Impaired Dnajb2 Response To Heat Shock In Fibroblasts From A Neuropathy Patient With Dnajb2/Hsj1 Mutation: Cystamine As A Potential Therapeutic Intervention, Raj K. Pradhan, Nikolas G. Kinney, Brigid K. Jensen, Hristelina Ilieva

Farber Institute for Neuroscience Faculty Papers

Background and Objectives: Neuropathy is a debilitating disorder characterized by peripheral nerve dysfunction and damage to sensory, motor, and autonomic neurons and their axons. While homozygous mutations in DNAJB2/HSJ1 have been linked to early-onset neuropathy, a heterozygous DNAJB2 c.823+6C>T was discovered in an adult patient with severe sensory–motor polyneuropathy. This mutation is predicted to affect both isoforms of the protein. DNAJB2 (HSP40), a key member of the heat shock protein family, plays a critical role in cellular protection and stress, including response to heat shock. DNAJB2 traffics unfolded proteins to another heat shock protein, HSP70, and activates its ATPase …

Intervertebral disc degeneration is a major risk factor contributing to chronic low back and neck pain. While the etiological factors for disc degeneration vary, age is still one of the most important risk factors. Recent studies have shown the promising role of SIRT6 in mammalian aging and skeletal tissue health, however its role in the intervertebral disc health remains unexplored. We investigated the contribution of SIRT6 to disc health by studying the age-dependent spinal phenotype of mice with conditional deletion of Sirt6 in the disc (AcanCreERT2; Sirt6fl/fl). Histological studies showed a degenerative phenotype in knockout mice …

Amyloid beta (Aβ42 and Aβ40) aggregation, along with neurofibrillary tangles, is one of the major neurotoxic events responsible for the onset of Alzheimer's disease. Many potent peptide-based inhibitors mainly focusing on central hydrophobic core Aβ16-20 (KLVFF) have been reported in recent years. Herein, we report pentapeptides -, based on the β-turn-inducing fragment Aβ19-23 (FFAED). The synthesis of peptides - was carried out using Fmoc/tBu-based solid-phase peptide synthesis technique, and it was found that pentapeptide potently inhibit the aggregation propensity of Aβ42, when incubated with it at 37 °C for 48 h. The aggregation inhibition study was conducted using thioflavin T-based …

Theoretical And Experimental Synthesis Of Β-Fluorinated Morphine Derivative For Selective Binding In Inflamed Tissue, Thu Doan, Amelia Bayha, Adel Martinez, Nayiri Alexander

Student Scholar Symposium Abstracts and Posters

Over the past two decades, the United States has experienced a substantial rise in opioid overdoses and related fatalities. Morphine is a frequent prescription for pain management following traumatic procedures and contributes to potential tolerance and dependency with prolonged use. By reconstructing the structure of morphine, adverse side effects may be eliminated. Previous studies have identified several morphine derivatives that, in theory, could result in fewer side effects. This was determined by a decreased pKₐ of the biochemically active amine group to promote the selective binding in peripheral opioid receptors within inflamed tissue (pH=6-6.5). This activation provides analgesia while central …

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

The mitochondrial phosphate carrier (mPiC), encoded by the nuclear gene SLC25A3, is synthesized with an N-terminus mitochondrial targeting sequence (MTS), enabling its import into the mitochondria. mPiC imports inorganic phosphate (Pi) into the mitochondrial matrix for ATP production and other matrix phosphorylation reactions, as well as regulates mitochondrial Ca2+ uptake and buffering of matrix Ca2+. PiC also imports copper (Cu), crucial to COX subunit holoenzyme assembly. Variants in SLC25A3 exist and lead to mPiC deficiency (MPCD), cause a rare autosomal recessive disease with no current cure; patients with MPCD usually die within the first …

Primary Prevention Of Nicotine Use In The Pediatric Population, Katie N. Ashton

Honors Capstones

This project explores how to prevent nicotine use, particularly e-cigarette use, among adolescents. Nicotine use has been on the rise since e-cigarette devices have been created. This makes it convenient to use nicotine products rather than going outside to light a cigarette. Nicotine use in adolescents is increasing as it is more convenient to use, and many may not understand the adverse health effects of using these devices. Any nicotine use can cause brain, heart, and lung damage. This project explores the factors that encourage adolescents to use nicotine such as the variety of flavors that e-cigarettes are made of …

Investigating The Ability Of The Norepinephrine-Dopamine Reuptake Inhibitor Nomifensine To Reverse The Effort-Related Effects Of Tetrabenazine In Female Rats, Vivien Daytree

