Transferability Of Barley Retrotransposon Primers To Analyze Genetic Structure In Iranian Hypericum Perforatum L. Populations, Razea Asadkhani Mamaghani, Seyed Abolghasem Mohammadi, Saeid Aharizad

Turkish Journal of Botany

Transferability of barley retrotransposon primers was investigated to analyze population structure in St. John's wort (Hypericum perforatum L.) based on inter-retrotransposon amplified polymorphism (IRAP). Seven long terminal repeat (LTR) retrotransposon primers derived from the barley genome were used to detect genetic polymorphism in eight Iranian populations and three cultivars (Helos, New Stem, and Topaz) of H. Perforatum based on IRAP analysis. Nine possible LTR primers/primer combinations successfully amplified fragments from the H. Perforatum genome. In total, 311 bands of 100-3000 base pairs were amplified, of which 244 were polymorphic. The number of polymorphic fragments ranged from 10 (Nikita/5'LTR-2) to 57 …

Genetic Diversity And Relationships Within And Among Onobrychis Species Using Molecular Markers, Mohammad Zarrabian, Mohammad Mahdi Majidi

Turkish Journal of Botany

Little is known about the taxonomic relationships of the genus Onobrychis (Fabaceae). To study inter- and intraspecific variations and the relationships among the species, 102 accessions (33 species) of the genus Onobrychis were evaluated using 22 inter-simple sequence repeat (ISSR) markers. Almost all the species belonging to the section Onobrychis clustered together. A similar pattern was found with the species belonging to the section Hymenobrychis, while those of the section Lophobrychis did not cluster together. The results indicate that the section Lophobrychis has a comparatively derived organization that can be attributed to the differences in their taxonomic delimitation. Geographical pattern …

Relationships And Generic Delimitation Of Eurasian Genera Of The Subtribe Asterinae (Astereae, Asteraceae) Using Molecular Phylogeny Of Its, Elena Korolyuk, Alexey Makunin, Tatiana Matveeva

Turkish Journal of Botany

The subtribe Asterinae (Astereae, Asteraceae) includes highly variable, often polyploid species. Recent findings based on molecular methods led to revision of its volume. However, most of these studies lacked species from Eurasia, where a lot of previous taxonomic treatments of the subtribe exist. In this study we used molecular phylogenetics methods with internal transcribed spacer (ITS) as a marker to resolve evolutionary relations between representatives of the subtribe Asterinae from Siberia, Kazakhstan, and the European part of Russia. Our reconstruction revealed that a clade including all Asterinae species is paraphyletic. Inside this clade, there are species with unresolved basal positions, …

Metabolic And Molecular-Genetic Regulation Of Proline Signaling And Itscross-Talk With Major Effectors Mediates Abiotic Stress Tolerance In Plants, Aryadeep Roychoudhury, Aditya Banerjee, Vikramjit Lahiri

Turkish Journal of Botany

Proline (Pro) accumulation is a common response of several plant species to combat abiotic stresses. Under stress conditions, Pro acts as an excellent compatible solute in the plant system, participating in the alleviation of stress sensitivity. Though the metabolic pathways associated with Pro are well studied, parts of its regulatory cascades are still not properly known. It has also been conjectured that epigenetic modifications regulate Pro metabolism during abiotic stress. Apart from Pro, the plant abiotic stress responses are essentially mediated by multiple effectors. Hence, proper analysis of the cross-talks of Pro with the other components of the abiotic stress …

Genetic Improvement Of Rice For Biotic And Abiotic Stress Tolerance, Mahmood Ur Rahman Ansari, Tayyaba Shaheen, Shazai Bukhari, Tayyab Husnain

