The Role Of Outer Inflammatory Protein A (Oipa) In Vaccination Of Thec57bl/6 Mouse Model Infected By Helicobacter Pylori, Mohaddese Mahboubi, Tahereh Falsafi, Majid Sadeghizadeh, Fatemeh Mahjoub

Turkish Journal of Medical Sciences

Background/aim: Outer inflammatory protein A (OipA) is an important adhesin of Helicobacter pylori. Our goal was to assess the role of OipA in protection of C57BL/6 mice against H. pylori. Materials and methods: C57BL/6 mice were mucosally immunized with recombinant OipA protein, OipA + propolis, propolis, and phosphate-buffered saline. After vaccination, anti-OipA IgA was measured. Mice were challenged three times with 5 x 107 CFU of the H. pylori B19 strain. Two weeks later, bacterial colonization and inflammation in the stomach was analyzed using standard methods. Results: The CFU number in the OipA group was significantly (P < 0.05) lower than that of the control. The CFU number in the OipA + propolis group was higher than those of the OipA and propolis groups. IgA titers were significantly higher (P < 0.0001) in the OipA group compared to the control and OipA + propolis groups. Propolis did not play an adjuvant effect but it interfered with the efficient vaccine effect of OipA. Conclusion: Results show the effect of vaccination by OipA in protection of the mouse model and the importance of OipA in H. pylori pathogenesis. OipA may be proposed as a suitable oral vaccine candidate against H. pylori infection; however, further study is required to determine adjuvant or adverse effects of propolis toward OipA.

Dysregulation Of The Dkk1 Gene In Pediatric B-Cell Acute Lymphoblastic Leukemia, Si̇nem Firtina, Özden Hatirnaz Ng, Yücel Erbi̇lgi̇n, Uğur Özbek, Müge Sayi̇toğlu

Turkish Journal of Medical Sciences

Background/aim: The canonical Wingless-type (WNT) pathway is involved in normal hematopoietic cell development and deregulated WNT signaling is implicated in the development of hematological malignancies. Dickkopf 1 (DKK1) acts as a modulator of the β-catenin regulated canonical pathway. Activation of DKK1 leads to apoptosis and growth suppression, whereas silencing by promoter hypermethylation results in abnormal WNT activation. The secreted inhibitor Dickkopf can antagonize WNT signaling by competitively binding to low density lipoprotein receptors (LRPs) 5 and 6. Materials and methods: We studied DKK1 gene promoter methylation and investigated DKK1, β-catenin, LRP5, and LRP6 mRNA levels in B-cell acute lymphoblastic leukemia …

Background/aim: The aim of the present study was to examine the anticancer effects of combination treatment with artemisinin and iron on the human gastric cancer cell line. Materials and methods: The AGS cell line was cultivated separately as a monolayer in culture flasks and different doses of 99% pure artemisinin with invariable doses of iron sulfate were added to the culture media and adhesive cells on the flask's bottom were stained with hematoxylin-eosin for a pathological assay. Results: Damage to cancerous cells increased dose dependently and it was higher in the combination treatment groups (artemisinin plus iron). The histopathological changes …

Objective

To evaluate the viscoelastic properties of two experimental BPA-free and one BisGMA-based orthodontic resin composite adhesives for bonding fixed retainers.

Methods

A commercially available BisGMA-based (TXA: Transbond LR) and two bisphenol A-free experimental adhesives (EXA and EXB) were included in the study. The viscoelastic behavior of the adhesives was evaluated under static and dynamic conditions at dry and wet states and at various temperatures (21, 37, 50 °C). The parameters determined were shear modulus (G), Young’s modulus (E) under static testing and storage modulus (G1), loss tangent (tan δ) and dynamic viscosity (n*) under …

Objectives

Postoperative pain management is of highest interest for patients undergoing maxillofacial surgery including microvascular reconstructive surgery. Currently, there is a lack of information regarding process and outcome of postoperative pain management after microvascular reconstruction.

Materials and methods

In a prospective clinical study, 31 adults were evaluated on the first postoperative day following microvascular reconstruction with a radial forearm flap using the standardized questionnaire of the Germany-wide project Quality Improvement in Postoperative Pain Management (QUIPS). It enables a standardized assessment of patients’ characteristics, pain parameters, outcome and pain therapy process parameters.

Results

Pain management consisted predominately of premedication with midazolam, …
