Use Of Two-Dimensional Agarose-Gel Analysis To Characterize Processing Of Uv-Irradiated Plasmids And The Composition Of The Replisome Following Uv-Induced Arrest, Harout Arthur Jeiranian 2012 Portland State University

Use Of Two-Dimensional Agarose-Gel Analysis To Characterize Processing Of Uv-Irradiated Plasmids And The Composition Of The Replisome Following Uv-Induced Arrest, Harout Arthur Jeiranian

Dissertations and Theses

In this thesis, I address two fundamental questions related to our understanding of how DNA damage is processed and repaired during replication. Using Two-dimensional (2-D) agarose gel analysis, I first examine whether DNA damage on plasmids introduced by transformation is processed in a manner similar to that observed on endogenously replicating plasmids and the chromosome. The original intent for using this approach was to develop a technique that could examine how different DNA adducts would be repaired in various sequence contexts. However, I found that distinct differences exist between the processing of DNA damage on transforming plasmids and the chromosome. …

Brassinosteroid-Mediated Stress Tolerance In Arabidopsis Thaliana, Tawhidur Rahman 2011 The University of Western Ontario

Brassinosteroid-Mediated Stress Tolerance In Arabidopsis Thaliana, Tawhidur Rahman

Electronic Thesis and Dissertation Repository

Brassinosteroids (BRs) are a group of steroidal plant hormones that are essential for proper plant development and also promote stress tolerance. Without BRs, plants are dwarfs and infertile. To understand the molecular mechanisms underlying BR-mediated stress tolerance, global gene expression analysis of untreated and 24-epibrassinolide (EBR)-treated Arabidopsis thaliana seedlings under non-stress and heat stress (HS) conditions was carried out. Microarray data analysis indicated that stress-related genes were predominant within the EBR up-regulated gene data set. Furthermore, several of these genes were abscisic acid (ABA) and jasmonic acid (JA) related. Measurements of endogenous hormones showed significant increases in the levels ABA …

Gata-Family Transcription Factors In Magnaporthe Oryzae, Cristian F. Quispe 2011 University of Nebraska-Lincoln

Gata-Family Transcription Factors In Magnaporthe Oryzae, Cristian F. Quispe

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

The filamentous fungus, Magnaporthe oryzae, responsible for blast rice disease, destroys around 10-30% of the rice crop annually. Infection begins when the specialized infection structure, the appressorium, generates enormous internal turgor pressure through the accumulation of glycerol. This turgor acts on a penetration peg emerging at the base of the cell, causing it to breach the leaf surface allowing its infection.

The enzyme trehalose-6- phosphate synthase (Tps1) is a central regulator of the transition from appressorium development to infectious hyphal growth. In the first chapter we show that initiation of rice blast disease requires a regulatory mechanism involving an …

Self-Assembled Rippling Structure Based On Metal-Elastomer Composite For Tunable Plasmonics, Fang-Tzu Chuang 2011 National Taiwan University

Self-Assembled Rippling Structure Based On Metal-Elastomer Composite For Tunable Plasmonics, Fang-Tzu Chuang

Fang-Tzu Chuang

We demonstrate that a periodic metallic wavy-structure can be easily fabricated on elastomeric polydymethylsiloxane (PDMS) substrates. The metallic wavy-structures are self-assembly formed by thermal evaporation of Ag on PDMS without any lithography processes. The size of the PDMS substrate with an aspect ratio (the ratio of length to width) higher than 2 are found to be necessary to self-assemble the one-dimensional metallic wavy-structure. The period of the wavy-structure is controllable by varing the thickness of the deposited silver film. The elastomeric- gratings can excite surface plasmon on Ag/air interfaces; the resonant wavelength can be tuned to a range of 3.5% …

Science Boot Camp For Librarians: Cpd On A Shoestring, Maxine G. Schmidt, Rebecca Reznik-Zellen

Maxine G Schmidt

Science Boot Camp for Librarians was envisioned as a casual but intensive immersion event into selected scientific subjects that employ networked computing capabilities for research and collaboration. The goal of the event is to provide librarians with networking opportunities, but more importantly, to give them some of the context and ocabulary of a discipline to enable them to better engage faculty and research scientists with regard to escience. A half-day is devoted to each of three topics chosen for that year’s camp. A local faculty member provides an overview of the research area, and a second describes a single project …

Muc4 Modulation Of Ligand-Independent Erbb2 Signaling, Goldi Attias Kozloski 2011 University of Miami

Muc4 Modulation Of Ligand-Independent Erbb2 Signaling, Goldi Attias Kozloski

Goldi A Kozloski

The membrane mucin Muc4 is a heterodimer, bi-functional glycoprotein complex that is normally expressed in epithelial tissue. Functional studies on the extracellular mucin subunit of Muc4 have shown that it acts to promote anti-adhesion properties by sterically interfering with cell-cell and cell-matrix interactions and that the extent of this effect is directly associated with the number of tandem repeats on this subunit. Functional studies on the transmembrane subunit of Muc4 have shown that this subunit participates in intracellular signaling through interaction with the receptor tyrosine kinase ErbB2. This role of Muc4 was shown to be mediated by stabilizing the heregulin …

Micrornas Are Independent Predictors Of Outcome In Diffuse Large B-Cell Lymphoma Patients Treated With R-Chop, Goldi Kozloski 2011 University of Miami

Micrornas Are Independent Predictors Of Outcome In Diffuse Large B-Cell Lymphoma Patients Treated With R-Chop, Goldi Kozloski

