Binding of sweet, umami, and bitter tastants to G protein-coupled receptors (GPCRs) in apical membranes of type II taste bud cells (TBCs) triggers action potentials that activate a voltage-gated nonselective ion channel to release ATP to gustatory nerves mediating taste perception. Although calcium homeostasis modulator 1 (CALHM1) is necessary for ATP release, the molecular identification of the channel complex that provides the conductive ATP-release mechanism suitable for action potential-dependent neurotransmission remains to be determined. Here we show that CALHM3 interacts with CALHM1 as a pore-forming subunit in a CALHM1/CALHM3 hexameric channel, endowing it with fast voltage-activated gating identical to that …

Fibromodulin Reduces Scar Size And Increases Scar Tensile Strength In Normal And Excessive-Mechanical-Loading Porcine Cutaneous Wounds, Wenlu Jiang, Kang Ting, Soonchul Lee, Janette N. Zara, Richard Song, Chenshuang Li, Eric Chen, Xinli Zhang, Zhihe Zhao, Chia Soo, Zhong Zheng

Departmental Papers (Dental)

Hypertrophic scarring is a major postoperative complication which leads to severe disfigurement and dysfunction in patients and usually requires multiple surgical revisions due to its high recurrence rates. Excessive-mechanical-loading across wounds is an important initiator of hypertrophic scarring formation. In this study, we demonstrate that intradermal administration of a single extracellular matrix (ECM) molecule—fibromodulin (FMOD) protein—can significantly reduce scar size, increase tensile strength, and improve dermal collagen architecture organization in the normal and even excessive-mechanical-loading red Duroc pig wound models. Since pig skin is recognized by the Food and Drug Administration as the closest animal equivalent to human skin, and …

Departmental Papers (Dental)

The pro-chondrogenic function of runt-related transcription factor 2 (Runx2) was previously considered to be dependent on direct binding with the promoter of Indian hedgehog (Ihh)—the major regulator of chondrocyte differentiation, proliferation, and maturation. The authors’ previous studies identified neural EGFL like 1 (Nell-1) as a Runx2-responsive growth factor for chondrogenic differentiation and maturation. In this study, it was further revealed that the pro-chondrogenic activities of Nell-1 also rely on Ihh signaling, by showing: i) Nell-1 significantly elevated Ihh signal transduction; ii) Nell-1 deficiency markedly reduced Ihh activation in chondrocytes; and iii) Nell-1–stimulated chondrogenesis was significantly reduced by the specific hedgehog …

Consideration For Care For Your Patient With Cerebral Palsy, Katlin R. Cannon, Melissa Stowe, Brooke Ryan 2018 Virginia Commonwealth University

Consideration For Care For Your Patient With Cerebral Palsy, Katlin R. Cannon, Melissa Stowe, Brooke Ryan

Dental Hygiene Student Scholarship

Cerebral palsy (CP) is a common developmental neurological disorder affecting about 2-3 children out of 1,000. CP is the result of infant brain damage or abnormal development resulting in impaired muscle control, coordination, tone, reflex, posture, and balance. These patients are unable to control motor movements of their muscles of mastication and facial expression, causing excessive drooling, clenching, bruxism, and other oral health-related issues. This lack of motor control affects their ability to swallow and often limits these patients to a liquid diet. This can lead to vitamin deficiencies and result in further developmental problems. As an example, a deficiency …

Cpap-Mad Combination Therapy For Cpap Intolerant Patients With Moderate To Severe Osa, Jacob Stadiem 2017 University of Nebraska Medical Center

Cpap-Mad Combination Therapy For Cpap Intolerant Patients With Moderate To Severe Osa, Jacob Stadiem

Theses & Dissertations

The treatment of obstructive sleep apnea (OSA) allows for a multidisciplinary approach between dentistry and medicine. Continuous positive airway pressure (CPAP) is the gold standard for OSA treatment, but the non-adherence rates are high. The mandibular advancement device (MAD) has a higher acceptance rate than the CPAP, however the efficacy is much lower in cases of moderate to severe OSA. This thesis attempted to evaluate if there was a change in compliance when a MAD was added as a combination therapy to moderate to severe OSA subjects that were currently non-compliant with their CPAP. In addition, the change in air …

Departmental Papers (Dental)

Sex hormones may contribute to the symptomatology of female-predominant temporomandibular disorders (TMDs) inflammatory pain. Pregnant women show less symptoms of TMDs than that of non-pregnant women. Whether progesterone (P4), one of the dominant sex hormones that regulates multiple biological functions, is involved in symptoms of TMDs remains to be explored. Freund's complete adjuvant were used to induce joint inflammation. We evaluated the behavior-related and histologic effects of P4 and the expression of tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6 in the synovial membrane. Primary TMJ synoviocytes were treated with TNF-α or IL-1β with the combination of P4. Progesterone receptor …

Foxo1 Deletion In Keratinocytes Improves Diabetic Wound Healing Through Mmp9 Regulation, Chenying Zhang, Jason Lim, Hyeran H. Jeon, Fanxing Xu, Chen Tian, Fang Miao, Alhassan Hameedaldeen, Dana T. Graves 2017 University of Pennsylvania

Foxo1 Deletion In Keratinocytes Improves Diabetic Wound Healing Through Mmp9 Regulation, Chenying Zhang, Jason Lim, Hyeran H. Jeon, Fanxing Xu, Chen Tian, Fang Miao, Alhassan Hameedaldeen, Dana T. Graves

Departmental Papers (Dental)

Keratinocyte migration is a key aspect of re-epithelialization during wound healing. Matric metalloproteinase 9 (MMP9) contributes to this process and deificiencies in the MMP9 lead to impaired healing. Inappropriate expression of MMP9 also contributes to impaired re-epithelialization. Previously we demonstrated that FOXO1 was activated in wound healing but to higher levels in diabetic wounds. To address mechanisms of impaired re-epithelialization we examined MMP0 expression in vivo in full thickness dermal scalp wounds creared in experimental K14.Cre+.Foxo1L/L mice with lineage-specific Cre recombinase deletion of floxed FOXO1 and compared the results to control littermates. MMP9 was induced during …

Effective And Ineffective Clinical Teaching And Instruction In Dental Hygiene School, Vicki Gianopoulos Pizanis 2017 University of New Mexico

Effective And Ineffective Clinical Teaching And Instruction In Dental Hygiene School, Vicki Gianopoulos Pizanis

Teacher Education, Educational Leadership & Policy ETDs

Introduction

Educators who teach in a clinical setting, primarily in healthcare fields, while having knowledge of the subject, having skills in what is taught, and having experience in the field, often have little-to-no training in teaching. This results in instructional practices being based upon intuition and experience. Becoming aware of how qualities and characteristics of clinical instructors affect adult learners’ educational experience through evidence-based research, and using its implications, would help promote more effective instruction and ultimately improve student learning.

Methodology

A mixed methods study using Grounded Theory and Critical Incidence technique was conducted to study junior student, senior student …

Orofacial Pain Overview: Getting Rid Of The Riddles, Andrew L. Young 2017 University of the Pacific

Orofacial Pain Overview: Getting Rid Of The Riddles, Andrew L. Young

Andrew L. Young

While most pains that find their way to the dental office are dental in origin, some have less familiar roots: muscular, joint, neurological, headache, sinus or psychological. All of these roots can at times masquerade as a toothache. The purpose of this article is to alert the dentist to the select features of each root that distinguish it from the rest, resulting in efficient and appropriate treatment.
