The SARS-Coronavirus (SARS-CoV) is the etiological agent of the severe acute respiratory syndrome (SARS). The SARS-CoV spike (S) glycoprotein mediates membrane fusion events during virus entry and virus-induced cell-to-cell fusion. Investigations, described herein, have focused on the genetic manipulation of the SARS-CoV S glycoprotein in order to delineate functional domains within the protein. This was accomplished by incorporating single point mutations, cluster-to-lysine and cluster-to-alanine mutations, as well as carboxyl terminal truncations into the protein and investigating these mutants in transient expression experiments. An Edwardsiella ictaluri strain with disruption of ureG was identified through the use of signature tagged insertion mutagenesis as being attenuated for virulence in the channel catfish host. Sequencing of the flanking regions surrounding the insert showed that the gene was part of a urease gene complex that included ureE, ureF, ureG, ureD, ureI, and an ammonium transporter homologue. The ureG gene encodes a GTP-binding accessory protein which is thought to function in energy-dependent urease assembly. Osteoarthritis is arguably the most common ailment in both dogs and people in the developed World. Treatment for osteoarthritis is currently symptomatic. Development of therapies designed at stopping the progression of osteoarthritis, require a method of evaluating efficacy. Objective analysis by measuring joint metabolism via clinical trials of the cross-over design is currently not possible with the methods utilized. Extracellular matrix degradation is a hallmark of osteoarthritis. The matrix metalloproteinases are major degradative enzymes. Intraoperative reductions in serum fentanyl levels in dogs with induced moderate hypothermia and transdermal fentanyl patches (TDF) in place has been documented. The impact of such reductions has not been evaluated nor has the anesthetic sparing effect of the TDF. Reductions in serum levels may be the result of either a decreased uptake of fentanyl from the dermal depot, or a biophase shift. Urokinase (uPA) and urokinase receptor (uPAR) are present in the urinary tract of people, mice, rats and cows. The presence of urokinase and urokinase receptor in the urinary tract of dogs has not been documented. Documenting the presence of uPA and uPAR in the urinary tract of healthy dogs will help determine the role these proteins play in pathology of the urinary tract in further research. Urine was collected from fifty-four healthy dogs. A fluorescence assay was developed using the fluorogenic substrate Z-Gly-Gly-Arg-AMC, HCl. Degenerative suspensory ligament desmitis (DSLD) is a debilitating disease of connective tissues seen in many breeds but has become prevalent in the Peruvian Pasohorse. DSLD is believed to be a genetic disorder caused by one primary founder and most likely has a recessive mode of inheritance although a dominant or co-dominant mode of inheritance has not been ruled out.

