Sarcocystis neurona is a protozoan parasite that causes the serious neurologic disease equine protozoal myeloencephalitis (EPM). The life cycle of S. neurona progresses through multiple developmental stages that differ morphologically and molecularly. The S. neurona merozoite surface is covered by multiple related proteins, which are orthologous to the surface antigen (SAG) gene family of Toxoplasma gondii. The SAG surface antigens in T. gondii and another related parasite Neospora caninum are life cycle stage-specific and seem necessary for parasite transmission and persistence of infection. Two types of placentitis were studied: ascending and nocardioform placentitis. Although the first diagnosis of nocardioform placentitis was made three decades ago, little is known about the disease, due to the lack of an experimental model. In attempt to develop a research model, Crossiela equi was inoculated through intrauterine, intravenous, intrapharyngeal, and oral routes, but none of the routes resulted in nocardioform placentitis. This may indicate that unidentified factors may play a role in disease pathogenesis and that simple presence of bacteria is not sufficient to induce nocardioform placentitis. Severe bacterial sepsis leads to a pro-inflammatory condition that can manifest as septic shock, multiple organ failure, and death. Neutrophils are critical for the rapid elimination of bacteria, however, the role of neutrophil chemoattractant CXCL1, pattern recognition receptors (PRRs)- NLR protein 3 (NLRP3) and alcohol in bacterial clearance during sepsis remains elusive. We demonstrate that CXCL1 plays a pivotal role in mediating host defense to polymicrobial sepsis following cecal ligation and puncture (CLP) in gene-deficient mice. CXCL1 appears to be essential for restricting bacterial outgrowth and preventing multiple organ failure and death in mice. Moreover, CXCL1 is essential for neutrophil …

Herpes simplex virus type 1 (HSV-1) is a ubiquitous neurotropic alphaherpesvirus transmitted by contact with mucocutaneous surfaces of infected individuals. HSV-1 enters the host by fusion of the viral envelope with the host cell plasma membrane, followed by translocation of the viral capsids to the nucleus where viral DNA is injected into the host cell nucleus to initiate viral replication. To generate infectious virions, newly assembled capsids travel to the cytoplasm and undergo a process called secondary envelopment by budding into cytoplasmic vesicles derived from the trans-Golgi network. Parasitology is an important area of veterinary medicine, but the risk factors for high parasite loads are not well-understood. Equine intestinal parasites can cause extensive disease and death. In the current study, the relationship between equine intestinal parasite loads and adherence to veterinary guidelines for equine management practices was studied. Satisfactory adherence to guidelines regarding food, pasture, and flooding management was related to lower parasite loads. Adherence to guidelines regarding deworming, quarantine, bedding, and water did not appear to lower parasite loads. Monocyte chemoattractant protein-1 is critical for monocyte recruitment to the lungs in response to bacterial infection. MCP-1 is also essential for protective neutrophil recruitment to the lungs during Escherichia coli and Klebsiella pneumoniae infection. Staphylococcus aureus pneumonia, specifically strain USA300, carries a high morbidity and mortality rate and is an important pathogen in hospital/ventilator and community acquired pneumonia. In the current study, we investigated the role of MCP-1 in pulmonary innate immunity to S. aureus in C57Bl/6, MCP-1-/- and MCP-1 AB blocked mice. Lidocaine, an amide local anesthetic agent, is commonly used in mammals, including humans. There is a general assumption that birds are more sensitive to lidocaine than mammals. Relatively low doses of lidocaine have been suggested to cause toxic effects in birds. While this information appears to be anecdotal, it has been perpetuated in the literature. Spayed heifers were developed into yearlings by grazing corn residue and bromegrass, followed by native range, and were finished on a common diet. Treatments were 2 lb or 5 lb of wet distillers grains with solubles (WDGS; DM basis) supplement on corn residue daily, and modified distillers grains with solubles (MDGS) fed at 0.6% BW daily or no MDGS during summer grazing. Feeding 5 lb increased winter ADG by 0.68 lb (year 1) or 0.40 lb (year 2) compared to 2 lb, and increased HCW after finishing. A finishing experiment evaluatedsubstitution of corn silage and modified distillers grains with solubles (MDGS) in place of corn. The treatment arrangementwas a 2 X 2 + 1 factorial with 15 or 45% corn silage and 20 or 40% MDGS plus a control containing 5% cornstalks and 40% MDGS. There were interactions between corn silage and MDGS for carcass adjusted performance. As corn silage inclusion was increased in the diet, F:G increased when fed with 20% MDGS, however there was no difference when fed with 40% MDGS.

