Understanding variation in host-associated microbial communities is important given the relevance of microbiomes to host physiology and health. Using 560 fecal samples collected from wild chimpanzees (Pan troglodytes) across their range, we assessed how geography, genetics, climate, vegetation, and diet relate to gut microbial community structure (prokaryotes, eukaryotic parasites) at multiple spatial scales. We observed a high degree of regional specificity in the microbiome composition, which was associated with host genetics, available plant foods, and potentially with cultural differences in tool use, which affect diet. Two related salmonids, Arctic char (Salvelinus alpinus) and lake whitefish (Coregonus clupeaformis) sampled from the high Arctic region of Nunavut, Canada are anadromous fish, migrating annually from the same ice-covered freshwater waterbodies to spend summers in the marine waters of the Arctic Ocean. Microbiota associated with the skin-associated mucus undergo community change coincident with migration, and irrespective of this turnover, antibiotic resistance was detected in mixed bacterial cultures initiated with mucus samples. The NS1 protein of influenza A virus (IAV) plays important roles in viral pathogenesis and host immune response. Through a proteomic approach, we have identified RuvB-like proteins 1 and 2 (RuvBL1 and RuvBL2) as interacting partners of the NS1 protein of IAVs. Infection of human lung A549 cells with A/PR/8/34 (PR8) virus resulted in reductions in the protein levels of RuvBL2 but not RuvBL1. Further studies with RuvBL2 demonstrated that the NS1-RuvBL2 interaction is RNA-independent, and RuvBL2 binds the RNA-binding domain of the NS1. The food industry currently focuses on wet food sanitation as opposed to low-moisture foods because of bacteria’s inability to reproduce in low water activity media. Background

Coagulase-negative staphylococci (CNS) survive as commensals of skin, anterior nares and external canals of human and were regarded as non-infectious pathogens. However, they are emerging as a major cause of nosocomial infectious due to their ability to form biofilms and high resistance to several classes of antibiotics. This study examines the biofilm forming abilities of 214 clinical CNS isolates using phenotypic and genotypic methods, and determines their antibiotic susceptibility patterns.

Four new sulfonyl phenoxides were synthesized through O-sulfonylation reaction of phenolic compounds with 5-chloro-1-ethyl-2-methylimidazole-4-sulfonyl chloride in good yield. FT-IR, 1H-NMR, 13C-NMR, and DEPT 135 NMR were carried out to characterize and the thin layer chromatography (TLC) confirm the purity. Antimicrobial activities of the sulfonyl phenoxides against Gram-positive (methicillin-susceptible Staphylococcus aureus, methicillin-resistant Staphylococcus aureus, and Bacillus subtilis), Gram-negative (Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae), and Candida albicans were carried out using the standard microbiological method. The antimicrobial activities were referenced to ciprofloxacin and itraconazole, antibacterial and antifungal drug respectively. The in vitro antimicrobial studies of 5-chloro-1-ethyl-2-methyl-1H-imidazole-4-sulfonyl(4-chloro-3-methyl)phenoxide and 5-chloro-1-ethyl-2-methyl-1H-imidazole-4-sulfonyl-2-methylphenoxide showed moderate activity …

The presence of Legionella pneumophila was assessed using a cultivation-based approach in New York City waterways, a freshwater portion of the lower Hudson River Estuary near Kingston NY, and in urban and suburban street water. Legionella pneumophila was detected in 51% of brackish New York City Estuary samples, most with concentrations near minimum detection (=>1 organism/ mL). In contrast, it was detected in 22% of suburban freshwater Hudson River Estuary samples. The research reported in this thesis was conducted to investigate whether protein hydrolysis associated with endogenous decay of biomass produced following the addition of a readily fermentable substrate is a potential source of precursors that can ultimately be transformed to toluene and p-cresol in environments undergoing enhanced bioremediation.

Phytophthora ramorum is the causal agent of Sudden Oak death (SOD), ramorum dieback and ramorum leaf blight which affect both forest environments and nurseries. This oomycete pathogen has had a huge economic impact on the nursery and lumber industry. Carbon monoxide (CO) has been exploited as a microbial energy source for much of life’s evolutionary history. A phylogenetically diverse array of microorganisms can oxidize CO using two distinct CO dehydrogenases, molybdenum-dependent (Mo-CODH) and nickel-dependent (Ni-CODH). Aerobes and facultative organisms contain Mo-CODHs which allow them to utilize oxygen as an electron acceptor in addition to alternatives such as nitrate and sulfate. Obligate anaerobic organisms contain Ni-CODHs, which oxidize CO at elevated concentrations, but cannot utilize oxygen. Cadmium (Cd) poisoning contributes to severe bodily detriments characterized by kidney failure, osteomalacia, and osteoporosis, otherwise known as Itai-Itai disease. The ingestion of cadmium contaminated foods primarily causes these conditions. There are no treatments to remove cadmium once absorbed, resulting in bioaccumulation. In this study, we are developing a method to manipulate the community of microorganisms (microbiome) living within the gastrointestinal (GI) tract of the fruit fly (Drosophila melanogaster), intending to use microorganisms to reduce the availability of ingested cadmium.

This paleopathological study aims to identify Mycobacterium tuberculosis complex (MTBC), Mycobacterium avium complex (MAC) and other Mycobacterium species in silico from skeletal samples that belonged to 28 Polish individuals in the Neolithic period under PRJNA422903 from the Sequence Read Archive (SRA). After next-generation sequencing (NGS), bioinformatics methods are heavily relied upon for identification of pathogens from complex samples. We implemented a bioinformatics pipeline, with custom-built databases, utilizing the following software tools: Trim Galore! and Kraken2. The introduction of the Chinese chestnut blight in 1904 decimated native American chestnut (Castanea dentata (Marshall) Borkh.) populations. In this study we aim to: 1) document the location of individual chestnuts in our vicinity; 2) document healthy versus blight infected individuals; 3) characterize putative differences in the soil microbiome between infected and non-infected individuals, as well as between chestnuts and hardwood neighbors. With this information we hope to provide new insights into mechanisms that may enhance blight and fungal resistance in American chestnuts through the understanding of the interactions between the trees and their surrounding soil microbiome.

In order to provide insight into potential therapeutic breakthroughs for the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), more research must be done to understand the structure and function of its proteins. The open reading frame 8 (ORF 8) accessory protein is particularly unstable on its own outside of the viral envelope but can be stabilized when bound to the small ubiquitin-related modifier (SUMO) protein.
