Antimicrobial Strategies For Topical Applications, Kelsey Marie Lopez 2020 Louisiana State University and Agricultural and Mechanical College

Antimicrobial Strategies For Topical Applications, Kelsey Marie Lopez

LSU Doctoral Dissertations

Bacteria can thrive in diverse environments and are often harmless or beneficial. Bacteria in digestive tracts is one example of beneficial bacteria; however, bacteria can be harmful and when this type proliferates, it can cause infections within hosts. Bacterial infections are easily treated with antibiotics in most cases. However, bacteria are also capable of developing mutations which could cause them to become multi-drug resistant and eventually, “superbugs.” Therefore, the development of novel antimicrobial agents and materials capable of combating drug-resistant bacteria is necessary. Research presented in this dissertation focuses on different strategies for minimizing and preventing topical bacterial infections using …

Microbial Community Structures In Three Bahamian Blue Holes, Meghan J. Gordon 2020 University of South Florida

Microbial Community Structures In Three Bahamian Blue Holes, Meghan J. Gordon

USF Tampa Graduate Theses and Dissertations

This study used 16S rDNA metagenomics and water chemistry to conduct an examination of microbial community dynamics and biogeochemistry in three physically adjacent, sunlit blue holes with variable hydrologic regimes on San Salvador Island, Bahamas. Church and Watling’s Blue Holes are holomictic with relatively clear waters, while Inkwell Blue Hole hosts density stratification and waters stained brown with tannins. Based on water color and clarity and physicochemical profiles, I hypothesized Church and Watling’s Blue Holes would be dominated by oxigenic photoautotrophs, and that the bottom layer of Inkwell would be characterized by euxinic (anoxic and sulfidic) conditions and host primarily …

Plant Defensin Antibacterial Mode Of Action Against Pseudomonas Species, Andrew E. Sathoff, Shawn Lewenza, Deborah A. Samac 2020 Dakota State University

Plant Defensin Antibacterial Mode Of Action Against Pseudomonas Species, Andrew E. Sathoff, Shawn Lewenza, Deborah A. Samac

Research & Publications

Background: Though many plant defensins exhibit antibacterial activity, little is known about their antibacterial mode of action (MOA). Antimicrobial peptides with a characterized MOA induce the expression of multiple bacterial outer membrane modifications, which are required for resistance to these membrane-targeting peptides. Mini-Tn5- lux mutant strains of Pseudomonas aeruginosa with Tn insertions disrupting outer membrane protective modifications were assessed for sensitivity against plant defensin peptides. These transcriptional lux reporter strains were also evaluated for lux gene expression in response to sublethal plant defensin exposure. Also, a plant pathogen, Pseudomonas syringae pv. syringae was modified through transposon mutagenesis to …

Dengue virus (DENV) is the most common and widespread arboviral infection worldwide. Though all four DENV serotypes cocirculate in nature, the clinicopathological framework of these serotypes is undefined in Pakistan. A cross-sectional, observational study was performed to document the circulation of various arboviruses in the Sindh region of Pakistan. Here we describe a population of patients diagnosed with DENV spanning a 2-year period. This study used an orthogonal system of NS1 antigen ELISA followed by RT-PCR for DENV detection and subtyping. A total of 168 NS1 positive patients were evaluated of which 91 patients were serotyped via RT-PCR. There was …

Microbiomes Of Caribbean Octocorals Vary Over Time But Are Resistant To Environmental Change, Mark Mccauley, Colin R. Jackson, Tamar Goulet

Faculty and Student Publications

The bacterial microbiome is an essential component of many corals, although knowledge of the microbiomes in scleractinian corals far exceeds that for octocorals. This study characterized the bacterial communities present in shallow water Caribbean gorgonian octocorals over time and space, in addition to determining the bacterial assemblages in gorgonians exposed to environmental perturbations. We found that seven shallow water Caribbean gorgonian species maintained distinct microbiomes and predominantly harbored two bacterial genera, Mycoplasma and Endozoicomonas. Representatives of these taxa accounted for over 70% of the sequences recovered, made up the three most common operational taxonomic units (OTUs), and were present in …

Contributions Of Gyra And Parc Mutations And Qnrs2 Acquisition To Ciprofloxacin Resistance In Aeromonas Veronii Hm21, Daniel J. Silverstein

Honors Scholar Theses

In recent years, ciprofloxacin resistant (CpR) Aeromonas veronii and A. hydrophila strains have been isolated from the wounds of patients receiving leech therapy. Genome comparisons of these CpR isolates revealed the presence of chromosomal mutations in gyrA and parC as well as the gain of qnrS2 on either a large, 34 kb, conjugatable, low-copy plasmid, pAv42, or on a small, 6.8 kb, high-copy plasmid, pAh1471. The minimum inhibitory concentration, MIC, for Cp of these clinical isolates ranged from 1 to ≥32 µg/mL and some harbored a qnrS2 containing plasmid. We wanted to assess the contributions of these factors in an …

Metabolic Network Analysis Of Filamentous Cyanobacteria, Daniel Alexis Norena-Caro 2020 Louisiana State University and Agricultural and Mechanical College

Metabolic Network Analysis Of Filamentous Cyanobacteria, Daniel Alexis Norena-Caro

LSU Doctoral Dissertations

Cyanobacteria were the first organisms to use oxygenic photosynthesis, converting CO2 into useful organic chemicals. However, the chemical industry has historically relied on fossil raw materials to produce organic precursors, which has contributed to global warming. Thus, cyanobacteria have emerged as sustainable stakeholders for biotechnological production. The filamentous cyanobacterium Anabaena sp. UTEX 2576 can metabolize multiple sources of Nitrogen and was studied as a platform for biotechnological production of high-value chemicals (i.e., pigments, antioxidants, vitamins and secondary metabolites). From a Chemical engineering perspective, the biomass generation in this organism was thoroughly studied by interpreting the cell as a microbial …

