Introduction Preterm birth complications are the most common cause of death in children under 5 years. The presence of multiple microorganisms and genital tract inflammation could be the common mechanism driving early onset of labour. South Africa has high levels of preterm birth, genital tract infections and HIV infection among pregnant women. We plan to investigate associations between the presence of multiple lower genital tract microorganisms in pregnancy and gestational age at birth. Bacterial vaginosis (BV), a dysbiosis of the vaginal microbiota, is a common coinfection with Chlamydia trachomatis (Ct), and BV-associated bacteria (BVAB) and their products have been implicated in aiding Ct evade natural immunity. Here, we determined if a non-optimal vaginal microbiota was associated with a higher genital Ct burden and if metronidazole, a standard treatment for BV, would reduce Ct burden or aid in natural clearance of Ct infection. Cervicovaginal samples were collected from women at enrollment and, if testing positive for Ct infection, at a follow-up visit approximately one week later. In the biosphere, limits for diversity among species, communities, and biomes are revealed through intensive and extensive field-based inventory and assembly of voucher specimens and associated informatics examined in a phylogenetic, historical, ecological, and biogeographic arena. Archival resources for specimens and information contribute to a cumulative view of faunal structure and assembly under a comparative umbrella. Ultimately, species definitions, and inclusive partitions among populations and lineages, are fundamental in articulating hypotheses that examine interactions about evolution, the nature of organisms, and the condition of environments across space and time. The ecology of the mesopelagic zone is an understudied area of the open ocean, with few diet studies even for dominant fish species. Since endoparasites are generally transmitted trophically, descriptions of the parasite communities within fishes can often provide additional insights on food web interactions. This project assesses the endoparasite faunal community of mesopredatory mesopelagic teleosts to better understand mesopelagic food webs in the South Atlantic Bight. Fishes in the Families Alepisauridae (lancetfishes), Gempylidae (snake mackerels), Bramidae (pomfrets), and Scombrolabracidae (longfin escolar) are frequent bycatch in the pelagic longline fishery targeting swordfish Xiphias gladius and thunnid tunas, yet the ecology …

The protozoan pathogen, Trypanosoma brucei, is the causative agent of sleeping sickness in humans and nagana in livestock in sub-Saharan Africa. T. brucei cycles between tsetse fly and mammalian hosts, and it is adapted to survive in diverse host tissues. Variant Surface Glycoprotein (VSG) plays a key role in immune evasion in the mammalian host. The VSG membrane anchor requires two myristates, 14-carbon saturated fatty acids (FAs) that are scarce in the host. T. brucei can synthesize FAs de novo, but also readily takes up exogenous FAs, despite lacking homologs to fatty acid uptake proteins found in other …

