Genome sequences are available for the human blood flukes, Schistosoma japonicum, S. mansoni and S. haematobium. Functional genomic approaches could aid in identifying the role and importance of these newly described schistosome genes. Transgenesis is established for functional genomics in model species, which can lead to gain- or loss-of-functions, facilitate vector-based RNA interference, and represents an effective forward genetics tool for insertional mutagenesis screens. Progress toward routine transgenesis in schistosomes might be expedited if germ cells could be reliably localized in cultured schistosomes. To the Editor: In 2009, an alveolar hydatid cyst, the intermediate stage of the cestode Echinococcus multilocularis, was detected in the liver of a dog from Quesnel, British Columbia (BC), Canada, 600 km west of the nearest known record of this parasite in central North America. Alveolar hydatid cysts normally occur in rodent intermediate hosts. However, humans can serve as aberrant intermediate hosts; cysts generally originate in the liver and, in about one third of cases, metastasize throughout the body. Opisthorchis viverrini is considered among the most important of the food-borne trematodes due to its strong association with advanced periductal fibrosis and bile duct cancer (cholangiocarcinoma). We investigated the relationship between plasma levels of Interleukin (IL)-6 and the risk of developing advanced fibrosis and bile duct cancer from chronic Opisthorchis infection. We show that IL-6 circulates in plasma at concentrations 58 times higher in individuals with advanced fibrosis than age, sex, and nearest-neighbor matched controls and 221 times higher in individuals with bile duct cancer than controls. Background
Schistosoma japonicum is a major public health concern in the Peoples' Republic of China (PRC), with about 800,000 people infected and another 50 million living in areas at risk of infection. Based on ecological, environmental, population genetic and molecular factors, schistosomiasis transmission in PRC can be categorised into four discrete ecosystems or transmission modes. It is predicted that, long-term, the Three Gorges Dam (TGD) will impact upon the transmission of schistosomiasis in the PRC, with varying degree across the four transmission modes.

Background: Recent findings of Plasmodium in African apes have changed our perspectives on the evolution of malarial parasites in hominids. However, phylogenetic analyses of primate malarias are still missing information from Southeast Asian apes. In this study, we report molecular data for a malaria parasite lineage found in orangutans.
Methodology/Principal Findings: We screened twenty-four blood samples from Pongo pygmaeus (Kalimantan, Indonesia) for Plasmodium parasites by PCR. The large extracellular loop of the Schistosoma mansoni tetraspanin, Sm-TSP-2, when fused to a thioredoxin partner and formulated with Freund's adjuvants, has been shown to be an efficacious vaccine against murine schistosomiasis. Moreover, Sm-TSP-2 is uniquely recognised by IgG1 and IgG3 from putatively resistant individuals resident in S. mansoniendemic areas in Brazil. In the present study, we expressed Sm-TSP-2 at high yield and in soluble form in E. coli without the need for a solubility enhancing fusion partner. Background: The majority of Haemosporida species infect birds or reptiles, but many important genera, including Plasmodium, infect mammals. Dipteran vectors shared by avian, reptilian and mammalian Haemosporida, suggest multiple invasions of Mammalia during haemosporidian evolution; yet, phylogenetic analyses have detected only a single invasion event. Until now, several important mammal-infecting genera have been absent in these analyses. This study focuses on the evolutionary origin of Polychromophilus, a unique malaria genus that only infects bats (Microchiroptera) and is transmitted by bat flies (Nycteribiidae).