Turkish Journal of Botany

Rice (Oryza sativa L.) is among the most important food crops that provide a staple food for nearly half of the world's population. Rice crops are prone to various types of stresses, both biotic and abiotic. Biotic stresses include insect pests, fungus, bacteria, viruses, and herbicide toxicity. Among abiotic stresses, drought, cold, and salinity are also well studied in rice. Various genes have been identified, cloned, and characterized to combat these stresses and protect rice crops. The identified genes are successfully transformed into rice plants to produce transgenic plants. These transgenic rice plants are being evaluated under field conditions in …

Turkish Journal of Botany

Sustainable potato production practices are crucial for food security and social sustainability in the future since potato is a highly nutritious food and it is considered as one of the most promising crops to reduce human hunger and poverty in the world due to its high yield potential. However, being a temperate crop, potato is exposed to various environmental stresses, including extended periods of drought and heat. The majority of potato genomics, transcriptomics, and transgenics studies concentrate on the characterization of molecular mechanisms governing cold hardiness of tubers and response and tolerance mechanisms against diseases. Likewise, potato breeding studies focus …

Soil Bacteria Conferred A Positive Relationship And Improved Salt Stress Tolerance In Transgenic Pea (Pisum Sativum L.) Harboring Na+/H+ Antiporter, Zahid Ali, Nasr Ullah, Saadia Naseem, Muhammad Inam Ul Haq, Hans Joerg Jacobsen

Turkish Journal of Botany

Among grain legumes, peas (Pisum sativum L.) are highly sensitive to salt stress. Acclimatization of plants to such conditions is mandatory. We provide improved salt stress tolerance response of transgenic pea plants overexpressing the Na+/H+ gene from Arabidopsis thaliana and a positive association with salt-tolerant plant growth-promoting rhizobacteria (PGPR). In addition to salt stress tolerance and phosphate solubilization, the selected rhizobacterial isolates were identified for indole acetic acid and proline production ability. Seed germination percentage in transgenic pea plants was significantly higher under NaCl challenge. The wild-type (WT) pea plants inoculated with known numbers of viable cells of salt-tolerant PGPR …

Turkish Journal of Botany

Bean is an edible grain legume species with the greatest selectivity in terms of ecological conditions. Increasing the yield per unit area depends on identification and breeding of species resistant to different ecological conditions. In this regard, we aimed to evaluate mRNA levels of the PvLEA3 gene and lipid peroxidation levels in 7 different bean varieties subjected to NaCl and PEG stresses. Lipid peroxidation analysis (MDA) and qRT-PCR analysis were performed with root and leaf tissues sampled at 3 and 27 h of stress conditions. All results showed that the PvLEA3 gene plays a key role in the defense mechanism …

Estimating Genetic Diversity Among Selected Cotton Genotypes And The Identificationof Dna Markers Associated With Resistance To Cotton Leaf Curl Disease, Ammad Abbas, Muhammad Atif Iqbal, Mehboob-Ur Rahman, Andrew H. Paterson

Turkish Journal of Botany

To the extent of our knowledge, applications of DNA markers in marker-assisted breeding of cotton are handicapped due to low genetic diversity in cotton germplasm. Cotton leaf curl disease, a disease of viral origin, has substantially depressed cotton production in Pakistan, and this disease is also an emerging threat to the neighboring cotton-growing countries like China and India. The present study was designed to identify DNA markers, predominately simple sequence repeats (SSRs), associated with tolerance and/or resistance to the disease. Based upon 2 years of disease-screening field experiments, a total of 10 cotton genotypes (five highly tolerant, four highly susceptible, …

Evaluation Of Metabolite Extraction Protocols And Determination Ofphysiological Response To Drought Stress Via Reporter Metabolites In Model Plant Brachypodium Distachyon, Özge Tatli, İlkem Emrah Ni̇kerel, Bahar Soğutmaz Özdemi̇r

Turkish Journal of Botany

Metabolomics aims to systematically gather (quantitative) information on metabolites in the cell and is commonly viewed as the "missing link" between genomics, transcriptomics, and physiology. Typical metabolomics platforms consist of two main steps, the quenching and extraction of metabolites from plant material and the (un)targeted quantification of the extracted metabolites. Brachypodium distachyon, a native grass species of the Mediterranean region, is an attractive model plant to study temperate crops. In recent years, despite an increasing interest in genomic and transcriptomic studies, metabolomics studies for B. Distachyon are still in their infancy. Drought, an abiotic stress factor, causes severe loss in …

