The 1990s saw the rapid reemergence of malaria in Amazonia, where it remains an important public health priority in South America. The Amazonian International Center of Excellence in Malaria Research (ICEMR) was designed to take a multidisciplinary approach toward identifying novel malaria control and elimination strategies. Based on geographically and epidemiologically distinct sites in the Northeastern Peruvian and Western Brazilian Amazon regions, synergistic projects integrate malaria epidemiology, vector biology, and immunology. The Amazonian ICEMR’s overarching goal is to understand how human behavior and other sociodemographic features of human reservoirs of transmission— predominantly asymptomatically parasitemic people—interact with the major Amazonian malaria …

Diversity And Phylogenetic Relationships Of ‘Tetraphyllidean’ Clade 3 (Cestoda) Based On New Material From Orectolobiform Sharks In Australia And Taiwan, Janine N. Caira, Kirsten Jensen
Harold W. Manter Laboratory of Parasitology: Library Materials
In an effort to expand knowledge of Clade 3—one of the ten clades that compose the non-monophyletic order ‘Tetraphyllidea’ all current members of which parasitise orectolobiform sharks—we targeted species of orectolobiform sharks that had not previously been examined for ‘tetraphyllidean’ cestodes. That work led to the discovery of three new species off Australia and Taiwan. Ambitalveolus gen. n. was erected to accommodate these species. Ambitalveolus costelloae gen. n. et sp. n., Ambitalveolus kempi sp. n., and Ambitalveolus penghuensis sp. n. differ from one another in scolex size, genital pore position, and number of marginal loculi, proglottids, and testes. Among ‘tetraphyllideans’, …

Genomic Approaches To Uncovering The Coevolutionary History Of Parasitic Lice [Review], Kevin P. Johnson
Harold W. Manter Laboratory of Parasitology: Library Materials
Next-generation sequencing technologies are revolutionizing the fields of genomics, phylogenetics, and population genetics. These new genomic approaches have been extensively applied to a major group of parasites, the lice (Insecta: Phthiraptera) of birds and mammals. Two louse genomes have been assembled and annotated to date, and these have opened up new resources for the study of louse biology. Whole genome sequencing has been used to assemble large phylogenomic datasets for lice, incorporating sequences of thousands of genes. These datasets have provided highly supported trees at all taxonomic levels, ranging from relationships among the major groups of lice to those among …

Amblyomma Quadricavum (Schulze, 1941) (Acari:Ixodidae): First Description Of The Nymphal Stage And Phylogenetic Assessment Of Its Taxonomic Status, Ashleigh V. Riggs
College of Graduate Studies: Theses & Dissertations
The hard tick (Acari: Ixodidae) Amblyomma quadricavum (Schulze, 1941) is a parasite of snakes and is endemic to the Caribbean region. Morphological characters of this species, namely its rudimentary eye structures, have led to debate about its taxonomic status. The species was originally assigned to the genus Aponomma. However, with revisions of the genus Aponomma, and the recent creation of new genera created for some basal former-Aponomma, it has been suggested that perhaps Amblyomma quadricavum might be a close relative of Robertsicus elaphensis, the Trans-Pecos rat snake tick of the southwest United States and New Mexico, another eyeless former-Aponomma species …

Biological Sciences Faculty Publications
Bacterial endosymbionts are abundantly found in both hard and soft ticks. Occidentia massiliensis, a rickettsial endosymbiont, was first identified in the soft tick Ornithodoros sonrai collected from Senegal and later was identified in a hard tick Africaniella transversale. In this study, we noted the presence of Occidentia species, designated as Occidentia-like species, in a soft tick O. turicata americanus. Sequencing and phylogenetic analyses of the two genetic markers, 16S rRNA and groEL confirmed the presence of Occidentia-like species in O. turicata americanus ticks. The Occidentia-like species was noted to be present in all developmental stages …
Avian Haemosporidian Blood Parasite Diversity, Prevalence, And Distribution In Michigan’S Western Upper Peninsula, Maria M. Ferrer
Avian Haemosporidian Blood Parasite Diversity, Prevalence, And Distribution In Michigan’S Western Upper Peninsula, Maria M. Ferrer
Dissertations, Master's Theses and Master's Reports
Avian haemosporidian parasites, known as avian malaria (phylum Apicomplexa) can diminish an individual bird’s fitness by causing parasitemia, anemia, and reduced survival. Climate change is predicted to increase the spread of malarial parasites into more northerly latitudes where little is known about community compositions of these parasites. I assessed the prevalence and diversity of haemosporidian parasites in the first-ever community-level sampling of malaria in songbirds across the western Upper Peninsula of Michigan. In total, 179 blood samples were collected from birds representing 17 species, at five locations in the Upper Peninsula, including a mature forest, an early successional forest, and …