The mucosal cytokine response of healthy humans to parasitic helminths has never been reported. We investigated the systemic and mucosal cytokine responses to hookworm infection in experimentally infected, previously hookworm naive individuals from non-endemic areas. We collected both peripheral blood and duodenal biopsies to assess the systemic immune response, as well as the response at the site of adult worm establishment. Our results show that experimental hookworm infection leads to a strong systemic and mucosal Th2 (IL-4, IL-5, IL-9 and IL-13) and regulatory (IL-10 and TGF-β) response, with some evidence of a Th1 (IFN-γ and IL-2) response. Parasite data were collected from unlogged and selectively logged lowland rain forests in Borneo. Scat from a female Viverra tangalunga (Malay civet) in the unlogged forest contained eggs of Capillaria spp., Trichuris sp., Mamomoganomus sp., and nematode larvae. Scat attributed to V. tangalunga contained reproductive products (eggs/oocysts/cysts/larvae) of Capillaria sp., Toxocara sp., Ancylostoma sp. (probably A. ceylanicum), Isospora spp., strongylate-type eggs (probably Ancylostoma spp.), Paragoni mus sp. and other Trematoda eggs, Monocystis sp. cysts, Gnathostoma sp. (probably G. spinigerum), the lungworm Viverrostrongylus brauni, pinworm-type eggs, Eimeria spp., Sarcocystis-type sporocyst, dermanysid mites and mesostigmatid mites. The goal of this research was to determine the extent of resistance that turkey roundworms, Ascaridia dissimilis, have developed to anti-parasitic chemicals used in commercial poultry operations. Roundworm infections in turkeys have resulted in monetary losses for the poultry industry for years, generally due to poor feed conversion. The infection itself is subclinical and many turkeys have a light to moderate worm burden. Since parasitisms are light, this leads to the infections being noticed only during processing. Candidatus Rickettsia andeanae was identified during an investigation of a febrile outbreak in northwestern Peru (2002). DNA sequencing from two ticks (Amblyomma maculatum, Ixodes boliviensis) collected during the investigation revealed a novel Rickettsia agent with similarity to the spotted fever group rickettsiae. Since then, Candidatus R. similar to andeanae has been detected in A. maculatum ticks collected in the southeastern and southcentral United States, Argentina, and Peru. Leishmania are protozoan parasites that are transmitted by a sand fly vector. These parasites affect not only humans but also wild animals including domestic dogs and rodents, which form an additional challenge and public health problem to control the disease. Leishmaniasis is an important disease with worldwide distribution, including Saudi Arabia, the Middle East, and other tropical and subtropical areas around the world. Due to the expansion of irrigation and agricultural activities, more exposure to sand fly occurs, which leads to the expansion of leishmaniasis infections as newly emerging disease.

Specimens originally identified as Arostrilepis horrida from the Nearctic are revised, contributing to the recognition of a complex of cryptic species distributed across the Holarctic region. Previously unrecognized species are described based on specimens in cricetid (Neotominae) and geomyid rodents. Arostrilepis mariettavogeae n. sp. in Peromyscus californicus from Monterey County, California, and Arostrilepis schilleri n. sp. in Thomomys bulbivorus from Corvallis, Oregon, are characterized. We report the distribution of 2 species of filarioid nematodes occurring in different hosts in the central region of South America. Molinema boliviensis n. sp. was recorded as a parasite of sigmodontine and echymyid rodents in Bolivia, and Litomosoides esslingeri was recorded in sigmodontine and ctenomyid rodents from Bolivia and Argentina. Molinema boliviensis n. sp. shares several similarities with other species reported in spiny rats; however, it can be easily differentiated by the presence of a flat anterior end, gradually tapering lappets and a tubercle present in posterior end, a short, uniform buccal capsule, an oval-shaped vagina vera, and a …

The Great American Interchange resulted in the mixing of faunistic groups with different origins and evolutionary trajectories that underwent rapid diversification in North and South America. As a result, groups of animals of recent arrival converged into similar habits and formed ecological guilds with some of the endemics. We present a reconstruction of the evolutionary events in Aspidoderidae, a family of nematodes that infect mammals that are part of this interchange, i.e., dasypodids, opossums, and sigmodontine, geomyid, and hystricognath rodents. We present the first reconstruction of the parasitoses among the people of the Loma San Gabriel culture, as represented by 36 coprolites excavated from the Cueva de los Muertos Chiquitos in Durango, Mexico. The coprolites date to approximately 1,400-yr ago. Species identified based on eggs recovered include the trematode Echinostoma sp., the tapeworms Hymenolepis sp. and Dipylidium caninum, and the nematodes Ancylostoma duodenale, Enterobius vermicularis, and Trichuris trichiura. After rehydration and screening, 2 methods were used to recover eggs from these samples including spontaneous sedimentation and flotation. Akodonema luzsarmientae n.g., n.sp. (Nemata: Metastrongyloidea) is described from the pulmonary arteries and heart from several individuals of "soft grass mouse," Akodon mollis (Rodentia: Cricetidae), collected in the region of Ancash, Peru. The new genus and species is distinguished by a reduction of the dorsal ray to two small widely separated papillae.

