Mechanistic Studies Of Carbon Steel Corrosion Inhibition By Cashew Nut Shell Liquid, J Y N Philip, J Buchweishaija, L L Mkayula Dec 2002

Mechanistic Studies Of Carbon Steel Corrosion Inhibition By Cashew Nut Shell Liquid, J Y N Philip, J Buchweishaija, L L Mkayula

Tanzania Journal of Science

The inhibition mechanism of the Cashew Nut Shell Liquid (CNSL) on SAE 1008 carbon steel corrosion in CO 2  saturated 3% NaCl solutions has been studied by weight loss, UV/VIS and electrochemical techniques. The phenoxide, R-Ar-O- ions from the CNSL inhibitor were found to be responsible for the reduction of the corrosion rate of the carbon steel. Also, it was observed that the surface charge of the carbon steel electrodes was positive with respect to the solutions containing CNSL inhibitor. It is likely that the mechanism of the CNSL inhibitor adsorption involved the electrostatic attraction between the positively charged metal …

Preparation And Characterization Of Activated Carbons From Rice Husks And Shells Of Palm Fruits, Jeg Mdoe, Ll Mkayula

Tanzania Journal of Science

Activated carbons from palm fruit shells (PFS) and rice husks (RH) were prepared using physical and chemical activation processes, respectively. The influence of activation time and temperature (in the case of PFS) and impregnation ratio and carbonization temperature (in the case of RH) on the properties of the resultant carbons were studied. In the case of PFS carbons, there were an increase in adsorption capacity and surface areas with increase in activation time and temperature. The surface area ranged between 292 and 642 m2g-1 at temperature range of between 1000 and 1200 K. In the case of RH carbons, surface …

Calcium Binding To Calmodulin Mutants Monitored By Domain-Specific Intrinsic Phenylalanine And Tyrosine Fluorescence, W. S. Vanscyoc, B. R. Sorensen, E. Rusinova, William Laws, J. B. A. Ross, M. A. Shea Nov 2002

Calcium Binding To Calmodulin Mutants Monitored By Domain-Specific Intrinsic Phenylalanine And Tyrosine Fluorescence, W. S. Vanscyoc, B. R. Sorensen, E. Rusinova, William Laws, J. B. A. Ross, M. A. Shea

Chemistry and Biochemistry Faculty Publications

Cooperative calcium binding to the two homologous domains of calmodulin (CaM) induces conformational changes that regulate its association with and activation of numerous cellular target proteins. Calcium binding to the pair of high-affinity sites (III and IV in the C-domain) can be monitored by observing calcium-dependent changes in intrinsic tyrosine fluorescence intensity (lambda(ex)/lambda(em) of 277/320 nm). However, calcium binding to the low-affinity sites (I and II in the N-domain) is more difficult to measure with optical spectroscopy because that domain of CaM does not contain tryptophan or tyrosine. We recently demonstrated that calcium-dependent changes in intrinsic phenylalanine fluorescence (lambdaex/lambdaem of …

Guanine And 7,8-Dihydro-8-Oxo-Guanine-Specific Oxidation In Dna By Chromium(V), Kent D. Sugden, Brooke Martin Oct 2002

Guanine And 7,8-Dihydro-8-Oxo-Guanine-Specific Oxidation In Dna By Chromium(V), Kent D. Sugden, Brooke Martin

Chemistry and Biochemistry Faculty Publications

The hexavalent oxidation state of chromium [Cr(VI)] is a well-established human carcinogen, although the mechanism of cancer induction is currently unknown. Intracellular reduction of Cr(VI) forms Cr(V), which is thought to play a fundamental role in the mechanism of DNA damage by this carcinogen. Two separate pathways of DNA damage, an oxidative pathway and a metal-binding pathway, have been proposed to account for the lesions observed in cell systems. We have used a model Cr(V) complex, N,N´-ethylenebis(salicylidene-animato)oxochromium(V) [Cr(V)-Salen], to investigate the oxidative pathway of DNA damage and to elucidate the lesions generated from this oxidation process. Reaction of Cr(V)-Salen with …