The phosphoprotein (P protein) of vesicular stomatitis virus (VSV) is an essential subunit of the viral RNAdependent RNA polymerase and has multiple functions residing in its different domains. In the present study, we examined the role of the hypervariable hinge region of P protein in viral RNA synthesis and recovery of infectious VSV by using transposon-mediated insertion mutagenesis and deletion mutagenesis. We observed that insertions of 19-amino-acid linker sequences at various positions within this region affected replication and transcription functions of the P protein to various degrees. Female B10.S mice are highly resistant to proteolipid protein (PLP) 139–151-induced experimental autoimmune encephalomyelitis (EAE) and depletion of PLP 139–151- reactive CD4+CD25+regulatory T (Treg) cells can slightly increase their EAE susceptibility. Although male B10.S mice are moderately susceptible to EAE, we report that depletion of Treg cells in male B10.S mice before immunization with PLP 139–151 renders them highly susceptible to severe EAE with more CNS neutrophil infiltrates than nondepleted controls. Increased susceptibility is associated with an enhanced PLP 139–151-specific T cell response and greater production of IFN-γ, IL-6, and IL-17. Infection of calves with bovine herpesvirus 1 (BHV-1) results in transient immunosuppression that may lead to bacterium-induced pneumonia and, occasionally, death. Although sensory neurons in the trigeminal ganglia (TG) are the primary site of BHV-1 latency, viral genomes are detected in the tonsils of latently infected calves. Dexamethasone (DEX) consistently induces reactivation from latency, and viral gene expression is detected in TG and tonsils. In sensory neurons of latently infected calves, the latency-related (LR) gene is abundantly expressed and is required for reactivation from latency. The objective of this study was to investigate coliform counts in feedlot cattle water and feed rations and their associations with management, climate, fecal material, and water Escherichia coli O157 using a cross-sectional study design. Coliform counts were performed on feed samples from 671 pens on 70 feedlots and on water samples from 702 pens on 72 feedlots in four U.S. states collected between May and August 2001. To obtain data about select zoonotic and other infectious diseases in free-ranging predators in five ecoregions in Nebraska, sera were collected from 67 coyotes (Canis latrans) and 63 raccoons (Procyon lotor)) from November 2002 through January 2003. For coyotes, antibodies were detected against canine distemper virus (CDV, 61%), Francisella tularensis) (32%), Rickettsia rickettsi) (13%), and flaviviruses (48%). None of the coyote sera had antibodies to Borrelia burgdorferi, Brucella canis), or six serovars of Leptospira interrogans). Recently, the protostrongylid nematode Parelaphostrongylus odocoilei has been reported in a new host species, thinhorn sheep (Ovis dalli). For the first time, we completed the life cycle of P. odocoilei in three Stone’s sheep (O. dalli stonei) and two thinhorn hybrids (O. dalli stonei × O. dalli dalli), each infected with 200 third-stage larvae from slugs (Deroceras laeve). The prepatent period ranged from 68 days to 74 days, and shedding of first-stage larvae (L1) peaked at >10,000 L1 per gram of feces between 90 and 110 days postinfection. A total of 75, …

Fearfulness reaction was examined using tonic immobility (TI) response and differential leucocyte counts as physiological indicators of distress from sire and dam brown layer lines. The study was performed on 20 male and 131 female chickens from the sire line and 24 male and 116 female chickens from the dam line. The duration of TI, the time interval until the bird righted itself, and the number of inductions (15 s periods of restraint) necessary to attain TI were recorded. The livers and lungs of 5596 sheep (443 local, 473 Romanian and 4680 Australian) slaughtered in Amman Central Abattoir during November-December 1999 were examined in routine meat inspection procedures for helminths. Dictyocaulus filaria, hydatid cyst, Fasciola hepatica and Dicrocoelium dendriticum were recovered from the examined sheep with variable prevalences. Dicrocoelium dendriticum is reported from the livers of imported sheep in Jordan for the first time.

This study was conducted to investigate the pre-weaning growth of crossbreed types. Fifteen genotypes (F_1 and B_1) were obtained by crossbreeding in the Animal Research Institute in Konya from 1989 to 1996. This goal of the present research was to determine the possibility of twice-yearly lambing in Awassi sheep. The research was appropriate in terms of ecological agriculture, animal breeding applications, and organic agricultural production with no impact on their health or behavior, or exposure to environmental conditions such as exogenous hormone applications. The purpose of this study was to investigate the prevalence of Salmonella serogroups in chicken meat. A total of 315 skins from the wing part of chicken carcasses were collected from 8 chicken carcass retailers. Salmonella isolation was performed as described in the Bacteriological Analytical Manual of the Food and Drug Administration, Center for Food Safety and Applied Nutrition. The sero-prevalence of contagious ecthyma (orf) in lambs and humans was studied in the Kars region of Turkey. For this purpose, serum samples were collected from 320 lambs from 5 different districts of the Kars region. Serum samples were also collected from 100 humans at risk of infection. The serum samples were tested for the presence of antibodies to orf virus by enzyme-linked immunosorbent assay (ELISA). The results showed that 52.81% and 5% of the serum samples collected from lambs and humans, respectively, were seropositive for orf virus. Total gastrectomy is widely used as a major surgical treatment for gastric cancer. The aims of this study were to investigate the possible effects of total gastrectomy on pancreas morphology, and to determine the immunolocalisation of chromogranin A (Cg A) and synaptophysin (SYP) antigens, described as specific tumour markers for neuroendocrine tumours. Ten rats underwent total gastrectomy via oesophago-duodenostomy. After a month the rats were killed, and tissue samples were taken from the pancreas, processed for light microscopy and stained with haematoxylin-eosin. The histochemical and ultrastructural effects of water Cu on the anterior latissimus dorsi (ALD) muscle of 120 newly hatched broilers were investigated in a 45-day experiment. Two doses of water Cu (250 mg Cu/kg water and 500 mg Cu/kg water) were given to the poults. Two distinct categories (IIIA and IIIB) of slow tonic fibers in the control groups were detected by enzyme reactions. The ultrastructure of the Z-line, mitochondrial content and sarcotubular content differed between the 2 fiber types. The effects of already accumulated heavy metals (Hg, Cd, Pb) in the body of tench on the 96-h LC_50 values of the respective heavy metals were studied. The body concentration of mercury, cadmium and lead was 0.011, 0.32 and 1.59 mg/g respectively, and their 96-h LC_50 values were 1.0, 6.5 and 300.0, ppm, respectively. The general accumulation order of heavy metals and their respective 96-h LC_50 values were Hg< Cd< Pb. It was observed that the fish with lowest body concentration of heavy metal showed the lowest 96-h LC_50 value of the respective heavy metal and vice versa. This close relationship showed that the previous body accumulation of heavy metals has a direct effect on the 96-h LC_50 values of the respective heavy metals and the susceptibility of fish.