Turkish Journal of Botany

In the present study, to determine the effects of β-estradiol on the ability of plants to tolerate lead toxicity, β-estradiol (10 μM) and lead (1.75 mM), singly or in combination, were exogenously applied to wheat seeds. Although lead resulted in a marked increase in the activities of antioxidant enzymes, including superoxide dismutase, guaiacol peroxidase, ascorbate peroxidase, and glutathione reductase (but not catalase), as well as an increase in the level of antioxidant compounds such as ascorbic acid and glutathione, this was insufficient to ameliorate the lead-induced oxidative injury or the superoxide anion, hydrogen peroxide, and malondialdehyde levels. However, β-estradiol was …

Functional Analysis Of Drought-Resistant Genes In Barley Using Vigs, Natalie Catherine Martin Jan 2015

Functional Analysis Of Drought-Resistant Genes In Barley Using Vigs, Natalie Catherine Martin

Honors Program Theses

The following study analyzed the role of the gamma-aminobutyric acid transaminase (GABAT) gene in the response of barley plants to drought stress. Expression of GABAT in barley was suppressed using virus-induced gene silencing (VIGS). In VIGS, an RNA virus is used to replicate a small portion of the target RNA. In this study, barley stripe mosaic virus (BSMV), that carries a piece of the GABAT gene to infect barley, was used. As the virus replicates inside infected barley plants, it also makes RNA that is complementary to GABAT messenger RNA (mRNA) that is normally made by the host. The presence …

Analysis Of Ipomoea (Morning Glory) Leaf Mutants, Abigail Anne Lee

Honors Program Theses

Ipomoea (morning glory) is the largest genus in the family Convolvulaceae. Cultivation of morning glory plants began in the late Edo period in Japan, and this horticultural success resulted in thousands of plants with varying floral displays. In addition to the different flower morphologies, leaf shape is highly variable within this family making it an ideal group in which to study leaf development. We selected four mutants that vary in lobe number and lobe depth: 1) Tokyo Standard (TKS1065; wild-type; 3 lobes), yellow maple (ym1018; 5 lobes), delicate maple (dlm620; 5 deep lobes), and maple willow …

Major challenges still exist in the cost-competitive production of cellulosic ethanol. Lignocellulose deconstruction using physiochemical pretreatments continues to be the most efficient way to remove lignin and expose cellulose for enzyme-mediated sugar reduction. Recent interest in the development of “value-added” lignin co-products and the desire to reduce the use of hazardous materials has prompted a resurgence of interest in alternative lignocellulosic feedstock and research into environment-friendly biological pretreatments. This article provides an overview of bioethanol processes and economics, standard and alternative feedstocks, physiochemical and biological pretreatments, and lignin chemistry. The chemistry and mode of action of ligninolytic and cellulolytic enzymes …

The Relationship Between Riparian Zone Width And Floristic Quality In Shenandoah County, Virginia, Jamie D. Smith Dec 2014

The Relationship Between Riparian Zone Width And Floristic Quality In Shenandoah County, Virginia, Jamie D. Smith

Masters Theses, 2010-2019

Riparian zones harbor an above average plant biodiversity. This biodiversity is threatened by invasive species and increasing human disturbance, the latter of which includes deforestation from agriculture and urban development. In this study, I examine relationship between the width of a forested riparian zone and the vegetation growing there. By using floristic quality assessment as a measure of anthropogenic disturbance, one can determine if wider riparian zones foster exclusion of non-native species while providing higher quality habitats for native plants. A randomized block design was used with three forested riparian treatments: deforested, moderately forested (woody vegetationstream), and extensively forested (woody …