Bats are an ecologically important taxon that can host zoonotic pathogens. Globally, many bat species are synanthropic and live closely with humans, often roosting in man-made structures. The spatial overlap between humans and bats creates opportunities for human-bat contact, which can lead to human exposure to bat-borne pathogens and conflicts that cause bat mortality. Despite this risk, little is known about the drivers and characteristics of these human-bat interactions in buildings and work is needed to understand this aspect of the wildlife-urban interface. Underinvestment in pathology and laboratory capacity caused by low visibility in research and in prioritization by public health leaders results in limited effective healthcare coverage and an estimated 1.1 million premature deaths annually in Low-and-Middle-Income Countries. Kenya’s public health laboratories provide a median 41% of the Essential Diagnostic List to their patients and in Kisumu County, as much as 44.2% of the population has little to no access to essential diagnostics. The government of Kisumu implemented the county Health Laboratory Strategic Plan 2018-2022 to address this public health challenge. Geographical distribution plays a major role in our understanding of marine biodiversity. Some marine fish trematodes have been shown to have highly restricted geographical distributions, while some are known to occur over very wide ranges; however, very few of these wide distributions have been demonstrated genetically. Here, we analyse species of the genus Schikhobalotrema (Haplosplanchnidae) parasitizing beloniforms from the tropical west Pacific, the eastern Pacific and the Gulf of Mexico (GoM). Molecular examination of representatives of Balaustium from several populations in SW Poland, performed using the sequence data from the mitochondrial cytochrome c oxidase subunit I, confirmed their common specific affiliation and identity with Balaustium murorum. The potential presence of distinct species in the studied material, preliminarily inferred from the discovery of clusters as a result of Principal Component Analysis exploring the metric data sets, was rejected due to the finding of only one haplotype, at intra- and inter-population sampling. D-shaped nematode eggs in the feces of Rangifer tarandus are expected to be oxyurid nematodes (Nemata: Oxyurida) of the genus Skrjabinema. The species S. tarandi is considered species-specific for this host. There is no consensus regarding the cross-infection of reindeer and sheep with S. ovis and S. tarandi. The drawings proposed by descriptors complicate differential diagnostics. Micrographs of S. tarandi eggs obtained via light microscopy and scanning electron microscopy as well as photographs of S. ovis eggs and drawings made on their basis are proposed to confidently distinguish between representatives of these two species, taking into account morphometric data. The emergence of fungicide resistance severely threatens crop production by limiting the availability and application of established fungicides. Therefore, it is urgent to identify new fungicidal targets for controlling plant diseases. Here, we characterized the function of a conserved homoserine O-acetyltransferase (HOA) from the rice blast fungus Magnaporthe oryzae that could serve as the candidate antifungal target. Deletion of the MoMET2 and MoCYS2 genes encoding HOAs perturbed the biosynthesis of methionine and S-adenyl methionine, a methyl group donor for epigenetic modifications, and severely attenuated the development and virulence of M. oryzae. Emerging infectious disease (EID) represents an existential threat to humanity. EIDs are increasing in frequency and impact because of climate change and other human activities. We are losing the battle against EIDs because of improper assessment of the risk of EID. This stems from adherence to a failed paradigm of pathogen-host associations that suggests EIDs ought to be both unpredictable and rare. That, in turn, leads to policies suggesting that crisis response is the best we can do. Real-time and phylogenetic assessments show EIDs to be neither rare nor unpredictable—this is the parasite paradox that shows the failures of the …

The distribution of avian haemosporidians of the genus Leucocytozoon in the Neotropics remains poorly understood. Recent studies confirmed their presence in the region using molecular techniques alone, but evidence for gametocytes and data on putative competent hosts for Leucocytozoon are still lacking outside highland areas. We combined morphological and molecular data to characterize a new Leucocytozoon species infecting a non-migratory red-legged seriema (Cariama cristata), the first report of a competent host for Leucocytozoon in Brazil. Parasites with complex, multi-host lifecycles often engage in host behavior manipulation to increase transmission between successive hosts. In intermediate fish hosts, previous research has measured increased frequency of conspicuous behaviors, decreased swimming performance, and reduced antipredator behavior, which would collectively increase the fish’s risk of predation. In ecosystems where this type of parasite increased trophic transmission (PITT) occurs, parasites can play a substantial role in food webs. In this study, I investigate how chronic versus acute exposure to the trematode Euhaplorchis sp. A. affects the antipredator behavior of the Gulf killifish Fundulus grandis. In Germany, the knowledge about ticks infesting bats is limited, and is restricted only to a few studies, most of them dating back decades. To further improve our knowledge on ticks parasitising bats, healthy and sick bats in central Germany were examined for ticks. In total 519 larvae and one nymph of Carios vespertilionis were collected from nine bat species: Eptesicus nilssonii, Eptesicus serotinus, Myotis daubentonii, Myotis myotis, Nyctalus leisleri, Pipistrellus nathusii, Pipistrellus pygmaeus, Pipistrellus pipistrellus, and Vespertilio murinus. A summer drenching program for sheep worm control is now recognised as a key cause of drench resistance in Western Australia. However, some small changes can increase the sustainability of pre-emptive control programs. Drenches in summer should always be given to younger, growing sheep (current and previous year’s age groups), whereas adult sheep should be drenched in autumn, or if drenched in summer, some should be left untreated (if drenching at all). Research shows that this reduces the chance of developing drench resistance but still provides effective worm control.

