Tetraspanin Kai1/Cd82 Inhibits Cell Migration-Related Cellular Events Via Reorganizing Actin Network, Wei Liu

To determine how tetraspanin KAI1/CD82, a tumor metastasis suppressor, inhibits cell migration, we assessed which motility-related cellular events are altered by KAI1/CD82 and how KAI1/CD82 regulates these events. We found that KAI1/CD82-overexpressing cancer cells exhibit various morphologies but typically display elongated cellular extensions and a lack of lamellipodia. Live imaging demonstrated that the formation of lamellipodia and retraction of extensions were deficient upon KAI1/CD82 overexpression.

The deficiency in developing motility-related cellular events was accompanied by defects in actin cortical network and stress fiber formations. Notably, actin polymerization was attenuated by KAI1/CD82. Although Rac1 activity was diminished upon KAI1/CD82 expression, Rac1 …

DNA topoisomerase I (Top1) is a highly conserved enzyme composed of four domains: a positively charged N-terminus; a DNA binding/core domain that circumscribes duplex DNA; a non-conserved linker domain; and a C-terminal/catalytic domain. Top1 catalyzes changes in DNA topology by transient cleavage of a single DNA strand and the concomitant formation of a phosphotyrosyl linkage between the enzyme and the 3’ DNA end. This covalent Top1-DNA complex is the binding site for camptothecin (CPT), which selectively inhibits religation of the cleaved DNA strand. CPT binding stabilizes the covalent complex, while the collision of replication forks with CPT-Top1-DNA adducts produces DNA …

A unifying meta-theory of nursing is suggested, building from the foundation of the AQAL framework. A definition of nursing as situated caring is presented, along with a historical discussion of nursing epistemology and theory for context. Implications for practice, education, and research are also discussed. A unifying meta-theory will enable nurses at both the practical and academic levels to appreciate the complexity and simplicity of nursing, allowing them to articulate confidently what we do and why we do it.

Tigecycline, a glycylcycline related to the tetracycline class of antibiotics, represents a new option for the treatment of complicated intra-abdominal and complicated skin and skin structure infections. It displays favorable activity in vitro against the most common causative Gram-positive, Gram-negative and anaerobic pathogens. In addition, tigecycline demonstrates activity against drug-resistant pathogens such as methicillin-resistant Staphylococcus aureus, vancomycin-resistant enterococci, and organisms (such as Escherichia coli and Klebsiella pneumoniae) producing extended-spectrum beta-lactamases. Tigecycline lacks activity in vitro against Pseudomonas and Proteus spp. In randomized clinical trials, tigecycline administered intravenously twice daily has demonstrated efficacy similar to comparators for a variety …

During the past few years, the AIDS campaign in Haiti has been targeting Vodou officiants and organizations. These awareness and training programmes in- form officiants about the transmission and prevention of AIDS, tests for HIV and anti- retroviral drugs, or even try to encourage them to become involved in a medical referral system. These culturalist interventions are grounded in an essentialist concept of culture that can have harmful effects on the targeted groups. The purpose of this study was to measure the influence of quercetin on plasma cytokines, leukocyte cytokine mRNA, and related variables in ultramarathoners competing in the 160-km Western States Endurance Run (WSER). Sixty-three runners were randomized to quercetin and placebo groups and under double-blinded methods ingested 1000 mg/day quercetin for 3 weeks before the WSER. Thirty-nine of the 63 subjects (n = 18 for quercetin, n = 21 for placebo) finished the race and provided blood samples the morning before the race and 15–30 min postrace. Pediatric adrenocortical cancer is extremely rare and often fatal (approximately 0.3-0.4 cases per million worldwide; 50% 5-year survival). The incidence of pediatric adrenocortical cancer in southern Brazil is 10-15 times higher than the worldwide incidence. Due to the rarity of adrenocortical cancer, especially in children, underlying gene dysregulation and mechanisms of tumorigenesis of the adrenal gland are very poorly described in the literature. However, it is well-known that the tumor suppressor p53, which is mutated in over 50% of all human cancers, is commonly mutated in pediatric adrenocortical cancer. Patients with unresectable cancers of the upper gastrointestinal tract often suffer severe symptoms due to pain, nausea and vomiting, weight loss, cachexia, and poor food tolerance. This can be related to gastric and duodenal cancers causing intrinsic obstruction of the intestinal lumen or pancreatic and biliary cancers causing extrinsic biliary compression. This article provides the first anatomically labeled, magnetic resonance imaging (MRI) -based atlas of the subadult and fetal Atlantic white-sided dolphin (Lagenorhynchus acutus) brain. It differs from previous MRI-based atlases of cetaceans in that it was created from images of fresh, postmortem brains in situ rather than extracted, formalin-fixed brains. The in situ images displayed the classic hallmarks of odontocete brains: fore-shortened orbital lobes and pronounced temporal width. Olfactory structures were absent and auditory regions (e.g., temporal lobes and inferior colliculi) were enlarged. In the subadult and fetal postmortem MRI scans, the hippocampus was identifiable, despite the relatively small size …