A Comparison Of The Low Mode And Monte Carlo Conformational Search Methods, Carol A. Parish, Rosina Lombardi, Kent Sinclair, Emelyn Smith, Alla Goldberg, Melissa Rappleye, Myrianne Dure

Chemistry Faculty Publications

The Low Mode (LM) and Monte Carlo (MC) conformational search methods were compared on three diverse molecular systems; (4R, 5S, 6S, 7R)-hexahydro-5,6-dihydroxy-1,3,4,7-tetrakis(phenylmethyl)-2H-1,3-diazapin-2-one (1), 2-methoxy-2-phenyl-2-triflouromethyl-N-α-methyl benzyl propanamide (2) and a trimeric 39-membered polyazamacrolide (3). We find that either method, or a combination of the methods, is equally efficient at searching the conformational space of the smaller molecular systems while a 50:50 hybrid of Low Mode and Monte Carlo is most efficient at searching the space of the larger molecular system.

Monte Carlo Simulation On The Indirect Exchange Interactions Of Co-Doped Zno Film, Abraham F. Jalbout, Hanning Chen, Scott L. Whittenburg Sep 2002

Monte Carlo Simulation On The Indirect Exchange Interactions Of Co-Doped Zno Film, Abraham F. Jalbout, Hanning Chen, Scott L. Whittenburg

Chemistry and Biochemistry Faculty Publications

Monte Carlo simulations using a three-dimensional lattice model studied the Ruderman–Kittel–Kasuya–Yosida (RKKY) indirect exchange interaction of doped magnetic Co ions in ZnOfilms. The results of the calculations show that the RKKY interaction in Co-doped ZnO is long ranged and its magnitude is proportional to (inverse of the distance from a central ion). The sign oscillates with a frequency that depends on the concentration of the carrier. The long-distance sum of the RKKY indirect exchange energies is positive indicating that these materials are ferromagnetic, in direct correlation with previously reported results.

Communicator-In-Chief: Presidential Use Of Television Past, Present, And Future, Jenna Wasson

Honors Theses

This thesis seeks to determine how television has changed as a communication medium for presidents over the past half century. An evaluation of the evolving ways presidents use television to communicate with and to build support from the American people has been conducted. Presidential communication strategies have been identified by drawing primarily from primary sources written by presidents and White House staff. Television technology and the television audience have changed over the years. Presidents have taken a more pro-active, aggressive role in their efforts to harness television for their own purposes. Why have these changes occurred? What impact have these …

Production Of A Monoclonal Antibody Against Benzo[Α]Pyrene Diol Epoxide Dna Adducts, Brian Peden Austin Apr 2002

Production Of A Monoclonal Antibody Against Benzo[Α]Pyrene Diol Epoxide Dna Adducts, Brian Peden Austin

Chemistry & Biochemistry Theses & Dissertations

Benzo[α]pyrene is a ubiquitous pollutant produced from the incomplete combustion of organic material such as fossil fuels. It is found in the workplace, urban air, drinking water, and the food supply. Recently, it has been proposed that benzo[α]pyrene may be the causative agent in the formation of lung adenocarcinomas among some Taiwanese women exposed to cooking oil fumes without adequate ventilation. In this study, calf thymus DNA was modified in vitro with benzo[α]pyrene-diol epoxide (BPDE) to a level consistent with that found in biological samples. This DNA of low modification was used as an immunogen in the production of a …

Faculty Publications, Chemistry

We report the thermal stability of wild type (WT) and 14 different variants of human copper/zinc superoxide dismutase (SOD1) associated with familial amyotrophic lateral sclerosis (FALS). Multiple endothermic unfolding transitions were observed by differential scanning calorimetry for partially metallated SOD1 enzymes isolated from a baculovirus system. We correlated the metal ion contents of SOD1 variants with the occurrence of distinct melting transitions. Altered thermal stability upon reduction of copper with dithionite identified transitions resulting from the unfolding of copper-containing SOD1 species. We demonstrated that copper or zinc binding to a subset of “WT-like” FALS mutants (A4V, L38V, G41S, G72S, D76Y, …

