In this work, we have made and characterized a pair of immunobiosensors for detecting the green fluorescent protein (GFP) in an aqueous matrix. An anti-GFP antibody-based biosensor was assembled to detect GFP, while a novel peptoid (N-substituted oligomers of glycine designated as IOS-1) biosensor was also assembled for GFP detection. A quartz crystal microbalance (QCM) gold sensor was used as the supporting substrate for self-assembly of the immunobiosensors. Gravimetric measurements of the QCM gold sensor during immunobiosensor construction and operation were available in real-time using a QCM instrument. As antibiotic resistance emerges stronger than ever, novel antimicrobial compounds are necessary to continue the infectious fight against disease and prevent millions of deaths by multidrug-resistant bacteria. One compound thought to be a natural source of antimicrobial activity is the chalcone. Abundant in fruits and vegetables, chalcones are versatile compounds that are easily synthesized via Claisen-Schmidt condensation and have demonstrated a wide-range of biological activity. Incidence of Clostridium difficile infection, (CDI) in patients receiving antibiotics is significant. In 2017, there were approximately 223,900 cases of CDI in hospitalized patients alone (Center for Disease Control [CDC], 2017). Despite enhanced infection control measures, CDI rates remain prevalent and are associated with increased costs to healthcare. An integrative literature review and synthesis was performed. Studies were analyzed to delineate if Lactobacillus containing probiotics, when administered concurrently with high risk antibiotics, decrease CDI rates for patients in the inpatient hospital setting. Death by drug poisoning is the leading cause of death due to injury in the state of Utah (UDOH, 2014). From 2013-2015, Utah ranked seventh in the nation for drug overdose deaths, the majority of which were opioid-related (CDC, 2018a). In 2017, the age-adjusted mortality rate from drug overdose in Utah was 15.5 per 100,000, putting the state above the national average of 14.9 per 100,000 (CDC, 2018b). Opioid overdose death rates in most of rural Utah far exceeds rates across the urban counties of the state. The higher rates of deaths in rural Utah is indeed concerning. Evidence indicates …

Selinexor is currently being used in several clinical trials for cancer and has shown anti-cancer effects in combination with other chemotherapies. Clinical trials evaluating the use of selinexor in the treatment of triple negative breast cancer are limited, but in vitro studies show promising results in its ability to kill triple negative breast cancer cells. The mechanism by which this occurs, however, is not understood. The goal of this research was to determine the mechanism by which selinexor kills BT- 549 triple negative breast cancer cells.

This study used gene expression analysis to explore the specific response to amodiaquine in HCT116 colorectal cancer cells. Real-time PCR revealed that 20 human stress and toxicity genes were upregulated by at least 3 fold in HCT116 colorectal cancer cells treated with AMQ. Of these 20 upregulated genes, 9 were found to be pro-apoptotic and 2 were DNA damage repair genes, which supports our original hypothesis that amodiaquine is inducing a stress response in these cells. Rhabdomyolysis is the rapid breakdown of myocytes in skeletal muscle. This condition can be triggered by a variety of things including crush injury, drugs, prolonged exercise, and venom. Although the causes can vary, the general pathophysiology of rhabdomyolysis is similar. Increased intracellular calcium ion levels instigate the breakdown of cellular and certain organelle membranes of muscle cells. Reactive oxygen species form, ATP is depleted, and the sarcoplasmic reticulum, which regulates muscle contraction, is also damaged. Necrosis ensues, leading to toxic intracellular components spreading throughout the body. Rhabdomyolysis can then lead to several complications including acute renal injury and heart arrhythmias, …

After genetic therapy failed because a suitable delivery system could not be found, researchers began searching for small organic molecules that could act as chaperones for CFTR. These molecules, known as modulators, allowed CFTR to be assembled correctly and function similarly to wild type CFTR. The aminoacyl-tRNA synthetases are an essential and universally distributed family of enzymes that plays a critical role in protein synthesis, pairing tRNAs with their cognate amino acids for decoding mRNAs according to the genetic code. Synthetases help to ensure accurate translation of the genetic code by using both highly accurate cognate substrate recognition and stringent proofreading of noncognate products. While alterations in the quality control mechanisms of synthetases are generally detrimental to cellular viability, recent studies suggest that in some instances such changes facilitate adaption to stress conditions. Impaired endothelial function and microvascular dysfunction are frequently observed in individuals with cardiovascular risk factors such as obesity, insulin resistance, and type 2 diabetes. Reduced nitric oxide (NO) bioavailability is considered to be central to the development of these risk factors. NO can be generated via enzymatic reaction via NO synthase which requires the amino acid arginine. Evidence suggests that the amino acid citrulline is more effective at increasing circulating arginine levels than arginine supplementation alone. The COVID-19 pandemic has resulted in increased need for diagnostic testing using reverse transcriptase real-time PCR (RT-PCR). An exponential increase in demand has resulted in a shortage of numerous reagents in particular those associated with the lysis buffer required to extract the viral RNA. Herein, we describe a rapid collective effort by hospital laboratory scientists, academic researchers and the biopharma industry to generate a validated lysis buffer. We have formulated a 4M Guanidinium thiocyanate (GITC)/ Triton X-100 Lysis buffer which provides comparable results with the recommended reagents. Imatinib is generally well tolerated, with mild common side effects such as nausea and vomiting, diarrhea, muscle cramps, fatigue, skin rash and edema; however, pulmonary complications are uncommon. A 73-year-old woman undergoing one month treatment with Imatinib for chronic myeloid neoplasm associated with eosinophilia was admitted for sudden alteration of her performance status, dyspnea at rest and productive cough. On clinical examination, the patient was hypoxic (oxygen saturation on room air was 87%), and auscultation of her lungs revealed diffuse bilateral fine crackles. Chronic migraine (CM) is characterized by the International Headache Society classification (ICHD-3) as headache that occurs for greater than or equal to 15 days a month, lasting 4 or more hours. For chronic migraine patients, the goal with prophylactic treatment is to prevent future attacks and reduce the migraine frequency and severity. OnabotulinumtoxinA is the most universally accepted medication for the treatment of chronic migraine; and if successful, reduces migraine frequency up to 50%. Gout is a chronic arthritis caused by the deposition of poorly soluble monosodium urate monohydrate (MSU) crystals in peripheral joints. Resident macrophages initiate inflammation in response to MSU mediated by NF-κB nuclear translocation and NLRP3 inflammasome activation. We investigated the role of CD44, a transmembrane receptor, in mediating MSU phagocytosis by macrophages. We used an antibody that sheds the extracellular domain (ECD) of CD44 to study the role of the receptor and its associated protein phosphatase 2A (PP2A) in macrophage activation.
