Genetic Analysis Of Flower Color Differences Between A Hummingbird-Pollinated And A Self-Pollinated Monkeyflower (Mimulus) Species, Caitlin Foster 2020 University of Connecticut

Genetic Analysis Of Flower Color Differences Between A Hummingbird-Pollinated And A Self-Pollinated Monkeyflower (Mimulus) Species, Caitlin Foster

University Scholar Projects

Flower color plays an important role in pollinator discrimination and speciation. Understanding the genetic contributions to flower color differences between two closely related species, Mimulus cardinalis and Mimulus parishii, can improve understanding of how they developed different pollination syndromes and diverged from a recent common ancestor. M. cardinalis is hummingbird-pollinated and has large, bright red flowers while M. parishii is self-pollinated and has small, pale pink flowers. An F2 hybrid population between these two species was created to establish a platform for analysis of the genetic architecture controlling the differences in anthocyanin pigmentation. Statistical analysis of anthocyanin concentration distribution …

Electronic Theses and Dissertations

This dissertation explores and expands the existing knowledge on the role of plant volatiles in facilitating seed priming and providing direct defense against herbivore. Although the roles of plant volatiles in priming a plant’s defenses and providing direct defense against pests is well known, information regarding their effects on seed and the fitness of future plants and direct toxicity to herbivores is understudied. This dissertation does a thorough examination of these two understudied aspects of plant volatiles and provides novel insight into the role of plant volatiles in seed priming and direct defense (Chapter I). Seeds in the soil can …

The Effects Of Titanium Dioxide Nanoparticles On The Growth And Development Of Sorghum Bicolor (L.) Moenech, Adam Gregory Shoemaker

Graduate Theses/Dissertations

Engineered nanoparticles (ENPs) have seen a drastic increase in their use over the past decade in various consumer products. ENPs will therefore enter terrestrial ecosystems and soils with increasing frequencies, yet research into the effects of ENPs on living organisms and crops is greatly lacking. Currently, there is only one major study reported on the effects of a single ENP, silver quantum dots, on Sorghum bicolor, the 5th largest crop in the world. I examined the effects of a commonly used metal oxide nanoparticle, titanium dioxide (TiO2), on the growth and development of sorghum grown in …

Control Of Volunteer Corn In Enlist Corn And Economics Of Herbicide Programs For Weed Control In Conventional And Multiple Herbicide-Resistant Soybean Across Nebraska, Adam Michael Striegel

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

With commercialization of multiple herbicide-resistant corn and soybean cultivars, producers have new management options for controlling herbicide-resistant weeds and volunteer corn. Corn-on-corn production systems are common in irrigated fields in southcentral Nebraska which can create issues with volunteer corn management in corn fields. Enlist corn contains a new multiple herbicide-resistant trait providing resistance to 2,4-D choline, glyphosate, and the aryloxyphenoxypropionate (FOPs). Field experiments were conducted in 2018 and 2019 at South Central Agricultural Laboratory near Clay Center, Nebraska with the objective to evaluate ACCase-inhibiting herbicides and herbicide application timing on volunteer corn control, Enlist corn injury, and yield. Glyphosate/glufosinate-resistant corn …

Buffalograss Diversity Assessment, Jesse Collin Marshall

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

Buffalograss [Buchloë dactylodes (Nutt.) Engelm. syn. Bouteloua dactyloides (Nutt.) Columbus] is a low input alternative turfgrass for the Great Plains region. Buffalograss is a dioecious stoloniferous warm season grass with wide variation of valuable traits and is well adapted for use in minimal maintenance landscapes. The obligate outcrossing nature of the species and wide variation of potentially valuable traits make buffalograss a prime candidate for cultivar improvement and genetic study. Limited information is available regarding buffalograss genetics. In effort to expand genetic resources around buffalograss, we developed buffalograss derived simple sequence repeat markers from previously available transcriptomes of buffalograss …