Malaria, caused by the apicomplexan Plasmodium spp. , is a major public health issue that impacts over one-third of the world’s population ,with Plasmodium vivax accounting for over 130 million clinical cases annually. The circumsporozoite protein (CSP) is the most abundant molecule on the surface of Plasmodium sporozoites and is considered a leading pre-erythrocytic stage vaccine candidate. CSP is essential for sporozoite maturation, migration, and invasion. Anti-CSP antibodies interrupt sporozoite migration and infection of hepatocytes thus reducing liver-stage burden. Some mites of the family Haemogamasidae can transmit a variety of zoonotic diseases and have important public health and safety implications. Currently, however, little attention has been paid to molecular data of Haemogamasidae species, limiting our understanding of their evolutionary and phylogenetic relationships. In this study, the complete mitochondrial genome of Eulaelaps huzhuensis was determined for the first time, and its genomic information was analyzed in detail. The mitochondrial genome of E. huzhuensis is 14,872 bp in length with 37 genes and two control regions. The base composition showed a distinct AT preference. The spotted fever group (SFG) of Rickettsia are zoonotic disease-causing pathogens, commonly transmitted by hard ticks to a wide range of hosts, including humans. Rickettsia conorii is the common SFG recognised in India, whereas most of the infections due to other group species go undifferentiated at the species level. Hence, this study was conducted to screen host-seeking ticks in the Western Ghats region, India, for the DNA of SFG Rickettsia. RTS,S/AS01, vaccine targeting malaria caused by Plasmodium falciparum, used in moderate to high malaria transmission, provides hope for pre-erythrocytic (PE) subunit malaria vaccine development. Despite this vaccine advance, efforts to develop an efficacious vaccine against the neglected parasite Plasmodium vivax have lagged. In addition, there exists a gap in our understanding of PE antigens that can induce broadly-neutralizing antibodies. Background: Understanding the geographic distribution of Rickettsia montanensis infections in Dermacentor variabilis is important for tick-borne disease management in the United States, as both a tick-borne agent of interest and a potential confounder in surveillance of other rickettsial diseases. Two previous studies modeled niche suitability for D. variabilis with and without R. montanensis, from 2002-2012, indicating that the D. variabilis niche overestimates the infected niche. This study updates these, adding data since 2012.

As part of an ongoing project to inventory the helminth parasites of amphibians from the state of Hidalgo, Central Mexico, specimens of Rhinella horribilis were collected from three municipalities: Eloxochitlán, Huehuetla, and San Felipe Orizatlán. A total of eight taxa of helminths were found: three digeneans (Haematoloechus sp., Langeronia macrocirra, and Mesocoelium danforthi) and five nematodes (Cosmocerca sp., Cruzia morleyi, Ochoterenella chiapensis, Oswaldocruzia subauricularis, and Rhabdias sp.). The highest species richness was recorded in Huehuetla. All reports of these species of helminths represent new locality records for Mexico.

Malaria remains a significant global health threat, responsible for 691,000 deaths in 2021. Artemisinin-based combination therapies have contributed significantly to decline of malaria disease burden. Upon activation by heme, artemisinins are believed to kill parasites by non-specifically alkylating proteins and causing widespread damage to the parasite. Artemisinin resistance is now prevalent in regions of Southeast Asia and resistance has recently emerged in Africa, where over 90% of all malaria deaths occur. Mutations in Kelch13 (K13) are believed to mediate artemisinin resistance by either reducing parasite hemoglobin digestion, and consequently artemisinin activation, or by increasing parasite stress response mechanisms. Toxoplasma gondii, the causative agent of toxoplasmosis, is an intracellular apicomplexan parasite. Toxoplasmosis is often asymptomatic, though, it can be debilitating or fatal to immunocompromised individuals and fetuses, being one of the leading causes of fetal mortality and birth defects. Toxoplasmosis is treated with a synergistic combination of drugs pyrimethamine and sulfadiazine; however, these drugs are only effective against the acute stage of infection, and the pyrimethamine is relatively cytotoxic, damaging bone marrow and causing noxious side effects. Toxoplasma gondii, an intracellular protozoan, is a globally prevalent parasite due to the infectivity of T. gondii with nearly 2.5 billion people affected worldwide. Toxoplasma gondii can cause toxoplasmosis, which is a disease acquired through the consumption of undercooked meat or found in the feces of felines, and often leads to profound neuropathology and potentially death. Currently, acute toxoplasmosis is treated with pyrimethamine and sulfadiazine, which is a combination of an antiprotozoal and a sulfonamide.