Factors Influencing Intestinal Parasite In Black-And-White Ruffed Lemurs (Varecia Variegata), Eliette Noromalala
All Master's Theses
Ecological factors, such as feeding behavior and climatic seasonality, might influence intestinal parasite infection and patterns in non-human primates. Here, we examined if frugivory levels and climate (i.e., temperature and rainfall) were associated with parasite patterns in black-and-white ruffed lemurs (Varecia variegata). Feeding data using 5-min focal sampling, climate data, and fecal samples from V. variegata (n = 23) at the “Mangevo” site were collected from February through May 2019. Microscopic examinations of fecal flotation and sedimentation were done to identify and count parasites. Nematoda (Callistoura vauceli, and Strongyloides sp.) Ciliophora (Balantidium sp.), Coccidia ( …

White-Nose Syndrome (WNS), caused by the psychrophilic pathogenic fungus Pseudogymnoascus destructans (Pd), has killed millions of bats in the eastern United States since its initial introduction in 2006 and recent expansion into the western U.S. Understanding factors that contribute to the spread of Pd and risk of infection is crucial for management of WNS. Bat ectoparasites, including bat mites and bat flies, are omnipresent in bat populations, yet the relationship between these ectoparasites and bat health is still unknown. We examined the relationship between bat ectoparasites and the skin microbiome in relation to WNS infection risk in Washington State bats. …

Comprehensive Study Of Human Pathogenic Trypanosomatids: From Chemotherapies To Disease Ecology, Felipe Rodriguez
Open Access Theses & Dissertations
The vector-borne diseases Leishmaniasis and Chagas disease, caused by the kinetoplastid parasites Leishmania spp. and Trypanosoma cruzi respectively, are among the most important parasitic diseases in the group of neglected tropical diseases. Even though the two diseases are endemic to the Americas, they affect millions of people worldwide. Leishmaniasis and Chagas disease cause a great array of symptoms and some of them can be fatal if left undiagnosed and untreated. Current treatment regimens are becoming less effective, vaccines are still not available, and diagnosis needs to be improved. Vector control has been responsible for a decrease of diseases in endemic …
Investigation Of Histomoniasis Prevention In Poultry, Lesleigh Beer
Investigation Of Histomoniasis Prevention In Poultry, Lesleigh Beer
Graduate Theses and Dissertations
Histomonas meleagridis is the etiological agent of histomoniasis, also commonly known as blackhead disease. This protozoal disease of poultry is detrimental to turkeys with flock mortalities often reaching 80-100%, although other gallinaceous birds are susceptible. Since the voluntary removal of nitarsone in 2015, the poultry industry is suffering with no approved prophylactics, therapeutics, or vaccines for this disease. The objectives of this dissertation were to evaluate multiple methods for prevention or control of histomoniasis, including dietary chemoprophylaxis and vaccination. Specifically, this research evaluated quinine as a chemoprophylactic candidate (Chapter 3) or live-attenuated H. meleagridis as vaccine candidates (Chapter 4) in …

