Background

Prostaglandins (PG), produced by the uterine endometrium, are key regulators of several reproductive events, including estrous cyclicity, implantation, pregnancy maintenance and parturition. Phospholipase A2 (PLA2) catalyzes the release of arachidonic acid from membrane phospholipids, the rate-limiting step in PG biosynthesis. The bovine endometrial (BEND) cell line has served as a model system for investigating regulation of signaling mechanisms involved in uterine PG production but information concerning the specific PLA2 enzymes involved and their role in regulation of this process is limited. Champagne coat color in horses is controlled by a single, autosomal-dominant gene (CH). The phenotype produced by this gene is valued by many horse breeders, but can be difficult to distinguish from the effect produced by the Cream coat color dilution gene (CR). Three sires and their families segregating for CH were tested by genome scanning with microsatellite markers. The CH gene was mapped within a 6 cM region on horse chromosome 14 (LOD = 11.74 for theta = 0.00). Retinal dopaminergic amacrine neurons (DA neurons) play a central role in reconfiguring retinal function according to prevailing illumination conditions, yet the mechanisms by which light regulates their activity are poorly understood. We investigated the means by which sustained light responses are evoked in DA neurons. Sustained light responses were driven by cationic currents and persisted in vitro and in vivo in the presence of L-AP4, a blocker of retinal ON-bipolar cells. Several characteristics of these L-AP4-resistant light responses suggested that they were driven by melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs), including long latencies, marked poststimulus persistence, and a peak …

Retinal cells which become ischemic will pass apoptotic signal to adjacent cells, resulting in the spread of damage. This occurs through open gap junctions. A class of novel drugs, based on primaquine (PQ), was tested for binding to connexin 43 using simulated docking studies. A novel drug has been synthesized and tested for inhibition of gap junction activity using R28 neuro-retinal cells in culture. Four drugs were initially compared to mefloquine, a known gap junction inhibitor. The drug with optimal inhibitory activity, PQ1, was tested for inhibition and was found to inhibit dye transfer by 70% at 10 μM. Retinal …

The Evaluation Of Selected 13 Polymorphic Microsatellite Markers For The Parentage Test Of Thai Holstein Cattle, Lerdchai Chintapitaksakul, Rapiphan Uavechanichkul

Nineteen families of 44 Thai Holstein cattle were evaluated for microsatellite characteristics and the parentage test. A total of 13 microsatellite markers were selected from the genome database and the genomic DNAs were isolated and used as templates for amplification. Multiplex PCR of these thirteen markers in a single reaction showed that only ten markers; b1, b2, b3, b4, b6, b7, b10, b11, b12 and b13 could amplify the fragments ranging from 5 to 12. The experiment was conducted to investigate the effects of renin-angiotensin blockade on renal function, renal norepinephrine (NE) contents and renal oxidative stress in Cyclosporine A (CsA) induced renal impairment in rats. Rats were assigned into three groups; group 1 (control group), receiving vehicle (propylene glycol) 1 ml/kg./day, subcutaneously (s/c). for 28 days; group 2 (CsA group), receiving CsA 15 mg/kg/day, s/c for 28 days; group 3 (CsA and losartan (LST) group), receiving CsA 15 mg/kg./day, s/c with the administration of LST 10 mg/k.g./day orally for 28 days. Treatment Of Infected Open Wounds On Two Dogs Using A Film Dressing Of Polysaccharide Extracted From The Hulls Of Durian (Durio Zibethinus Murr.): Case Report, Piyarat Chansiripornchai, Sunanta Pongsamart

The Thai Journal of Veterinary Medicine

There have been many reports on the efficacy of the polysaccharide peptidoglycan (PG) isolated from durian hulls in assisting wound healing in pig, dog, and cat skin. In this report, a two - year - old and a three -year - old female mongrel dogs with infected open wounds and skin necrosis were treated with PG film dressing. Complete wound healing in both dogs occurred within 14 weeks of treatment. PG film dressing may effectively promotes wound healing. The results show the benefit of using PG film dressings in pharmaceutical and veterinary applications.