Department of Agronomy and Horticulture: Faculty Publications

Seed oils have proved recalcitrant to modification for the production of industrially useful lipids. Here, we demonstrate the successful metabolic engineering and subsequent field production of an oilseed crop with the highest accumulation of unusual oil achieved so far in transgenic plants. Previously, expression of the Euonymus alatus diacylglycerol acetyltransferase (EaDAcT) gene in wild-type Arabidopsis seeds resulted in the accumulation of 45 mol% of unusual 3-acetyl-1,2- diacyl-sn-glycerols (acetyl-TAGs) in the seed oil (Durrett et al., 2010 PNAS 107:9464). Expression of EaDAcT in dgat1 mutants compromised in their ability to synthesize regular triacylglycerols increased acetyl-TAGs to 65 mol%. Camelina and soybean …

The Mechanism Of Lhcp Insertion Into Thylakoid Membranes, Larae Brown Dec 2014

The Mechanism Of Lhcp Insertion Into Thylakoid Membranes, Larae Brown

Graduate Theses and Dissertations

The light harvesting chlorophyll a/b-binding proteins (LHCPs) are the most abundant membrane proteins. LHCP is a nuclear encoded protein which is targeted to the thylakoid membranes by chloroplast signal recognition particles (cpSRP). Insertion into thylakoid membranes is facilitated by the cpSRP receptor cpFtsY and the Alb3 translocase. Work here focused on understanding the molecular events of LHCP insertion into the thylakoid membranes. Specifically, we sought to develop a tool to detect the insertion of the lumen-localized loop of LHCP into thylakoid membranes, which relies on cleavage of the loop by a thylakoid lumen processing protease. We also sought to understand …

Functional Genomics And Microbiome Profiling Of The Asian Longhorned Beetle (Anoplophora Glabripennis) Reveal Insights Into The Digestive Physiology And Nutritional Ecology Of Wood Feeding Beetles, Erin D. Scully, Scott M. Geib, John E. Carlson, Ming Tien, Duane Mckenna, Kelli Hoover

Department of Agronomy and Horticulture: Faculty Publications

Background: Wood-feeding beetles harbor an ecologically rich and taxonomically diverse assemblage of gut microbes that appear to promote survival in woody tissue, which is devoid of nitrogen and essential nutrients. Nevertheless, the contributions of these apparent symbionts to digestive physiology and nutritional ecology remain uncharacterized in most beetle lineages.

Results: Through parallel transcriptome profiling of beetle- and microbial- derived mRNAs, we demonstrate that the midgut microbiome of the Asian longhorned beetle (Anoplophora glabripennis), a member of the beetle family Cerambycidae, is enriched in biosynthetic pathways for the synthesis of essential amino acids, vitamins, and sterols. Consequently, the midgut …

Academic, Government & Associated Organizational Research Resources

Situated at the northwest end of Santa Cruz County and occupying circa 30 square miles of sharply contrasted terrain, the Scott Creek Watershed concentrates within its geomorphological boundaries, at least 10-12% of California's flora, both native and introduced. Paradoxically, the use/abuse that the watershed has sustained over the past 140+ years, has not necessarily diminished the biodiversity and perhaps parallels the naturally disruptive but biologically energizing processes (fire, flooding, landslides and erosion), which have also been historically documented for the area for +60 years. Department of Agronomy and Horticulture: Faculty Publications

Background:

The homeodomain leucine zipper (HD-Zip) transcription factor family is one of the largest plant specific superfamilies, and includes genes with roles in modulation of plant growth and response to environmental stresses. Many HD-Zip genes are characterized in Arabidopsis (Arabidopsis thaliana), and members of the family are being investigated for abiotic stress responses in rice (Oryza sativa), maize (Zea mays), poplar (Populus trichocarpa) and cucumber (Cucmis sativus). Findings in these species suggest HD-Zip genes as high priority candidates for crop improvement.

Results:

In this study we have identified members of …