During plant-pathogenic fungi and host plants interactions, numerous pathogen-derived proteins are secreted resulting in the activation of the unfolded protein response (UPR) pathway. For efficient trafficking of secretory proteins, including those important in disease progression, the cytoplasmic coat protein complex II (COPII) exhibits a multifunctional role whose elucidation remains limited. Here, we discovered that the COPII cargo receptor MoErv29 functions as a target of MoHac1, a previously identified transcription factor of the UPR pathway. In Magnaporthe oryzae, deletion of MoERV29 severely affected the vegetative growth, conidiation and biotrophic invasion of the fungus in susceptible rice hosts. We demonstrated that MoErv29 …

Ticks (Acari: Ixodidae) Parasitizing Migrating And Local Breeding Birds In Finland, Jani J. Sormunen, Tero Klemola, Eero J. Vesterinen
Harold W. Manter Laboratory of Parasitology: Library Materials
Ticks are globally renowned vectors for numerous zoonoses, and birds have been identified as important hosts for several species of hard ticks (Acari: Ixodidae) and tick-borne pathogens. Many European bird species overwinter in Africa and Western Asia, consequently migrating back to breeding grounds in Europe in the spring. During these spring migrations, birds may transport exotic tick species (and associated pathogens) to areas outside their typical distribution ranges. In Finland, very few studies have been conducted regarding ticks parasitizing migrating or local birds, and existing data are outdated, likely not reflecting the current situation. Consequently, in 2018, we asked volunteer …

Natterer In Neotropical Nematoda: Species Described By Rudolphi, Diesing, And Molin, Ricardo Guerrero
MANTER: Journal of Parasite Biodiversity
Between 1817 and 1835 Johann Natterer collected 1,729 samples of endoparasitic helminths in Brazil and southern Venezuela. Of the 323 names that were assigned to the collected nematode specimens, 163 are still valid and accepted species, 84 are still doubtful, and 76 must be rejected. In this work, each name is analyzed and correlated to the literature to clarify its taxonomic status.
The purpose of this review of the material collected by Natterer is to establish a complete list of all described species; their current status; and whether they are valid species, synonyms, or nomen nudum as well as to …

Assemblages Of Acari In Shallow Burials: Mites As Markers Of The Burial Environment, Of The Stage Of Decay And Of Body‑Cadaver Regions, Jas K. Rai, Brian J. Pickles, M. Alejandra Perotti
Harold W. Manter Laboratory of Parasitology: Library Materials
The burial of a cadaver results in reduced arthropod activity and disruptions in colonisation patterns. Here, the distribution and diversity of mite taxa was studied across decomposition stages of shallowly buried pig carcasses (Sus scrofa domesticus). In total 300 mites (88 species) were collected from three pig shallow graves compared to 129 mites (46 species) from control (bare) soil samples at the same depth. A successional pattern of Acari higher taxa and families was observed, and species richness and biodiversity fluctuated throughout decomposition, whereas active decay showed the greatest biodiversity. Mesostigmata mites were the most abundant in ‘cadaver …

First Detection Of Colletotrichum Fructicola (Ascomycota) On Horsehair Worms (Nematomorpha), Mattia De Vivo, Wen-Hong Wang, Ko-Hsuan Chen, Jen-Pan Huang
Harold W. Manter Laboratory of Parasitology: Library Materials
Fungal members of Colletotrichum (Ascomycota) were found to be associated with Chordodes formosanus, one of the three currently known horsehair worm (Nematomorpha) species in Taiwan. The fungi were identified as Colletotrichum fructicola, which is mostly known as a plant pathogen, through the use of the nuclear ribosomal internal transcribed spacer and partial large subunit (nrITS + nrLSU) and glyceraldehyde-3-phosphate dehydrogenase (GAPDH) DNA sequences. To our knowledge, this report represents both the first records for Colletotrichum associated with hairworms and for fungi on Nematomorpha. These findings expand the knowledge on the ecological relationships of both clades.

