One-third of our worldwide fruit, nut and vegetable production is completely dependent on the existence of pollinators. Commercial honey bees, Apis mellifera, have an annual economic value of $15 billion in the U.S. Additionally, their honey is valued at $150 million annually. In Fall 2006, commercial beekeepers observed sudden mass disappearances of whole colonies. By Spring 2007, the condition, dubbed Colony Collapse Disorder (CCD), raised environmental and fiscal concerns while the cause(s) continued to be sought. Forensic examinations of hive samples did not reveal a singular cause of CCD. The results pointed to a collection of detrimental factors affecting …

The adoption of regenerative farming practices is gaining traction, but the costs and benefits are not often considered on a systems level. Encouraging biodiversity and soil health is the goal of many agricultural practices used in regenerative farming; regenerative systems employ practices which abide by the two main principles of increasing biodiversity and decreasing disturbance, with the goal of encouraging ecosystem functioning to minimize inputs and maximize the productivity of a farm. Assessing the risk of transgenic crop products is essential when determining the safety of a crop for deregulation and commercialization. The Organization of Economic and Cooperative Development (OECD), International Standards Organization (ISO), and governmental regulatory agencies require a battery of tests to demonstrate the safety of a GM product against several surrogate species of organisms that perform various ecosystem services. Assays are performed using toxicology methods established for pesticides. Lytopylus is species-rich genus of Agathidinae (Hymenoptera: Braconidae: Agathidinae), but limited information of the genus has been published. Morphological, molecular, and ecological data were compared to resolve the species limits of Lytopylus reared from caterpillars collected the Area de Conservación Guanacaste (ACG) in Costa Rica. Molecular data were initially used to hypothesize species limits, and morphological and host use data were employed to make a final decision when molecular data was indecisive, e.g., when the genetic difference between species was slight. Across animals, a male's fitness is largely dictated by his ability to fertilize eggs; and there exists a plethora of male adaptations associated with increasing fertilization success. In the nursery web spider, Pisaurina mira, males restrain females prior to and during copulation by wrapping them with silk. Previous research demonstrates that copulatory silk wrapping reduces a male's chance of being sexually cannibalized and increases the number of sperm transfer opportunities (termed insertions) that a male can achieve within a mating. While avoiding cannibalism provides an obvious survival benefit to males, the impact of insertion number on male fitness remains …

Blow flies (Diptera: Calliphoridae) are usually the first insects to colonize human remains. By determining the time of colonization, a postmortem interval (PMI), or “time of death”, can be estimated. To develop more accurate PMI estimates, it is important for forensic entomologists to understand the cues that Blow flies use to locate vertebrate remains. The purpose of this study was to determine whether Blow flies use visual cues, in addition to olfactory cues, to locate carrion. Phytophagous insects make up over one quarter of described species on Earth, and this incredible diversity seems directly linked to feeding on plants. Comparative studies of sister groups have shown shifts to herbivory are consistently associated with increased species diversity in insects, but the reasons for this diversification remain unclear. While other explanations, such as decreased extinction rates or influences on population structure, exist, one prominent hypothesis suggests shifts and subsequent adaptation to novel host plants can lead to the evolution of reproductive barriers.

