To test the efficacy of RNA interference (RNAi) as a method for target-site screening in Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae) larvae, genes were identified and tested for which clear RNAi phenotypes had been identified in the Coleopteran model, Tribolium castaneum. Here the cloning of the D. v. vergifera orthologs of laccase 2 (DvvLac2) and chitin synthase 2 (DvvCHS2) is reported. Injection of DvvLac2-specific doublestranded RNA resulted in prevention of post-molt cuticular tanning, while injection of DvvCHS2- specific dsRNA reduced chitin levels in midguts. Silencing of both DvvLac2 and DvvCHS2 was confirmed by RT-PCR and quantitative RT-PCR. As in T. …

The higher fungi exhibit a dichotomy with regard to urea utilization. The hemiascomycetes use urea amidolyase (DUR1,2), whereas all other higher fungi use the nickel-containing urease. Urea amidolyase is an energy-dependent biotincontaining enzyme. It likely arose before the Euascomycete/Hemiascomycete divergence c. 350 million years ago by insertion of an unknown gene into one copy of a duplicated methylcrotonyl CoA carboxylase (MccA). The dichotomy between urease and urea amidolyase coincides precisely with that for the Ni/Co transporter (Nic1p), which is present in the higher fungi that use urease and is absent in those that do not. Candida albicans causes candidiasis, secretes farnesol, and switches from yeast to hyphae to escape from macrophages after phagocytosis. However, before escape, macrophages may respond to C. albicans’ pathogen-associated molecular patterns (PAMPs) through toll-like receptor 2 (TLR2) and dectin-1 receptors by expressing cytokines involved in adaptive immunity, inflammation, and immune regulation. Therefore, macrophages and the RAW264.7 macrophage line were challenged with C. albicans preparations of live wild-type cells, heat-killed cells, a live mutant defective in hyphae formation, a live mutant producing less farnesol, or an isolate producing farnesoic acid instead of farnesol. Soil organic carbon (SOC) is directly related to soil organic matter (SOM), and is important in the functionality and stability of terrestrial ecosystems. Soil organic carbon is also an important pool for C storage and small changes of SOC may cause significant gains and losses in the large scale carbon cycle. The mechanisms by which SOC is transported and stored however are not widely studied. Escherichia coli isolates (72 commensal and 10 O157:H7 isolates) were compared with regard to physiological and growth parameters related to their ability to survive and persist in the gastrointestinal tract and found to be similar. We propose that nonhuman hosts in E. coli O157:H7 strains function similarly to other E. coli strains in regard to attributes relevant to gastrointestinal colonization.

The use of ocean observatories is expanding with the potential for collecting serial data with high temporal resolution at multiple sites within an ecosystem. Integration of underwater video cameras in observatory systems allow observation of vagile species and are useful tools for observing variations in behavior over time. Much of the decline of the U.S. sheep industry has been attributed to losses caused by predators. Most predatory losses are inflicted on lambs rather than ewes. Losses have historically ranged from 4-8% of lamb crops, inflicting significant financial loss on ranchers. However, most research providing data on sheep predation is over 20 years old. Changes in the sheep industry as well as predation rates may make previous loss rates inapplicable to current conditions. Spatiotemporal variability in the diatom and dinoflagellate community structure and chlorophyll a (chl a) concentrations in Tumaco Bay during the 1993-2005 period was related clearly to seasonal and interannual variability in environmental conditions due to the migration of the Inter-tropical Convergence Zone (ITZC) and the influence of El Nino Southern Oscillation (ENSO) events. A total of 134 species of diatoms that belong to 57 genera, and 78 species of dinoflagellates that belong to 25 genera were identified during the survey. The small mammal communities of central Africa are not well understood, and the southwestern section of Gabon has not been previously surveyed except for some recent work in Monts Doudou. At five sites within the Gamba Complex we set out to 1) document the species of terrestrial rodents (Muridae) and shrews (Soricidae) weighing less than 100 g, 2) compare the community composition between inland and coastal sites, and 3) evaluate our sampling protocols. Using a combination of pitfall lines, Sherman live traps, and snap traps we captured 721 individuals of 12 rodent and 10 shrew species in 15,792 trap-nights. The …