Molecular And Cellular Characterizations Of Glucosylceramide Transferase (Synthase) In Giardia Lamblia, Vanessa Enriquez
Open Access Theses & Dissertations
Giardiasis, caused by an intestinal protozoan, Giardia lamblia, is a major public health problem worldwide. Giardia infection is spread through the fecal-oral route via contaminated water and food. It is estimated that over 280 million people are infected with Giardia across the globe. Giardia is a non-invasive parasite and consists of a two-stage life cycle: trophozoites and cysts. Ingested cysts undergo excystation in the proximal small intestine, releasing two trophozoites (excyzoites). Upon reaching the distal small intestine and colonizing there, trophozoites transform into cysts by encystation. It has been reported earlier that the process of encystation induced the synThesis of …

Nematode Biodiversity In Lincoln, Nebraska's Tallgrass Prairie Corridor, Abigail Borgmeier
Department of Plant Pathology: Dissertations, Theses, and Student Research
The tallgrass prairie was once one of the most diverse grasslands on the planet, however over 95% has been converted to agriculture in the past 150 years. The remaining prairie is still host to a wide variety of plants, insects, mammals, and soil-dwelling microorganisms, creating an incredibly complex ecosystem. The first aim of this study is to compare three approaches for the measurement of nematode diversity within a 10-mile-long protected prairie habitat corridor near Lincoln, Nebraska. The methods are 1) a traditional morphological analysis of 150 nematodes per study site, 2) a metabarcoding analysis using the 18S genetic marker of …

In The Search For Novel Treatments For Chagas' Disease Using Cutting-Edge Imaging Technology, Karsten Dieter Amezcua Winter
Open Access Theses & Dissertations
Chagas disease (ChD) is caused by Trypanosoma cruzi (T. cruzi), an intracellular protozoan parasite. ChD has a global mortality of 15,000 annual deaths, and approximately, 8-10 million people are infected. There is growing concern in the United States as autochthonous cases of ChD have been reported in the southern region. The two available treatments are only partially effective and highly toxic. N-aroyl derivatives and α, β-unsaturated ketones have been previously tested against Leishmania, a closely related parasite, demonstrating selective toxicity towards the microorganism. The objective of this study is to evaluate a drug library consisting of 21 α, β-unsaturated ketones …

Tick Saliva And The Alpha-Gal Syndrome: Finding A Needle In A Haystack, Surendra Raj Sharma, Shahid Karim
Faculty Publications
Ticks and tick-borne diseases are significant public health concerns. Bioactive molecules in tick saliva facilitate prolonged blood-feeding and transmission of tick-borne pathogens to the vertebrate host. Alpha-gal syndrome (AGS), a newly reported food allergy, is believed to be induced by saliva proteins decorated with a sugar molecule, the oligosaccharide galactose-⍺-1,3-galactose (α-gal). This syndrome is characterized by an IgE antibody-directed hypersensitivity against α-gal. The α-gal antigen was discovered in the salivary glands and saliva of various tick species including, the Lone Star tick (Amblyomma americanum). The underlying immune mechanisms linking tick bites with α-gal-specific IgE production are poorly understood and are …
Diversity And Host Specificity Of Nycteribiid Bat Flies (Diptera: Nycteribiidae) In Kenya, Taylor Verrett
Diversity And Host Specificity Of Nycteribiid Bat Flies (Diptera: Nycteribiidae) In Kenya, Taylor Verrett
Masters Theses & Specialist Projects
Bat flies (Diptera: Nycteribiidae and Streblidae) are hematophagous ectoparasites of bats distributed globally. Members of Nycteribiidae are morphologically constrained relative to streblids, and are united by their lack of wings, dorso-ventrally compressed bodies, and a primary distribution across the Eastern hemisphere. Bats are principal reservoirs of infectious diseases, including viral zoonoses of important consideration to human health, but the overall high host specificity of bat flies has largely been thought to curb their potential as inter-specific vectors of bat-borne pathogens. However, nycteribiid diversity and host associations remain critically understudied in some geographic regions, and rare examples of nycteribiid bat flies …

Parasites are organisms that live inside or on another species, called the host. Parasites depend on their hosts for food and a place to live. They may harm the host in small or large ways. Parasitism is the most common mode of life on Earth. Humans, other animals, and all plants have parasites, usually two or more kinds. Even parasites can have parasites. There are millions of species of parasites, and scientists discover new ones every day. Parasite specimens are stored in museums all around the world. One of the world’s largest collections is in the H. W. Manter Laboratory …

