Premise of the Study: Insect-resistant Bacillus thuringiensis (Bt) maize is widely cultivated, yet few studies have examined the interaction of symbiotic arbuscular mycorrhizal fungi (AMF) with different lines of Bt maize. As obligate symbionts, AMF may be sensitive to genetic changes within a plant host. Previous evaluations of the impact of Bt crops on AMF have been inconsistent, and because most studies were conducted under disparate experimental conditions, the results are difficult to compare. Vegetation and climate both play integral roles in water availability, particularly for arid to semi-arid regions. Changes in these variables can lead to extreme shortages in water for regions that rely on water for crop irrigation (i.e., the Great Plains). The objective of this study is to evaluate the impacts of vegetation on water availability in the Republican River basin in central Nebraska. Decreases in streamflow have been observed in the river basin for many years and, as a result, an invasive riparian plant species (Phragmites australis) is being removed in an effort to reduce evapotranspiration and reclaim …

Organoamine-silica hybrids were prepared by a co-condensation of tetraethoxysilane (TEOS) and different aminoalkoxysilanes, namely, 3-aminopropyltrimethoxysilane, N-(3(trimethoxysilyl)propyl)-ethylenediamine and 3[2(2-amino)ethylamino]propyl trimethoxysilane, at different ratios using cashew nut shell liquid (CNSL) components as templates. Of the major components of CNSL tested, only cardanol was found to be efficient as a template as it yielded 66% of the hybrid. The prepared materials were characterized by diffuse reflectance Fourier Transform Infrared (FTIR), Atomic Force Microscopy and acid titration. Results indicated that indeed the organoamine groups were attached onto the silica surface, with a maximum loading of 2.9 mmol organic group per g silica. The materials …

Dalbergia melanoxylon is a plant with valuable wood in the world and therefore is over harvested for timber while its regeneration is very low. The propagation efforts by techniques such as tissue culture or mycorrhiza have not been investigated which instigated conduction of this study. Soil and cuttings were collected from Kilwa, Kilosa and Babati for rooting test, potting media and soil characterization. The results showed that fresh soil improved rooting characteristics while sterilized soil did not due to the presence of mycorrhiza in fresh soil compared to none in sterilized media. Background: Sphingosine kinase (SPHK) and phospholipaseD(PLD) produce different lipid mediators involved in abscisic acid (ABA) response.
Results: Ablation of SPHKs and PLDα1 attenuates ABA-induced production of LCBPs and PA. Phyto-S1P closes stomata in sphk1, sphk2, but not in pldα1, whereas PA closes stomata in all mutants.
Conclusion: SPHK acts upstream of PLDα1, whereas PLDα1 promotes SPHK.
Significance: The roles of lipid messengers in the ABA signaling pathway are clarified.

There is rising interest in modeling the noncentrosomal cortical microtubule cytoskeleton of plant cells, particularly its organization into ordered arrays and the mechanisms that facilitate this organization. In this review, we discuss quantitative models of this highly complex and dynamic structure both at a cellular and molecular level. We report differences in methodologies and assumptions of different models as well as their controversial results. Our review provides insights for future studies to resolve these controversies, in addition to underlining the common results between various models. Superficially, plants seem so simple. Rooted in place, they do not move around. And while plant growth is a dynamic process, without time-lapse photography, growth events are so imperceptibly slow that, to us impatient humans, plants seem both immobile and static. Nevertheless, there is a lot going on inside the plant body, and this is especially true for the events of reproduction that play out inside flowers and fruits. Colonization and lesion development by virulent, asymptomatic and hypovirulent isolates of Sclerotinia homoeocarpa on nonwounded (NW) and wounded (W) leaves of creeping bentgrass were characterized. Hypovirulent and asymptomatic isolates contain the fungal virus, Ophiostoma mitovirus 3a, and virulent isolates are virus-free. On NW leaves, all isolates infected leaves with appressoria along cell walls and through stomata by 48 hours post-inoculation (hpi). Inter- and intracellular hyphae formed on heavily colonized, NW leaves at 96 hpi. Wound-inoculated grass had a colonization front characterized by inter- and intracellular hyphal colonization within nonsymptomatic tissues at 8 hpi by direct infection of the wound …

In angiosperms, sucrose uptake transporters (SUTs) have important functions especially in vascular tissue. Here we explore the evolutionary origins of SUTs by analysis of angiosperm SUTs and homologous transporters in a vascular early land plant, Selaginella moellendorffii, and a non-vascular plant, the bryophyte Physcomitrella patens, the charophyte algae Chlorokybus atmosphyticus, several red algae and fission yeast, Schizosaccharomyces pombe. Plant SUTs cluster into three types by phylogenetic analysis. Bundling of microtubules (MTs) is critical for the formation of complex MT arrays. In land plants, the interphase cortical MTs form bundles specifically following shallow-angle encounters between them. To investigate how cells select particular MT contact angles for bundling, we used an in vitro reconstitution approach consisting of dynamic MTs and the MT-cross-linking protein MAP65-1. We found that MAP65-1 binds to MTs as monomers and inherently targets antiparallel MTs for bundling. Dwell-time analysis showed that the affinity of MAP65-1 for antiparallel overlapping MTs is about three times higher than its affinity for single MTs and parallel overlapping MTs. We also …

Insect herbivory can have a major influence on plant reproduction, and potentially drive selection for strategies that reduce or resist herbivore effects. I used a combination of field experiments and ecological modeling to examine how modifications in the patterns and timing of reproductive investment might ameliorate the consequences of herbivore damage for plant reproduction. I performed experiments to examine how changes in reproductive effort after apical damage and reduction of insect herbivory affected seed production in two thistles native to Nebraska. The grasses, Poaceae, are one of the largest and most successful angiosperm families. Like many radiations of flowering plants, the divergence of the major grass lineages was preceded by a whole-genome duplication (WGD), although these events are not rare for flowering plants. By combining identification of syntenic gene blocks with measures of gene pair divergence and different frequencies of ancient gene loss, we have separated the two subgenomes present in modern grasses. A foliar fungicide efficacy timing trial was conducted at the University of Nebraska-Lincoln South Central Agricultural Laboratory near Clay Center, NE. DeKalb corn hybrid DKC 62-54, rating of “good” (6 out of 9) for gray leaf spot (GLS) and “excellent” (2 out of 9) for common rust (CR), was planted on 2 May in 30 in. rows with a target population of 32,000 plants/A.
