Defining genetic diversity in the wake of the release of several Mycobacterium avium subsp. paratuberculosis (MAP) genome sequences has become a major emphasis in the molecular biology and epidemiology of Johne’s disease research. These data can now be used to define the extent of strain diversity on the farm. However, to perform these important tasks, researchers must have a way to distinguish the many MAP isolates/strains that are present in the environment or host to enable tracking over time. Johne’s disease (JD), or paratuberculosis is a fatal enteritis of animals caused by infection with Mycobacterium avium subspecies paratuberculosis (Map). There may be a long subclinical phase with no signs of clinical disease.

Diagnosis of JD is problematic and no test can reliably detect sub-clinical disease. Th1 responses to Map are believed to be activated first with a later switch to Th2 responses and progression to clinical disease. Detection of a cell-mediated response, indicated by interferon gamma (IFN-􏰁) produced in response to mycobacterial antigens, may give an early indication of infection. Although most pests of soybeans, Gycine max (L.), in the Northern Great Plains are managed using pesticides, farm management practices that encourage biodiversity offer promising long-term, sustainable solutions for controlling insect and weed pests profitably. The recent invasion of the Northern Great Plains by the soybean aphid (Aphis glycines Matsumura; Hemiptera: Aphididae) has had potentially important implications for insect communities in soybeans of this region, although recent descriptions of this regional community are scarce. This thesis studies intergenerational communication in farm families during the succession planning process. The research focuses on intergenerational relationships between fathers and sons. Due to the lack of research on intergenerational communication in farm families, this thesis examines the role communication plays in this relationship, and how communication affects the succession planning process of passing down the family farm. Communication accommodation theory is used to understand the communication processes taking place within the father-son relationship. Rhizoctonia blight is a common and serious disease of many turfgrass species. The most widespread causal agent, Thanatephorus cucumeris (anamorph: R. solani), consists of several genetically different subpopulations. In addition, Waitea circinata varieties zeae, oryzae and circinata (anamorph: Rhizoctonia spp.) also can cause the disease. Accurate identification of the causal pathogen is important for effective management of the disease. It is challenging to distinguish the specific causal pathogen based on disease symptoms or macroscopic and microscopic morphology. Food security and land required for food production largely depend on rate of yield gain of major cereal crops. Previous projections of food security are often more optimistic than what historical yield trends would support. Many econometric projections of future food production assume compound rates of yield gain, which are not consistent with historical yield trends. Here we provide a framework to characterize past yield trends and show that linear trajectories adequately describe past yield trends, which means the relative rate of gain decreases over time. A large array of literature has revealed the complexity of export performance analysis. Using the case of Ghana, this dissertation, divided into three essays, seeks to provide the methodological guidance, empirical and simulation evidence necessary to analyze a country’s export performance. The choice of Ghana was motivated by the country’s growth experience, strong export and agricultural sectors and implemented reforms and programs since 1983. The meat goat industry is one of the fastest growing agricultural sectors in the United States. However, there has been limited research on consumers’ preferences for goat meat; therefore, a choice experiment was used to assess consumer preferences for goat meat and/or live goats. A national online survey was conducted from April 27, 2012 to May 4, 2012. Respondents were asked to complete either the goat meat choice experiment, live goat choice experiment or both. This dissertation examines a movement in Western North Carolina to build a local food system, one grounded in the conditions and relationships of place. In 2000, Mountain Family Farms launched the Local Food Campaign to raise public awareness about the region's farms and farming heritage, to educate consumers about the benefits of buying food grown by local farms, and, ultimately, to build markets for locally grown food to sustain the region's farms.