Individually fed cattle were adapted to high grain diets with a traditional grain adaptation program or one of two RAMP® adaptation programs. RAMP programs adapted cattle to a finishing diet gradually over 28 days in four steps or directly without an adaptation. Feed intake variance among d was greater for traditionally adapted cattle compared to either RAMP program, but DMI was not different during the adaptation period. Over the 138-day period, feedlot performance and carcass traits were not affected by adaptation treatment. The effects of feeding a complete pelleted feed to newly received steer calves (585 ± 4 lb; n = 1318 ) was compared to a control ration consisting of 32% (DM basis) wet or modified distillers grains, 32% alfalfa hay, 32% dry-rolled corn, and 4% supplement. The pelleted complete feed consisted of 35% corn residue and a blend of grain byproducts and minerals. Feeding the complete pelleted feed increased DMI but decreased ADG, thereby reducing feed efficiency. The pelleted feed numerically reduced morbidity. Feeding a complete pellet consisting of corn residue appears to be a viable option for receiving calves …

Yearling Angus heifers at a commercial ranch in the Nebraska Sandhills were randomly assigned to one of two progestin-based fixed-time AI protocols (MGA or 14-day CIDR) to compare pregnancy rates. Heifers had similar fixed-time AI pregnancy rates between MGA and 14-day CIDR. A similar proportion of MGA and 14-day CIDR heifers displayed a second estrus; however, heifers previously synchronized with MGA tended to have a greater second AI pregnancy rate. Overall pregnancy rate was similar between MGA and 14-day CIDR treatments. The MGA system was the more cost effective synchronization protocol in this study.

Cattle fed a de-oiled wet distiller’s grains plus solubles (WDGS) diet were compared to cattle fed corn or traditional (full-fat) WDGS diets to determine effects on discoloration, oxidation, and tenderness of beef aged for seven and 21 days. At seven days of aging, dietary treatment had no effect on discoloration. At 21 days of aging, beef from cattle fed de-oiled WDGS had less oxidation than the corn control and several of the full-fat WDGS treatments. Although tenderness increased with aging and retail display, dietary treatment had no effect on tenderness. These findings suggest that these dietary treatments, followed by a …

The objective of this trial was to evaluate the effect of feeding different concentrations of wet distillers grains during winter backgrounding and either modified wet distillers grains or Sweet Bran® during the finishing phase on ground beef color and lipid oxidation. After a 14 day aging period, ground beef patties were made and placed in a simulated retail display for seven days. Cattle were grazed without or with energy supplementation of wet distillers grains with solubles (WDGS) during backgrounding, and were finished on a corn-based diet with or without 35% WDGS. Ground beef patties were made from shoulder clods, cooked, and stored in a refrigerated or frozen state. Cattle supplemented with WDGS had greater lipid oxidation in cooked ground beef patties regardless of finishing diet or storage type.

The community composition of the fecal microbiota was compared between beef steers which were high-shedders and low-shedders of Shiga toxin-producing Escherichia coli. Based on Shannon and Chao 1 diversity indices, the high-shedders had a more diverse fecal bacterial community than the low-shedding steers. Members of the genus Prevotella were observed as being more abundant in the low-shedders compared to the high-shedders, while Succinivibrio were more abundant in the high-shedders. Isolation of specific bacteria which are significantly more abundant in low-shedders may pave the way to developing direct-fed microbials which are effective in reducing STEC shedding among high-shedding beef steers.

Seven research trials conducted over five years at the University of Nebraska–Lincoln were summarized to determine how animal performance changes through the finishing period on a live and carcass weight basis. Live weight, carcass weight, carcass ADG, and carcass feed efficiency all changed quadratically throughout the feeding period; live ADG and live feed efficiency declined linearly. During times of negative profit margins, optimal profitability for steers marketed on a live-basis occurred by selling early, whereas optimal profitability was achieved by feeding steers marketed on a carcass-basis longer.