Child sexual abuse (CSA), a social problem of endemic proportions, has existed in all historical eras and societies (Conte 1994; Fergusson and Mullen 1999; Wekerle and Wolfe 1996; Wolfe 1999). Since antiquity, anecdotal records (e.g., legal, artistic, philosophical, and literary accounts) have documented activities that would today be classified as CSA (deMause 1974; Kahr 1991; Olafson, Corwin, and Summit 1993). For instance, a sizable portion of adults in ancient Greek and Roman cultures openly engaged in what is now considered pederasty or rape (deMause 1974; Kahr 1991). Fishes are capable of regenerating sensory hair cells in the inner ear after exposure to excessive noise. However, a time course of auditory hair cell regeneration has not been characterized for zebrafish, nor has gene expression following noise exposure. To establish a time course of hair cell regeneration, adult zebrafish were exposed to a 100 Hz pure tone at 179 dB re 1 |u.Pa RMS for 36 hours, and then allowed to recover for 0 to 14 days before morphological analysis. Long chain omega 3 (n-3) fatty acids, eicosapentaenoic (EPA) and/or docosahexaenoic acid (DHA), have been shown to suppress growth of most cancer cells. In vivo, alpha linolenic acid (ALA, 18:3n-3) can be converted to EPA or DHA. We hypothesized that substituting canola oil (10% ALA) for the corn oil (1% ALA) in the diet of cancer bearing mice would slow tumor growth by increasing n-3 fatty acids in the diet. Knee bracing is used at all levels of athletics to prevent or support existing knee injuries; however, little research has been conducted on the effects of knee braces on athletic performance. Therefore, the purpose of this study was to examine how two different types of knee braces, semi-customizable braces and knee sleeves, affect vertical jumping performance. BACKGROUND: Perioperative myocardial injury (PMI) remains a major cause of perioperative morbidity and mortality but clinical strategies to prevent PMI are still uncertain.

METHODS AND RESULTS: We comprehensively searched PubMed for major research articles concerning clinical strategies to prevent PMI. The key findings are as follows: (1) the American College of Cardiology/American Heart Association guideline update for perioperative cardiovascular evaluation for noncardiac surgery is very useful to stratify cardiac risk preoperatively; (2) cardiac troponin has emerged as a biomarker to diagnose postoperative PMI and to predict clinical outcomes; (3) coronary revascularization before noncardiac surgery probably would provide cardiac protection in …

Nursing schools in the United States have not been teaching evidence-based practices for safe patient handling, putting their graduates at risk for musculoskeletal disorders (MSDs). The specific aim of this study was to translate research related to safe patient handling into the curricula of nursing schools and evaluate the impact on nurse educators and students' intentions to use safe patient handling techniques. Nurse educators at 26 nursing schools received curricular materials and training; nursing students received the evidence-based curriculum module. There were three control sites. African American (AA) grandmothers who parent their grandchildren may experience burden related to these responsibilities. Little is known about perceived caregiver burden and its influence on AA grandmother health and service needs. The purpose of this descriptive, correlational research study was to investigate relationships among personal characteristics, information need, perceived burden, perceived health, service need, and service use in AA grandmothers who are the primary caregivers for one or more of their biological grandchildren. Recruited through churches and community centers, 93 AA grandmothers, ages 36 to 78, participated in this study. These grandmothers were primary caregivers for 211 grandchildren. The …
