment (phase II clinical study). Grammatophyllum speciosum Blume (Orchidaceae) is a large-sized orchid in Southeast Asia. The aim of this study was to evaluate G. speciosum pseudobulb extract for anti-aging activities, both in vitro and in vivo. In vitro anti-aging activity was performed using non-cell and cell-based assays. In vitro study, G. speciosum pseudobulb extract exhibited a high content of gastrodin, which was used as an active analytical marker. Phytochemical study of Dendrobium palpebrae Lindl. (Orchidaceae) afforded ten pure compounds. One of them was characterized as a new phenanthrene-phenanthraquinone derivative named dendropalpebrone, and nine known compounds were identified as gigantol, lusianthridin, nobilone, 3,7-dihydroxy-2,4,8-trimethoxyphenanthrene, 2,5-dihydroxy-4,9-dimethoxyphenanthrene, moscatilin, scoparone, 4,5,4'-trihydroxy-3,3'-dimethoxybibenzyl and dendroflorin. All of the isolated compounds were initially screened for their free radical scavenging activity via oxygen radical absorbance capacity assay, DPPH radical scavenging assay, deoxyribose degradation assay, and assay for intracellular antioxidant activity in H2O2-induced RAW 264.7 murine macrophage cells. Phytochemical study of the methanol extract from Dendrobium infundibulum (Orchidaceae) led to isolation of nine pure compounds including two new compounds (dendroinfundin A and dendroinfundin B) and seven known compounds (ephemeranthol A, moscatilin, aloifol I, batatasin III, 3,3'-dihydroxy-4,5-dimethoxybibenzyl, 3,4'-dihydroxy-3',4,5-trimethoxybibenzyl and dendrosinen B). Their structures were determined from their spectroscopic data. All compounds were then examined for their lipase and alpha-glucosidase inhibitory activities. Dendrosinen B (IC50 = 295.0±37.9 µM) showed moderate inhibitory activity against lipase when compared with orlistat (IC50 = 31.4±0.6 nM). In this study, the EtOAc extract of Dendrobium parishii Rchb. f. was separated to obtain two new compounds including a bibenzyl derivative, 4,3′,4′-trihydroxy-3,5-dimethoxybibenzyl, and a bibenzyl-dihydrophenanthrene derivative, (−)-dendroparishiol, and five known compounds including flavanthrinin, moscatilin, 4,5,4′-trihydroxy-3,3′-dimethoxybibenzyl, dendrocandin E and asiatic acid. The structures of all of the isolated compounds were determined by analysis of spectroscopic data (NMR and HR-ESI-MS). They were then evaluated for antioxidant activities using DPPH free radical scavenging activity, deoxyribose degradation and oxygen radical absorbance capacity assays. Chemical investigation of the leaves and stems of Pseuduvaria fragrans resulted in the isolation of seven pure compounds, including a new benzophenone C-glycoside named pseuduvarioside and six known compounds comprising (-)-guaiol, (+)-isocorydine, cyathocaline, isoursuline, N-trans-feruloyltyramine and N-trans-coumaroyltyramine. The structure determination of the isolated compounds was carried out by analysis of their UV, HRS-ESI-MS and NMR data in comparison with previously reported values. All the isolates were evaluated for their α-glucosidase inhibitory activity. Only N-trans-feruloyltyramine and N-trans-coumaroyltyramine exhibited strong activity with IC50 values of 3.58 µM and 0.58 µM, respectively, as compared with the drug acarbose (IC50 985.6 µM).