Goldi A Kozloski

PURPOSE:
Diffuse large B-cell lymphoma (DLBCL) heterogeneity has prompted investigations for new biomarkers that can accurately predict survival. A previously reported 6-gene model combined with the International Prognostic Index (IPI) could predict patients' outcome. However, even these predictors are not capable of unambiguously identifying outcome, suggesting that additional biomarkers might improve their predictive power.
EXPERIMENTAL DESIGN:
We studied expression of 11 microRNAs (miRNA) that had previously been reported to have variable expression in DLBCL tumors. We measured the expression of each miRNA by quantitative real-time PCR analyses in 176 samples from uniformly treated DLBCL patients and correlated the results to …

Localized Shape Resonance On Silver Film Perforated By H-Shaped And More Complex Shaped Hole Arrays, Fang-Tzu Chuang 2011 National Taiwan University

Localized Shape Resonance On Silver Film Perforated By H-Shaped And More Complex Shaped Hole Arrays, Fang-Tzu Chuang

Fang-Tzu Chuang

The experimental results of light transmission through periodic array of H-shaped hole and more complicated hole which is a combination of multiple U shape are demonstrated. The observations indicate that the localized shape resonance in the longest resonant length of unfolded U-shaped part of the hole always appears. However, localized modes resonant in smaller U-shaped length don't always appear. Localized mode with non-U-shaped resonant path cannot be seen in our sample. In addition, localized mode with different order and resonant path can be excited by different polarized light.

The Role Of Dawn Song In Tree Swallows And Its Place In The Diversity Of Oscine Song Learning, Benjamin Nichols Taft 2011 University of Massachusetts Amherst

The Role Of Dawn Song In Tree Swallows And Its Place In The Diversity Of Oscine Song Learning, Benjamin Nichols Taft

Open Access Dissertations

Aspects of the behavioral ecology of bird song learning are examined in three parts. First, an approach from image analysis is extended to allow rapid, quantitative description of animal sounds. In this approach, sounds are summarized as sets of time-frequency-amplitude landmarks. Second, the role of dawn song in tree swallow (Tachycineta bicolor) breeding biology is examined. Song syllable sharing among tree swallows was found to be high among birds nesting at the same site, but sharing was lower between birds nesting at different sites. When birds nested at different sites, the distance between those sites was not related …

The Impact Of Tricaine Methanesulfonate, 2-Phenoxyethanol, And Carvone-Methyl Salicylate On The Innate Immune Response Of Zebrafish (Danio Rerio), Charles R. Wulff 2011 Colby College

The Impact Of Tricaine Methanesulfonate, 2-Phenoxyethanol, And Carvone-Methyl Salicylate On The Innate Immune Response Of Zebrafish (Danio Rerio), Charles R. Wulff

Honors Theses

Anesthesia plays a vital role in the maintenance of aquaculture species, where it is used to minimize stress during complex handling tasks such as transport, assessment, and harvesting. However, anesthetics have been shown to suppress the innate immune response, which could impact immunity and increase risk of infection. Tricaine methanesulfonate (MS-222) and 2-Phenoxyethanol (2-PE) represent two of the most commonly used anesthetics in aquaculture, with R-(+)-carvone, in the form of carvone-methyl salicylate (CMS) has recently been proposed as an alternative anesthetic for food fish. These three anesthetics were used to assess the influence of anesthetics on the immune system of …

Science For The Environment: Examining The Allocation Of The Burden Of Uncertainty, Elisa Vecchione 2011 LSHTM, UK

Science For The Environment: Examining The Allocation Of The Burden Of Uncertainty, Elisa Vecchione

Elisa Vecchione

The aim of this paper is to review the basic literature on scientific uncertainty in its statistical paradigm in order to provide enlightenment on one pivotal facet of the precautionary principle, i.e. the allocation of the burden of proof to demonstrate that an activity is not harmful to the environment. The purpose is not to explain a new theory of statistical inference, but to show how regulatory policymaking that is properly informed by scientific expertise and designed to avoid one type of error, may actually make other errors more likely and thus expose the public to danger. This problem is …

Zootecnia

Masters

The aim of this thesis was to investigate the effect of the VEGF family of growth factors and
cytokines TNF- α and IL-1β on oBRB function. The human RPE line ARPE-19 was
employed as an in vitro model of the oBRB. Two-Dimensional gel electrophoresis was
employed to study the effect of growth factors on the RPE proteome. The barrier properties of
the oBRB were evaluated by expression of the TJ proteins occludin or Zo-1, or claudins-1, 2,
3 and 4, determined by western blotting analysis. Claudins 2, 3 and 4 were found to be
significantly up-regulated by TNF-α, while interleukin …

Mechanistic And Signaling Analysis Of Muc4-Erbb2 Signaling Module: New Insights Into The Mechanism Of Ligand-Independent Erbb2 Activity, Goldi Kozloski 2010 University of Miami

Mechanistic And Signaling Analysis Of Muc4-Erbb2 Signaling Module: New Insights Into The Mechanism Of Ligand-Independent Erbb2 Activity, Goldi Kozloski

Goldi A Kozloski

The membrane mucin Muc4 is aberrantly expressed in numerous epithelial carcinomas and is currently used as a cancer diagnostic and prognostic tool. Muc4 can also potentiate signal transduction by modulating differential ErbB2 phosphorylation in the absence and in the presence of the ErbB3 soluble ligand heregulin (HRG-beta1). These features of Muc4 suggest that Muc4 is not merely a cancer marker, but an oncogenic factor with a unique-binding/activation relationship with the receptor ErbB2. In the present study, we examined the signaling mechanisms that are associated with the Muc4-ErbB2 module by analyzing ErbB2 differential signaling in response to Muc4 expression. Our study …