Calorimetric Study Of Block-Copolymers Of Poly(N-Butyl Acrylate) And Gradient Poly(N-Butyl Acrylate-Co-Methyl Methacrylate), A I. Buzin, M Pyda, P Costanzo, K Matyjaszewski, B Wunderlich

Chemistry and Biochemistry

The nanophase separation in diblock and triblock copolymers consisting of immiscible poly(n-butyl acrylate) (block A) and gradient copolymers of methyl methacrylate (MMA) and n-butyl acrylate (n BA) (block M/A) were investigated by means of their heat capacity, Cp, as a function of the composition of the blocks M/A and temperature. In all copolymers studied, both blocks are represented by their Cp and glass transition temperature, Tg, as well as the broadening of the transition-temperature range. The low-temperature transition of the blocks A is always close to that of the …

Induction Of Myocarditis And Valvuluitis In Lewis Rats By Different Epitopes Of Cardiac Myosin And Its Implications In Rheumatic Carditis, Jeffrey E. Galvin, Mark E. Hemric, Stanley D. Konsanke, Stephen M. Factor, Anthony Quinn, Madeleine W. Cunningham

Faculty Publications and Presentations

Immune responses against cardiac myosin and group A streptococcal M protein have been implicated in the pathogenesis of rheumatic heart disease. Although cardiac myosin is known to produce myocarditis in susceptible animals, it has never been investigated for its role in production of valvular heart disease, the most serious sequelae of group A streptococcal infection in acute rheumatic fever. In our study, cardiac myosin induced valvulitis in the Lewis rat, and epitopes responsible for production of valvulitis were located in the rod region. Human and rat cardiac myosins induced severe myocarditis in the Lewis rats as expected. A purified S2 …

Sources And Diagenetic Alteration Of Organic Matter In Coastal Waters At The Vicinity Of Dar Es Salaam, Tanzania, Ann Muzuka Dec 2001

Sources And Diagenetic Alteration Of Organic Matter In Coastal Waters At The Vicinity Of Dar Es Salaam, Tanzania, Ann Muzuka

Tanzania Journal of Science

The stable isotope compositions of organic carbon (OC) and nitrogen, contents of OC and nitrogen, and C/N ratios for four short cores, which were recovered from Msansani Bay (WO 57), off Msimbazi River (WO 76) and Dar es Salaam harbour (WO 62 and WO 74), are used to document temporal variation in the sources and diagenetic alteration of organic matter (OM). The δ15N and δ13C values at sites WO 57, WO 62 and WO 74 show a trend of down-core decrease to the base of the core, and such a down-core trend could be related to diagenetic loss of compounds …

Physicochemical Characteristics Of Some Locally Manufactured Edible Vegetable Oils Marketed In Dar Es Salaam, F N Ngassapa, O C Othman

Tanzania Journal of Science

The physicochemical properties (refractive index, saponification value, Free Fatty Acid (FFA) value, iodine value, acid value, peroxide value and tocopherol content) of seven edible vegetable oils manufactured in Tanzania, were assessed using standard procedures of the Association of Official Analytical Chemists (AOAC). Also determined were the levels of these properties with respect to shelf storage of the products. When compared with standard values recommended by the Codex Alimentarius Commission of FAO/WHO and with other published results, the locally produced vegetable oils were found to be of fair quality but of poor storage stability.

Intrasteric Inhibition Of Atp Binding Is Not Required To Prevent Unregulated Autophosphorylation Or Signaling By The Insulin Receptor, Mark Frankel, Ararat J. Ablooglu, Joseph W. Leone, Elena Rusinova, J. B. A. Ross, Robert L. Heinrikson, Ronald A. Kohanski Jul 2001