Crossbred steers (n = 64) were grazed on warm- or cool-season grass-dominated pastures, without or with energy supplementation of wet distillers grains with solubles (WDGS), and were finished on a corn-based diet with or without 35% WDGS. Finishing on corn increased desirable flavor notes and decreasedundesirable flavor notes in both L. dorsi and B. femoris steaks. In addition, grazing on warm-season grasses increased the prevalence of undesirable flavors but was often dissipated by the addition WDGS supplementation. Longer aging periods tended to increase the prevalence of undesirable flavors, especially in B. femoris steaks. It is recommended producers provide WDGS supplementation, …

To determine sequence characteristics and phylogenetic relationships of the nonstructural (NS) gene, the NS genes of 9 avian influenza (AI) viruses were sequenced and compared with other Iranian AI H9N2 viruses available in GenBank. The similarity among the earliest Iranian H9N2 isolate (A/chicken/Iran/ZMT-101/1998) and recent (2007–2011) studied isolates was low (94%). Some of the studied viruses revealed substitutions in RNA-binding and effector domains as well as antigenic regions of the NS protein. Recently studied isolates have a high similarity (97.4%-98.7%) with some Pakistani isolates (like A/chicken/Sawabi/NARC-2434/2006) in the NS gene. The aim of the present study was to compare the effectiveness of cefuroxime, herd-specific autovaccine, and cefuroxime/herd-specific autovaccine treatments in eliminating S. aureus from the milk of 45 cows with subclinical mastitis. In our early work, one mutation (A898G, which caused Ser32Gly (S32G) change of mature peptide) was identified in the goat BMP15 gene and the allele G was associated with high litter size in Jining Grey goats (the goat breed with the highest fecundity in China). The aim of this study was to find polymorphisms in the actin alpha 1 gene (ACTA1) in horses belonging to different breeds and utility types and to compare them. The genomic region of the actin alpha 1 gene was positively selected in purebred horses. By sequencing part of the ACTA1 gene, 2 polymorphic sites located in intron 5 (G > T) and intron 6 (G > C) were found. The objective of this study was to evaluate the genetic and phenotypic parameters for daily milk yield (DMY) in Black Bengal does. Genetic parameters were estimated using a residual maximum likelihood procedure, fitting an animal model while ignoring maternal genetic or permanent environmental effects. The least squares mean for DMY was 354.76 g. DMY was significantly (P < 0.01) affected by parity, litter size, and season of kidding. The heritability of DMY was low (0.15) and the fact that the trait can only be measured in females suggests that progeny testing may be the best sire selection method for this trait.

The present study compared superovulation and embryo recovery rates applied repeatedly during breeding and nonbreeding seasons in the same 20 Kivircik sheep. Trials 1 and 3 were performed during the breeding season and the trial 2 was performed in the nonbreeding season. We synchronized the sheep's estrus cycles and induced a superovulatory response with porcine follicle stimulating hormone (pFSH) injections. The aim of the study was to use hexapeptide combinatorial libraries to evaluate the influence of high-abundance protein depletion on low-abundance protein enrichment, protein concentration, and 2D gel quality in plasma and serum proteomes of growing piglets and calves. Depleted and nondepleted samples of plasma/serum were resolved using 2D electrophoresis (2DE). Bioinformatic analysis of the 2D gels showed an increased number of detected spots on each gel after depletion of excess high-abundance proteins in calf plasma (+26.46%) and in piglet serum (+49.40%). Twenty male Barbados lambs (3 months old) were used to evaluate the effects of sodium bicarbonate (NaHCO_3) supplementation on growth performance and carcass characteristics of lambs fed concentrate diets in controlled-environment chambers set at 20 °C and 30 °C. Phenotypic and genetic parameters among udder traits (udder depth, UD; median suspensory line, MS; fore udder attachment, FUA; rear udder height, RUH; rear udder width, RUW), final score (FS), and milk production traits (305-day milk yield, 305 DM; 305-day milk-fat yield, 305 DF; 305-day milk-protein yield, 305 DP) were estimated using a derivative-free restricted maximum likelihood procedure with an animal model.
