Cardiovascular disease is the leading cause of death in women in the United States. Aging is a primary risk factor for the development of cardiovascular disease as well as cardiovascular-related morbidity and mortality. Aging is a universal process that all humans undergo; however, research in aging is limited by cost and time constraints. H5N1 avian influenza is a significant global concern with the potential to become the next pandemic threat. Recombinant subunit vaccines are an attractive alternative for pandemic vaccines compared to traditional vaccine technologies. In particular, polyanhydride nanoparticles encapsulating subunit proteins have been shown to enhance humoral and cell-mediated immunity and provide protection upon lethal challenge. In this work, a recombinant H5 hemagglutinin trimer (H5₃) was produced and encapsulated into polyanhydride nanoparticles. The studies performed indicated that the recombinant H5₃ antigen was a robust immunogen. Excessive free radical production will lead to oxidative stress. One of the indicators used to determine stress oxidative is malondialdehyde (MDA). Hylocereus polyhizus peel is potential used natural exogenous antioxidant. Catalase enzyme is one of the endogenous intracellular antioxidant that can be reduced by oxidative stress conditions. Soybean extract has known for its antiaging potential. The addition of soybean extract was predicted can influence the physical stability of the cream. Therefore, this research was aimed to test the physical stability of cream contained soybean extract in different concentration (2%, 4%, 6%, and 8%). The physical stability test including the storage for eight weeks at room temperature, high temperature (40°±2°C), and low temperature (4°±2°C). Stability parameters were the organoleptic observation, pH, globul size, viscosity, and cycling test. Those four cream formula showed good stability in organoleptic, pH, globul size, viscosity, and cycling test.

Inhibition of CDK2 activity in G1 and S phases of the cell cycle can promote selective apoptosis of cancer cells through the E2F1 pathway. Currently available CDK inhibitors target the ATP binding pocket and result in lack of specificity for the cell cycle vs. the transcriptional CDKs. It has been shown that a peptide HAKRRLIF derived from the tumor suppressor p21 binds to the cyclin binding groove (CBG) and selectively inhibits cell cycle CDKs (CDK2/Cyclin A, CDK2/Cyclin E and CDK4/Cyclin D). Polymer based nanoparticle formulations have been shown to increase drug bioavailability and/or reduce drug adverse effects. Nonsteroidal anti-inflammatory drugs (e.g. celecoxib) reduce prostaglandin synthesis and cause side effects such as gastrointestinal and renal complications. The aim of this study was to formulate celecoxib entrapped poly lactide-co-glycolide based nanoparticles through a solvent evaporation process using didodecyldimethylammonium bromide or poly vinyl alcohol as stabilizer. • Geriatric patients (> 65 years of age) have a 3-5% incremental increase in annual occurrence of trauma making it a top five leading cause of death in this population.

• The mortality rate ranges from 15-30%, which is 4-5 times higher than younger patients.

• In 2011, data from the Pennsylvania Trauma Systems Foundation (PTSF) demonstrated nearly 32% of injured patients statewide and nearly 50% of the trauma patients at Thomas Jefferson University Hospital (TJUH) were > 65 years.

Many patients go without access to contemporary or traditional Western medicine due to lack of quality health insurance. Thus, more patients are turning to other less expensive, alternative forms of treatment. Our analysis of several different community-pharmacies in Maine, Indiana, Ohio, and Kentucky showed that many practicing pharmacists know little about these types of alternative therapies, and make little effort answering questions and/or making a safe recommendation about these products. Nano-scale particles sized 10—400 nm administered systemically preferentially extravasate from tumor vasculature due to the enhanced permeability and retention effect. Therapeutic success remains elusive, however, because of inhomogeneous particle distribution within tumor tissue. Insufficient tumor vascularization limits particle transport and also results in avascular hypoxic regions with non-proliferating cells, which can regenerate tissue after nanoparticle-delivered cytotoxicity or thermal ablation. The protective mechanisms of microglia cells help to maintain central nervous system (CNS) homeostasis and function.1 Microglia are innate immune cells that constantly survey their surrounding CNS microenvironment for pathogens, damaged cells, and inflammatory molecules. Sugars from our diet, including glucose and fructose, combine with endogenous proteins non-enzymatically and form advanced glycation-end products (AGEs).2 AGEs are shown to produce reactive oxygen species, leading to inflammation and cellular damage that may be mediated by microglia.3 Fructose consumption has become increasingly prevalent within the American diet, as it is a lower cost sweetener.4 Microglia become activated and phagocytic in the presence of …

Objective: As a result of the heightened reactivity fructose demonstrates compared to glucose and our current knowledge of glucose advanced glycation end-products, the aim of this research was to further elucidate the proinflammatory pathways involved in the response to fru-AGE exposure, including the effects of fru-AGEs on cell-surface receptor expression. We hypothesized that once microglia were activated in response to fru-AGE exposure, there would be an increase in the expression of RAGE and TLR4 to facilitate the proinflammatory cascade.