Kohanski

Chemistry and Biochemistry Faculty Publications

Receptor tyrosine kinases may use intrasteric inhibition to suppress autophosphorylation prior to growth factor stimulation. To test this hypothesis we made an Asp1161Ala mutant in the activation loop that relieved intrasteric inhibition of the unphosphorylated insulin receptor (IR) and its recombinant cytoplasmic kinase domain (IRKD) without affecting the activated state. Solution studies with the unphosphorylated mutant IRKD demonstrated conformational changes and greater catalytic efficiency from a 10-fold increase in kcat and a 15-fold-lower Km ATP although Km peptide was unchanged. Kinetic parameters of the autophosphorylated mutant and wild-type kinase domains were virtually identical. The Asp1161Ala mutation increased the …

Complex Effects Arising In Smoke Plume Simulations Due To Inclusion Of Direct Emissions Of Oxygenated Organic Species From Biomass Combustion, Sherri A. Mason, Richard J. Field, Robert J. Yokelson, Michael A. Kochivar, Mark R. Tinsley, Darold E. Ward, Wei Min Hao Jun 2001

Complex Effects Arising In Smoke Plume Simulations Due To Inclusion Of Direct Emissions Of Oxygenated Organic Species From Biomass Combustion, Sherri A. Mason, Richard J. Field, Robert J. Yokelson, Michael A. Kochivar, Mark R. Tinsley, Darold E. Ward, Wei Min Hao

Chemistry and Biochemistry Faculty Publications

Oxygenated volatile organic species (oxygenates), including HCOOH, H2CO, CH3OH, HOCH2CHO (hydroxyacetaldehyde), CH3COOH, and C6H5OH, have recently been identified by Fourier transform infrared measurements as a significant component of the direct emissions from biomass combustion. These oxygenates have not generally been included in the hydrocarbon-based initial emission profiles used in previous photochemical simulations of biomass combustion smoke plumes. We explore the effects of oxygenates on this photochemistry by using an established initial emission hydrocarbon profile and comparing simulation results obtained both with and without addition of the above six …

Shape Imprinting Due To Variable Disulfide Bonds In Polyacrylamide Gels, Andrew B. Greytak, Alexander Y. Grosberg, Toyoichi Tanaka Jun 2001

Shape Imprinting Due To Variable Disulfide Bonds In Polyacrylamide Gels, Andrew B. Greytak, Alexander Y. Grosberg, Toyoichi Tanaka

Faculty Publications

Through the use of variable disulfide crosslinkers, we have created polyacrylamide gels whose shape can be altered after polymerization. N,N'-bisacryloylcystamine is incorporated as a crosslinker, along with a smaller amount of a permanent crosslinker. After polymerization, the disulfide bonds are cleaved into thiols through reduction. By reoxidizing the thiols with the gel held in a new macroscopic shape, a new set of disulfide bonds is formed, and the gel is forced to adopt the new shape. Retension of the new shape improves with greater distortion from the original shape, as well as with increased concentration of variable …

Intrinsically Bent Dna In The Promoter Regions Of The Yeast Gal1–10 And Gal80 Genes, Ralph C. Bash, Jeff Vargason, P. Shing Ho, D. Lohr Jan 2001

Intrinsically Bent Dna In The Promoter Regions Of The Yeast Gal1–10 And Gal80 Genes, Ralph C. Bash, Jeff Vargason, P. Shing Ho, D. Lohr

Faculty Publications - Department of Biological & Molecular Science

Circular permutation analysis has detected fairly strong sites of intrinsic DNA bending on the promoter regions of the yeast GAL1–10 and GAL80 genes. These bends lie in functionally suggestive locations. On the promoter of the GAL1–10 structural genes, strong bends bracket nucleosome B, which lies between the UASG and the GAL1 TATA. These intrinsic bends could help position nucleosome B. Nucleosome B plus two other promoter nucleosomes protect the TATA and start site elements in the inactive state of expression but are completely disrupted (removed) when GAL1–10 expression is induced. The strongest intrinsic bend (;70°) lies at the downstream edge …

Production Of A Novel Copper-Binding Ligand By Marine Synechococcus (Cyanobacteria) In Response To Toxic Concentrations Of Copper, Arunsi C. Brown, Andrew S. Gordon

