Double polyethylene Quonset-style greenhouses offergrowers and farmers a more cost-effective structure than glass- or acrylic-covered structures for growing plants off-season. These greenhouses are especially applicablefor producing off-season high value crops such as fruits and vegetables for local markets. To support this production alternative, this publication contains a list of greenhouse components, the start-up costs associated with these items and an estimate of the number of person hours required to assemble and outfit a 24-foot-by-72-foot double polyethylene, air-inflated greenhouse structure. Soybeans [Glycine max (L.) Merr.] are susceptible to many diseases including fungal diseases such as soybean sudden death syndrome (SDS). Several studies reported SDS resistance quantitative trait loci (QTL) on the soybean genome using different recombinant inbred line (RIL) populations and low density genetic linkage maps. High density exclusively single nucleotide polymorphisms-based (SNP-based) maps were not yet reported in soybean. Genome-wide structural and gene content variations are hypothesized to drive important phenotypic variation within a species. Structural and gene content variations were assessed among four soybean (Glycine max) genotypes using array hybridization and targeted resequencing. Many chromosomes exhibited relatively low rates of structural variation (SV) among genotypes. However, several regions exhibited both copy number and presence-absence variation, the most prominent found on chromosomes 3, 6, 7, 16, and 18. Interestingly, the regions most enriched for SV were specifically localized to gene-rich regions that harbor clustered multigene families. Increasing seed yield is an important breeding goal of soybean [Glycine max (L.) Merr.] improvement efforts. Due to the small number of ancestors and subsequent breeding and selection, the genetic base of current soybean cultivars in North America is narrow. Soybean rust (SBR), caused by Phakopsora pachyrhizi Sydow, is one of the most economically important and destructive diseases of soybean [Glycine max (L.) Merr.] and the discovery of novel SBR resistance genes is needed because of virulence diversity in the pathogen. Gene expression is controlled by the complex interaction of transcription factors binding to promoters and other regulatory DNA elements. One common characteristic of the genomic regions associated with regulatory proteins is a pronounced sensitivity to DNase I digestion. We generated genome-wide high-resolution maps of DNase I hypersensitive (DH) sites from both seedling and callus tissues of rice (Oryza sativa). Approximately 25% of the DH sites from both tissues were found in putative promoters, indicating that the vast majority of the gene regulatory elements in rice are not located in promoter regions. Transcription factors (TFs) play important roles in plants. However, there is no systematic study of their structures and functions of most TFs in plants. Here, we performed template-based structure prediction for all TFs in Arabidopsis thaliana, with their full-length sequences as well as C-terminal and N-terminal regions. A total of 2,918 model structures were obtained with a high confidence score. We find that TF families employ only a smaller number of templates for DNA-binding domains (DBD) but a diverse number of templates for transcription regulatory domains (TRD). An experiment was conducted for 210 days to demonstrate the role of vegetable-fish culture integration in the growth, yields and economic benefits of fish and vegetables. Two 200 m2 earthen fishponds were stocked with Nile tilapia Oreochromis niloticus at 20,000 fish fingerlings per hectare. Pond A was fertilized with chicken manure and stocked fish fed on 35% crude protein supplementary diet referred to here as treated fish pond (TFP). Makurunge woodland is part of the major vegetation component covering coastal forest landscape in Tanzania that has been severely affected by anthropogenic disturbance. The present study determined the effects of anthropogenic disturbance on biomass, diversity, plant communities and plant species distribution pattern using nested sample plots systematically established along transects. Six major vegetation communities with different biomass were found. Plant species diversity ranged from 2.5 to 3.3 and was high in scrub forests, riverine and thicketed habitats, although the difference among these habitats was not significant (P>0.05). Plants like other living organisms respond differently under different environmental conditions. An elevated level of heavy metals is one of the stresses which results into three classes of plants depending on their heavy metal content. The classes of plant species according to their accumulated heavy metals around North Mara Gold Mine were not known. Rapeseed oil which is a triglyceride had undergone chemical modification on its structure to obtain hydroxyl groups. The hydroxylated chemical structure was crosslinked using methylene-pphenyl diisocyanate to produce a thermoset rapeseed oil resin. The cross linking process was monitored in situ using the Attenuated Total Internal Reflectance Fourier Transform-Infrared spectroscopy and the thermogravimetric analysis techniques. The infrared technique was also used to monitor chemical groups of the unmodified rapeseed oil. Cellular and molecular aspects of abiotic stress responses in Arabidopsis thaliana subjected to cold, drought, and high salinity and in two photosynthetic green alga, Chlamydomonas reinhardtii and Coccomyxa sp. C-169, subjected to nitrogen deprivation were investigated. Cold, drought, and high salinity can negatively affect plant growth and crop production. The first research aimed at determining the physiological functions of the stress-responsive Arabidopsis thaliana RD29A and RD29B genes. Cold, drought, and salt induced both genes; the promoter of RD29Awas found to be more responsive to drought and cold stresses, whereas the promoter of RD29B was highly responsive to salt stress. …

This dissertation combines investigation of the large-scale responses of bryophyte species diversity and distribution with small-scale physiological adaptations to global change. These two areas of inquiry are linked because one way to predict plant species responses to global change is to examine their distribution across current ecological gradients produced by factors such as latitude and elevation. By examining these biogeographic patterns one can identify those species that have a narrow tolerance and therefore are most sensitive to change. Plant mitochondrial genomes are composed of unusually complex structures, due to active recombination at numerous repeated sequences in the genome. The maintenance of mitochondrial genome stability is under the control of identifiable nuclear genes. In plants, three nuclear genes (MSH1, RECA3 and OSB1) have been shown to participate in recombination surveillance and the suppression of illegitimate recombination in mitochondria. Disruption of these loci in Arabidopsis results in reproducible mitochondrial genome rearrangements. Soil communities associated with specific plant species affect individual plants' growth and competitive ability. Limited evidence suggests that unique soil communities can also differentially influence growth and competition at the ecotype level. Previous work with Arabidopsis thaliana has shown that accessions produce distinct and reproducible rhizosphere bacterial communities, with significant differences in both species composition and relative abundance. We tested the hypothesis that soil communities uniquely affect the growth and reproduction of the plant accessions with which they are associated. Epigenetic variation describes heritable differences that are not attributable to changes in DNA sequence. There is the potential for pure epigenetic variation that occurs in the absence of any genetic change or for more complex situations that involve both genetic and epigenetic differences. Methylation of cytosine residues provides one mechanism for the inheritance of epigenetic information.