Honors Scholar Theses

Anergia, lassitude, and psychomotor retardation are core symptoms of motivational dysfunction that often accompany Major Depressive Disorder (MDD). While selective serotonin reuptake inhibitors effectively alleviate mood and anxiety symptoms, they remain relatively ineffective at alleviating motivational deficits. Compounds that elevate extracellular dopamine (DA) have shown promise in addressing these symptoms. Bupropion, a norepinephrine-dopamine reuptake inhibitor (NDRI), increases DA and is effective at attenuating motivational symptoms in patients. Another NDRI, nomifensine, has demonstrated potential antidepressant properties in preclinical research. Effort-based choice tasks, such as the FR5/chow choice task, serve as animal models for studying motivational dysfunction. Rats choose between exerting effort …

Computationally Assessing The Specificity Of Receptor Tyrosine Kinase Inhibitors, Htoo Say 2025 University of Nebraska-Omaha

Computationally Assessing The Specificity Of Receptor Tyrosine Kinase Inhibitors, Htoo Say

Theses/Capstones/Creative Projects

Receptor tyrosine kinases (RTKs) are vital to cell signaling processes such as growth, survival, and differentiation. Dysregulation through mutation or overexpression often contributes to cancer, making RTKs key therapeutic targets for tyrosine kinase inhibitors (TKIs). However, the low specificity of many TKIs leads to off-target effects. This project uses molecular docking to evaluate the binding affinities and selectivity of FDA-approved TKIs across 18 RTK subtypes. The dockings conducted revealed significant variability in binding affinities, with Sorafenib exhibiting strong specificity for EphB2 (-7.6 kcal/mol) and poor compatibility with EphA7 and ErbB4 (-3.0 kcal/mol). These findings underscore the role of specific molecular …

Ivak, Daniel Chandler, John Tkaczynski, Thomas M. Keck, Comfort A. Boateng, Eleni Papadopoulos, Barry D. Waterhouse, Rachel L. Navarra

Rowan-Virtua Research Day

The consequences of modulating subtypes of G protein coupled dopamine receptors are broadly studied for their role in decision making behavior, specifically in situations involving uncertain risk and reward. The actions of dopamine D4 receptors (D4Rs) on these complex processes are the least understood, particularly due to a lack of available compounds with suitable selectivity for D4Rs. Further, current research often fails to evaluate how modulating specific dopamine receptor subtypes may affect locomotor activity and anxiety-like behavior as confounding factors prior to assessing their function in behavioral assays of higher order decision making. The goal of the current study was …

Investigating Fatty Acid Influence On West Nile Virus In Mosquitoes And Birds, Hayley Moree

Honors Program: Senior Projects (Public)

Vector-borne diseases, including Zika virus (ZIKV), Dengue virus (DENV), and West Nile virus (WNV), pose considerable global health threats, accounting for over 400 million infections and more than 700,000 deaths annually. Many vector-borne diseases are within the Flaviviridae family. Despite this, effective treatments have not been found for some flaviviruses. Flaviviruses exploit host cellular machinery to facilitate their replication and survival, reprogramming metabolic pathways, including lipid metabolism. Lipids play essential roles in the viral life cycle, providing structural components for the viral envelope, enabling membrane rearrangements for replication complexes, and supplying energy resources necessary for particle assembly.

Targeting lipid metabolism …

Hydralazine is a vasodilator commonly used to treat high blood pressure. Even with the safety profile of Hydralazine, it is necessary to know that it may lead to Hydralazine-Induced Vasculitis. Here, we discuss how this rare side effect can present in an 80-year-old female with a past medical history of hypertension who presented to the emergency department with complaints of progressive shortness of breath on exertion and while lying supine. Further evaluation revealed a large right-sided pleural effusion with consolidation at the right lower lobe. The patient’s dyspnea worsened, and the surgical team placed a chest tube. The patient also …

Symptomatic Treatment Of Cannabis Withdrawal Syndrome: A Review Of The Existing Literature, Aria Sen, Dale Shober 2025 Rowan University

Symptomatic Treatment Of Cannabis Withdrawal Syndrome: A Review Of The Existing Literature, Aria Sen, Dale Shober

Rowan-Virtua Research Day

Despite its public reputation as a recreational substance that does not cause physical withdrawal symptoms, cannabis withdrawal syndrome (CWS) was added to the DSM-5 in 2013. Existing research would suggest that almost half of regular cannabis users will experience symptoms such as mood symptoms, insomnia, and gastrointestinal distress. For more than 20 years, the rates of cannabis use have continued to trend upward due to expanding legislation on the recreational use of marijuana. Currently, there are no FDA approved treatments for CWS. Considering this trend, the lack of a well-formed treatment protocol for CWS will have a growing impact on …