Insecticide resistance is a global problem. Insecticide resistance management is very important, considering the time, effort, and cost of discovering and developing a new insecticide. There are diverse resistance mechanisms, but enhanced detoxification through overexpression of cytochrome P450s and target site insensitivity through mutation in insecticide binding site are the two most common mechanisms. The xenobiotic detoxification is divided into three successive phases (I, II and III), which ensures the metabolism and excretion of the detrimental toxins. Despite advances in mosquito management, mosquito-borne disease in the United States is still of relevant public health concern and vector control is a top priority in preventing transmission of pathogens. Insecticide barrier sprays have become a common tool for suppression of mosquitoes in single-homeowner backyards. The application of the synthetic pyrethroid, lambda-cyhalothrin to perimeter vegetation with a backpack sprayer has been shown to significantly suppress mosquito levels for around 6 weeks. Traditional methods of pest control are threatened by the development of insecticide resistance, both to traditional insecticides and Bt toxins. Discovery of RNA interference (RNAi) has created opportunities to develop new insect control mechanisms. However, RNAi responses appear to be robust in coleopteran pests, but other orders, e.g. Lepidoptera and Hemiptera, present varied or ineffective RNAi responses. Current delivery strategies for double-stranded RNA (dsRNA) include microinjection, ingestion, and soaking. These approaches have benefits and problems. The emerald ash borer (EAB, Agrilus planipennis Fairmaire) (Coleoptera: Buprestidae) is a non-native wood boring beetle that is causing extensive ash (Fraxinus spp.) mortality in eastern North America, affecting both urban and wildland forests and drastically altering forest structure and composition. As EAB-induced ash mortality progresses, native arthropod associates of ash forests are impacted by the effects of rapid and broad scale tree mortality. These include loss of food source, increased canopy gap formation, alterations in litter inputs causing shifting temperature and moisture regimes on the forest floor, and significant accumulation of coarse woody debris.

The crapemyrtle bark scale, Acanthococcus lagerstroemiae (Kuwana) (Hemiptera: Eriococcidae), is an exotic pest on crapemyrtles, Lagerstroemia spp. (Myrtales: Lythraceae). Because of its recent arrival in the US, little is known about its biology and ecology. The purpose of my thesis was to improve the knowledge about A. lagerstroemiae in four aspects involving its thermal tolerance, physiological adaptations to cold temperatures, temperature-dependent development and host range. Thermal tolerance was determined to understand how temperature extremes constrain the distribution of A. lagerstroemiae in the US. Mosquito control districts in Louisiana focus their efforts on Culex quinquefasciatus, the primary vector of West Nile virus in the southern United States, with rigorous larvicide treatments. However, the development of resistant populations of Cx. quinquefasciatus in response to extensive insecticide application has been demonstrated repeatedly. Examining changes in insecticide susceptibility and larvicide efficacy in real world scenarios can help inform mosquito control districts as to whether or not their treatments are killing mosquitoes. Phylogenetic studies suggest that historically all paper wasps (Vespidae: Polistinae) in North America have tropical origins. However, paper wasps have adapted to thrive in temperate conditions. Twospotted spider mites, Tetranychus urticae, is an important agricultural pest of many field crops worldwide. This study investigated the impacts of imidacloprid seed treatments on populations of twospotted spider mites while also investigating if exogenous applications of jasmonic acid can offset any hormone modulating effects caused by seed treatments. Imidacloprid seed treatments significantly increased cumulative adult mite days in 2013 but not 2015 or 2016 in the field. Applications of 10 millimolar jasmonic acid did not reduce mite severity or injury in all field trials. Imidacloprid seed treatments significantly increased all spider mite life stages in the laboratory while applications …

The Formosan subterranean termite (FST), Coptotermes formosanus, is an invasive urban pest in the United States. Colonies of the FST are dependent on the symbiotic gut protozoa for cellulose digestion in the workers’ guts, and the gut bacterial community is known to provide essential nutrients to the termite. The objectives of this PhD research were to develop and evaluate paratransgenesis and phage therapy for termite control. During this study, a termite gut bacterium: Trabulsiella odontotermitis was genetically engineered and was evaluated as a ‘Trojan horse’ for paratransgenesis. Sweetpotato weevil (SPW), Cylas formicarius elegantulus (Summer), is the most damaging root-feeding insect of sweetpotato, Ipomoea batatas L. (Lam), worldwide. The efficacy and compatibility of host plant resistance and insecticide control were evaluated in this dissertation. cv. Beauregard is the most susceptible cultivar for oviposition and offspring performance of SPW compared to cvs. Evangeline and Murasaki. The oviposition preference was shaped by the larval experience of SPW, providing evidence in support of the Hopkins’ Host-Plant Selection Principle. Although oviposition on cv. Murasaki was reduced, the egg capacity of SPW developed on cv. Murasaki was not decreased, indicating an adaptive behavior …