The structure of blood cells and platelets in 11 macaques (2 stump-tailed macaques, 5 long-tailed macaques, 1 rhesus macaque and 3 pig-tailed macaques) were studied using light and transmission electron microscopy (TEM). Erythrocytes were non-nucleated, biconcave disks without cytoplasmic organelles. Neutrophils had numerous small specific granules. Eosinophils contained specific granules with a dense matrix and a pale crystalloid body. Basophils revealed large round specific granules. Lymphocytes possessed scant cytoplasm with some mitochondria and azurophilic granules. Monocytes contained a large number of mitochondria, azurophilic granules and endoplasmic reticulum. To investigate the effects of pentoxifylline (PTX) to enhance the motility and fertilization capacity of semen samples with the low-motile sperm in Asian elephants, fourteen semen collection attempts in 9 elephant bulls by manual stimulation were undertaken and eleven ejaculates fitted the criteria of investigation (0-30% motility). They were divided into two groups: poor-motile (0-9% motility) and low-motile (10-30% motility) sperm groups. The Correlation Between Plasma L-Carnitine And Growth Performance In Suckling Pigs In Respect Of Mrna Expression Of A Carnitine Transporter, Octn2, In Lactating Porcine Mammary Tissues, Boonrit Thongsong, Sarinee Kalandakanond-Thongsong

The Thai Journal of Veterinary Medicine

Two objectives of this study were to investigate the weekly change in plasma L-carnitine concentration during the 28 day suckling period and to determine the relative mRNA expression level of a L-carnitine transporter, OCTN2, in porcine mammary tissue at early and peak lactation. The results showed that the L-carnitine concentration of piglets after giving birth was 3.94±0.13 mg/L. L-carnitine levels on day 7 were significantly decreased (p<0.05) compared to those on the 1st day. On day 14, L-carnitine level increased significantly (p<0.05) to the concentration of 3.99±0.55 mg/L and maintained this level until weaning day. As with the increase in plasma L-carnitine concentration 1 week following suckling age, the body weight also increased significantly (p<0.05). The linear correlation of suckling ages and L-carnitine levels has a significant and positive correlation (R2Adj=0.1328, p<0.05). The mRNA expression of OCTN2 found in these lactating sow mammary tissues showed the relative abundance of this carnitine transporter. It tended to be decreased in lactating mammary tissue of sows at their peak compared to those at early lactation (p=0.065). Therefore, the significant change or maintenance in plasma L-carnitine concentration and body weight gain during suckling period provided an insight into the effects of L-carnitine in suckling piglets. Additionally, the mRNA expression of OCTN2 in the lactating mammary cells will be further investigated in cellular and molecular regulatory mechanisms to transport L-carnitine from dams to their piglets via the mammary glands.

Thioredoxin (TRx) is known to control redox homeostasis in cells. In recent years, a specific TRx binding protein called thioredoxin binding protein-2 (TBP-2) was found in other cell types and it appeared to negatively regulate TRx bioavailability and thereby control TRx biological function. In view of the sensitivity of lens transparency to redox status, proper regulation of TRx bioavailability is of the utmost importance. BACKGROUND: Identification of biomarkers among thousands of genes arrayed for disease classification has been the subject of considerable research in recent years. These studies have focused on disease classification, comparing experimental groups of effected to normal patients. Related experiments can be done to identify tissue-restricted biomarkers, genes with a high level of expression in one tissue compared to other tissue types in the body.

The susceptibility of wild ruminants, especially cervids, to bovine viral diarrhea virus (BVDV) has remained an enigma. Two white-tailed deer (Odocoileus virginianus) were submitted to the Animal Disease Research and Diagnostic Laboratory (ADRDL) in the fall of 2003 by the South Dakota Game Fish and Parks for chronic wasting disease (CWD) testing. Both animals were CWD negative. The animals were necropsied and histopathology, viral antigen detection, and virus isolation were performed. A noncytopathic (NCP) BVDV was isolated from the lungs and several other tissues of both animals. Formalin-fixed ear notches from both animals were positive for BVDV antigen by immunohistochemistry. …

In swine, the most common and severe enterotoxigenic Escherichia coli (ETEC) infections are caused by strains that express K88 (F4)+ fimbriae, heat-labile enterotoxin (LT), heat-stable enterotoxin b (STb), and enteroaggregative E. coli heat-stable toxin 1. This was the first infected herd identified since Minnesota’s Accredited Free (AF) status was obtained in 1976. Subsequent testing identified infection in white-tailed deer and several cattle herds adjacent to this index herd. In February 2008, Minnesota declared its 11th infected cattle herd, resulting in the downgrade of the entire State’s TB status to Modified Accredited (MA).

The amount of new information of the last seven years regarding Lawsonia intracellularis sensitivity to antimicrobial drugs, prevention/control based on treatment and vaccination, diagnosis and, finally, genome is amazing. Nevertheless, not much is known about the epidemiology of the disease, mainly regarding the source of infection, resistance of the bacteria in the environment and possible biological vectors that could spread the infection among farms. It was learned that as few as 1,000 to 10,000 bacteria are enough to induce infection and fecal shedding two to three weeks after inoculating susceptible pigs.