Crested wheatgrass (Agropyron cristatum [L.] Gaertn.) is one of the most commonly seeded grass species in the western United States and dominates thousands of hectares in the Great Basin. Although many degraded Wyoming big sagebrush (Artemisia tridentata ssp. wyomingensis) plant communities have been seeded with crested wheatgrass, successional pathways, influence of soil attributes, and cultivation history on the vegetation of these communities have not been fully characterized. Drug release and its relationship with kinetic and thermodynamic parameters of drug sorption onto biodegradable and biocompatible fibers made of polylactic acid (PLA), starch acetate (SA), wheat gluten (WG) and soy protein (SP) have been studied using Diclofenac, 5 Fluorouracil (5-Fu) and Metformin as drug models. Two years ago on a Saturday morning, I was asked quite pointedly by two friends how I could be both spiritual and a scientist. My questioning friends felt that these qualities were incompatible. It turned out their church taught that scientists were ‘the enemy’ because scientists believe in evolution. Since that time, Letters to the Editor regarding evolution have attracted my attention. One animal beautifully adapted to the viscous asphalt of the La Brea tar pits is the oil fly Helaeomyia petrolei (Syn. Psilopa). As a normal part of its carnivorous existence the oil fly larval guts are filled with tar, with no adverse effects. Surface sterilized larvae 5 contained ca. 2 x 10 heterotrophic bacteria per larva. These bacteria have been identified as a mixture of enteric bacteria, most commonly Providencia rettgeri, and Acinetobacter spp. These bacteria were clearly growing because their numbers in the larval guts were 100 to 1,000 times greater than in free oil/asphalt. Field studies were conducted during the summers of 2007 and 2008 at the Agricultural Research and Education Complex, Western Kentucky University, Warren County, KY and commercial production fields in Caldwell County, KY, Warrick County IN, and Vanderburgh County, IN. The goals of these studies were to further validate the Duncan grain yield model, the Russell aboveground biomass model, and to study the effect of inconsistent spacing within rows on Zea mays L. yield. All plants contain primary and secondary compounds. Primary compounds are needed by plants and herbivores for maintenance, growth, and reproduction, while secondary compounds play roles as diverse as protecting plants from ultraviolet radiation, defenses against herbivores, pollination attraction, and stress resistance. Secondary compounds have nutritional and medicinal benefits for herbivores as well, especially when eaten in diverse combinations that complement one another. While complementarities among secondary compounds are an important but little understood area of plant-herbivore interactions, even less is known about how the sequences of eating plants with different compounds affects foraging behavior, though they may be critical. Candida albicans is a dimorphic fungus that can interconvert between yeast and filamentous forms. Its ability to regulate morphogenesis is strongly correlated with virulence. Tup1, a transcriptional repressor, and the signaling molecule farnesol are both capable of negatively regulating the yeast to filamentous conversion. Based on this overlap in function, we tested the hypothesis that the cellular response to farnesol involves, in part, the activation of Tup1. Tup1 functions with the DNA binding proteins Nrg1 and Rfg1 as a transcription regulator to repress the expression of hypha-specific genes. The tup1/tup1 and nrg1/nrg1 mutants, but not the rfg1/rfg1 mutant, failed to …

Women's perceptions of technological changes in textiles and apparel and how the changes have affected daily life were examined using the transmissive reminiscence technique. Technological advances do contribute to use of less time and effort and offer a greater variety of products, but the products often have a shorter life expectancy or are kept in inventory for a shorter time because it is more expedient to replace rather than repair or recycle textile and clothing items. Care must be taken to cultivate cohesive relationships within the family and teach values that were once inherent in textiles/ apparel tasks. Otherwise advances …

Colby College Watershed Study: Pattee Pond (2008, 1992)
The Colby Environmental Assessment Team (CEAT) investigated the impact of land use patterns on the water quality of Pattee Pond in Winslow, Maine, during the summer and fall of 2008. Physical, chemical, and biological characteristics of water quality were analyzed to evaluate the current health of the lake. Data collected were compared with previous studies conducted by the Maine Department of Environmental Protection (DEP) and CEAT to examine changes in water quality and land use over time. Trends in Pattee Pond water quality suggest an improvement since the 1970s. However, the lake is still experiencing algal blooms resulting from phosphorus …

Echinacea purpurea is an herbal supplement used to reduce symptoms of common colds and flu-like illnesses. As of right now, it is not well understood what chemical components in the plant are immunologically active. The four main potentially immunologically active chemical components in E. purpurea are caffeic acid derivatives, alkamides, polyacetylenes, and polysaccharides. The geological makeup of Maine’s mountains in the western part of the state, and the exposed coastline provide opportune areas to capture wind and convert it into energy. The information included in this poster will suggest the most likely areas for wind development based on a number of factors as recommended by the American Wind Energy Association.

Sexual signals are commonly used among animals to enhance reproductive success. Chemical signals such as pheromones are commonly used to attract mates. Little is known, though, about how these signals are received or interpreted by the target. I studied the water spider, Dolomedes scriptus in the pisaurid family. Most Dolomedes species only make webs as a nursery for their young. However, females often create silk drag lines, potentially serving for species recognition. In some Dolomedes species, females who are previously mated are known to cannibalize most approaching males. Males might avoid cannibalism by avoiding mated females, particularly if they can …

Candida albicans is a diploid fungus that undergoes a morphological transition between budding yeast, hyphal, and pseudohyphal forms. The morphological transition is strongly correlated with virulence and is regulated in part by quorum sensing. Candida albicans produces and secretes farnesol that regulates the yeast to mycelia morphological transition. Mutants that fail to synthesize or respond to farnesol could be locked in the filamentous mode. To test this hypothesis, a collection of C. albicans mutants were isolated that have altered colony morphologies indicative of the presence of hyphal cells under environmental conditions where C. albicans normally grows only as yeasts.