The assembly of single-amplified genomes (SAGs) and metagenome-assembled genomes (MAGs) has led to a surge in genome-based discoveries of members affiliated with Archaea and Bacteria, bringing with it a need to develop guidelines for nomenclature of uncultivated microorganisms. The International Code of Nomenclature of Prokaryotes (ICNP) only recognizes cultures as ‘type material’, thereby preventing the naming of uncultivated organisms. In this Consensus Statement, we propose two potential paths to solve this nomenclatural conundrum. One option is the adoption of previously proposed modifications to the ICNP to recognize DNA sequences as acceptable type material; the other option creates a nomenclatural code …

3,4-Dihydroxyphenylacetate 2,3-Dioxygenase Modulates The Ability Of Enterobacter Hormaechei To Induce Cryptococcus Neoformans Melanization, Amy Nelson

Honors Scholar Theses

In this study, we performed transposon mutagenesis to create a library of Enterobacter hormaechei mutants and developed a melanization assay to identify mutants that could not induce the typical melanization phenotype in Cryptococcus neoformans. Relevant phenotype labels included: “hypomelanizer”, a total lack of melanization; “hypermelanizer”, an increased melanization; or “abnormal”, an increased melanization with complete color change of the assay plate to a reddish-brown. Genomic sequencing of 47 mutants and bioinformatic analysis allowed us to pinpoint the transposon insertion site in each E. hormaechei mutant to identify the genes that were affected. A single mutant that induced the abnormal …

Analysis Of Biofilm Remediation Capacity For Octenyl Succinic Anhydride (Osa), A Bioactive Food Starch Modifier Compound, Matthew R. Borglin

Master's Theses

Matthew R. Borglin

This thesis demonstrates efficacy of Octenyl Succinic Anhydride (OSA), as a biofilm sanitizer. Biofilms allow bacteria to adhere to solid surfaces with the use of excreted polymeric compounds. For example, surfaces found in food production or processing facilities such as the interior of a raw milk holding tank, are some of the most susceptible to biofilm contamination. When present, biofilms can cause a variety of negative effects, which include; reduction of product shelf life, corrosion, and outbreaks of foodborne illnesses. The close association of biofilms with the majority of foodborne illness cases led the US Environmental Protection …

Analysis And Optimization Of Colorimetric Nanosensors For Rapid Detection Of Microbes In Water, Ruby A. Lang 2020 California Polytechnic State University, San Luis Obispo

Analysis And Optimization Of Colorimetric Nanosensors For Rapid Detection Of Microbes In Water, Ruby A. Lang

Master's Theses

Access to safe water is a basic human right recognized by the United Nations General Assembly in 2010 (WHO, 2020). However, a least 2.2 billion people globally still are without safely managed water services meaning they use a drinking water source that can be contaminated with faeces (WHO, 2020). With such a pressing global health issue, it is clear that improvement to water systems is important and required in the Agenda 2030 Sustainable Development Goals (SDGs). However, to improve water systems and prove they are safe water sources, water quality testing must occur. A solution to this issue is the …

Molecular Roles Of Small Inner Membrane Proteins In Escherichia Coli Cell Envelope Integrity, Aaron Mychack 2020 CUNY Graduate Center

Molecular Roles Of Small Inner Membrane Proteins In Escherichia Coli Cell Envelope Integrity, Aaron Mychack

Dissertations, Theses, and Capstone Projects

The biological membrane is an essential, defining feature of all cells. Biological membranes comprise phospholipid bilayers as well as a complement of proteins which are unique to a given organism. These proteins play a central role in dictating the biochemical state of the cell’s internal cytoplasm by controlling selective passage of solutes in and out of the cell, transducing signals in response to extracellular stimuli, and controlling the biogenesis of the bilayer itself which is critical towards barrier function. For most bacteria, the periphery of the cell is multi-layered, including both a biological membrane as well as a peptidoglycan cell …

Isolation Of Plastic-Degrading Fungi From Landfill And Determining The Selected Plastics Biodegradation Capability, Mohan Priya

Student Works (2020-2029)

Polyethylene Terephthalate (PET), Low Density Polyethylene (LDPE) and High Density Polyethylene (HDPE) are the most important polymers used predominantly in present world. Since synthetic polymers hardly undergo degradation and remain in the environment for a very long time, up to 1000 years, these drawbacks increase the percentage of solid wastes in landfill and threaten the environment. To date, Municipal Solid Waste (MSW) landfill is the most favored disposal method especially among the developing countries. The aim of present study is to evaluate the potential of fungus isolated from landfilled plastics to degrade PET, LDPE and HDPE. Besides, effect of different …

Abundance And Distribution Of Microplastics And Phthalic Acid Esters In Selected Mangrove Areas In Peninsular Malaysia, Anuar Norkhairah

Student Works (2020-2029)

Contamination of microplastics in mangrove has been receiving growing attention as experts reckon 30 billion tonnes of plastics will accumulate in 2050. The vast increase in plastic commodities will increase microplastics abundance in mangroves as mangroves are substantially sinks and traps for plastic debris. Due to persistent properties and high surface to volume ratio of microplastics, Phthalic Acid Esters (PAEs) can be taken up by microplastics, representing a grave threat to the ecosystems. This study investigated the abundance and distribution of microplastics and PAEs in selected mangrove areas in Peninsular Malaysia. The objectives of this study are to identify the …