The Red Blood Cell Membrane Proteins In Rabbits With Experimental Ketosis, Sefa Çeli̇k, Hi̇lal Karagül

This study investigated the effects of experimentally induced ketosis on erythrocyte membrane proteins in rabbits by sodium dodecyl sulfate/polyacrylamide gel electrophoresis (SDS-PAGE). The animals were allocated to 2 groups; ketosis and control. The experiment was lasted 13 days. Food was withheld for 5 days in the ketosis group, and then the animals were fed for 8 days. The control group was fed standard rabbit rations. Erythrocyte membrane proteins were isolated from the blood, and analyzed by SDS-PAGE using 10% (w/v) acrylamide monomer. The effects of the age of the hen, genotype, rearing system and dietary structure on egg and serum cholesterol content and some other characteristics of eggs were investigated in laying hens. However, only the findings related to genotype and rearing system are given. Cataract surgery was planned on 41 ERG positive eyes of 25 dogs of which 9 had unilateral and 16 had bilateral cataracts. Phacofragmentation and aspiration surgery was performed on 32 eyes of 20 dogs (and ECCE was performed on 4 eyes and ICCE was performed on 5 eyes of 5 dogs) of different breeds, sexes and ages. Vision restoration and complications were evaluated postoperatively in the short term (4 weeks). Mean age of the 20 dogs was 7.3 years. The Prevalence Of Strongyle Infections And Persistent Efficacy Of Pyrantel Embonate, Ivermectin And Moxidectin In Turkish Horses, Veli̇ Yilgör Çirak, Ender Güleğen, Christian Bauer

Strongyle infection was detected in 68% of 320 horses from 9 farms in western Turkey. Egg counts per gram of faeces (EPG) were more than 950 in half of the animals. The persistent efficacy of 3 different anthelmintics to suppress cyathostome EPG was compared on a horse farm. One group of horses was treated with moxidectin on day 0, and the 2 other groups received pyrantel embonate or ivermectin on days 0 and 70. Pyrantel embonate reduced the EPG by >90% for 2 weeks; thereafter egg counts steadily increased. Ivermectin reduced the EPG by >98% for up to 6 weeks. …

Conjugated linoleic acid (CLA) is a group of geometrical and positional isomers of linoleic acid (C18:2, cis-9, cis-12). In contrast to linoleic acid, double bonds in CLA are usually located at positions 9 and 11 or 10 and 12 and each double bond can be either in the cis or trans configuration. Meat and dairy products from ruminant animals (such as milk, butter, yogurt and cheese) are the principal natural sources of CLA in the human diet. The endocrine cells of gastrointestinal tract of the Oreochromis niloticus were investigated using immunohistochemical techniques. 8 antisera were tested and 3 immunoreactivities were detected: Serotonin, glucagon and somatostatin immunoreactive cells. Substance P, insulin, gastrin, calcitonin gene related peptide (CGRP) and calbindin immunoreactive cells were not found.
