Production And Characterization Of Monoclonal Antibodies Against Botulinum Toxin Produced In Nicotiana Benthamiana, Kornchanok Sangprasat

Botulism is an uncommon but serious neurological condition caused by the botulinum toxin produced by the gram-positive bacterium named Clostridium botulinum. Common symptoms of botulism include muscle cramps, nausea, vomiting, diarrhea, difficulty swallowing, respiratory failure, muscle weakness, and potentially death. Currently, two types of antitoxin are used: equine-derived heptavalent antitoxin and human-derived immunoglobulin (BabyBIG®). However, treatment with heptavalent antitoxin may lead to hypersensitivity reactions, while BabyBIG®, which is produced in low yield, is used only for children. Background: Despite significant progress in diabetes research, there remain significant health disparities between Hispanic/Latino patients in the United States, particularly those with lower socioeconomic status, compared to non-Hispanic white patients. Previous studies have identified potential reasons behind these disparities, including the need for a multifaceted approach that addresses social determinants of health (SDOH) faced by patients. Social cognitive theory (SCT) stipulates that interventions that promote self-management by improving knowledge, self-efficacy, and problem-solving skills can be successful in improving health outcomes. Natural products are of great significance to the pharmaceutical industry in the development of new drugs. Diquinane or bicyclo[3.3.0]octane is a conspicuous structural unit existing in the carbo-frameworks of a wide range of natural products such as alkaloids and terpenoids. These diquinane-containing molecules not only exhibit intriguing architectures, but also showcase a broad spectrum of significant bioactivities, which draw widespread attention from the global synthetic community. This study aimed to investigate the drug-polymer interactions between simvastatin (SIM) and three polymers [PVP-VA, HPMC-AS, and Soluplus® (SOL)] in amorphous solid dispersions (ASDs) prepared by hot melt extrusion (HME), and to understand the implications on physical stability. ASDs are of significant interest within the pharmaceutical industry for improving drug bioavailability. However, the amorphous nature of a drug in an ASD presents physical stability challenges. Pharmacogenomics research has predominantly focused on populations of European ancestry, limiting the application for diverse populations such as American Indian and Alaska Native (AIAN) communities. Our community-centric study aims to understand perspectives of utilizing pharmacogenomics to guide tobacco cessation in an AIAN community using a survey with qualitative and quantitative components. We assessed participant (n=273) tobacco usage and cessation history, pharmacogenomics knowledge, and perceptions of utilizing pharmacogenomics in the context of tobacco cessation. We found that the majority of participants (92%) were aware of risks associated with tobacco usage and believe it to be a problem within their community (76%). …

Although considered a rare form of nonmelanoma skin cancer, Merkel Cell Carcinoma incidence rate has increased to 0.74 per 100,000 and projections expect this number to steadily rise over the next decades and to reach an average of 3,285 cases per year in 2025. MCC incidence increases significantly with age. Among those aged 40-44, it is 0.44, it rises to 1.0/100,000 in the 60-64 age group and soars to 9.8/100,000 among those ≥85 years and above. Prostate cancer is the leading cancer in men. Continued tobacco use after prostate cancer diagnosis may affect a patient’s treatment outcomes, quality of life, and mortality. Despite the potential negative outcomes, 10-20% of prostate cancer patients continue to use tobacco after their cancer diagnosis. Abstract Cannabis extract, one of the herbal medicine products on the national list of essential medicines, offers various therapeutic benefits; however, it has unpleasant taste, and the products are available as liquid preparation which can significantly reduce patient compliance and drug effectiveness. The objectives of this study were to develop film formulations and characterizations composed of tetrahydrocannabinol and taste-masking polymers and develop tetrahydrocannabinol-coated granules via a fluidized bed-coating. Eudragit E100 (E100) and Kollidon VA64 (KVA64) were used as taste-masking polymers. Carpaine, a major alkaloid present in Carica papaya leaves, has been shown to increase platelet counts in patients suffering from thrombocytopenia. Numerous commercial papaya leaf products are available, but few provide proper bioactive ingredient information. We present herein a technique for rapid screening of carpaine in these products using DART-MS. The results indicate that carpaine was detected in various forms (powder, solution) of papaya leaves. Its presence was confirmed by examining the mass pattern when conducted on a standard solution at both low and high voltages (+10 V and +90 V), using MS1 and MS2 data obtained from LC-QTOF-MS/MS. BACKGROUND: Neuropsychiatric disorders, such as mood disorders, schizophrenia, and Alzheimer's disease (AD) and related dementias, are associated to significant morbidity and mortality worldwide. The pathophysiological mechanisms of neuropsychiatric disorders remain to be fully elucidated, which has hampered the development of effective therapies. The Renin Angiotensin System (RAS) is classically viewed as a key regulator of cardiovascular and renal homeostasis. While the concurrent use of multiple prescriptions (polypharmacy) is common for those with chronic conditions, professional ownership is lacking when it comes to tapering or discontinuing potentially inappropriate medications. Our growing elderly population is especially vulnerable to negative risks associated with polypharmacy. Pharmacists are highly educated and skilled healthcare professionals working as critical touchpoints with patients; yet they are not empowered to deprescribe potentially inappropriate medications directly. This qualitative research study examined the perceived barriers and facilitators pharmacists encounter when pursuing deprescribing recommendations and initiatives with prescribers, healthcare staff, patients, and caregivers. This article provides an early analysis of the potential for creating future biosimilar competition for cell and gene therapies (CGTs) to lower prices and improve patient access, building on a unique set of interviews with relevant experts. Our discussion addressed regulatory, manufacturing, intellectual property, and market size challenges. Due to CGTs’ complexity, meeting the regulatory requirement of ‘high similarity with no clinically meaningful differences’ will be difficult. Gene therapies are likely better candidates for biosimilar development than cell therapies. Biosimilarity should be met when gene therapy biosimilars contain the same genetic sequence as a reference product, and the variability in …