Peer tutoring services for a pathophysiology and therapeutics course have historically been made available to pharmacy students, but they were greatly underutilized. The role of peer tutors had been exclusively to hold office hours during which students could ask questions or review material. Peer tutoring services were updated and expanded based on the needs of the students. The objectives of this project were to develop, implement, and assess the impact of peer-led organized weekly review sessions in a pathophysiology and therapeutics course.

HMG-CoA reductase inhibitors, also known as statins, are a ubiquitous class of medication used for lowering cholesterol. In-vitro and animal studies have suggested that statins can activate osteoblast differentiation and have anabolic effects on bones; however, observational and experimental studies in humans have shown conflicting results.1-5 The exact mechanism of statins on bone growth is unknown; however, there are several hypotheses. The “Lipid Hypothesis” (Figure 1) suggests that lipid oxidation leads to activation of PPARγ, and production of isoprostanes including isoPGF2α and isoPGEα. Chlorpromazine is a phenothiazine antipsychotic which is often used in hospice and palliative care to treat hiccups, delirium, and nausea. With the discontinuation of the commercial oral solution concentrate, there is a need to prepare this product by extemporaneous compounding. This study was initiated to identify an easy-to-prepare formulation for the compounding pharmacist. A stability study was also conducted to select the proper storage conditions and establish the beyond-use date. Lunasin is a 44 amino acid peptide derived from the soybean seed that has been shown to have cancer chemopreventive and chemotherapeutic properties. In this study, we investigated the potential utility of lunasin as a chemotherapeutic in a melanoma model. Initial studies showed that lunasin has little activity against established melanoma cell lines in vitro using adherent culture methods; however, lunasin’s in vitro activity was significantly higher in non-adherent colony-forming assays in soft agar and oncosphere assays. Objective. To describe an interprofessional communication course in an academic health sciences center and to evaluate and compare interpersonal and interprofessional communication self-efficacy beliefs of medical, nursing, and pharmacy students before and after course participation, using Bandura’s self-efficacy theory as a guiding framework.

Design. First-year nursing (n=36), first-year medical (n=73), and second-year pharmacy students (n=83) enrolled in an interprofessional communication skills development course voluntarily completed a 33-item survey instrument based on Interprofessional Education Collaborative (IPEC) core competencies prior to and upon completion of the course during the fall semester of 2012.

Using prior biological data, pharmacophore models were made for hCNT1, hCNT3, hENT1, and hENT4. The hCNT3 and hCNT1 pharmacophore were used to select compounds for biological testing. The NBMPR analogue and dipyridamole analogue hENT1 pharmacophores were compared to each other and to a combined pharmacophore for hENT1. The dipyridamole analogue pharmacophore better predicted non-nucleoside small molecule inhibitors, and as such appears to be the better tool for aiding in the design of new small molecule inhibitors. The hCNT3 pharmacophore failed to select active compounds and as such must be redesigned. The hCNT1 pharmacophore succeeded in identifying two moderately active compounds …

Chronic obstructive pulmonary disease (COPD) was found to be the fourth leading cause of death in 2004, but current treatments can only relieve the symptoms and improve the quality of life for the patients. Glucocorticoids have been broadly applied in other inflammatory conditions with promising therapeutic effects, but performed poorly in COPD. The phenomenon called glucocorticoid resistance is responsible for glucocorticoid insensitivity in COPD. Dystrophia myotonica (DM1), one of the most common forms of muscular dystrophy, is caused by a repeated trinucleotide expansion in the DMPK gene. This mutation results in the accumulation of toxic cellular RNA transcripts. Spliceosome-mediated RNA trans-splicing (SMaRT) technology is a form of gene therapy that possesses the potential to correct these toxic RNA transcripts and thus cure the disease. Despite its promise, prior research applications of SMaRT technology to DM1 have been hampered by poor efficiency and have not been validated in a relevant model of the disease. Osteoporosis, a major public health problem in the U.S., is characterized by low bone mass and structural deterioration of bone tissue. Since the imbalance between bone formation and bone resorption during bone remodeling has been documented to be a major factor in the pathogenesis of osteoporosis, it is crucial to identify novel genes and/or novel functions of known genes that regulate the formation and activity of bone forming osteoblasts and bone resorbing osteoclasts.