Resistance of several commercial rice varieties widely grown in Louisiana was assessed against the rice water weevil (Lissorhoptrus oryzophilus Kuschel), the major insect pest of rice in the United States. A five-year field study was conducted to identify resistant genotypes among conventional inbred, herbicide tolerant, conventional hybrid, and herbicide tolerant hybrid varieties. Resistance was evaluated in two different locations to enable selection of rice varieties with wide adaptability over diverse environments. Evolution of resistance in target pest populations is a major threat to the sustainability of transgenic crops expressing Bacillus thuringiensis (Bt) proteins. Fall armyworm, Spodoptera frugiperda (J.E. Smith), is a cross-crop target pest of Bt corn, Bt cotton, and Bt soybean. This pest, thus far, is the only target pest species that has developed field resistance to Bt crops in multiple areas across countries. Cry2Ab2 is a common Bt protein expressed in transgenic corn and cotton targeting lepidopteran pests including S. frugiperda. We determined the sex-ratio of 1,095 plant bugs (Hemiptera: Miridae) taken from 60 individual UV light-trap samples in Clark County, Arkansas over a two year period. We found that of the 21 taxa in which a sex-ratio determination could be made, 61.9% of them (13 of 21) contained a majority (over 50%) of males. Three taxa were exclusively represented by males, while two taxa were exclusively represented by females. Although taxa dependent, our data indicate that male mirids are, in general, more frequently encountered in UV light-traps. Modeling Microorganism Transmission With Madagascar Hissing Cockroaches: An Inquiry Activity, Hillary Guzik, Kessler Mccoy-Simandle

Students will test Madagascar hissing cockroach’s capacity as a vector for transmission of microorganisms. By comparing a cockroach exposed to human contact (handled by students) and a cockroach with limited exposure (not handled), students can assess the ability of cockroaches to transmit microorganisms from one location (hands) to another (agar plate where the microorganism will be grown). This will allow students to determine if the Madagascar hissing cockroach, the classroom pet, is a potential vector for microorganisms. Students then will be able to question and relate the concept of insects and objects as vectors for common pathogen transfer.

Background: Aphid infestation of switchgrass (Panicum virgatum) has the potential to reduce yields and biomass quality. Although switchgrass-greenbug (Schizaphis graminum; GB) interactions have been studied at the whole plant level, little information is available on plant defense responses at the molecular level.
Results: The global transcriptomic response of switchgrass cv Summer to GB was monitored by RNA-Seq in infested and control (uninfested) plants harvested at 5, 10, and 15 days after infestation (DAI). The increased use of thiamethoxam seed treatments for controlling pests such as the bean leaf beetle, Cerotoma trifurcata (Forster) (Coleoptera: Chrysomelidae), suggests the need for methods to measure and monitor the development of resistance to thiamethoxam. The objectives of this study were to develop a bioassay method that can be used to monitor bean leaf beetle susceptibility to thiamethoxam, and to quantify the relative concentrations of thiamethoxam and clothianidin in early growth stage soybean tissue treated with thiamethoxam as a seed treatment. Soybean aphid, Aphis glycines (Hemiptera: Aphididae), is the most economically important soybean [Glycine max (L.) Merr.] pest of North America. Multiple studies have identified soybean expressing antibiosis and/or antixenosis; however, soybean tolerance remains underexplored. Tolerance to soybean aphid injury was previously identified in soybean KS4202. This research examined the yield response of KS4202 infested with soybean aphid at specific plant stages and identified at what plant stage tolerance initiates. Drosophila suzukii (Matsumura) is an invasive species that is a devastating pest of soft-skinned fruit crops. Although much effort has been directed toward developing traps and attractants to monitor for D. suzukii, current monitoring tools do not reliably predict fruit infestation. The objective of this study was to determine if D. suzukii females at different developmental stages are differentially attracted to monitoring traps with fermentation-based baits and ripe fruits.