Resilience To Large, “Catastrophic” Wildfires In North America's Grassland Biome, Victoria M. Donovan, Dirac Twidwell, Daniel R. Uden, Tsegaye Tadesse, Brian D. Wardlow, Christine H. Bielski, Matthew O. Jones, Brady W. Allred, David E. Naugle, Craig R. Allen

Department of Agronomy and Horticulture: Faculty Publications

Wildfires are ecosystem‐level drivers of structure and function in many vegetated biomes. While numerous studies have emphasized the benefits of fire to ecosystems, large wildfires have also been associated with the loss of ecosystem services and shifts in vegetation abundance. The size and number of wildfires are increasing across a number of regions, and yet the outcomes of large wildfire on vegetation at large‐scales are still largely unknown. We introduce an exhaustive analysis of wildfire‐scale vegetation response to large wildfires across North America's grassland biome. We use 18 years of a newly released vegetation data set combined with 1,390 geospatial …

A Contribution To The Characterization Of The Diversity Of Ectomycorrhizal Fungi Associated With American Chestnut At The Utc Fortwood Street Nursery, Colton Jones 2020 University of Tennessee at Chattanooga

A Contribution To The Characterization Of The Diversity Of Ectomycorrhizal Fungi Associated With American Chestnut At The Utc Fortwood Street Nursery, Colton Jones

Honors Theses

Ectomycorrhizas play several essential roles in the biosphere and have immeasurable implications on the ecosystems in which they exist. Much has been discovered about the relationships between ectomycorrhizal fungi and the trees with which they associate, but there is still much to learn. Due to the nature of ectomycorrhizal morphology, DNA analysis is frequently required in order to accurately identify the fungal partner. Some ectomycorrhizal fungi produce above-ground fruiting bodies that presumably contain the same DNA sequences as the fungi encapsulating corresponding plant root tips below the soil; these fruiting bodies have been frequently observed growing in nursery containers at …

Modification, Verification Of Sequence And Optimization Of Expression Of P297f An Inactive Mutant Of Flavonol Specific Glucosyltransferase From Grapefruit (Cp3gt), Sarah Fox

Undergraduate Honors Theses

Citrus fruits are widely consumed and can offer various health benefits. One enzyme found in grapefruits, Citrus paradisi flavonol specific 3-O-glucosyltransferase (CP3GT), catalyzes the addition of glucose to one specific flavonoid class and at only one site. These flavonoids are plant secondary metabolites that can be used in a variety of plant functions including signaling and protection. The only class of flavonoids that CP3GT glucosylates is flavonols, and this specificity is of interest to study for potential benefits in biotechnology and enzyme modeling. In order to study this enzyme and its structure, a variety of mutants were created using site-directed …

Subcellular Localization Of Tobacco Sabp2 Under Normal And Stress Conditions, Sanjeev Das 2020 East Tennessee State University

Subcellular Localization Of Tobacco Sabp2 Under Normal And Stress Conditions, Sanjeev Das

Undergraduate Honors Theses

Subcellular Localization of Tobacco SABP2 under Normal and Stress Conditions

Salicylic acid (SA), a phytohormone, plays an important role in plant physiology. SA mediated innate immune pathway is an important pathway for plant immunity against pathogens. Plants resisting pathogen infection synthesize higher levels of Methyl Salicylate (MeSA), which is then converted to SA by the esterase activity of Salicylic Acid Binding Protein 2 (SABP2). The high level of the converted SA leads to enhanced pathogen resistance. The study of subcellular localization of a protein is critical in explaining its potential biochemical functions. SABP2 tagged with eGFP was expressed transiently in …

Student Works (2020-2029)

Studies on genetic variation in plants had become extensively employed in research for plant improvement and conservation. Due to their sessile nature, plants are exposed to various environmental stressors such as exposure to enhanced levels of ultraviolet (UV), ionizing and non-ionizing radiations. Exposure to these harmful radiations will result in various damages, ranging from DNA and chromosomal aberrations to phenotypic abnormalities. However, plants have evolved efficient DNA repair mechanisms to detect and repair any DNA damages caused by the exposure to these harmful stressors, therefore ensuring their survival. In this study, the effect of gamma radiation (as a source of …