Host-Parasite Interactions Within Food Webs, Adam Zvanut Hasik
Graduate Theses and Dissertations
Parasitism is one of the most common life history strategies employed in nature, yet the effects of parasites are often thought to be minimal, and the vast majority of studies fail to consider parasites and their effects on host organisms. This is likely a problem, as the magnitude of parasite-mediated effects on their hosts can be quite large. Additionally, the effects of parasites are known to extend beyond the host to affect other species interactions. I used a series of approaches to gain a more integral understanding of host-parasite interactions by studying (1) the effects of parasites on biotic interactions …

Development And Validation Of An Isothermal Amplification Assay For Eastern Equine Encephalitis Virus, Mikayla D. Maddison
USF Tampa Graduate Theses and Dissertations
Eastern Equine Encephalitis virus (EEEV) is a neurotrophic alphavirus for which there is no effective treatment or vaccine for humans. Periodic outbreaks in the Eastern United States represent an ongoing public health problem; Florida serves as the reservoir for EEEV for the rest of the country. Reverse transcription-polymerase chain reaction (RT-PCR) is the current gold standard for molecular diagnostic testing of the presence of EEEV in vectors. However, RT-PCR is technically complex and can be difficult for mosquito control districts to utilize. In order to provide a simple, cost-effective alternative for mosquito surveillance and control, a novel one step reverse …

The Effects Of Infliximab On The Fibrosis Response Of Three-Spine Stickleback, Mita Kale
Honors Scholar Theses
This study was conducted to determine if the monoclonal antibody drug infliximab could effectively suppress the fibrosis immune response in three-spine stickleback fish. Successful suppression of this response could allow for further study of cestode infection and growth without the presence of fibrosis. Infliximab’s efficacy was investigated through conducting two intraperitoneal injection experiments and examinations of the fibrosis in the body cavity of the euthanized stickleback. We used immune adjuvant alum to induce a fibrosis response without the presence of the S. solidus tapeworm parasite. Ultimately, the differences in fibrosis levels between the treatment groups that received alum and the …

Plasmodium malariae and Plasmodium vivax are protozoan parasites that can cause malaria in humans. They are genetically indistinguishable from, respectively, Plasmodium brasilianum and Plasmodium simium, that is, parasites infecting New World non-human primates in South America. In the tropical rainforests of the Brazilian Atlantic coast, it has long been hypothesized that P. brasilianum and P. simium in platyrrhine primates originated from P. malariae and P. vivax in humans. A recent hypothesis proposed the inclusion of Plasmodium falciparum into the transmission dynamics between humans and non-human primates in the Brazilian Atlantic tropical rainforest. Herein, we assess the occurrence of human …

Toxoplasma gondii is a globally-distributed obligate intracellular parasite and the causative agent of toxoplasmosis in humans, a biphasic disease often culminating in lifelong chronic infection. Though often mild, toxoplasmosis can lead to severe clinical presentation in immunocompromised patients and developing fetuses. Current therapeutic options are severely limited, with a combinatorial pyrimethamine/sulfadiazine regimen acting as the current standard of care for toxoplasmosis; however, both of these compounds can elicit serious adverse reactions in patients, and, additionally, are only effective against the acute stage of infection. To address these limitations, screenings of several analogs derived from the marine natural product marinopyrrole A …

An updated and increased compilation of georeferenced tick locations in Germany is presented here. This data collection extends the dataset published some years ago by another 1448 new tick locations, 900 locations of which were digitized from literature and 548 locations are published here for the first time. This means that a total of 3492 georeferenced tick locations is now available for Germany. The tick fauna of Germany includes two species of Argasidae in the genera Argas and Carios and 19 species of Ixodidae in the genera Dermacentor, Haemaphysalis, and Ixodes, altogether 21 tick species. In addition, three species …