A new species of amphistome digenean from the stomach and intestine of Podocnemis expansa (Pelomedusidae), of the tropical rain forest, from the State of Pará, Brazil, is described and allocated to a new genus (Oriximinatrema noronhae). The new species is characterized by the presence of an esophageal bulb, an esophageal extension uncovered by an extension of the pharyngeal sacs, a well-developed cirrus sac, post-bifurcal genital sucker, a ventro-terminal acetabulum with an anterior lip, and medium-sized eggs. This is the first report of a Dadayiinae trematode infecting a reptilian host.

Traditional concepts of the Bering Land Bridge as a zone of predominantly eastward expansion from Eurasia and a staging area for subsequent colonization of lower latitudes in North America led to early inferences regarding biogeographic histories of North American faunas, many of which remain untested. Here we apply a host–parasite comparative phylogeographical (HPCP) approach to evaluate one such history, by testing competing biogeographic hypotheses for five lineages of host-specific parasites shared by the collared pika (Ochotona collaris) and American pika (Ochotona princeps) of North America. Collections to explore helminth diversity among free-ranging ungulates in the North American Arctic revealed the occurrence of a third male, or "davtiani," morphotype for Teladorsagia boreoarcticus. Designated as T. boreoarcticus forma (f.) minor B, the males occurred with T. boreoarcticus f. major and T. borearcticus f. minor A in endemic populations of muskoxen (Ovibos moschatus wardi) and barrenground caribou (Rangifer tarandus groenlandicus) on Victoria Island, Nunavut, Canada, and in muskoxen and Peary caribou (Rangifer tarandus pearyi) on Banks Island, Northwest Territories, Canada. Marshallagia lichtenfelsi sp. n. is a dimorphic ostertagiine nematode occurring in the abomasum of mountain goats, Oreamnos americanus, from the Western Cordillera of North America. Major and minor morphotype males and females are characterized and distinguished relative to the morphologically similar Marshallagia marshalli/Marshallagia occidentalis from North America and Marshallagia dentispicularis, along with other congeners, from the Palearctic region. The configuration of the convoluted and irregular synlophe in the cervical region of males and females of M. lichtenfelsi is apparently unique, contrasting with a continuous and parallel system of ridges among those species of Marshallagia, including …

The animals traditionally studied by parasitologists range from protozoa to arthropods, and include all types of internal and external worms. This study analyses ectoparasites found on Mongolian bats between 2008 and 2011. We examined 12 different bat species, with a total of 23 ectoparasite species present. Apart from reporting distributions, we also discuss specific host-parasite relationships. Owing to recent taxonomic changes splitting the Myotis mystacinus-group into several new taxa, their corresponding ectoparasite fauna could also be addressed in detail. Introducing ectoparasitic insects at length elsewhere (Scheffler et al. 2010), this paper focuses on the analysis of parasitic Acari. Additional findings for Spinturnicidae (wing mites) and Macronyssidae broadened the spectrum of known parasites. Altogether, the knowledge of bat ectoparasites from Mongolia …

Mosquitoes (Diptera: Culicidae) are vectors of disease in the adult stage, but understanding the factors affecting distributions of the immature stages is important to the understanding and control of adult populations. Discarded automobile tires comprise important larval mosquito habitats. The Asian tiger mosquito (Aedes albopictus) and the southern house mosquito (Culex quinquefasciatus) are two medically important species commonly found in tires, but factors affecting their larval distributions in tires have not been studied, nor have their interspecific interactions. Over the last decade, small noncoding RNA molecules such as microRNAs (miRNAs) have emerged as critical regulators in the expression and function of eukaryotic genomes. It has been suggested that viral infections and neurological disease outcome may also be shaped by the influence of small RNAs. This has prompted us to suggest that HIV infection alters the endogenous miRNA expression patterns, thereby contributing to neuronal deregulation and AIDS dementia.
