Identification Of Genetic And Antigenic Variation And Evolution Pattern Among Influenza A And B Viruses In Thailand, Nungruthai Suntronwong Jan 2020

Identification Of Genetic And Antigenic Variation And Evolution Pattern Among Influenza A And B Viruses In Thailand, Nungruthai Suntronwong

Chulalongkorn University Theses and Dissertations (Chula ETD)

Seasonal influenza viruses commonly cause respiratory disease and have a considerable impact on public health threats which annually estimates 3 to 5 million cases of severe illness worldwide. The triggering of the seasonal influenza epidemic results from a complex interplay between viral, host and external factors such as climate. Although vaccination is an effective tool for influenza prevention and its complications, the composition of vaccine strain has been changed every year due to influenza constantly evolving. Here, we aim to examine the association between influenza activity and local climate factors, host immunity, genetic and antigenic variation and evolution pattern among …

Chulalongkorn University Theses and Dissertations (Chula ETD)

Liver cancer is one of the most cancer-related mortality with high incidence worldwide. Hepatocellular carcinoma (HCC) is the most common liver cancer type. Currently, Liquid biopsy is known as a non-invasive biomarker tool that specifically represents tumor appearance, leading to improved patient outcomes. Therefore, the novel biomarkers of liquid biopsy in Thai HCC patients are still unexplored and needed for precision medicine. The main aims of this study were First, to explore genetic profiles from liquid biopsies of circulating cell-free DNA (cfDNA) and peripheral blood mononuclear cells (PBMCs) using next-generation sequencing (NGS) and identify novel biomarkers for HCC from these …

Investigation Of Cytotoxic Activity Of Full-Length And Truncated Colicin N On Human Cancer Cells, Methawee Duangkaew

Chulalongkorn University Theses and Dissertations (Chula ETD)

Bacteria, exposed to stress from the environment or competition, release toxins called bacteriocins. Bacteriocins are classified by their mode of cytotoxicity such as pore former, endonuclease, etc. Colicins are the member of bacteriocins produced by Escherichia coli (E.coli). Among several types of colicins cytotoxicity, Pore-forming colicins has been reported in both bacteria and various type of cancer cells. Colicin N is also a pore-forming colicin and the anticancer activity of colicin N has not been extensively reported. Here we examined the cytotoxic effects of colicin N and its domain on cancer cells. The expression and purification of recombinant full-length and …

Chemical Constituents With Alpha-Glucosidase Inhibitory Activity From Dendrobium Braianense And Dendrobium Kentrophyllum, Mia Indria Permatasari

Chulalongkorn University Theses and Dissertations (Chula ETD)

To investigate the phytochemicals of the entire plants of Dendrobium braianense brought about in the isolation of three bibenzyls, which included chrysotoxin, moscatilin and gigantol. From the whole plant of D. kentrophyllum three flavonoids, namely, quercetin, kaempferol and rutin were isolated, the structures of which were decided by means of analysis of spectroscopic data to compare the values already reported. They were assessed for their alpha-glucosidase inhibitory activity. Gigantol, quercetin and kaempferolwere found to exhibit strong alpha-glucosidase inhibitory activity when compared with the positive control acarbose, while chrysotoxin, moscatilin and rutin were devoid of activity.

Development Of Hyaluronic Acid-Based Nanogel Platform For Improved Stability And Polyvalent Ligand Display Of Poly(I:C) As An Effective Immunostimulatory Adjuvant In Cancer Immunotherapy, Nararat Kotcharat

Chulalongkorn University Theses and Dissertations (Chula ETD)

The aim of this research was to develop a nanogel formulation-based on modified natural polymer, hyaluronic acid (HA), as a biodegradable material for adjuvant delivery. Polyinosinic:polycytidylic acid (poly(I:C)) have been approved by FDA as a promising adjuvant candidate for the TLR3 (Toll-Like Receptor 3) activation. However, it suffers from being poor stability and is subjected to rapid enzymatic hydrolysis in serum, so that it requires high administered dose leading to adverse effects. To augment the adjuvant stability and protection from the degradation, the nano-particulate carriers were herein designed with self-assembly of HA scaffold grafted with poly (N-isopropylacrylamide), or pNIPAM. The …

Comparison Of Area Under The Curves For Vancomycin From One- And Two-Compartment Models Using Sparse Data, Nyein Hsu Maung Jan 2020

Comparison Of Area Under The Curves For Vancomycin From One- And Two-Compartment Models Using Sparse Data, Nyein Hsu Maung

Chulalongkorn University Theses and Dissertations (Chula ETD)

Vancomycin pharmacokinetics has been described by 1- and 2-compartment models. One-compartment models built from routine monitoring data, which were mainly trough samples, are commonly used to predict area under the curves (AUC), the useful indicator for vancomycin efficacy. The question stands whether AUCs from 1-compartment models with sparse data can sufficiently represent the true AUC. This study aimed to compare AUCs from 1- and 2-compartment models using sparse data. A previously published model was used to simulate full individual profiles for 100 patients. From these data, the reference AUC (AUCref) was calculated and two depleted datasets (trough-only and peak-trough) were …

Chulalongkorn University Theses and Dissertations (Chula ETD)

The incidence of metastasis stage crucially contributes to high recurrence and mortality rate in lung cancer patients. Unfortunately, no available treatment inhibits migration, a key metastasis process in lung cancer. In this study, the effect of 22-O-(N-Boc-L-glycine) ester of renieramycin M (22-Boc-Gly-RM), a semi-synthetic amino ester derivative of bistetrahydroisoquinolinequinone alkaloid isolated from Xestospongia sp., on migratory behavior of human lung cancer cells was investigated. Following 24 h of treatment, 22-Boc-Gly-RM at non-toxic concentrations (0.5-1 µM) effectively restrained motility of human lung cancer H460 cells assessed through wound healing, transwell migration and multicellular spheroid models. The capability to invade through matrix …

Phytochemical Screening, In Vitro Antimalarial Activity And Genetic Diversity Of Selected Asteraceae Medicinal Plants Indigenous To Thailand, Desy Liana Jan 2020

Phytochemical Screening, In Vitro Antimalarial Activity And Genetic Diversity Of Selected Asteraceae Medicinal Plants Indigenous To Thailand, Desy Liana

Chulalongkorn University Theses and Dissertations (Chula ETD)

The emergence of artemisinin resistance leaded the effort to find the new antimalarial drug or artemisinin activity booster. Due to the chance that secondary metabolites can be evolutionary conserved, combining phylogeny with ethnobotanical data for screening antimalarial activity may be helpful to predict bioactivity and minimize the expenditure and time for laboratory research. The aimed of this study is screening the antimalarial activity, phytochemicals and genetic diversity of selected Asteraceae medicinal plants generated by combinatorial phylogeny and ethnobotanical data. 733 medicinal plants were obtained from literature search however only 340 taxa were met the inclusion and exclusion criteria hence these …