Virginia Journal of Science

Marine Synechococcus spp. are extremely sensitive to copper toxicity. Some strains have been shown to produce high-affinity, extracellular ligands of unknown structure which form complexes with free cupric ion. They are also known to produce metallothioneins (MT) in response to cadmium and zinc stress. In the present study, marine Synechococcus PCC 73109 (Agmenellum quadruplicatum BG-1) (Van Baalen) was exposed to three concentrations of CuSO4 for various times. Size exclusion chromatography, atomic absorption spectrophotometry, and reverse phase HPLC were used to isolate an intracellular copper binding ligand of low molecular weight (< 6,500 Da). The ligand was detected after exposure to ≥ 8 μM CuSO4 for 2 hr in BG-11 medium. …

Use Of Py-Gc/Ms Analysis Techniques In Animal Waste Management: A Preliminary Survey Of Dairy Manures, Daniel L. Vaughn, Michael A. Kruge Jan 2001

Use Of Py-Gc/Ms Analysis Techniques In Animal Waste Management: A Preliminary Survey Of Dairy Manures, Daniel L. Vaughn, Michael A. Kruge

Department of Earth and Environmental Studies Faculty Scholarship and Creative Works

The increasing practice of industrial-scale agriculture tends to concentrate large masses of animal waste in relatively compact areas, potentially leading to excessive release of polluting nutrients into waterways during major storms. Anaerobic treatment conditions are generally favored to conserve nitrate N as an agricultural commodity. However, overall N contents in waste are often in excess of crop fertilization needs: storing excess N in soluble nitrate form increases pollution potential. Thus the perceived needs of agriculture and society-at-large become at odds. Organic nitrogen forms (e.g., proteins) are more environmentally stable and are less subject to unintentional release. Although U.S. farmers tend …

Analysis Of Wine For Penicillin, Gary Morris, James Yuan, Roy Williams Jan 2001

Analysis Of Wine For Penicillin, Gary Morris, James Yuan, Roy Williams

Chemistry & Biochemistry Faculty Publications

This study addresses the question of whether the antibiotic Penicillin, which is produced by the common mold Penicillium notatum, could possibly become a contaminate of wine during the fermentation process. The significance of this study is related to the potential health effects this agent might produce in those consumers who have an allergic response to Penicillin. It has been estimated that between 6% and 8% of the American population is subject to this type of allergic response. A method is developed for the detection of penicillin in wines using high-pressure liquid chromatography. We demonstrate that penicillin G hydrolyzes rapidly …

Identification Of The Domains Of Urer, An Arac-Like Transcriptional Regulator Of The Urease Gene Cluster In Proteus Mirabilis, Carrie A. Poore, Christopher Coker, Jonathan D. Dattelbaum, Harry L.T. Mobley

Chemistry Faculty Publications

Proteus mirabilis urease catalyzes the hydrolysis of urea to CO2 and NH3, resulting in urinary stone formation in individuals with complicated urinary tract infections. UreR, a member of the AraC family, activates transcription of the genes encoding urease enzyme subunits and accessory proteins, ureDABCEFG, as well as its own transcription in the presence of urea. Based on sequence homology with AraC, we hypothesized that UreR contains both a dimerization domain and a DNA-binding domain. A translational fusion of the leucine zipper dimerization domain (amino acids 302 to 350) of C/EBP and the C-terminal half of UreR …

Heavy Metals And Organic Pollutants In Sediments Of Dar Es Salaam Harbour Prior To Dredging In 1999, J F Machiwa

Tanzania Journal of Science

Heavy metals and organic contaminants were analysed in the sediments of the inner area of the Dar es Salaam harbour. Complementary analytical parameters, such as, the organic carbon content and the silt/clay fraction of the sediments showed good positive correlation (r = 0.64). Stations that had high content of fine grain material in the sediment also indicated relatively high level of organic carbon and pollutants. The Florida criteria (MacDonald 1993) for assessment of pollution of tropical marine sediments was adopted in oredr to evaluate the extent of pollution in Dar es Salaam harbour sediments. The Florida criteria is one of …