Abstract: Accurately monitoring pharmacy inventory in real-time is vital to providing safe and cost-effective medical care at hospitals of any size. At smaller critical access hospitals, in particular, effective inventory management requires solutions to unique problems posed by floorspace, budget, and workflow limitations. In 2022, Providence Health System (PHS) began systemwide implementation of both a standardized inventory management solution (IMS) and an AI-assisted medication ordering software (MOS). Abstract: Approximately 10% of patients with recent percutaneous intervention (PCI) have concomitant atrial fibrillation (AF) and others may have concomitant venous thromboembolism (VTE). This makes choosing the optimal antithrombotic regimen challenging. This study was conducted to evaluate the subchronic toxicity of the leaf extract of ki hampelas. The extract was orally administered to male and female Sprague-Dawley rats at doses of 50, 200, and 400 mg/kg bodyweight (BW) per day for 28 days. Lysosomes and the endoplasmic reticulum (ER) are Ca2+ stores mobilized by the second messengers NAADP and IP3, respectively. Here, we establish Ca2+ signals between the two sources as fundamental building blocks that couple local release to global changes in Ca2+. Cell-wide Ca2+ signals evoked by activation of endogenous NAADP-sensitive channels on lysosomes comprise both local and global components and exhibit a major dependence on ER Ca2+ despite their lysosomal origin. Knockout of ER IP3 receptor channels delays these signals, whereas expression of lysosomal TPC2 channels accelerates them. Osteoporosis represents a significant global public health issue, particularly among the aging population. Its incidence reaches 18.3% of the total population, with the highest prevalence observed in elderly postmenopausal women. A key factor in osteoporosis is the decreased expression level of estrogen receptor alpha (ERα), attributed to its degradation by the ubiquitin ligase protein complex Cullin4B (CUL4B), DNA damage binding 1 (DDB1), and aryl hydrocarbon receptor (AhR), collectively known as CUL4BAhR. Acanthus ilicifolius L contains compounds exhibiting antiosteoporosis activity, primarily by inhibiting osteoclastogenesis via RANKL. Rosella, also known as Hibiscus sabdariffa L., is a plant that contains anthocyanin compounds in its calyx, which are known for their antioxidant properties. These compounds give the plant a captivating red color and can be used as natural dyes. This study aimed to exploit anthocyanins in rosella calyx as natural dyes and antioxidants in lip cream product. The formulations employed extracts from rosella calyx, a viscous extract from maceration in 96% ethanol (F1) and a commercially available powdered rosella extract (F2). Dragon fruit, known as Hylocereus undantus (HU), has traditionally been utilized in Banyuwangi, East Java, Indonesia, for addressing gout and rheumatism. Despite its empirical use, there is a lack of reported pharmacological research on this plant. This study aimed to explore the properties of HU extract in treating hyperuricemia, inflammation, and pain. Adenine-potassium oxonate-induced mice were employed to evaluate anti-hyperuricemia activity, measuring uric acid, blood urea nitrogen (BUN), and creatinine serum levels. The completion of disease treatment at the health center was important for payment of Capitation Based on Fulfillment of Service Commitment (PCFSC/KBPKP) and the Non-Specialist Case Outpatient Referral (NSOR/RRNS) was one of the key indicators. This study aimed to assess public health center readiness in the completion of disease treatment for those stipulated in Ministerial Decree number 514 of the year 2015. This study used Health Facility Research (Rifaskes) data conducted in 2019 with a total of 9831 health centers as observations.
